1FHY3 and FAR1 Integrate Light Signals with the miR156-SPL Module-Mediated Aging Pathway to Regulate Arabidopsis Flowering显示文摘In response to competition for light from their neighbors,shade-intolerant plants flower precociously to ensure reproductive success and survival.However,the molecular mechanisms underlying this key developmental switch are not well understood.Here,we show that a pair of Arabidopsis transcription factors essential for phytochrome A signaling,FAR-RED ELONGATED HYPOCOTYL3(FHY3)and FAR-RED IMPAIRED RESPONSE1(FAR1),regulate flowering time by integrating environmental light signals with the miR156-SPL module-mediated aging pathway.We found that FHY3 and FAR1 directly interact with three flowering-promoting SQUAMOSA-PROMOTER BINDING PROTEIN-LIKE(SPL)transcription factors,SPL3,SPL4,and SPL5,and inhibit their binding to the promoters of several key flowering regulatory genes,including FRUITFUL(FUL),LEAFY(LFY),APETALA1(AP1),and MIR172C,thus downregulating their transcript levels and delaying flowering.Under simulated shade conditions,levels of SPL3/4/5 proteins increase,whereas levels of FHY3 and FAR1 proteins decline,thus releasing SPL3/4/5 from FHY3/FAR1 inhibition to allow activation of FUL,LFY,AP1,and MIR172C and,consequently,early flowering.Taken together,these results unravel a novel mechanism whereby plants regulate flowering time by integrating environmental cues(such as light conditions)and an internal developmental program(the miR156-SPL module-mediated aging pathway).Yurong Xie Qin Zhou Yongping Zhao Quanquan Li Yang Liu Mengdi Ma Baobao Wang Rongxin Shen Zhigang Zheng Haiyang Wang 2020Molecular Plant2020,13,3:8
2DELLA proteins interact with FLC to repress flowering transition显示文摘Flowering is a highly orchestrated and extremely ct critical process in a plant's life cycle. Previous study has ademonstrated that SUPPRESSOR OF OVEREXPRESSION OF pCONSTANS 1(SOC1) and FLOWERING LOCUS T(FT) integrate m-I the gibberellic acid(GA) signaling pathway and vernalization higpathway in regulating flowering time, but detailed molecular Hmechanisms remain largely unclear. In GA signaling pathway,DELLA proteins are a group of master transcriptional regulators, while in vernalization pathway FLOWERING LOCUS C(FLC) is a core transcriptional repressor that down-regulates the expression of SOC1 and FT. Here, we report that DELLA proteins interact with FLC in vitro and in vivo, and the LHRI domains of DELLAs and the C-terminus of MADS domain of FLC are required for these interactions.Phenotypic and gene expression analysis showed that mutation of FLC reduces while over-expression of FLC enhances the GA response in the flowering process. Further,DELLA-FLC interactions promote the repression ability of FLC on its target genes. 4Transcriptome dynamics of hippocampal neurogenesis in macaques across the lifespan and aged humans显示文摘Whether adult hippocampal neurogenesis(AHN)persists in adult and aged humans continues to be extensively debated.A major question is whether the markers identified in rodents are reliable enough to reveal new neurons and the neurogenic trajectory in primates.Here,to provide a better understanding of AHN in primates and to reveal more novel markers for distinct cell types,droplet-based single-nucleus RNA sequencing(snRNA-seq)is used to investigate the cellular heterogeneity and molecular characteristics of the hippocampi in macaques across the lifespan and in aged humans.All of the major cell types in the hippocampus and their expression profiles were identified.The dynamics of the neurogenic lineage was revealed and the diversity of astrocytes and microglia was delineated.In the neurogenic lineage,the regulatory continuum from adult neural stem cells(NSCs)to immature and mature granule cells was investigated.A group of primate-specific markers were identified.We validated ETNPPL as a primate-specific NSC marker and verified STMN1 and STMN2 as immature neuron markers in primates.Furthermore,we illustrate a cluster of active astrocytes and microglia exhibiting proinflammatory responses in aged samples.The interaction analysis and the comparative investigation on published datasets and ours imply that astrocytes provide signals inducing the proliferation,quiescence and inflammation of adult NSCs at different stages and that the proinflammatory status of astrocytes probably contributes to the decrease and variability of AHN in adults and elderly individuals.Wei Wang Mengdi Wang Meng Yang Bo Zeng Wenying Qiu Qiang Ma Xiaoxi Jing Qianqian Zhang Bosong Wang Chonghai Yin Jiyao Zhang Yuxin Ge Yufeng Lu Weizhi Ji Qian Wu Chao Ma Xiaoqun Wang 2022Cell Research2022,32,8:2
5Structural modification of BODIPY: Improve its applicability显示文摘BODIPY has been considered a potential scaffold because of their neutral total charge, sharp absorption,and emission with high fluorescence quantum yield. However, the drawback of emission wavelength at less than 600 nm and hydrophobicity limit its application. One of the extremely interesting properties of BODIPY is that small modifications to their structures could be able to tune their properties, mainly including the absorption/emission wavelength and the hydrophilicity. 7Seco-cyclic phorbol derivatives and their anti-HIV-1 activities显示文摘Phorbol esters are recognized for their dual role as anti-HIV-1 agents and as activators of protein kinase C(PKC).The efficacy of phorbol esters in binding with PKC is attributed to the presence of oxygen groups at positions C20,C3/C4,and C9 of phorbol.Concurrently,the lipids located at positions C12/C13 are essential for both the anti-HIV-1 activity and the formation of the PKC-ligand complex.The influence of the cyclopropane ring at positions C13 and C14 in phorbol derivatives on their anti-HIV-1 activity requires further exploration.This research entailed the hydrolysis of phorbol,producing seco-cyclic phorbol derivatives.The anti-HIV-1 efficacy of these derivatives was assessed,and the affinity constant(Kd)for PKC-δprotein of selected seco-cyclic phorbol derivatives was determined through isothermal titration calorimetry.The findings suggest that the chemical modification of cyclopropanols could affect both the anti-HIV-1 activity and the PKC binding affinity.Remarkably,compound S11,with an EC50 of 0.27μmol·L^(−1) and a CC50 of 153.92μmol·L^(−1),demonstrated a potent inhibitory effect on the intermediate products of HIV-1 reverse transcription(ssDNA and 2LTR),likely acting at the viral entry stage,yet showed no affinity for the PKC-δprotein.These results position compound S11 as a potential candidate for further preclinical investigation and for studies aimed at elucidating the pharmacological mechanism underlying its anti-HIV-1 activity.HUANG Xiaolei HUANG Xusheng LI Qirun MA Mengdi CUI Yadong YANG Liumeng WANG Haibo LUO Ronghua CHEN Jinglei YANG Jingxuan LIN Jinrong LI Duxin ZHENG Yongtang ZHANG Jian 2024Chinese Journal of Natural Medicines2024,22,4:0

9JARID2 coordinates with the NuRD complex to facilitate breast tumorigenesis through response to adipocyte-derived leptin显示文摘Background Proteins containing the Jumonji C(JmjC)domain participated in tumorigenesis and cancer progression.However,the mechanisms underlying this effect are still poorly understood.Our objective was to investigate the role of Jumonji and the AT-rich interaction domain-containing 2(JARID2)—a JmjC family protein—in breast cancer,as well as its latent association with obesity.Methods Immunohistochemistry,The Cancer Genome Atlas,Gene Expression Omnibus,and other databases were used to analyze the expression of JARID2 in breast cancer cells.Growth curve,5-ethynyl-2-deoxyuridine(EdU),colony formation,and cell invasion experiments were used to detect whether JARID2 affected breast cancer cell proliferation and invasion.Spheroidization-based experiments and xenotumor transplantation in NOD/SCID mice were used to examine the association between JARID2 and breast cancer stemness.RNA-sequencing,Kyoto Encyclopedia of Genes and Genomes,and Gene Set Enrichment Analysis were used to identify the cell processes in which JARID2 participates.Immunoaffinity purification and silver staining mass spectrometry were conducted to search for proteins that might interact with JARID2.The results were further verified using co-immunoprecipitation and glutathione S-transferase(GST)pull-down experiments.Using chromatin immunoprecipitation(ChIP)sequencing,we sought the target genes that JARID2 and metastasis-associated protein 1(MTA1)jointly regulated;the results were validated by ChIP-PCR,quantitative ChIP(qChIP)and ChIP-reChIP assays.A coculture experiment was used to explore the interactions between breast cancer cells and adipocytes.Results In this study,we found that JARID2 was highly expressed in multiple types of cancer including breast cancer.JARID2 promoted glycolysis,lipid metabolism,proliferation,invasion,and stemness of breast cancer cells.Furthermore,JARID2 physically interacted with the nucleosome remodeling and deacetylase(NuRD)complex,transcriptionally repressing a series of tumor suppressor genes such as BRCA2 DNA repair associated(BRCA2),RB transcriptional corepressor 1(RB1),and inositol polyphosphate-4-phosphatase type II B(INPP4B).Additionally,JARID2 expression was regulated by the obesity-associated adipokine leptin via Janus kinase 2/signal transducer and activator of transcription 3(JAK2/STAT3)pathway in the breast cancer microenvironment.Analysis of various online databases also indicated that JARID2/MTA1 was associated with a poor prognosis of breast cancer.Conclusion Our data indicated that JARID2 promoted breast tumorigenesis and development,confirming JARID2 as a target for cancer treatment.Wei Liu Yi Zeng Xinhui Hao Xin Wang Jiaxiang Liu Tianyang Gao Mengdi Wang Jingyao Zhang Miaomiao Huo Ting Hu Tianyu Ma Die Zhang Xu Teng Hefen Yu Min Zhang Baowen Yuan Wei Huang Yunkai Yang Yan Wang 2023Cancer Communications2023,43,10:0
10Synthesis and anti-HIV activities of phorbol derivatives显示文摘In this study,37 derivatives of phorbol esters were synthesized and their anti-HIV-1 activities evaluated,building upon our previous synthesis of 51 phorbol derivatives.12-Para-electron-acceptor-trans-cinnamoyl-13-decanoyl phorbol derivatives stood out,demonstrating remarkable anti-HIV-1 activities and inhibitory effects on syncytia formation.These derivatives exhibited a higher safety index compared with the positive control drug.Among them,12-(trans-4-fluorocinnamoyl)-13-decanoyl phorbol,designated as compound 3c,exhibited the most potent anti-HIV-1 activity(EC_(50)2.9 nmol·L^(−1),CC50/EC_(50)11117.24)and significantly inhibited the formation of syncytium(EC_(50)7.0 nmol·L^(−1),CC50/EC_(50)4891.43).Moreover,compound 3c is hypothesized to act both as an HIV-1 entry inhibitor and as an HIV-1 reverse transcriptase inhibitor.Isothermal titration calorimetry and molecular docking studies indicated that compound 3c may also function as a natural activator of protein kinase C(PKC).Therefore,compound 3c emerges as a potential candidate for developing new anti-HIV drugs.HUANG Xiaolei TANG Chengrun HUANG Xusheng YANG Yun LI Qirun MA Mengdi ZHAO Lei YANG Liumeng CUI Yadong ZHANG Zhenqing ZHENG Yongtang ZHANG Jian 2024Chinese Journal of Natural Medicines2024,22,2:0
11Three-dimensional nanoscale vortex line visualization and chiral nanostructure fabrication of tightly focused multi-vortex beams via direct laser writing显示文摘Optical singularity is pivotal in nature and has attracted wide interest from many disciplines nowadays,including optical communication,quantum optics,and biomedical imaging.Visualizing vortex lines formed by phase singularities and fabricating chiral nanostructures using the evolution of vortex lines are of great significance.In this paper,we introduce a promising method based on two-photon polymerization direct laser writing(2PP-DLW)to record the morphology of vortex lines generated by tightly focused multi-vortex beams(MVBs)at the nanoscale.Due to Gouy phase,the singularities of the MVBs rotate around the optical axis and move towards each other when approaching the focal plane.The propagation dynamics of vortex lines are recorded by 2PP-DLW,which explicitly exhibits the evolution of the phase singularities.Additionally,the MVBs are employed to fabricate stable three-dimensional chiral nanostructures due to the spiral-forward property of the vortex line.Because of the obvious chiral features of the manufactured nanostructures,a strong vortical dichroism is observed when excited by the light carrying orbital angular momentum.A number of applications can be envisioned with these chiral nanostructures,such as optical sensing,chiral separation,and information storage.MENGDI LUO JISEN WEN PENGCHENG MA QIUYUAN SUN XIANMENG XIA GANGYAO ZHAN ZHENYAO YANG LIANG XU DAZHAO ZHU CUIFANG KUANG XU LIU 2024Photonics Research2024,12,1:0
