| 5 | The intracellular mechanism of alpha-fetoprotein promoting the proliferation of NIH 3T3 cells显示文摘AIM The existence and properties of alpha-fetoprotein (AFP) receptor on the surface of NIH 3T3 cells and the effects of AFP on cellular signal transduction pathway were investigated. METHODS The effect of AFP on the proliferation of NIH 3T3 cells was measured by incorporation of 3H-TdR. Receptor-binding assay of 125I-AFP was performed to detect the properties of AFP receptor in NIH 3T3 cells. The influences of AFP on the [cAMP]i and the activities of protein kinase A (PKA) were determined. Western blot was used to detect the change of K-ras P21 protein expression. RESULTS The proliferation of NIH 3T3 cells treated with 0-80 mg/L of AFP was significantly enhanced. The Scatchard analysis indicated that there were two classes of binding sites with KD of 2.722×10-9M (Bmax=12810 sites per cell) and 8.931× 10-SM (Bmax=l19700 sites per cell) respectively. In the presence of AFP (20 mg/L), the content of cAMP and activities of PKA were significantly elevated . The level of K-ras P21 protein was upregulated by AFP at the concentration of 20 mg/L. The monoclonal antibody against AFP could reverse the effects of AFP on the cAMP content, PKA activity and the expression of K-ras p21 gene. | 6 | Morphological study on colonic pathology in ulcerative colitis treated by moxibustion显示文摘AIM To observe the therapeutic effect ofmoxibustion on ulcerative colitis and itsinfluence on the colonic mucosal morphology.METHODS Forty-six patients with ulcerativecolitis were randomly divided into themoxibustion with herbal medicine underneathgroup and the western medicine group.Thirtypatients were treated with the abovemoxibustion and 16 patients with Salicylayefapyridine(SASP).The colonic mucosa of 13patients in the moxibustion group was observedby colonoscopy before and after the treatment.Mucin was also analyzed by H.E and AB-PASstaining.RESULTS Seventeen patients were clinicallycured,12 were improved and 1 unchanged in themoxibustion group.In the control group,5patients were clinically cured,7 improved and 4unchanged.Thirteen patients with active UCwere taken as the subjects for histopathologicanalysis in this study.The colonic mucosallesions were remarkably improved and thecharacteristic of the mucin also changed.Inmost sections,the chronic inflammation ofmucosa was geatly ameliorated(P<0.01).Theinflammatory cell infiltratation much decreased and neutrophils,disapeared in most sections(P<0.001).The goblet cells significantlyincreased(P<0.001);crypt paracrypt abscessor mucosal ulceration was seen(P<0.001).CONCLUSION The rate of cure of ulcerative colitis by moxibustion with herbal medicine beneath is superior to that by SASP. | 7 | Isolation and Identification of Cancer Stem-Like Cells from Murine Melanoma Cell Lines显示文摘In current study,cancer stem-like cells in the murine melanoma B16F10 cells were investigated.CD phenotypes of the B16F10 cells were analyzed by flow cytometry,and the specific CD phenotype cells from the B16F10 cells were isolated by MACS.Then we used colony formation assay in soft agar media,the cell growth assay in serum-free culture media as well as the tumorigenicity investigation of the specific CD phenotype cells in C57BL/6 mice, respectively,to identify cancer stem-like cells in the B16F10 cells.The results showed that the B16F10 cells could form spherical clones in serum-free culture media,and the rate of clonegenesis of CD133 + ,CD44 + and CD44 + CD133 + cells was higher than that of CD133 - ,CD44 - and CD44 + CD133 - cells in soft agar media,respectively. | 9 | Association between Vitamin D Insufficiency and the Risk for Gestational Diabetes Mellitus in Pregnant Chinese Women显示文摘Objective To investigate the association between vitamin D deficiency and risk of gestational diabetes mellitus(GDM) in pregnant Chinese women.Methods A nested case-control study was conducted.Clinical and biochemical data were analyzed for 200 subjects with GDM and 200 subjects with normal glucose tolerance(NGT).Results The median(interquartile range) serum 25-hydroxyvitamin D(25OHD) levels were 22.39(17.67,29.38) and 25.86(19.09,34.88) nmol/L in the GDM and NGT groups,respectively.Rates of 25OHD deficiency or insufficiency were significantly higher in the GDM group than in the NGT group.Subjects with 25OHD levels <25 nmol/L had a 1.8-fold higher risk of GDM compared with subjects with higher vitamin D levels.In the GDM group,serum 25OHD was independently associated with HbA1c and insulin resistance after adjusting for confounding factors.In the NGT group,serum 25OHD was independently associated with fasting plasma glucose and systolic blood pressure after adjusting for maternal age and other confounding factors.Conclusion 25OHD insufficiency is very common in Chinese women.Low 25OHD status may be associated with insulin resistance and act as a risk factor for GDM. | WANG Ou NIE Min HU Ying Ying ZHANG Kui LI Wei PING Fan LIU Jun Tao CHEN Li Meng XING Xiao Ping | 2012 | Biomedical and Environmental Sciences2012,25,4: | 21 |
| 10 | Submucosal tunneling endoscopic resection: An effective and safe therapy for upper gastrointestinal submucosal tumors originating from the muscularis propria layer显示文摘AIM To evaluate the effectiveness and safety of submucosal tunneling endoscopic resection(STER) and compare its outcomes in esophageal and cardial submucosal tumors(SMTs) of the muscularis propria(MP) layer.METHODS From May 2012 to November 2017, 173 consecutive patients with upper gastrointestinal(GI) SMTs of the MP layer underwent STER. Overall, 165 patients were included, and 8 were excluded. The baseline characteristics of the patients and SMTs were recorded. The en bloc resection rate, complete resection rate,residual rate, and recurrence rate were calculated to evaluate the effectiveness of STER, and the complication rate was recorded to evaluate its safety. Effectiveness and safety outcomes were compared between esophageal and cardial SMTs.RESULTS One hundred and twelve men and 53 women with a mean age of 46.9 ± 10.8 years were included. The mean tumor size was 22.6 ± 13.6 mm. Eleven SMTs were located in the upper esophagus(6.7%), 49 in the middle esophagus(29.7%), 46 in the lower esophagus(27.9%), and 59 in the cardia(35.7%). Irregular lesions accounted for 48.5% of all lesions. STER achieved an en bloc resection rate of78.7%(128/165) for GI SMTs with an overall complication rate of 21.2%(35/165).All complications resolved without intervention or were treated conservatively without the need for surgery. The en bloc resection rates of esophageal and cardial SMTs were 81.1%(86/106) and 72.1%(42/59), respectively(P = 0.142), and the complication rates were 19.8%(21/106) and 23.7%(14/59), respectively,(P =0.555). The most common complications for esophageal SMTs were gas-related complications and fever, while mucosal injury was the most common for cardial SMTs.CONCLUSION STER is an effective and safe therapy for GI SMTs of the MP layer. | 11 | Report on Childhood Obesity in China (10): Association of Sleep Duration with Obesity显示文摘Objectives To explore the association of sleep duration with obesity among children in urban areas of China.Methods A total of 6 576 children(3 293 boys and 3 283 girls) aged 7-11 years were randomly selected from 36 primary schools in 6 metropolitan cities in China.A 7-day Physical Activity Recall was used to assess the sleep duration and physical activity level.The height,weight,waist circumference(WC) and percentage of body fat(%BF,as determined by bioelectrical impedance analysis technique) were measured by following the standardized operation procedures.The information on demography,lifestyle and eating habits was collected with a self-administered questionnaire from participants and their parents.Results The average sleep duration per night in the children was 9.7 h with the decreasing trends along with the increase of age(P<0.05).The sleep duration was negatively associated with body mass index(BMI) and WC in both boys and girls after adjustment for confounders(β value-0.23 and-0.82 for boys,-0.24 and-0.91 for girls,respectively,P<0.01).However,no significant association of sleep duration with %BF was found.Children who slept less than 9.0 h per night had a higher risk for overweight and obesity(OR=1.29,95% CI:1.01,1.64) and abdominal obesity(OR=1.38,95% CI:1.04,1.83) as compared with those who slept for 10.0-10.9 h.Conclusions Short sleep duration is associated with obesity.It is important to ensure adequate sleep duration of children and foster their healthy lifestyle at an early stage of life. | MENG Li Ping LIUAi Ling HU Xiao Qi ZHANG Qian DU Song Ming FANG Hong Yun MA Jun XU Gui Fa LI Ying GUO Hong Wei DU Lin MA Guan Sheng | 2012 | Biomedical and Environmental Sciences2012,25,2: | 16 |
| 12 | Selective laser melting of NiTi alloy with superior tensile property and shape memory effect显示文摘It is a challenge to develop complex-shaped Ni Ti shape memory alloy parts by traditional processing methods, due to the poor machinability of Ni Ti alloy. It is reported that selective laser melting(SLM) of additive manufacturing could overcome this problem. However, the reported SLM-produced Ni Ti exhibits poor tensile ductility due to the inner defects and adverse unidirectional columnar grains from SLM process. In this work, the defect-less SLM-Ni Ti with nondirective columnar grains was fabricated by optimizing the intraformational laser scanning length and interformational laser scanning direction. The obtained lath-shaped SLM-Ni Ti sample exhibits tensile strain of 15.6%, more than twice of the reported maximum result(7%). The natural gas is mainly composed of methane (88.99%–98.01%), and the dryness coefficient varies between 0.908 and 0.997. The gas generally displays positive alkane carbon and hydrogen isotopic series. The geochemical characteristics and gas-source correlation indicate that the gases stored in the 5^(th) member of the Upper Triassic Xujiahe Formation are coal-type gases which are derived from source rocks in the stratum itself. The gases reservoired in the 4^(th) member of the Xujiahe Formation and Jurassic strata in the Xinchang gas field are also coal-type gases that are derived from source rocks in the 3^(rd) and 4^(th) members of the Xujiahe Formation. The gases reservoired in the 2^(nd) member of the Upper Triassic Xujiahe Formation are mainly coal-type gases with small amounts of oil-type gas that is derived from source rocks in the stratum itself. This is accompanied by a small amount of contribution brought by source rocks in the Upper Triassic Ma'antang and Xiaotangzi formations. | 19 | Efficacy and safety of Shenyankangfu Tablet,a Chinese patent medicine,for primary glomerulonephritis:A multicenter randomized controlled trial显示文摘Background:Shenyankangfu Tablet(SYKFT)is a Chinese patent medicine that has been used widely to decrease proteinuria and the progression of chronic kidney disease.Objective:This trial compared the efficacy and safety of SYKFT,for the control of proteinuria in primary glomerulonephritis patients,against the standard drug,losartan potassium.Design,setting,participants and intervention:This was a multicenter,double-blind,randomized,controlled clinical trial.Primary glomerulonephritis patients,aged 18-70 years,with blood pressure≤140/90 mmHg,estimated glomerular filtration rate(eGFR)>45 mL/min per 1.73 ㎡,and 24-hour proteinuria level of 0.5-3.0 g,were recruited in 41 hospitals across 19 provinces in China and were randomly divided into five groups:SYKFT,losartan potassium 50 mg or 100 mg,SYKFT plus losartan potassium 50 mg or 100 mg.Main outcome measu res:The primary outcome was change in the 24-hour proteinuria level,after 48 weeks of treatment.Results:A total of 735 participants were enrolled.The percent decline of urine protein quantification in the SYKFT group after 48 weeks was 8.78%±2.56%(P=0.006)more than that in the losartan 50 mg group,which was 0.51%±2.54%(P=1.000)less than that in the losartan 100 mg group.Compared with the losartan potassium 50 mg group,the SYKFT plus losartan potassium 50 mg group had a 13.39%±2.49%(P<0.001)greater reduction in urine protein level.Compared with the losartan potassium 100 mg group,the SYKFT plus losartan potassium 100 mg group had a 9.77%±2.52%(P=0.001)greater reduction in urine protein.With a superiority threshold of 15%,neither was statistically significant.eGFR,serum creatinine and serum albumin from the baseline did not change statistically significant.The average change in TCM syndrome score between the patients who took SYKFT(-3.00[-6.00,-2.00])and who did not take SYKFT(-2.00[-5.00,0])was statistically significant(P=0.003).No obvious adverse reactions were observed in any group.Conclusion:SYKFT decreased the proteinuria and improved the TCM syndrome scores of primary glomerulonephritis patients,with no change in the rate of decrease in the eGFR.SYKFT plus losartan potassium therapy decreased proteinuria more than losartan potassium therapy alone.Trial registration number:NCT02063100 on ClinicalTrials.gov. | Jie Wu Shu-wei Duan Hong-tao Yang Yue-yi Deng Wei Li Ya-ni He Zhao-hui Ni Yong-li Zhan Shan Lin Zhi-yong Guo Jun Zhu Jing-ai Fang Xu-sheng Liu Li-hua Wang Rong Wang Nian-song Wang Xiao-hong Cheng Li-qun He Ping Luo Shi-ren Sun Ji-feng Sun Ai-ping Yin Geng-ru Jiang Hong-yu Chen Wen-hu Liu Hong-li Lin Meng Liang Lu Ma Ming Chen Li-qun Song Jian Chen Qing Zhu Chang-ying Xing Yun Li Ji-ning Gao Rong-shan Li Ying Li Hao Zhang Ying Lu Qiao-ling Zhou Jun-zhou Fu Qiang He Guang-yan Cai Xiang-mei Chen | 2021 | Journal of Integrative Medicine2021,19,2: | 8 |
| 20 | Flow stress equation for multipass hot-rolling of aluminum alloys显示文摘A series of simple axisymmetric compression tests were carried out on the computer servo controlled Gleeble 1 500 machine when strain rates ranged between 0.05 25 s -1 and deformation temperature 300 500 ℃. The results show that flow stress is related to the Zener Hollonom parameter Z and strain, as well as the static recrystallization fraction between passes during multipass hot deformation of 5182 aluminum alloy. Hence, a modified exponential flow stress equation was presented by considering the values of ln A and β as functions of strain, and by using the uniform softening method and incorporating the static recrystallization fraction between passes to consider the effects of residual strain during multipass hot rolling of 5182 aluminum alloy.
