3OsGRF4 controls grain shape, panicle length and seed shattering in rice显示文摘Traits such as grain shape, panicle length and seed shattering, play important roles in grain yield and harvest. In this study, the cloning and functional analysis of PANICLE TRAITS 2(PT2), a novel gene from the Indica rice Chuandali(CDL), is reported. PT2 is synonymous with Growth-Regulating Factor 4(OsGRF4), which encodes a growth-regulating factor that positively regulates grain shape and panicle length and negatively regulates seed shattering. Higher expression of OsGRF4 is correlated with larger grain, longer panicle and lower seed shattering. A unique OsGRF4 mutation, which occurs at the Osmi RNA396 target site of OsGRF4, seems to be associated with high levels of OsGRF4 expression, and results in phenotypic difference. Further research showed that OsGRF4 regulated two cytokinin dehydrogenase precursor genes(CKX5 and CKX1) resulting in increased cytokinin levels,which might affect the panicle traits. High storage capacity and moderate seed shattering of OsGRF4 may be useful in high-yield breeding and mechanized harvesting of rice. 4Investigation of the combustion deterioration of an automotive diesel engine under transient operation显示文摘With increasingly stringent emission regulations and demand for fuel economy by the public,the combustion and emission problems of automotive diesel engines during transient operation have become vital and urgent issues.In this study,combustion deterioration has been experimentally analyzed using a heavy-duty turbocharged diesel engine running under transient conditions(constant speed and increasing torque).Optimization of the transient combustion process was performed by adjusting the fuel injection parameters.The results indicated that the notable combustion deterioration relative to steady state operation while transient was a function of the delay in the air-supply to the turbocharged engine,and took the form of combustion phasing delay,resulting in rapidly increasing smoke emission and fuel consumption.However,the delay in combustion phasing can be controlled by advancing the fuel injection timing,effectively increasing thermal efficiency.Unfortunately,smoke and NO x emissions increased at the same time.The deterioration in combustion phasing can also be improved by increasing injection pressure,resulting in decreased smoke emission while NO x emission increased.It is worth noting that the effective thermal efficiency first increased and then decreased as fuel injection pressure increased during transient operation.ZHANG LongPing LIU ZhongChang TIAN Jing SUN ShiJie 2014Science China(Technological Sciences)2014,57,3:9
5A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. 7Effect of Ningmitai capsule plus sertraline on patients with premature ejaculation and enlarged seminal vesicles:A randomized clinical trial显示文摘OBJECTIVE: To investigate the effects of Ningmitai capsule combined with sertraline on patients with premature ejaculation(PE) and an increased anterior-posterior diameter(APD) of the seminal vesicles(SVs).METHODS: Sixty men with acquired PE were enrolled and randomly divided into two groups. The combined group was treated with Ningmitai capsule and sertraline, while the control group was treated with sertraline alone. Main outcomes were measured using the premature ejaculation diagnostic tool(PEDT), APD of SVs, and Clinical Global Impression of Change questionnaire and compared before and after 3 months of treatment.RESULTS: Comparing after treatment with before treatment outcomes within each group, the PEDT score was significantly reduced in the combined group(12.1 ± 2.5 vs 8.6 ± 3.2, P < 0.001, respectively) and control group(12.9 ± 2.6 vs 10.3 ± 1.6, P <0.001, respectively). Furthermore, the PEDT score after treatment was significantly lower in the combined compared with control group(8.6 ± 3.2 vs10.3 ± 1.6, P = 0.011, respectively). The APD of SVs in the combined group was significantly decreased after treatment [(10.8 ± 2.4) vs(12.9 ± 2.2) mm, P =0.001], while the APD of SVs in the control group was equivalent before and after treatment. The treatment response rate was not significantly higher in the combined compared with control group.CONCLUSION: These results indicated that the effect of Ningmitai capsule combined with sertraline was better than that of sertraline alone for the treatment of PE patients exhibiting an increased APD of SVs. 9Regulation of pathological BBB restoration via nanostructured ROS-responsive glycolipid-like copolymer entrapping siVEGF for glioblastoma targeted therapeutics显示文摘Glioblastoma(GBM)is one of the malignant brain tumors with high mortality and no curative treatments.Abnormally elevated vascular endothelial growth factor(VEGF)in GBM seriously disrupts the blood brain barrier(BBB)with an increased permeability,resulting in poor outcome and prognosis.RNAi interference has shown strong potential to inhibit VEGF expression,thus it is necessary to development an effective and safe gene delivery system possessing the ability to cross the BBB and target GBM cells.This study aims to explore the anti-GBM effect of angiopep-2(Ap)peptide modified reactive oxygen species(ROS)cleavable thioketal(TK)linked glycolipid-like nanocarrier(CSTKSA)delivering anti-VEGF siRNA(R),termed as Ap-CSTKSA/R complexes.Ap functionalized modification produced an enhanced cellular uptake and a stronger bio-distribution of Ap-CSTKSA/R complexes in U87 MG cells and brain tumor tissues,respectively.Ap-CSTKSA/R complexes exhibited great superiority in GBM growth inhibition and finally translated into the longest survival period mainly via receptor-mediated targeting delivery,VEGF gene silencing accompanied with remarkable angiogenesis inhibition,and suppressed expression of caveolin-1 which is involved in BBB functional regulation in the occurrence and treatment of GBM.The study indicated that Ap functionalization on ROS-responsive glycolipid-like copolymer exhibits a promising and effective gene delivery platform for GBM targeted treatment.Lijuan Wen Yu Peng Kai Wang Zhihua Huang Shiying He Ruiwen Xiong Longping Wu Fengtian Zhang Fuqiang Hu 2022Nano Research2022,15,2:2
10Balancing selection and wild gene pool contribute to resistance in global rice germplasm against planthopper显示文摘Interactions and co-evolution between plants and herbivorous insects are critically important in agriculture.Brown planthopper(BPH)is the most severe insect of rice,and the biotypes adapt to feed on different rice genotypes.Here,we present genomics analyses on 1,520 global rice germplasms for resistance to three BPH biotypes.Genome-wide association studies identified 3,502 single nucleotide polymorphisms(SNPs)and 59 loci associated with BPH resistance in rice.We cloned a previously unidentified gene Bph37 that confers resistance to BPH.The associated loci showed high nucleotide diversity.Genome-wide scans for trans-species polymorphisms revealed ancient balancing selection at the loci.The secondarily evolved insect biotypes II and III exhibited significantly higher virulence and overcame more rice varieties than the primary biotype I.In response,more SNPs and loci evolved in rice for resistance to biotypes II and III.Notably,three exceptional large regions with high SNP density and resistance-associated loci on chromosomes 4 and 6 appear distinct between the resistant and susceptible rice varieties.Surprisingly,these regions in resistant rice might have been retained from wild species Oryza nivara.Our findings expand the understanding of long-term interactions between rice and BPH and provide resistance genes and germplasm resources for breeding durable BPH-resistant rice varieties.Cong Zhou Qian Zhang Yu Chen Jin Huang Qin Guo Yi Li Wensheng Wang Yongfu Qiu Wei Guan Jing Zhang Jianping Guo Shaojie Shi Di Wu Xiaohong Zheng Lingyun Nie Jiaoyan Tan Chaomei Huang Yinhua Ma Fang Yang Xiqin Fu Bo Du Lili Zhu Rongzhi Chen Zhikang Li Longping Yuan Guangcun He 2021Journal of Integrative Plant Biology2021,63,10:2

13Studies of Improving the Frequency of Indica Rice Transformation by Biolistic Bombardment显示文摘In order to improve the frequency of indica rice transformation by biolistic bombardment, suitable culture conditions for embryonic calli,an optimal selection scheme for resistant calli and seedlings, and optimum bombardment parameters a investigated by using 14 commercially important indica rice cultivars. The main results show that the CC medium with 36g/L mannitol is a scheme subculture medium in which the browning of indica rice calli can be mitigated significantly; The concentration of 30~40mg/L Hyg or 150~200mg/L G418 or 10~20 mg/L Basta is suitable for selection of resistant calli; The transformation parameters of 100μg gold powder absorbing 0.2μg DNA per shot and 900 psi helium pressure and 6 cm bombardment distance and bombarded twice for each plate give the best result; Keeping the target calli on osmotic medium containing 60g/L mannitol from 12 ~24h before bombardment to 24~48h after it can increase the efficiencies of transformation .
