2A microRNA encoded by HSV-1 inhibits a cellular transcriptional repressor of viral immediate early and early genes显示文摘Viral microRNAs are one component of the RNA interference phenomenon generated during viral infection. They were first identified in the Herpesviridae family, where they were found to regulate viral mRNA translation. In addition, prior work has suggested that Kaposi's sarcoma-associated herpesvirus (KSHV) is capable of regulating cellular gene transcription by miRNA. We demonstrate that a miRNA, hsv1-mir-H27, encoded within the genome of herpes simplex virus 1 (HSV-1), targets the mRNA of the cellular transcriptional repressor Kelch-like 24 (KLHL24) that inhibits transcriptional efficiency of viral immediate-early and early genes. The viral miRNA is able to block the expression of KLHL24 in cells infected by HSV-1. 3A comparative study of the characteristics of two Coxsackie A virus type 16 strains (genotype B)显示文摘Coxsackie A virus is one of the major pathogens associated with hand, foot and mouth disease (HFMD). The etiological characteristics of Coxsackie A virus type 16 (CA16) are thought to correlate with the pathological process of its infection. Two CA16 strains that were isolated from a severe HFMD patient presented with different plaque forms. This observation, along with biological analysis, indicated that the differences in the strains' biological characteristics, such as proliferation kinetics and immunogenicity, correlate with differences in their pathogenicity toward neonatal mice. Furthermore, these differences are thought to be associated with the sequence of the 5′ non-coding region of the viral genome and the VP1 structural region sequence. 4Transcriptional regulation of the Herpes Simplex Virus 1α-gene by the viral immediate-early protein ICP22 in association with VP16显示文摘Herpes Simplex Virus 1 (HSV1) is capable of inducing two forms of infection in individuals, and the establishment of which type of infection occurs is linked to the transcriptional activation of viral α genes. One of the HSV1 α genes, ICP22, is known to have multiple functions during virus replication, but its distinct roles are still unclear. This study showed that ICP22 functions as a general repressor for certain viral and cellular promoters, and this transcriptional repression by ICP22 is independent of the specific upstream promoter element, as shown using the CAT enzyme assay system. Further work also found that VP16 interfered with ICP22 mediated transcriptional repression of the viral α4 gene, through interactions with specific elements upstream of the α4 gene promoter. 5Pingwei capsules improve gastrointestinal motility in rats with functional dyspepsia显示文摘OBJECTIVE: To investigate the mechanism of Ping-wei capsules(PWC) in improving gastrointestinal m otility in rats with functional dyspepsia(FD).METHODS: We established an FD model by stimu-lating semi-starvation rats via tail damping, provo-cation, and forced exercise fatigue. The FD model group was further divided into five groups accord-ing to the treatment received: normal saline, dom-peridone, low-dose PWC, mid-dose PWC, or highdose PWC. The effect of PWC on FD was evaluated by measuring gastrointestinal motility. Changes in leptin and cholecystokinin(CCK) were detected through enzyme-linked immunosorbent assay, re-verse transcription-polymerase chain reaction, and immunohistochemistry.RESULTS: PWC significantly increased gastrointesti-nal m otility in FD rats. Furthermore, PWC signifi-cantly increased CCK m RNA and protein concentra-tions in the duodenum and antrum, decreased leptin protein concentrations in the duodenum, an-trum, and hypothalamus, and decreased CCK pro-tein concentration in the hypothalamus.CONCLUSION: PWC improves gastrointestinal mo-tor function in FD rats by decreasing the leptin con-centration in serum and the brain-gut axis, and by increasing the CCK concentration in gastrointesti-nal tissue. 6HSV-1 stimulation-related protein HSRG1 inhibits viral gene transcriptional elongation by interacting with Cyclin T2显示文摘The protein encoded by HSRG1(HSV-1 stimulation-related gene 1) is a virally induced protein expressed in HSV-1-infected cells.We have already reported that HSRG1 is capable of interacting with transcriptional regulator proteins.To further analyze the effects of HSRG1 on the regulation of viral gene transcription,we expressed the HSRG1 protein in transfected cells and found that it postpones the proliferation of HSV-1.CAT(chloramphenicol acetyltransferase) assays also revealed that HSRG1 reduces transcription from HSV-1 promoters.Yeast two-hybrid and immunoprecipitation assays indicated that HSRG1 interacts with Cyclin T2,the regulatory subunit of P-TEFb,which is required for transcription elongation by RNA Pol II(RNAP II) ,and that amino acid residues 1-420 in Cyclin T2 are important for binding with HSRG1.Fluorescence assays suggested that the cellular localizations of those two proteins are influenced by their interaction.Further analyses with CAT assays revealed that HSRG1 inhibits the transcriptional activation by Cyclin T2 of viral promoters.Our results suggested that the inhibitory effects of HSRG1 on viral replication and proliferation are probably induced by its binding to Cyclin T2.Therefore,it is likely that HSRG1 inhibits viral gene transcriptional elongation by interacting with Cyclin T2.WU WenJuan YU Xian LI WeiZhong GUO Lei LIU LongDing WANG LiChun LI QiHan 2011Science China(Life Sciences)2011,54,4:3
7Analysis of HSV-I ICP22 effects on HCMV major immediate-early promoter structure显示文摘The human cytomegalovirus (HCMV) major immediate-early (MIE) promoter has strong transcriptional promoting capability. Its cis-acting regulatory elements form a special structure in this region that is repeated multiple times; the biological significance of these elements and their different compositions in the transcriptional promoting process remain unclear. Our results demonstrate that the HSV-I MIE protein ICP22 can generate strong repression of many viral and cellular promoters and enhancers. We further studied the transcriptional effects of ICP22 on structural elements and mutations in various HCMV MIE promoters by using a CAT assay. In spite of different transcriptional effects of all the ele- ments in the presence of ICP22, the transcriptional efficiencies exhibited by mutations generated by different compositions and an entire HCMV promoter, are not the simple sum of the functions of these elements. Furthermore, the transcriptional activities of specific sequences were not affected by the presence of ICP22. 8Immediate-early gene product ICP22 inhibits the trans-transcription activating function of P53-mdm-2显示文摘As a product of HSVI immediate-early gene, ICP22 is capable of interacting with various cellular tran-scriptive and regulatory molecules during viral infection so as to impact the normal cellular molecular mechanism. ICP22 expressed in transfected cells can push the cells’ entering into S phase with binding to mdm-1 promoter region and impact its trans-transcription activating effect by P53. 9An analysis of major scientific problems and research paths of Gulong shale oil in Daqing Oilfield, NE China显示文摘After the preliminary basic research on the problems encountered during the production period of Gulong shale oil in the Songliao Basin, NE China, and the scientific exploration, the special characteristics of Gulong shale oil in terms of reservoir space, phase distribution, flow pattern, and mineral evolution are proposed. The main results are as follows :(1) The source of organic matter, mechanism of hydrocarbon generation and expulsion, and key factors affecting shale oil abundance;(2) The types and structural characteristics of the reservoir and their contribution to porosity and permeability;(3) The mineral origin and evolution of minerals and their influence on reservoir availability, sensitivity, and compressibility;(4) The rock mechanical characteristics and fracture propagation law of Gulong shale;(5) The shale oil products, phase change law and main control factors of adsorption and desorption conversion of Gulong shale oil;(6) The mechanism of shale oil-liquid, solid-liquid gas interaction and enhanced oil recovery. Three key research suggestions are proposed to realize the large-scale economic utilization of the Gulong shale oil as follows:(1) Deepen research on the mechanism of oil and gas generation and discharge, storage and transportation, to guide the selection of geological sweet spots of shale oil;(2) Deepen research on the compressibility and fracture initiation mechanism to support the selection of engineering sweet spots and optimization of engineering design;(3) Deepen research on the fluid interaction mechanism under reservoir conditions, os us to guide the optimization of development schemes and the selection of EOR technologies. 10A cellular response protein induced during HSV-1 infection inhibits viral replication by interacting with ATF5显示文摘Studies of herpes simplex virus type 1(HSV-1)infection have shown that many known and unknown cellular molecules involved in viral proliferation are up-regulated following HSV-1 infection.In this study,using two-dimensional polyacrylamide gel electrophoresis,we found that the expression of the HSV-1 infection response repressive protein(HIRRP,GI 16552881)was up-regulated in human L02 cells infected with HSV-1.HIRRP,an unknown protein,was initially localized in the cytoplasm and then translocated into the nucleus of HSV-1-infected cells.Further analysis showed that HIRRP represses HSV-1proliferation by inhibiting transcription of the viral genome by interacting with the cellular transcription factor,ATF5,via its N-terminal domain.ATF5 represses the transcription of many host genes but can also act as an activator of genes containing a specific motif.We found that ATF5 promotes the proliferation of HSV-1 via a potential mechanism by which ATF5 enhances the transcription of viral genes during the course of an HSV-1 infection;HIRRP then induces feedback repression of this transcription by interacting with ATF5.WU LianQiu ZHANG XueMei CHE YanChun ZHANG Ying TANG SongQing LIAO Yun NA RuiXiong XIONG XiangLin LIU LongDing LI QiHan 2013Science China(Life Sciences)2013,56,12:2

20Sedimentary diagenesis of rudist shoal and its control on reservoirs: A case study of Cretaceous Mishrif Formation, H Oilfield, Iraq显示文摘Based on the core, cast thin section, whole rock analysis, conventional physical properties and high pressure mercury intrusion test, the sedimentary diagenesis characteristics of rudist shoal in Cretaceous Mishrif Formation of H Oilfield, Iraq and its control on the reservoir were studied. The rudist shoal of the Mishrif Formation develops in the high-stand systems tract and is distributed in the high places of paleogeomorphology on the edge of platform with strong hydrodynamic force. According to the relative sea level changes, lithologic evolution and sedimentary structure characteristics of the rudist shoal, the single rudist shoal is divided into four lithologic sections: A, B, C and D, that is, low-angle cross-bedding pelletoids-rudist packstone, low-angle cross-bedding and parallel bedding arene-rudist grainstone, parallel bedding rudist gravel limestone, and horizontal bedding carbonaceous mudstone. The complete sedimentary sequence of a single rudist shoal is often disrupted. Several rudist shoals superimpose to form thick rudist shoal sediment. The single rudist shoal thickness and lithologic sections assemblage change regularly in vertical direction. The rudist shoal has the characteristics of 'strong dissolution, weak cementation and strong compaction', forming pore-type reservoir with intergranular pores, intergranular dissolved pores, mold pores, and dissolved pores. With mainly coarse pore throats larger than 5 μm, the reservoir is of medium-high porosity and high permeability. There is lithological reverse cycles inside single shoals and between single shoals, with content of mud crystals decreasing from the bottom to the top, dissolution increasing, cementation decreasing in strength, pore throats getting larger, and physical properties turning better.
