| 1 | Safety and Efficacy of Physical Thermal Ablation Combined Sorafenib for Hepatocellular Carcinoma:A Meta-analysis显示文摘Background and Aims:To compare the efficacy and safety of physical thermal ablation(PTA),including radiofrequency ablation(RFA)and microwave ablation(MWA),combined with sorafenib and physical thermal ablation alone for the control and treatment of hepatocellular carcinoma(HCC)according to the available literature.Methods:Comprehensive searches were performed on PubMed,Embase,CNKI,the Cochrane Library,China Biomedical Literature Database(known as CBM),Weipu Journal,and Wanfang Database.Meta-analysis was performed using Revman 5.3 software.Results:A total of 15 studies,consisting of 2,227 HCC patients,were selected and included in this meta-analysis.Compared with the RFA-alone group,the patients in the RFA+sorafenib group had longer 1-,2-,and 3-year overall survival(all p<0.05),better overall efficacy(p<0.0001),longer radiofrequency interval(p<0.001),and lower 2-year recurrence rate(p=0.02).The 1-year overall survival(p=0.003)and overall efficacy(p=0.002)of the MWA+sorafenib group were also higher than those of the MWA-alone group.The incidences of adverse reactions in the RFA+sorafenib group,such as hand-foot skin reactions(p<0.001),diarrhea and constipation(p=0.0001),hypertension(p=0.009),and alopecia(p<0.001),were significantly higher than those in the RFA-alone group.Conclusions:RFA or MWA combined with sorafenib has produced a better therapeutic effect on HCC than physical thermal ablation alone;however,adverse reactions have been obvious.It is necessary to evaluate the safety of combination therapy,and pay close attention to the adverse reactions that develop in patients. | Mengdi Jin Qiong Yu Yahui Liu Weiling Xu Xueqi Fu Bai Ji | 2021 | Journal of Clinical and Translational Hepatology2021,9,2: | 6 |
| 2 | NLP1 reciprocally regulates nitrate inhibition of nodulation through SUNN-CRA2 signaling in Medicago truncatula显示文摘Most legume plants can associate with diazotrophic soil bacteria called rhizobia,resulting in new root organs called nodules that enable N2 fixation.Nodulation is an energy-consuming process,and nodule number is tightly regulated by independent systemic signaling pathways controlled by CLE/SUNN and CEP/CRA2.Moreover,nitrate inhibits legume nodulation via local and systemic regulatory pathways.In Medicago truncatula,NLP1 plays important roles in nitrate-induced inhibition of nodulation,but the relationship between systemic and local pathways in mediating nodulation inhibition by nitrate is poorly understood.In this study,we found that nitrate induces CLE35 expression in an NLP1-dependent manner and that NLP1 binds directly to the CLE35 promoter to activate its expression.Grafting experiments revealed that the systemic control of nodule number involves negative regulation by SUNN and positive regulation by CRA2 in the shoot,and that NLP1’s control of the inhibition of rhizobial infection,nodule development,and nitrogenase activity in response to nitrate is determined by the root.Unexpectedly,grafting experiments showed that loss of CRA2 in the root increases nodule number at inhibitory nitrate levels,probably because of CEP1/2 upregulation in the cra2 mutants,suggesting that CRA2 exerts active negative feedback regulation in the root. | Zhenpeng Luo Jie-shun Lin Yali Zhu Mengdi Fu Xiaolin Li Fang Xie | 2021 | Plant Communications2021,2,3: | 5 |

| 5 | The construction and optimization of engineered yeast chassis for efficient biosynthesis of 8-hydroxygeraniol显示文摘Microbial production of monoterpenoid indole alkaloids(MIAs)provides a sustainable and eco-friendly means to obtain compounds with high pharmaceutical values.However,efficient biosynthesis of MIAs in heterologous microorganisms is hindered due to low supply of key precursors such as geraniol and its derivative 8-hydroxygeraniol catalyzed by geraniol 8-hydroxylase(G8H).In this study,we developed a facile evolution platform to screen strains with improved yield of geraniol by using the SCRaMbLE system embedded in the Sc2.0 synthetic yeast and confirmed the causal role of relevant genomic targets.Through genome mining,we identified several G8H enzymes that perform much better than the commonly used CrG8H for 8-hydroxygeraniol production in vivo.We further showed that the N-terminus of these G8H enzymes plays an important role in cellular activity by swapping experiments.Finally,the combination of the engineered chassis,optimized biosynthesis pathway,and utilization of G8H led to the final strain with more than 30-fold improvement in producing 8-hydroxygeraniol compared with the starting strain.Overall,this study will provide insights into the construction and optimization of yeast cells for efficient biosynthesis of 8-hydroxygeraniol and its derivatives. | Yu Zhang Mengdi Yuan Xinxin Wu Qiuhui Zhang Yuzhu Wang Liming Zheng Tsan-Yu Chiu Huiming Zhang Lei Lan Feng Wang Ying Liao Xuemei Gong Shirui Yan Yun Wang Yue Shen Xian Fu | 2023 | mLife2023,2,4: | 0 |
