| 1 | WIP1 Phosphatase Plays a Critical Neuroprotective Role in Brain Injury Induced by High-Altitude Hypoxic Inflammation显示文摘The hypobaric hypoxic environment in highaltitude areas often aggravates the severity of inflammation and induces brain injury as a consequence. However, the critical genes regulating this process remain largely unknown. The phosphatase wild-type p53-induced phosphatase 1(WIP1) plays important roles in various physiological and pathological processes, including the regulation of inflammation in normoxia, but its functions in hypoxic inflammation-induced brain injury remain unclear.Here, we established a mouse model of this type of injury and found that WIP1 deficiency augmented the release of inflammatory cytokines in the peripheral circulation and brain tissue, increased the numbers of activated microglia/macrophages in the brain, aggravated cerebral histological lesions, and exacerbated the impairment of motor and cognitive abilities. | 2 | Branched glycerol dialkyl glycerol tetraethers and paleoenvironmental reconstruction in Zoigê peat sediments during the last 150 years显示文摘Branched glycerol dialkyl glycerol tetraethers (GDGTs) were analyzed in peat samples taken from the Zoigê grassland, located on the eastern margin of the Qinghai-Tibetan Plateau. The mean annual air temperature (MAAT) and pH in the 210 Pb-dating peat core were reconstructed based on the cyclization of branched tetraethers (CBT) and methylation of branched tetraethers (MBT) indices for the last 150 years. The results showed that the MAAT and pH values involved three stages of variation over the last 150 years. The pH was stable and the MAAT decreased slowly by about 1°C from 1851 to 1927. The pH decreased rapidly by 0.6 and the MAAT decreased significantly by 2.5°C between 1927 and 1979. The pH and MAAT increased by 0.8 and 5°C, respectively, since the 1980s. The reconstructed temperatures correspond well to the mean winter temperatures reconstructed using tree-rings from the Jiuzhaigou Valley, situated at the boundary between the Qinghai-Tibetan Plateau and the Sichuan Basin. The estimated MAAT was similar to the temperatures reconstructed using tree-rings, indicating an increasing trend, as shown by local instrumental records from 1957 to 2001. | 3 | A fundamental theorem for eco-environmental surface modelling and its applications显示文摘We propose a fundamental theorem for eco-environmental surface modelling(FTEEM) in order to apply it into the fields of ecology and environmental science more easily after the fundamental theorem for Earth’s surface system modeling(FTESM). The Beijing-Tianjin-Hebei(BTH) region is taken as a case area to conduct empirical studies of algorithms for spatial upscaling, spatial downscaling, spatial interpolation, data fusion and model-data assimilation, which are based on high accuracy surface modelling(HASM), corresponding with corollaries of FTEEM. The case studies demonstrate how eco-environmental surface modelling is substantially improved when both extrinsic and intrinsic information are used along with an appropriate method of HASM. Compared with classic algorithms, the HASM-based algorithm for spatial upscaling reduced the root-meansquare error of the BTH elevation surface by 9 m. The HASM-based algorithm for spatial downscaling reduced the relative error of future scenarios of annual mean temperature by 16%. The HASM-based algorithm for spatial interpolation reduced the relative error of change trend of annual mean precipitation by 0.2%. The HASM-based algorithm for data fusion reduced the relative error of change trend of annual mean temperature by 70%. The HASM-based algorithm for model-data assimilation reduced the relative error of carbon stocks by 40%. | 4 | High-precision measurement of mercury isotope ratios of atmospheric deposition over the past 150 years recorded in a peat core taken from Hongyuan, Sichuan Province, China显示文摘High-precision 210Pb dating technology was applied to a peat core with a time span of about 150 years that was taken from Hongyuan, Sichuan Province, China. The concentrations of total mercury (Hg) and stable isotope compositions of mercury in the peat core were measured using a LUMEX 915 instrument and multi-collector inductively coupled plasma mass spectrometer, respectively. Total mercury (Hg) concentrations in the peat core had a clearly increasing trend from the bottom to top of the core while δ 202Hg values (relative to NIST 3133) of peat had a decreasing trend. The total mercury (Hg) concentration varied from 16.7 to 101.3 ng/g and the δ 202Hg values ranged from –0.44‰± 0.14‰ to –1.45‰± 0.22‰. | 6 | Development and utilization of a new chemically-induced soybean library with a high mutation density显示文摘Mutagenized populations have provided important materials for introducing variation and identifying gene function in plants. In this study, an ethyl methanesulfonate(EMS)-induced soybean(Glycine max) population,consisting of 21,600 independent M_2 lines, was developed.Over 1,000 M_(4(5))families, with diverse abnormal phenotypes for seed composition, seed shape, plant morphology and maturity that are stably expressed across different environments and generations were identified. Phenotypic analysis of the population led to the identification of a yellow pigmentation mutant, gyl, that displayed significantly decreased chlorophyll(Chl) content and abnormal chloroplast development. Sequence analysis showed that gyl is allelic to Minn Gold, where a different single nucleotide polymorphism variation in the Mg-chelatase subunit gene(ChlI1a) results in golden yellow leaves. A cleaved amplified polymorphic sequence marker was developed and may be applied to marker-assisted selection for the golden yellow phenotype in soybean breeding. | 7 | Tourist Behaviors in Wetland Park:A Preliminary Study in Xixi National Wetland Park,Hangzhou,China显示文摘Public education as well as tourism and leisure are the two important functions of national wetland park, and tourist behaviors act as the guiding factor in the national wetland park construction and management.As the first national wetland park in China, Xixi National Wetland Park, located in Hangzhou City, Zhejiang Province, will give constructive instructions to the construction and management of other wetland parks and ecotourism resorts.This paper analyzed the tourist behaviors in Xixi National Wetland Park.By the observation, in-depth interview and questionnaire, the results show that tourists in wetland parks are different from those in the other common destinations, and they are also different from those strict eco-tourists.In fact, the tourists in wetland park have unique characteristics, such as the intense perception to the functions of wetland ecosystem and the environment impact behaviors.Those tourists are preferred to natural landscapes in their motivations and expectations.Wetland landscapes are the main image to those tourists in the Xixi National Wetland Park.Tourist expectation, experience and satisfaction are all critical factors for the success of the planning and management in national wetland parks.Based on the results, some measures for the sustainable development of wetland parks, including to optimize wetland theme landscape and tourism products, to improve tourist facilities and services, to enhance quality of visitor experiences, and to implement total quality management based on tourist characteristics, are given to national wetland park ecotourism. | PAN Lili CUI Lijuan WU Ming | 2010 | Chinese Geographical Science2010,20,1: | 5 |

| 9 | Cardiomyocyte-like differentiation of human bone marrow mesenchymal stem cells after exposure to 5-azacytidine in vitro显示文摘Objective To investigate the potential of adult mesenchymal stem cells (MSCs) derived from human bone marrow to undergo cardiomyogenic differentiation after exposure to 5-azacytidine (5-aza) in vitro. Methods A small bone marrow aspirate was taken from the iliac crest of human volunteers, and hMSCs were isolated by 1.073g/mL Percoll and propagated in the right cell culturing medium as previously described. The phenotypes of hMSCs were characterized with the use of flow cytometry. The hMSCs were cultured in cell culture medium (as control) and medium mixed with 5-aza for cellular differentiation. We examined by immunohistochemistry at 21 days the inducement of desmin, cardiac-specific cardiac troponin I (cTnI), GATA 4 and connexin-43 respectively. Results The hMSCs are fibroblast-like morphology and express CD44+ CD29+ CD90+ / CD34- CD45- CD31- CD11a. After 5-aza treatment, 20-30% hMSCs connected with adjoining cells and coalesced into myotube structures after 14days. Twenty-one days after 5-aza treatment, immunofluorescence showed that some cells expressed desmin,GATA4, cTnI and connexin-43 in 5,10 μmol/L 5-aza groups, but no cardiac specific protein was found in neither 3μmol/L 5-aza group nor in the control group. The ratio of cTnI positively stained cells in 10 μmol/L group was higher than that in 5 μmol/L group (65.3 ± 4.7% vs 48.2 ± 5.4%, P < 0.05). Electron microscopy revealed that myofilaments were formed. The induced cells expressed cardiac-myosin heavy chain (MyHC) gene by reverse transcription-polymerase chain reaction (RT-PCR). Conclusions Theses findings suggest that hMSCs from adult bone marrow can be differentiated into cardiac-like muscle cells with 5-aza inducement in vitro and the differentiation is in line with the 5-aza concentration. | 17 | Protective effects of sodium butyrate on rotavirus inducing endoplasmic reticulum stress-mediated apoptosis via PERK-eIF2αsignaling pathway in IPEC-J2 cells显示文摘Background:Rotavirus(RV)is a major pathogen that causes severe gastroenteritis in infants and young animals.Endoplasmic reticulum(ER)stress and subsequent apoptosis play pivotal role in virus infection.However,the protective mechanisms of intestinal damage caused by RV are poorly defined,especially the molecular pathways related to enterocytes apoptosis.Thus,the aim of this study was to investigate the protective effect and mechanism of sodium butyrate(SB)on RV-induced apoptosis of IPEC-J2 cells.Results:The RV infection led to significant cell apoptosis,increased the expression levels of ER stress(ERS)markers,phosphorylated protein kinase-like ER kinase(PERK),eukaryotic initiation factor 2 alpha(eIF2α),caspase9,and caspase3.Blocking PERK pathway using specific inhibitor GSK subsequently reversed RV-induced cell apoptosis.The SB treatment significantly inhibited RV-induced ERS by decreasing the expression of glucose regulated protein 78(GRP78),PERK,and eIF2α.In addition,SB treatment restrained the ERS-mediated apoptotic pathway,as indicated by downregulation of C/EBP homologous protein(CHOP)mRNA level,as well as decreased cleaved caspase9 and caspase3 protein levels.Furthermore,siRNA-induced GPR109a knockdown significantly suppressed the protective effect of SB on RV-induced cell apoptosis.Conclusions:These results indicate that SB exerts protective effects against RV-induced cell apoptosis through inhibiting ERS mediated apoptosis by regulating PERK-eIF2αsignaling pathway via GPR109a,which provide new ideas for the prevention and control of RV. | Ye Zhao Ningming Hu Qin Jiang Li Zhu Ming Zhang Jun Jiang Manyi Xiong Mingxian Yang Jiandong Yang Linyuan Shen Shunhua Zhang Lili Niu Lei Chen Daiwen Chen | 2021 | Journal of Animal Science and Biotechnology2021,12,4: | 2 |
