1Hepatic fibrosis: It is time to go with hepatic stellate cell-specifictherapeutic targets显示文摘Hepatic fibrosis is a pathological lesion, characterized by the progressive accumulation of extracellular matrix(ECM) in the perisinusoidal space and it is a major problem in chronic liver diseases. Phenotypic activation of hepatic stellate cells(HSC) plays a central role in the progression of hepatic fibrosis. Retardation of proliferation and clearance of activated HSCs from the injured liver is an appropriate therapeutic strategy for the resolution and treatment of hepatic fibrosis. 2Stem cells for liver tissue repair:Current knowledge and perspectives显示文摘Stem cells from extra-or intrahepatic sources have been recently characterized and their usefulness for the generation of hepatocyte-like lineages has been demonstrated. Therefore, they are being increasingly considered for future applications in liver cell therapy. In that field, liver cell transplantation is currently regarded as a possible alternative to whole organ transplantation, while stem cells possess theoretical advantages on hepatocytes as they display higher in vitro culture performances and could be used in autologous transplant procedures. However, the current research on the hepatic fate of stem cells is still facing difficulties to demonstrate the acquisition of a full mature hepatocyte phenotype, both in vitro and in vivo. Furthermore, the lack of obvious demonstration of in vivo hepatocyte-like cell functionality remains associated to low repopulation rates obtained after current transplantation procedures. The present review focuses on the current knowledge of the stem cell potential for liver therapy. We discuss the characteristics of the principal cell candidates and the methods to demonstrate their hepatic potential in vitro and in vivo. 3Use of mesenchymal stem cells to treat liver fibrosis:Current situation and future prospects显示文摘Progressive liver fibrosis is a major health issue for which no effective treatment is available,leading to cirrhosis and orthotopic liver transplantation.However,organ shortage is a reality.Hence,there is an urgent need to find alternative therapeutic strategies.Cellbased therapy using mesenchymal stem cells(MSCs) may represent an attractive therapeutic option,based ontheir immunomodulatory properties,their potential to differentiate into hepatocytes,allowing the replacement of damaged hepatocytes,their potential to promote residual hepatocytes regeneration and their capacity to inhibit hepatic stellate cell activation or induce their apoptosis,particularly via paracrine mechanisms.The current review will highlight recent findings regarding the input of MSC-based therapy for the treatment of liver fibrosis,from in vitro studies to pre-clinical and clinical trials.Several studies have shown the ability of MSCs to reduce liver fibrosis and improve liver function.However,despite these promising results,some limitations need to be considered.Future prospects will also be discussed in this review.Silvia Berardis Prenali Dwisthi Sattwika Mustapha Najimi Etienne Marc Sokal 2015World Journal of Gastroenterology2015,21,3:22
4Phenolic and flavonoid contents, antioxidant and antimicrobial activities of leaf extracts from ten Algerian Ficus carica L. varieties显示文摘Objective: To determine the total phenolic and flavonoid contents, antioxidant and antimicrobial activities of methanolic leaf extracts of ten Algerian fig(Ficus carica L.)varieties(uniferous, biferous and caprifig tree).Methods: Phenolics were extracted by Soxhlet method and analyzed by the Folin–Ciocalteu colorimetric method. Flavonoids were determined by aluminum trichloride assay and the antioxidant capacity was determined by the 2,2-diphenyl-1-picrylhydrazyl radical scavenging assay. The antimicrobial activity was studied with the disc diffusion method and a macrodilution broth method was used to determine the minimal inhibitory concentrations and minimal lethal concentrations.Results: The mean extract yield was 14.10% ± 0.66%(n = 10). Leaf extract of biferous followed by uniferous varieties had the highest total phenolic contents [(52.296 ± 5.232)and(48.973 ± 2.015) mg gallic acid equivalent/g of dry plant extract respectively], flavonoids [(14.388 ± 0.333) and(14.136 ± 1.082) mg quercetin equivalent/g of dry plant extract] and antioxidant capacity [IC50(798.754 ± 108.590) and(825.004 ± 110.835) mg/m L]. Antioxidant capacity of fig leaves was significantly correlated with phenolic contents(r = 0.748). These extracts showed bactericidal activity and moderate antifungal activity, and the minimal inhibitory concentrations and minimal lethal concentrations were determined on Bacillus cereus and Staphylococcus aureus.Conclusions: All tested extracts contain phenolic compounds and exhibited an antioxidant activity and an antimicrobial effect against Gram-positive and Gram-negative bacteria. 5玄武岩-聚丙烯混杂纤维混凝土硫酸盐腐蚀试验研究显示文摘通过对单掺和三种不同混杂比例混掺的方式,以体积掺加率0.3%、0.6%、0.9%、1.2%将玄武岩、聚丙烯纤维掺入普通C30混凝土中形成混杂纤维混凝土,对其进行7d、14d、28d龄期的抗硫酸盐腐蚀性能试验研究。结果表明,对基体混凝土在龄期为7d的抗压强度耐蚀系数提高最为显著的是单掺纤维系列与1:1、1:2混杂纤维系列纤维混凝土。各系列纤维对基体混凝土14d耐蚀系数提高的最佳纤维掺加率在0.3%附近;单掺聚丙烯纤维系列、单掺玄武岩纤维系列对基体28d耐蚀系数提高的最佳纤维掺加率分别在0.3%、0.9%附近。总体上混杂纤维系列纤维混凝土的抗硫酸静泡腐蚀能力优于单掺纤维系列纤维混凝土。王学志 贺晶晶 邹浩飞 郑淑文 Mustapha Abdulhadi 2014混凝土与水泥制品2014,,5:15
6Metastatic gastric cancer treatment: Second line and beyond显示文摘Advanced gastric cancer(a GC), not amenable to curative surgery, is still a burdensome illness tormenting afflicted patients and their healthcare providers. Whereas combination chemotherapy has been shown to improve survival and tumor related symptoms in the frontline setting, second-line therapy(SLT) is subject to much debate in the scientific community, mainly because of the debilitating effects of GC, which would impede the administration of cytotoxic therapy. Recent data has provided sufficient evidence for the safe use of SLT in patients with an adequate performance status. Taxanes, Irinotecan and even some Fluoropyrimidine analogs were found to provide a survival advantage in this subset of patients. Most importantly, quality of life measures were also improved through the use of adequate therapy. Even more pertinent were the findings involving antiangiogenic agents, which would add measurable improvements without significantly jeopardizing the patients' well-being. Further lines of therapy are cause for much more debate nowadays, but specific targeted agents have shown considerable promise in this context. 7Liver cell transplantation for Crigler-Najjar syndrome type Ⅰ: Update and perspectives显示文摘Liver cell transplantation is an attractive technique to treat liver-based inborn errors of metabolism. The feasibility and efficacy of the procedure has been demonstrated, leading to medium term partial metabolic control of various diseases. Crigler-Najjar is the paradigm of such diseases in that the host liver is lacking one function with an otherwise normal parenchyma. The patient is at permanent risk for irreversible brain damage. The goal of liver cell transplantation is to reduce serum bilirubin levels within safe limits and to alleviate phototherapy requirements to improve quality of life. Preliminary data on Gunn rats, the rodent model of the disease, were encouraging and have led to successful clinical trials. Herein we report on two additional patients and describe the current limits of the technique in terms of durability of the response as compared to alternative therapeutic procedures. 8Hepatocyte cryopreservation:Is it time to change the strategy?显示文摘Liver cell transplantation presents clinical benefit in patients with inborn errors of metabolism as an alternative,or at least as a bridge,to orthotopic liver transplantation.The success of such a therapeutic approach remains limited by the quality of the transplanted cells.Cryopreservation remains the best option for long-term storage of hepatocytes,providing a permanent and sufficient cell supply.However, isolated adult hepatocytes are poorly resistant to such a process,with a significant alteration both at the morphological and functional levels.Hence,the aim of the current review is to discuss the state of the art regarding widely-used hepatocyte cryopreservation protocols,as well as the assays performed to analyse the post-thawing cell quality both in vitro and in vivo. The majority of studies agree upon the poor quality and efficiency of cryopreserved/thawed hepatocytes as compared to freshly isolated hepatocytes.Intracellular ice formation or exposure to hyperosmotic solutionsremains the main phenomenon of cryopreservation process,and its effects on cell quality and cell death induction will be discussed.The increased knowledge and understanding of the cryopreservation process will lead to research strategies to improve the viability and the quality of the cell suspensions after thawing.Such strategies,such as vitrification,will be discussed with respect to their potential to significantly improve the quality of cell suspensions dedicated to liver cell-based therapies.Xavier Stéphenne Mustapha Najimi Etienne M Sokal 2010World Journal of Gastroenterology2010,16,1:9
9Silibinin induces hepatic stellate cell cycle arrest via enhancing p53/p27 and inhibiting Akt downstream signaling protein expression显示文摘BACKGROUND: Proliferation of hepatic stellate cells(HSCs) plays a pivotal role in the progression of liver fibrosis consequent to chronic liver injury. Silibinin, a flavonoid compound,has been shown to possess anti-fibrogenic effects in animal models of liver fibrosis. This was attributed to an inhibition of cell proliferation of activated HSCs. The present study was to gain insight into the molecular pathways involved in silibinin anti-fibrogenic effect. METHODS: The study was conducted on LX-2 human stellate cells treated with three concentrations of silibinin(10, 50 and 100 μmol/L) for 24 and 96 hours. At the end of the treatment cell viability and proliferation were evaluated. Protein expression of p27, p21, p53, Akt and phosphorylated-Akt was evaluated by Western blotting analysis and Ki-67 protein expression was by immunocytochemistry. Sirtuin activity was evaluated by chemiluminescence based assay. RESULTS: Silibinin inhibits LX-2 cell proliferation in doseand time-dependent manner; we showed that silibinin upregulated the protein expressions of p27 and p53. Such regulation was correlated to an inhibition of both downstream Akt and phosphorylated-Akt protein signaling and Ki-67 protein expression. Sirtuin activity also was correlated to silibinininhibited proliferation of LX-2 cells. 10Emotion processing in Parkinson's disease: a blood oxygenation level-dependent functional magnetic resonance imaging study显示文摘Parkinson's disease is a neurodegenerative disorder caused by loss of dopamine neurons in the substantia nigra pars compacta. Tremor, rigidity, and bradykinesia are the major symptoms of the disease. These motor impairments are often accompanied by affective and emotional dysfunctions which have been largely studied over the last decade. The aim of this study was to investigate emotional processing organization in the brain of patients with Parkinson's disease and to explore whether there are differences between recognition of different types of emotions in Parkinson's disease. We examined 18 patients with Parkinson's disease(8 men, 10 women) with no history of neurological or psychiatric comorbidities. All these patients underwent identical brain blood oxygenation level-dependent functional magnetic resonance imaging for emotion evaluation. Blood oxygenation level-dependent functional magnetic resonance imaging results revealed that the occipito-temporal cortices, insula, orbitofrontal cortex, basal ganglia, and parietal cortex which are involved in emotion processing, were activated during the functional control. Additionally, positive emotions activate larger volumes of the same anatomical entities than neutral and negative emotions. Results also revealed that Parkinson's disease associated with emotional disorders are increasingly recognized as disabling as classic motor symptoms. 12Risk modification of colorectal cancer susceptibility by interleukin-8-251T>A polymorphism in Malaysians显示文摘AIM: To investigate the allele and genotype frequencies and associated risk of interleukin (IL)-8-251T>A polymorphism on colorectal cancer (CRC) susceptibility risk. METHODS: Peripheral blood samples of 255 normal controls and 255 clinically and histopathologically confirmed CRC patients were genotyped for IL-8-251T>A polymorphism employing allele-specific polymerase chain reaction. The relative association of variant allele and genotypes with CRC susceptibility risk was determined by calculating the odds ratios (ORs). Corresponding χ 2 tests on the CRC patients and controls were carried out and 95% confidence intervals (CIs) were determined using Fisher's exact test. The allele frequencies and its risk association were calculated using FAMHAP, haplotype association analysis software. RESULTS: On comparing the frequencies of genotypesof patients and controls, the homozygous variant AA was significantly higher in CRC patients (P = 0.002) compared to controls. Investigation on the association of the polymorphic genotypes with CRC susceptibility risk, showed that the homozygous variant IL-8 -251AA had a significantly increased risk with OR 3.600 (95% CI: 1.550-8.481, P = 0.001). In the case of allele frequencies, variant allele A of IL-8 -251 showed a significantly increased risk of CRC predisposition with OR 1.32 (95% CI: 1.03-1.69,P = 0.003). 15Modeling of fiber bridging in fluid flow for well stimulation applications显示文摘Accurate acid placement constitutes a major concern in matrix stimulation because the acid tends to penetrate the zones of least resistance while leaving the low-permeability regions of the formation untreated.Degradable materials(fibers and solid particles)have recently shown a good capability as fluid diversion to overcome the issues related to matrix stimulation.Despite the success achieved in the recent acid stimulation jobs stemming from the use of some products that rely on fiber flocculation as the main diverting mechanism,it was observed that the volume of the base fluid and the loading of the particles are not optimized.The current industry lacks a scientific design guideline because the used methodology is based on experience or empirical studies in a particular area with a particular product.It is important then to understand the fundamentals of how acid diversion works in carbonates with different diverting mechanisms and diverters.Mathematical modeling and computer simulations are effective tools to develop this understanding and are efficiently applied to new product development,new applications of existing products or usage optimization.In this work,we develop a numerical model to study fiber dynamics in fluid flow.We employ a discrete element method in which the fibers are represented by multi-rigid-body systems of interconnected spheres.The discrete fiber model is coupled with a fluid flow solver to account for the inherent simultaneous interactions.The focus of the study is on the tendency for fibers to flocculate and bridge when interacting with suspending fluids and encountering restrictions that can be representative of fractures or wormholes in carbonates.The trends of the dynamic fiber behavior under various operating conditions including fiber loading,flow rate and fluid viscosity obtained from the numerical model show consistency with experimental observations.The present numerical investigation reveals that the bridging capability of the fiber–fluid system can be enhanced by increasing the fiber loading,selecting fibers with higher stiffness,reducing the injection flow rate,reducing the suspending fluid viscosity or increasing the attractive cohesive forces among fibers by using sticky fibers.Mehdi Ghommem Mustapha Abbad Gallyam Aidagulov Steve Dyer Dominic Brady 2020Petroleum Science2020,17,3:4

18Surfactant-decorated graphite nanoplatelets(GNPs) reinforced aluminum nanocomposites: sintering effects on hardness and wear显示文摘The exceptional properties of graphene make it ideal as a reinforcement to enhance the properties of aluminum matrices and this critically depends on uniform dispersion. In this study, the dispersion issue was addressed by sonication and non-covalent surface functionalization of graphite nanoplatelets(GNPs) using two types of surfactant: anionic(sodium dodecyl benzene sulfate(SDBS)) and non-ionic polymeric(ethyl cellulose(EC)). After colloidal mixing with Al powder, consolidation was performed at two sintering temperatures(550 and 620°C). The structure, density, mechanical and wear properties of the nanocomposite samples were investigated and compared with a pure Al and a pure GNPs/Al nanocomposite sample. Noticeably, EC-based 0.5 wt% GNPs/Al samples showed the highest increment of 31% increase in hardness with reduced wear rate of 98.25% at 620°C, while a 22% increase in hardness with reduced wear rate of 96.98% at 550°C was observed, as compared to pure Al. 19Mathematical modeling of COVID-19 epidemic with effect of awareness programs显示文摘Severe acute respiratory syndrome coronavirus 2(SARS-COV-2)is a novel virus that emerged in China in late 2019 and caused a pandemic of coronavirus disease 2019(COVID-19).The epidemic has largely been controlled in China since March 2020,but continues to inflict severe public health and socioeconomic burden in other parts of the world.One of the major reasons for China’s success for the fight against the epidemic is the effectiveness of its health care system and enlightenment(awareness)programs which play a vital role in the control of the COVID-19 pandemic.Nigeria is currently witnessing a rapid increase of the epidemic likely due to its unsatisfactory health care system and inadequate awareness programs.In this paper,we propose a mathematical model to study the transmission dynamics of COVID-19 in Nigeria.Our model incorporates awareness programs and different hospitalization strategies for mild and severe cases,to assess the effect of public awareness on the dynamics of COVID-19 infection.We fit the model to the cumulative number of confirmed COVID-19 cases in Nigeria from 29 March to 12 June 2020.We find that the epidemic could increase if awareness programs are not properly adopted.We presumed that the effect of awareness programs could be estimated.Further,our results suggest that the awareness programs and timely hospitalization of active cases are essential tools for effective control and mitigation of COVID-19 pandemic in Nigeria and beyond.Finally,we perform sensitive analysis to point out the key parameters that should be considered to effectively control the epidemic.Salihu Sabiu Musa Sania Qureshi Shi Zhao Abdullahi Yusuf Umar Tasiu Mustapha Daihai He 2021Infectious Disease Modelling2021,6,1:3
20Thermal Analysis of a Phase Change Material Based Heat Sink for Cooling Protruding Electronic Chips显示文摘This work aims to numerically study the melting natural convection in a rectangular enclosure heated by threediscreet protruding electronic chips. The heat sources generate heat at a constant and uniform volumetric rate. Apart of the power generated in the heat sources is dissipated to a phase change material (PCM, n-eicosane withmelting temperature, Tm=36℃). Numerical investigations were carried out in order to examine the effects of theplate thickness on the maximum temperature of electronic components, the percentage contribution of plate heatconduction on the total removed heat and temperature profiles in the plate.
