| 3 | Polarization modulation with optical lock-in detection reveals universal fluorescence anisotropy of subcellular structures in live cells显示文摘The orientation of fluorophores can reveal crucial information about the structure and dynamics of their associated subcellular organelles.Despite significant progress in super-resolution,fluorescence polarization microscopy remains limited to unique samples with relatively strong polarization modulation and not applicable to the weak polarization signals in samples due to the excessive background noise.Here we apply optical lock-in detection to amplify the weak polarization modulation with super-resolution.This novel technique,termed optical lock-in detection super-resolution dipole orientation mapping(OLID-SDOM),could achieve a maximum of 100 frames per second and rapid extraction of 2D orientation,and distinguish distance up to 50 nm,making it suitable for monitoring structural dynamics concerning orientation changes in vivo.OLID-SDOM was employed to explore the universal anisotropy of a large variety of GFP-tagged subcellular organelles,including mitochondria,lysosome,Golgi,endosome,etc.We found that OUF(Orientation Uniformity Factor)of OLID-SDOM can be specific for different subcellular organelles,indicating that the anisotropy was related to the function of the organelles,and OUF can potentially be an indicator to distinguish normal and abnormal cells(even cancer cells).Furthermore,dual-color super-resolution OLID-SDOM imaging of lysosomes and actins demonstrates its potential in studying dynamic molecular interactions.The subtle anisotropy changes of expanding and shrinking dendritic spines in live neurons were observed with real-time OLID-SDOM.Revealing previously unobservable fluorescence anisotropy in various samples and indicating their underlying dynamic molecular structural changes,OLID-SDOM expands the toolkit for live cell research. | Meiling Guan Miaoyan Wang Karl Zhanghao Xu Zhang Meiqi Li Wenhui Liu Jing Niu Xusan Yang Long Chen Zhenli Jing Micheal Q.Zhang Dayong Jin Peng Xi Juntao Gao | 2022 | Light(Science & Applications)2022,11,1: | 1 |

| 5 | A petunia transcription factor, PhOBF1, regulates flower senescence by modulating gibberellin biosynthesis显示文摘Flower senescence is commonly enhanced by the endogenous hormone ethylene and suppressed by the gibberellins(GAs)in plants.However,the detailed mechanisms for the antagonism of these hormones during f lower senescence remain elusive.In this study,we characterized one up-regulated gene PhOBF1,belonging to the basic leucine zipper transcription factor family,in senescing petals of petunia(Petunia hybrida).Exogenous treatments with ethylene and GA3 provoked a dramatic increase in PhOBF1 transcripts.Compared with wild-type plants,PhOBF1-RNAi transgenic petunia plants exhibited shortened f lower longevity,while overexpression of PhOBF1 resulted in delayed flower senescence.Transcript abundances of two senescence-related genes PhSAG12 and PhSAG29 were higher in PhOBF1-silenced plants but lower in PhOBF1-overexpressing plants.Silencing and overexpression of PhOBF1 affected expression levels of a few genes involved in the GA biosynthesis and signaling pathways,as well as accumulation levels of bioactive GAs GA1 and GA3.Application of GA3 restored the accelerated petal senescence to normal levels in PhOBF1-RNAi transgenic petunia lines,and reduced ethylene release and transcription of three ethylene biosynthetic genes PhACO1,PhACS1,and PhACS2.Moreover,PhOBF1 was observed to specifically bind to the PhGA20ox3 promoter containing a G-box motif.Transient silencing of PhGA20ox3 in petunia plants through tobacco rattle virus-based virus-induced gene silencing method led to accelerated corolla senescence.Our results suggest that PhOBF1 functions as a negative regulator of ethylene-mediated f lower senescence by modulating the GA production. | Xiaotong Ji Ziwei Xin Yanping Yuan Meiling Wang Xinyi Lu Jiaqi Li Yanlong Zhang Lixin Niu Cai-Zhong Jiang Daoyang Sun | 2023 | Horticulture Research2023,10,4: | 0 |
| 6 | Porous microtubes of nickel-cobalt double oxides as non-enzymatic hydrogen peroxide sensors显示文摘Non-enzymatic electrochemical sensors for the determination of hydrogen peroxide(H_(2)O_(2))have attracted more and more concerns.A series of nickel and cobalt double oxides(Ni_(x)Co_(y)-DO)with the different ratios of Ni/Co have been prepared by a polyol-mediated solvothermal method for H_(2)O_(2)detection.The obtained products exhibit honeycomb-like open porous microtubes constituted with the low-dimensional nanostructured Ni_(x)Co_(y)-DO blocks after the calcination treatment.Compared with nickel oxides,the introduced Co ions in Ni_(x)Co_(y)-DO can induce the production of surficial oxygen vacancies,and further enhance the electrode surface activity.In particular,the NiCo-DO sample(with an atomic ratio of Ni/Co=4:3)shows the richest surficial oxygen vacancies and presents the highest H_(2)O_(2)detection activity among all the as-prepared samples,demonstrating an excellent sensitivity of698.60μAL mmol^(-1)cm^(-2)(0~0.4 mmol/L),low detection limit(0.28μmol/L,S/N=3),as well as long stability,high selectivity and good reproducibility.This work lends a new impetus to the potential application of double metal oxides for the next generation of non-enzymatic sensors. | Qin Li Jin Niu Meiling Dou Zhengping Zhang Feng Wang | 2021 | Chinese Chemical Letters2021,32,3: | 0 |
| 7 | Histone lactylation driven by mROS-mediated glycolytic shift promotes hypoxic pulmonary hypertension显示文摘Increased mitochondrial reactive oxygen species(mROS)and glycolysis have been established in pulmonary hypertension(PH).However,the effect of elevated mROS on glycolytic shift and how increased glycolysis promotes hypoxic pulmonary artery smooth muscle cell(PASMC)proliferation and vascular remodeling remain elusive.Here,we reported that hypoxia-induced mROS inhibit HIF-1αhydroxylation and further trigger PASMC glycolytic switch through the upregulated HIF-1α/PDK1&PDK2/p-PDH-E1αaxis,which facilitates lactate accumulation and histone lactylation.Through H3K18la and HIF-1αChIP–seq analysis,we found that the enhanced histone lactylation of HIF-1αtargets,such as Bmp5,Trpc5,and Kit,promotes PASMC proliferation.Knockdown of Pdk1&2 blunts lactate production,histone lactylation marks,and PASMC proliferation.Moreover,pharmacological intervention with lactate dehydrogenase inhibitor diminishes histone lactylation and ameliorates PASMC proliferation and vascular remodeling in hypoxic PH rats.Taken together,this study provides proof of concept for anti-remodeling therapy through lactate manipulation. | Jian Chen Meiling Zhang Yanjie Liu Shihong Zhao Yanxia Wang Meng Wang Wen Niu Faguang Jin Zhichao Li | 2022 | Journal of Molecular Cell Biology2022,14,12: | 0 |
