| 1 | Roles of the canonical myomiRs mi R-1,-133 and-206 in cell development and disease显示文摘Micro RNAs are small non-coding RNAs that participate in different biological processes, providing subtle combinational regulation of cellular pathways, often by regulating components of signalling pathways. Aberrant expression of mi RNAs is an important factor in the development and progression of disease. The canonical myomi Rs(mi R-1,-133 and-206) are central to the development and health of mammalian skeletal and cardiac muscles, but new findings show they have regulatory roles in the development of other mammalian non-muscle tissues, including nerve, brain structures, adipose and some specialised immunological cells. Moreover, the deregulation of myomi R expression is associated with a variety of different cancers, where typically they have tumor suppressor functions, although examples of an oncogenic role illustrate their diverse function in different cell environments. | 2 | 基于低成本聚甲基丙烯酸甲酯(PMMA)电泳芯片的微型DNA分析仪显示文摘在低成本的聚甲基丙烯酸甲酯(PMMA)电泳芯片上,利用双通道共聚焦激光诱导荧光检测系统实现了单链DNA快速高效的分离检测.选用0.8mm厚度的PMMA薄片加工微管道,一方面降低了检测限,另一方面提高了散热性能.通过线性聚丙烯酰胺筛分胶液以及使用纤维素衍生物对微管道表面进行动态修饰等条件的优化,芯片完成了高分辨率、高重现性的短串联重复序列(STR)等位基因的快速分型检测.两个STR位点D13S317和CSF1PO的等位基因分型标准物(allelicladder)和实际样本的PCR扩增产物均在3min内达到了基线分离,表明低成本的PMMA电泳芯片在法医学及临床医学领域的基因分析方面具有良好的发展潜能. | 杨茜 熊强 姜成涛 李彩霞 孙爱梅 KEITH MITCHELSON 邢婉丽 叶健 程京 | 2009 | 生物化学与生物物理进展2009，36，4： | 3 |

| 4 | 一种检测小鼠转录因子活性的微阵列芯片的构建及应用显示文摘转录因子是一类具有序列特异性的双链DNA结合蛋白.在哺乳动物细胞中,一个转录因子可以调控多个靶基因,一个基因也同时受到多个转录因子的调控,从而形成复杂的基因表达网络调控机制.在蛋白质水平检测特定状态下组织或细胞中的转录因子活性对深入研究基因表达调控的分子机制具有重要的意义.针对TRANSFAC数据库提供的226个小鼠转录因子的PSSM,设计了240个转录因子结合探针,构建了可以在蛋白质水平同时检测约200个小鼠转录因子的微阵列芯片,实验结果显示:a.针对含有不同DNA结合结构域的7个转录因子,进行依赖于抗体的转录因子活性检测,结果证明该芯片具有良好的特异性;b.针对NFκB转录因子纯品,芯片的检测灵敏度可以达到0.5nmol/L,线性范围为0.5~20nmol/L,表明芯片具有较高的灵敏度和良好的定量能力;c.针对经典的IKK,JNK信号通路和JAK/STAT信号通路,该芯片可以检测到其关键转录因子的活性变化,证明芯片的可靠性;d.针对小鼠前脂肪细胞系3T3-L1的分化,该转录因子活性芯片不但检测到了已经报道的活性发生变化的转录因子,如PPAR家族和C/EBP家族外,还发现了以前没有报道过的SRF等,充分显示了芯片技术的高通量优势和发现新数据的能力. | 孙义民 曾令琴 张岩 魏丽 K R MITCHELSON 张亮 程京 | 2009 | 生物化学与生物物理进展2009，36，2： | 2 |
