| 1 | Photoprotective Effects of Hydroxychloroqine and TCMs on Human Keratinocytes Damaged from Ultraviolet Irradiation显示文摘Objective: To investigate damage effects of ultraviolet irradiation on eternal keratinocyte-HaCaT cells and to evaluate photo-protective efficiency of hydroxychloroqine and Traditional Chinese Medicines ( epigallocatechingallate[EGCG], baikal skullcap root and szechwan lovge rhizome) on HaCaT ceils damaged by middle wave ultraviolet(UVB) irradiation. Methods: Subconfluent HaCaT cells were sham or UVB irradiated and treated with above TCM agents. The damage degree of HaCaT cells was observed by a light microscop. Cell growth was recorded by cellcount and cellular activity was detected by MTT method. The secretion amount of IL-6 and TNF-α was measured by ELISA. Results: The irradiation damage of HaCaT cells was depended on the irradiated dosages and cellular activity was reduced by 36%-80%, with a maximum decrease over 90% after 72 h. The intervention of the above drugs may increase the cellular activity by 10%-72%. The photo-protective efficiency was more apparent in F_A^G (from 1.19+0.07 to 1.28 + 0.06, P < 0.01 ) than that in hydroxychloroqine ( from 0.43 + 0.04 to 0.96 + 0.04, P < 0.05). The other two tested drugs also showed photo-protective effect( from 0.44 + 0.07 to 1.21 -+ 0.02, P < 0.05). As to cytokine secretion, EGCG could decline the secretion amount of IL-6 and TNF.a apparently. Hydroxychloroqine and baikal skullcap root baikal skullcap root could only reduce the secretion of ID6. The secretion of IL-6 and TNF-α could not be inhibitedby szechwan lovge rhizome. Conclusion : The injury effect of UVB irradiation on cultured keratinocytes is dose-dependent and the tested drugs have photo-protective potency. | 2 | Protective effect of Yiguanjian decoction against DNA damage on concanavalin A-induced liver injury mice model显示文摘OBJECTIVE:To investigate the inhibitory effect of Yiguanjian decoction(YD) on DNA damage in Concanavalin A(Con A)-induced liver injury mice model and to explain the possible mechanism.METHODS:Totally 120 male BALB/c mice were randomly divided into 6 groups,20 mice each:normal group,model group,Bifendate group,YD low dose group,YD middle dose group and YD high dose group.Except normal group,liver injury model induced by Con A was established.While modeling,each mouse in YD group was given YD(0.4 m L/20 g per day) by intragastric administration(0.13 g YD for YD low dose group;0.26 g for YD middle dose group;0.52 g for YD high dose group).Bifendate group was given Bifendate(0.2 gnd model·kg-1 grou·d-1) by gavage.Normal group ap were fed with same volume of physiological saline daily.After 8 weeks,the serum alanine transaminase(ALT)and aspartate transaminase(AST) were tested.The hematoxylin-eosin staining was used to evaluate the grade of liver inflammation and liver fibrosis stage.Hepatocellular DNA damage was detected by single cell gel electrophoresis technology.The protein expression of tumor necrosis factor-α(TNF-α),Bax and Mut T Homolog 1(MTH1) was detected by western blotting and enzyme linked immunosorbent assay.Bax m RNA and MTH1 m RNA were detected by Real-time Polymerase Chain Reaction(PCR).RESULTS:YD can improve the degree of liver inflammation and fibrosis in the liver of chronic hepatitis mice,the dose effect relationship is remarkable(P < 0.05).YD can reduce liver cell DNA damage.The difference between YD middle dose group and model group was statistically significant(P < 0.05).YD middle dose group had decreased the protein expression of TNF-α in the mice liver of immunological liver injury(P < 0.05).YD can increase the protein expression of Bax(P < 0.05).Compared with normal group,the protein expression of MTH1 was decreased(P < 0.05),but there was no statistical significance between YD group and model group(P >0.05).YD can increase the m RNA expression of Bax and MTH1(both P < 0.05).CONCLUSION:YD can effectively inhibit the DNA damage in immunological liver injury mice,the mechanism may be that it can decrease the TNF-αand increase the Bax and MTH1 expression. | Tian Mengxi Liu Wenlan You Hongjie Zhao Qingzhou Ouyang Luodan Gao Biane Zhang Xin Che Niancong | 2016 | Journal of Traditional Chinese Medicine2016,36,4: | 7 |
| 3 | Construction of ecological security patterns and ecological restoration zones in the city of Ningbo,China显示文摘Studying an ecological restoration zoning process under the background of ecological security patterns is of great significance to the rapid adjustment and optimization of a landscape pattern.In this study,a remote sensing ecological index and a morphological spatial pattern analysis method were used to assess the quality of habitats and identify ecological sources in the city of Ningbo;ecological corridors,ecological pinch points,and ecological barrier points were extracted by using a circuit theory to construct ecological security patterns and ecological restoration zones.The results indicate:(1)There were 47 ecological sources,and 83 key ecological corridors in Ningbo,and the ecological land area was about 1898.39 km^(2),accounting for 19.89%of the total study area.(2)The ecological source areas were distributed in“one patch and three belts”,and the low-resistance ecological corridors were concentrated in southern Yuyao city,western Haishu district,and central and western Fenghua district;the ecological network in the western and southern regions was dense.(3)There were four types of ecological restoration zones that need to be established,which were prioritized restoration zones,prioritized protection zones,key conservation zones,and general conservation zones distributed hierarchically from inner part towards outside.(4)Ninghai county,Yuyao city,and Fenghua district had large ecological land areas,however,prioritized restoration and protection zones in Ninghai and Fenghua were also large.The analysis results are expected to provide a reference for optimizing a territorial ecological space in a city. | ZHANG Haitao LI Jialin TIAN Peng PU Ruiliang CAO Luodan | 2022 | Journal of Geographical Sciences2022,32,4: | 7 |

| 5 | Theranostic nanomedicine by surface nanopore engineering显示文摘Theranostic nanomedicine that integrates diagnostic and therapeutic agents into one nanosystem has gained considerable momentum in the field of cancer treatment. Among diverse strategies for achieving theranostic capabilities, surface-nanopore engineering based on mesoporous silica coating has attracted great interest because of their negligible cytotoxicity and chemically active surface that can be easily modified to introduce various functional groups(e.g.,-COOH,-NH_2,-SH, etc.) via silanization, which can satisfy various requirements of conjugating biological molecules or functional nanoparticles. In addition,the nanopore-engineered biomaterials possess large surface area and high pore volume, ensuring desirable loading of therapeutic guest molecules. In this review, we comprehensively summarize the synthetic procedure/paradigm of nanopore engineering and further broad theranostic applications. Such nanopore-engineering strategy endows the biocompatible nanocomposites(e.g., Au,Ag, graphene, upconversion nanoparticles, Fe_3O_4, MXene, etc.) with versatile functional moieties, which enables the development of multifunctional nanoplatforms for multimodal diagnostic bio-imaging, photothermal therapy, photodynamic therapy,targeted drug delivery, synergetic therapy and imaging-guided therapies. | 7 | Ecosystem service valuation of bays in East China Sea and its response to sea reclamation activities显示文摘Ecosystem service values(ESVs)of bays and their response to sea reclamation are of great practical importance for forming bay eco-compensation policy and extension of blue economic space.Based on land use information of bays collected during the period of 1990–2015,the spatiotemporal evolution of ESVs of 12 main bays in East China Sea and their response to sea reclamation activities over the past 25 years were quantitatively analyzed.The analysis results indicate that ESVs of bays in East China Sea showed a continuous downward trend and the whole ecosystem was continuously degraded,in which the degradation degree of ESV in the southern bays was higher than that in the northern bays.Spatial zoning of ESVs of bays in East China Sea was remarkable,showing a continuous downward trend from low-value to high-value zone.Spatial variation of ESVs of each bay was also significant,expanding from a city and from inland to the coast,which suggests that human activities,mainly reclamation,have become main agents for ESV evolution of bays in East China Sea.ESVs of bays have a significant response to sea reclamation,manifested as a significant negative correlation between ESV and reclamation intensity.The correlation in the southern bays was stronger than that in the northern bays,which was caused by different effects of sea reclamation modes on ESV evolution of muddy and bedrock bays.A negative effect of sea reclamation activities on bay ecosystem was hysteretic.Therefore,an attention should be paid to dynamic monitoring and early warning of development status in offshore areas,ecosystem-level reclamation control policy,and coastal wetland reserves planning.Moreover,the spatial coupling mechanism study between bay ecosystem service demand and its service supply capacity should be strengthened to realize systematic regulation of bay ecological security pattern. | LIU Ruiqing XU Hao LI Jialin PU Ruiliang SUN Chao CAO Luodan JIANG Yimei TIAN Peng WANG Lijia GONG Hongbo | 2020 | Journal of Geographical Sciences2020,30,7: | 2 |

Herein, a bio-mimetic radiosensitized platform based on a natural in-situ oxygen-evolving photosynthetic cyanobacteria combined with two-dimensional (2D) bismuthene with high atomic-number (Z) components, is designed and engineered to effectively modulate the radiotherapy-resistant hypoxic tumor environment and achieve sufficient radiation energy deposition into tumor. Upon the exogenous sequential irradiation of 660 nm laser and X-ray beam, continuous photosynthetic oxygen evolution by the cyanobacteria and considerable generation of reactive oxygen species by the 2D bismuthene radiosensitizer substantially augmented the therapeutic efficacy of radiotherapy and suppressed the in vivo tumor growth, as demonstrated on both LLC-lung tumor xenograft-bearing C57/B6 mice model and 4T1-breast tumor xenograft-bearing Balb/c mice model, further demon-strating the photosynthetic hypoxia-alleviation capability and radiosensitization performance of the engineered biomimetic radiosensitized platform.
