1Hydrogen sutfide protects colon cancer cells from chemopreventative agent β-phenylethyl isothiocvanate induced apoptosis显示文摘AIM: Hydrogen sulfide (H2S) is a prominent gaseous constituent of the gastrointestinal (GI) tract with known cytotoxic properties. Endogenous concentrations of H2S are reported to range between 0.2-3.4 mmol/L in the GI tract of mice and humans. Considering such high levels we speculate that, at non-toxic concentrations, H2S may interact with chemical agents and alter the response of colonic epithelium cells to such compounds. The GI tract is a major site for the absorption of phytochemical constituents such as isothiocyanates, flavonoids, and carotenoids, with each group having a role in the prevention of human diseases such as colon cancer. The chemopreventative properties of the phytochemical agent β-phenyethyl isothiocyanate (PEITC) are well recognized. However, little is currently known about the physiological or biochemical factors present in the GI tract that may influence the biological properties of ITCs. The current study was undertaken to determine the effects of H2S on PEITC mediated apoptosis in colon cancer cells.METHODS: Induction of apoptosis by PEITC in human colon cancer HCT116 cells was assessed using classic apoptotic markers namely SubG1 population analysis,caspase-3 like activity and nuclear fragmentation and condensation coupled with the MTT (3-(4,5-dimethylthiazol2-yl)-2,5-diphenyl tetrasodium bromide) viability assay and LDH leakage.RESULTS: PEITC significantly induced apoptosis in HGT116cells as assessed by SubG1 population formation, nuclear condensation, LDH leakage and caspase-3 activity after 24 h, these data being significant from control groups (P<0.01). In contrast, co-treatment of cells with physiological concentrations of H2S (0.1-1 mmol/L) prevented PEITC mediated apoptosis as assessed using the parameters described.CONCLUSION: PEITC effectively induced cell death in the human adenocarcinoma cell line HCT116 in vitro through classic apoptotic mechanisms. However, in the presence of H2S, apoptosis was abolished. 2Dredging effects on selected nutrient concentrations and ecoenzymatic activity in two drainage ditch sediments in the lower Mississippi River Valley显示文摘Agricultural drainage ditches are conduits between production acreage and receiving aquatic systems.Often overlooked for their mitigation capabilities,agricultural drainage ditches provide an important role for nutrient transformation via microbial metabolism.Variations in ecoenzyme activities have been used to elucidate microbial metabolism and resource demand of microbial communities to better understand the relationship between altered nutrient ratios and microbial activity in aquatic ecosystems.Two agricultural drainage ditches,one in the northeast portion of the Arkansas Delta and the other in the lower Mississippi Delta,were monitored for a year.Sediment samples were collected prior to each ditch being dredged(cleaned),and subsequent post-dredging samples occurred as soon as access was available.Seasonal samples were then collected throughout a year to examine effects of dredging on selected nutrient concentrations and ecoenzymatic activity recovery in drainage ditch sediments.Phosphorus concentrations in sediments after dredging decreased 33–66%,depending on ditch and phosphorus extraction methodology.Additionally,ecoenzymatic activity was significantly decreased in most sediment samples after dredging.Fluorescein diacetate hydrolysis activity,an estimate of total microbial activity,decreased 56–67%after dredging in one of the two ditches.Many sample sites also had significant phosphorus and ecoenzymatic activity differences between the post-dredge samples and the year-long follow-up samples.Results indicate microbial metabolism in dredged drainage ditches may take up to a year or more to recover to pre-dredged levels.Likewise,while sediment nutrient concentrations may be decreased through dredging and removal,runoff and erosion events over time tend to quickly replenish nutrient concentrations in replaced sediments.Understanding nutrient dynamics and microbial metabolism within agricultural drainage ditches is a crucial step toward addressing issues of nutrient enrichment in aquatic receiving systems,especially those contributing to the Gulf of Mexico.Matt Moore Martin A.Locke Michael Jenkins Robert W.Steinriede Daniel S.McChesney 2017International Soil and Water Conservation Research2017,5,3:1

10Reservoir construction from the Newberry Volcano EGS Demonstration显示文摘The Newberry Volcano EGS Demonstration in central Oregon tests recent technological advances designed to reduce the cost of power generated by EGS in a hot,dry well(NWG 55-29) drilled in 2008. An EGS reservoir was created by injecting large volumes of cold water,causing existing fractures to slip in shear(known as hydroshearing) generating the seismic waves that can be used to map fracture location and size. At the Newberry Demonstration the final injectivity ranged between 1.4 and 1.7 L/s/MPa a ~6x improvement over the initial injectivity of the well.
