| 1 | Regulation of RIG-I-like receptor-mediated signaling:interaction between host and viral factors显示文摘Retinoic acid-inducible gene I(RIG-l)-like receptors(RLRs)are RNA sensor molecules that play essential roles in innate antiviral immunity.Among the three RLRs encoded by the human genome,RIG-1 and melanoma differentiation-associated gene 5,which contain N-terminal caspase recruitment domains,are activated upon the detection of viral RNAs in the cytoplasm of virus-infected cells.Activated RLRs induce downstream signaling via their interactions with mitochondrial antiviral signaling proteins and activate the production of type Ⅰ and Ⅲ interferons and inflammatory cytokines.Recent studies have shown that RLR-mediated signaling is regulated by interactions with endogenous RNAs and host proteins,such as those involved in stress responses and posttranslational modifications.Since RLR-mediated cytokine production is also involved in the regulation of acquired immunity,the deregulation of RLR-mediated signaling is associated with autoimmune and autoinflammatory disorders.Moreover,RLRmediated signaling might be involved in the aberrant cytokine production observed in coronavirus disease 2019.Since the discovery of RLRs in 2004,significant progress has been made in understanding the mechanisms underlying the activation and regulation of RLR-mediated signaling pathways.Here,we review the recent advances in the understanding of regulated RNA recognition and signal activation by RLRs,focusing on the interactions between various host and viral factors. | Koji Onomoto Kazuhide Onoguchi Mitsutoshi Yoneyama | 2021 | Cellular & Molecular Immunology2021,18,3: | 8 |
| 2 | Immunological milieu in the peritoneal cavity at laparotomy for gastric cancer显示文摘AIM:To investigate the immunological repertoire in the peritoneal cavity of gastric cancer patients.METHODS:The peritoneal cavity is a compartment in which immunological host-tumor interactions can occur.However,the role of lymphocytes in the peritoneal cavity of gastric cancer patients is unclear.We observed 64 patients who underwent gastrectomy for gastric cancer and 11 patients who underwent laparoscopic cholecystectomy for gallstones and acted as controls.Lymphocytes isolated from both peripheral blood and peritoneal lavage were analyzed for surface markers of lymphocytes and their cytokine production by flow cytometry.CD4+CD25high T cells isolated from the patient's peripheral blood were co-cultivated for 4 d with the intra-peritoneal lymphocytes,and a cytokine assay was performed.RESULTS:At gastrectomy,CCR7 -CD45RA-CD8+ef-fector memory T cells were observed in the peritoneal cavity.The frequency of CD4+CD25 high T cells in both the peripheral blood and peritoneal cavity was elevated in patients at advanced stage[control vs stage Ⅳin the peripheral blood:6.89(3.39-10.4)vs 15.34(11.37-19.31),P<0.05,control vs stageⅣin the peritoneal cavity:8.65(5.28-12.0)vs 19.56(14.81-24.32),P<0.05].On the other hand,the suppression was restored with CD4+CD25high T cells from their own peripheral blood.This study is the first to analyze lymphocyte and cytokine production in the peritoneal cavity in patients with gastric cancer.Immune regulation at advanced stage is reversible at the point of gastrectomy.CONCLUSION:The immunological milieu in the peritoneal cavity of patients with advanced gastric cancer elicited a Th2 response even at gastrectomy,but this response was reversible. | Akira Yoneda Shinichiro Ito Seiya Susumu Mitsutoshi Matsuo Ken Taniguchi Yoshitsugu Tajima Susumu Eguchi Takashi Kanematsu Yasuhiro Nagata | 2012 | World Journal of Gastroenterology2012,18,13: | 5 |
| 3 | Does the root to shoot ratio show a hormetic response to stress? An ecological and environmental perspective显示文摘Root/shoot(R/S)ratio is an important index for assessing plant health,and has received increased attention in the last decades as a sensitive indicator of plant stress induced by chemical or physical agents.The R/S ratio has been discussed in the context of ecological theory and its potential importance in ecological succession,where species follow different strategies for above-ground growth for light or below-ground competition for water and nutrients.We present evidence showing the R/S ratio follows a biphasic dose–response relationship under stress,typical of hormesis.The R/S ratio in response to stress has been widely compared among species and ecological succession classes.It is constrained by a variety of factors such as ontogeny.Furthermore,the current literature lacks dose-response studies incorporating the full dose–response continuum,hence limiting scientific understanding and possible valuable application.The data presented provide an important perspective for new-generation studies that can advance current ecological understanding and improve carbon storage estimates by R/S ratio considerations.Hormetic response of the R/S ratio can have an important role in forestry for producing seedlings with desired characteristics to achieve maximum health/productivity and resilience under plantation conditions. | Evgenios Agathokleous Regina G.Belz Mitsutoshi Kitao Takayoshi Koike Edward J.Calabrese | 2019 | Journal of Forestry Research2019,30,5: | 5 |
| 4 | Helicobacter heilmannii sensu stricto-related gastric ulcers:A case report显示文摘A spiral bacterium(SH9),morphologically different from Helicobacter pylori(H.pylori),was found in a 62-yearold woman’s gastric mucosa.Gastroscopic examination revealed multiple gastric ulcers near the pyloric ring;mapping gastric biopsy showed mild mononuclear infiltration with large lymphoid follicles in the antrum,without corpus atrophy.Urea breath test and H.pylori culture were negative,but Giemsa staining of biopsies revealed tightly coiled bacteria that immunostained with anti-H.pylori antibody.Sequencing of SH9 16S rRNA and the partial urease A and B subunit genes showed that the former sequence had highest similarity(99%;1302/1315 bp)to Helicobacter heilmannii(H.heilmannii)sensu stricto(H.heilmannii s.s.)BC1 obtained from a bobcat,while the latter sequence confirmed highest similarity(98.3%;1467/1493 bp)to H.heilmannii s.s.HU2 obtained from a human.The patient was diagnosed with multiple gastric ulcers associated with H.heilmannii s.s.infection.After triple therapy(amoxicillin,clarithromycin,and lansoprazole)with regimen for eradicating H.pylori,gastroscopy showed ulcer improvement and no H.heilmannii s.s.upon biopsy. | Takehisa Matsumoto Masatomo Kawakubo Taiji Akamatsu Naohiko Koide Naoko Ogiwara Seiko Kubota Mitsutoshi Sugano Yoshiyuki Kawakami Tsutomu Katsuyama Hiroyoshi Ota | 2014 | World Journal of Gastroenterology2014,20,12: | 4 |
