| 1 | Androgen receptor: structure, role in prostate cancer and drug discovery显示文摘Androgens and androgen receptors (AR) play a pivotal role in expression of the male phenotype. Several diseases, such as androgen insensitivity syndrome (AIS) and prostate cancer, are associated with alterations in AR functions. Indeed, androgen blockade by drugs that prevent the production of androgens and/or block the action of the AR inhibits prostate cancer growth. However, resistance to these drugs often occurs after 2-3 years as the patients develop castration-resistant prostate cancer (CRPC). In CRPC, a functional AR remains a key regulator. Early studies focused on the functional domains of the AR and its crucial role in the pathology. The elucidation of the structures of the AR DNA binding domain (DBD) and ligand binding domain (LBD) provides a new framework for understanding the functions of this receptor and leads to the development of rational drug design for the treatment of prostate cancer. | 8 | Adhesion of PBO Fiber in Epoxy Composites显示文摘The high mechanical and thermal performance of poly p-phenylene-2, 6-benzobisoxazole(PBO) fiber provides great potential applications as reinforcement fibers for composites. A composite of PBO fiber and epoxy resin has excellent electrical insulation properties, therefore, it is considered to be the best choice for the reinforcement in high magnetic field coils for pulsed magnetic fields up to 100 T. However, poor adhesion between PBO fiber and matrix is found because of the chemically inactive and/or relatively smooth surface of the reinforcement fiber preventing efficient chemical bonding in the interface, which is a challenging issue to improve mechanical properties. Here, we report the surface modification of PBO fibers by ultraviolet (UV) irradiation, O2 and NH3 plasma, as well as acidic treatments. The interfacial adhesion strength values of all the treatments show the similar level as determined for aramid fibers by pull-out tests, a significant impact on fiber-matrix-adhesion was not achieved. The surface free energy and roughness are increased for both sized and extracted fibers after plasma treatments together with maleic anhydride grafting. The sized fiber shows marginal improvement in adhesion strength and no change in fiber tensile strength because of the barrier effect of the finish. For the extracted fiber, different surface treatments either show no apparent effect or cause reduction in adhesion strength. Atomic force microscopy (AFM) topography analysis of the fracture surfaces proved adhesive failure at the fiber surface. The fiber surface roughness is increased and more surface flaws are induced, which could result in coarse interface structures when the treated fiber surface has no adequate wetting and functional groups. The adhesion failure is further confirmed by similar adhesion strength and compression shear strength values when the fiber was embedded in various epoxy resins with different temperature behavior. The tensile strength of fiber is sensitive to surface treatment conditions as revealed by a bimodal Weibull statistical distribution analysis. | 13 | Burden of onchocerciasis-associated epilepsy:first estimates and research priorities显示文摘Background:Since the 1990s,evidence has accumulated of an increased prevalence of epilepsy in onchocerciasisendemic areas in Africa as compared to onchocerciasis-free areas.Although the causal relationship between onchocerciasis and epilepsy has yet to be proven,there is likely an association.Here we discuss the need for disease burden estimates of onchocerciasis-associated epilepsy(OAE),provide them,detail how such estimates should be refined,and discuss the socioeconomic impact of OAE,including a cost-estimate for anti-epileptic drugs.Main body:Providing OAE burden estimates may aid prevention of epilepsy in onchocerciasis-endemic areas by inciting and informing collaboration between onchocerciasis control programmes and mental health services.Epilepsy not only massively impacts the health of those affected,but it also carries a high socioeconomic burden for the households and communities involved.We used previously published geospatial estimates of onchocerciasis in Africa and a separately published logistic regression model quantifying the association between onchocerciasis and epilepsy to estimate the number of OAE cases.We then applied disability weights for epilepsy to quantify the burden in terms of years of life lived with disability(YLD)and estimate the cost of treatment.We estimate that in 2015 roughly 117000 people were affected by OAE across onchocerciasis-endemic areas previously under the African Programme for Onchocerciases control(APOC)mandate where OAE has ever been reported or suspected,and another 264000 persons in onchocerciasis-endemic areas where OAE has never been investigated before.The total number of YLDs due to OAE was 39300 and 88700 in these areas respectively,based on a weighted mean disability weight of 0.336.The burden of OAE is approximately 13%of the total YLDs attributable to onchocerciasis and 10%of total YLDs attributable to epilepsy.We estimated that by 2015 the total costs of treatment with antiepileptic drug for OAE cases would have been a minimum of 12.4 million US$.Conclusions:These estimates suggest a considerable health,social and economic burden of OAE in Africa.The treatment and care for people with epilepsy,especially in hyperendemic onchocerciasis areas with high epilepsy prevalence thus requires more financial and human resources. | Natalie V.S.Vinkeles Melchers Sarah Mollenkopf Robert Colebunders Michael Edlinger Luc E.Coffeng Julia Irani Tresor Zola Joseph N.Siewe Sake Jde Vlas Andrea S.Winkler Wilma A.Stolk | 2018 | Infectious Diseases of Poverty2018,7,1: | 3 |
