3c-Fos overexpression increases the proliferation of human hepatocytes by stabilizing nuclear Cyclin D1显示文摘AIM: To investigate the effect of stable c-Fos overexpression on immortalized human hepatocyte (IHH) proliferation. METHODS: IHHs stably transfected with c-Fos (IHH-Fos) or an empty vector (IHH-C) were grown in medium supplemented with 1% serum or stimulated with 10% serum. Cell proliferation was assessed by cell counts, 3H-thymidine uptake and fl ow cytometry analyses. The levels of cell cycle regulatory proteins (Cyclin D1, E, A) cyclin dependent kinases (cdk) cdk2, cdk4, cdk6, and their inhibitors p15, p16, p21, p27, to- tal and phosphorylated GSK-3β and epidermal growth factor receptor (EGF-R) were assayed by Western blot- ting. Analysis of Cyclin D1 mRNA levels was performed by reverse transcription-polymerase chain reaction and real-time polymerase chain reaction (PCR) analysis. Stability of Cyclin D1 was studied by cycloheximide blockade experiments. RESULTS: Stable c-Fos overexpression increased cell proliferation under low serum conditions and resulted in a two-fold increase in [3H]-thymidine incorpora- tion following serum addition. Cell cycle analysis by fl ow cytometry showed that c-Fos accelerated the cell cycle kinetics. Following serum stimulation, Cyclin D1 was more abundantly expressed in c-Fos overexpressing cells. Cyclin D1 accumulation did not result from increased transcriptional activation, but from nuclear stabilization. Overexpression of c-Fos correlated with higher nuclear levels of inactive phosphorylated GSK- 3β, a kinase involved in Cyclin D1 degradation and higher levels of EGF-R mRNA, and EGF-R protein compared to IHH-C both in serum starved, and in serum stimulated cells. Abrogation of EGF-R signalling in IHH- Fos by treatment with AG1478, a specif ic EGF-R tyrosine kinase inhibitor, prevented the phosphorylation of GSK-3β induced by serum stimulation and decreased Cyclin D1 stability in the nucleus. CONCLUSION: Our results clearly indicate a positive role for c-Fos in cell cycle regulation in hepatocytes. 4KAP Survey of Turkish University Students on Sexuality,reproductive Health,Drug Addiction and Violence显示文摘Objective To investigate the attitudes, behaviours and reflections of the university students, on sexuality, reproductive health, violence, smoking, alcohol and drug use, and to find out their expectations from a youth health service. Methods A total of 5 300 students (1 880 girls, 3 420 boys) from various faculties of Kyrykkale University were enrolled into this cross-sectional study. Students com- pleted the self-administered questionnaire as one of the researchers was present at the classroom. Research monitors five categories of priority health-risk behaviours among youth and young adults. Results Mean age of 3 420 male (64.5%) and 1 880 female (35.5%) students were 20.3 years. Among all 4 380 (82.9%) students [1 350 girls (71.8%), 3 030 boys (88.5%)] had some kinds of information about sexuality, however this dropped to 2 730 students (51.5%) within the context of adequacy. Friends were the major source (34%) for the first information on sexuality. Using a condom was the mostly heard method of contraception (46.9%) and interestingly 1.1% of the students had no knowledge on any of the protection methods. Among all 25.3% of the students (33.0% boys, 11.4% girls) had some kind of sexual intercourse, and within this group 58.8% experienced his/her first sexual relationship at 18 years of age and over; 53.9% were protected in all intercourses. Using a condom was the main way of protection for boys (63.7%) and pills were for the girls (73.9%). None of the female students mentioned having an abortion but 8% had friends who had undergone abortion. Tobacco use was 35.1% among the group, and 8.7% of the male and 3.6% of the female students were using drugs. Although 67.4% students claimed they never witnessed violence between their parents, 43.2% witnessed violence among their siblings, 67.0% beween friends, 72.6% in the streets and 64.2% in the school. About 30.0% of the students reported having weight problems, and 14.7% admitted visiting a doctor for that problem. If there would be a service provided for the adolescents in the university, 67% of thestudents would attend in case of a problem. Past experiences were important for us and we saw that among all 8% of the participants had some kind of sexual problem, within this group 50.7% felt uncomfortable during the visit and only 60.4% were satisfied with the care of the physician. 6The role of intracellular protein O-glycosylation in cell adhesion and disease显示文摘Post-translational protein modification,including phosphorylation,is generally quick and reversible,facilitating rapid biologic adjustments to altered cellular physiologic demands.In addition to protein phosphorylation,other post-translational modifications have been identified.Intracellular protein O-glycosylation,the addition of the simple sugar O-linked N-acetylglucosamine (O-GlcNAc) to serine/threonine residues,is a relatively recently identified post-translational modification that has added to the complexity by which protein function is regulated.Two intracellular enzymes,O-GlcNAc transferase and O-GlcNAcase,catalyze the addition and removal,respectively,of O-GlcNAc to serine and threonine side-chain hydroxyl groups.Numerous proteins,including enzymes,transcription factors,receptors and structural proteins have been shown to be modified by intracellular O-glycosylation.In this review,the mechanism and relevance of O-GlcNAc protein modification are discussed in the context of cell adhesion and several representative diseases.Meryem Bektas David S.Rubenstein 2011The Journal of Biomedical Research2011,25,4:2
