1Soybean AP1 homologs control flowering time and plant height显示文摘Flowering time and plant height are key agronomic traits that directly affect soybean(Glycine max)yield.APETALA1(AP1)functions as a class A gene in the ABCE model for floral organ development,helping to specify carpel,stamen,petal,and sepal identities.There are four AP1 homologs in soybean,all of which are mainly expressed in the shoot apex.Here,we used clustered regularly interspaced short palindromic repeats(CRISPR)–CRISPR-associated protein 9 technology to generate a homozygous quadruple mutant,gmap1,with loss-of-function mutations in all four GmAP1 genes.Under short-day(SD)conditions,the gmap1 quadruple mutant exhibited delayed flowering,changes in flower morphology,and increased node number and internode length,resulting in plants that were taller than the wild type.Conversely,overexpression of GmAP1a resulted in early flowering and reduced plant height compared to the wild type under SD conditions.The gmap1 mutant and the overexpression lines also exhibited altered expression of several genes related to flowering and gibberellic acid metabolism,thereby providing insight into the role of GmAP1 in the regulatory networks controlling flowering time and plant height in soybean.Increased node number is the trait with the most promise for enhancing soybean pod number and grain yield.Therefore,the mutant alleles of the four AP1 homologs described here will be invaluable for molecular breeding of improved soybean yield.Liyu Chen Haiyang Nan Lingping Kong Lin Yue Hui Yang Qingsong Zhao Chao Fang Haiyang Li Qun Cheng Sijia Lu Fanjiang Kong Baohui Liu Lidong Dong 2020Journal of Integrative Plant Biology2020,62,12:9
2Predictive factors associated with gefitinib response in patients with advanced non-small-cell lung cancer(NSCLC)显示文摘Purpose: A number of different clinical characteristics have been reported to singly correlate with therapeutic activity of epidermal growth factor receptor(EGFR) tyrosine kinase inhibitors(TKIs) in advanced non-small-cell lung cancer(NSCLC). This study aimed to identify predictive factors associated with prognostic benefits of gefitinib.Patients and methods: EGFR gene typing in 33 advanced NSCLC patients received gefitinib(250 mg/day) were analyzed with mutant-enriched PCR assay. Gefitinib response was evaluated with potential predictive factors retrospectively.Results: The overall objective response rate(ORR) and median progression-free survival(PFS) in the 33 patients treated by gefitinib were 45.5% and 3.0(2.0-4.0) months. The ORR and median PFS in EGFR gene mutation patients were significantly higher/longer than those in EGFR gene wild-type patients(P<0.01). Similarly, the ORR and median PFS in non-smoker patients were significantly higher/longer than those in smoker patients(P<0.05, P<0.01, respectively). However, no difference for ORR and median PFS occurred between male and female patients. Logistic multivariate analysis showed that only EGFR mutated gene was significantly associated with the ORR(P<0.01). Both EGFR mutated gene and non-smoker were the major factors that contributed to PFS(P<0.05). 3FT5a interferes with the Dt1-AP1 feedback loop to control flowering time and shoot determinacy in soybean显示文摘Flowering time and stem growth habit determine inflorescence architecture in soybean, which in turn influences seed yield. Dt1, a homolog of Arabidopsis TERMINAL FLOWER 1(TFL1), is a major controller of stem growth habit, but its underlying molecular mechanisms remain unclear.Here, we demonstrate that Dt1 affects node number and plant height, as well as flowering time,in soybean under long-day conditions. The b ZIP transcription factor FDc1 physically interacts with Dt1, and the FDc1-Dt1 complex directly represses the expression of APETALA1(AP1). We propose that FT5 a inhibits Dt1 activity via a competitive interaction with FDc1 and directly upregulates AP1. Moreover, AP1 represses Dt1 expression by directly binding to the Dt1 promoter, suggesting that AP1 and Dt1 form a suppressive regulatory feedback loop to determine the fate of the shoot apical meristem. 5Polarized holographic lithography system for high-uniformity microscale patterning with periodic tunability显示文摘Periodic microscale array structures play an important role in diverse applications involving photonic crystals and diffraction gratings.A polarized holographic lithography system is proposed for patterning high-uniformity microscale two-dimensional crossed-grating structures with periodic tunability.Orthogonal two-axis Lloyd’s mirror interference and polarization modulation produce three sub-beams,enabling the formation of two-dimensional crossed-grating patterns with wavelength-comparable periods by a single exposure.The two-dimensional-pattern period can also be flexibly tuned by adjusting the interferometer spatial positioning.Polarization states of three sub-beams,defining the uniformity of the interference fringes,are modulated at their initial-polarization states based on a strict full polarization tracing model in a three-dimensional space.A polarization modulation model is established considering two conditions of eliminating the unexpected interference and providing the desired identical interference intensities.The proposed system is a promising approach for fabricating high-uniformity two-dimensional crossed gratings with a relatively large grating period range of 500–1500 nm.Moreover,our rapid and stable approach for patterning period-tunable two-dimensional-array microstructures with high uniformity could be applicable to other multibeam interference lithography techniques.Gaopeng Xue Qihang Zhai Haiou Lu Qian Zhou Kai Ni Liyu Lin Xiaohao Wang Xinghui Li 2021Microsystems & Nanoengineering2021,7,2:2
6Virtual&augmented reality for biological microscope in experiment education显示文摘Background Mixed-reality technologies,including virtual reality(VR)and augmented reality(AR),are considered to be promising potential tools for science teaching and learning processes that could foster positive emotions,motivate autonomous learning,and improve learning outcomes.Methods In this study,a technology-aided biological microscope learning system based on VR/AR is presented.The structure of the microscope is described in a detailed three-dimensional(3D)model,each component being represented with their topological interrelationships and associations among them being established.The interactive behavior of the model was specified,and a standard operating guide was compiled.The motion control of components was simulated based on collision detection.Combined with immersive VR equipment and AR technology,we developed a virtual microscope subsystem and a mobile virtual microscope guidance system.Results The system consisted of a VR subsystem and an AR subsystem.The focus of the VR subsystem was to simulate operating the microscope and associated interactive behaviors that allowed users to observe and operate the components of the 3D microscope model by means of natural interactions in an immersive scenario.The AR subsystem allowed participants to use a mobile terminal that took a picture of a microscope from a textbook and then displayed the structure and functions of the instrument,as well as the relevant operating guidance.This flexibly allowed students to use the system before or after class without time and space constraints.The system allowed users to switch between the VR and AR subsystems.Conclusions The system is useful for helping learners(especially K-12 students)to recognize a microscope's structure and grasp the required operational skills by simulating operations using an interactive process.In the future,such technology-assisted education would be a successful learning platform in an open learning space.Xiang ZHOU Liyu TANG Ding LIN Wei HAN 2020Virtual Reality & Intelligent Hardware2020,2,4:1

11Rapid and sensitive detection of urinary KIM-1 using fully printed photonic crystal microarrays显示文摘Urinary kidney injury molecule 1(uKIM-1)serves as a reliable marker for the early diagnosis of acute kidney injury(AKI).The rapid and facile detection of changes in uKIM-1 is essential for early AKI diagnosis,ultimately improving the prognosis of patients.In this study,we developed a fully printed photonic crystal-integrated microarray with photonic crystal-enhanced fluorescence properties,which can detect uKIM-1 levels at the point-of-care.We confirmed its efficacy in the early diagnosis of AKI using clinical urine specimens.Direct quantitative detection of uKIM-1 was achieved within 10 min.The lowest limit of detection is 8.75 pg·mL^(-1) with an accuracy of 94.2%.The diagnostic efficacy was validated using 86 clinical urine samples,highlighting the high sensitivity and stability of the photonic crystal microarray.Consequently,a facile and reliable immunoassay was designed and prepared for the rapid quantitative detection of uKIM-1,which is crucial for the early identification and convenient detection of AKI in hospital or community settings.Rapid,convenient,cost-effective,and long-term monitoring of changes in uKIM-1 levels can assist clinicians in making timely adjustments to treatment regimens,preventing the transition from AKI to chronic kidney disease(CKD),improving the quality of life of patients with AKI,and reducing healthcare costs.It highlights the advantages of utilizing urine samples as a noninvasive and easily accessible medium for early detection and monitoring of kidney-related conditions.Yang Liu Xuwei He Zewei Lian Qian Guo Jimei Chi Xiaoxue Lin Liyue Zhang Zheng Liu Yingyuan Liu Meng Su Keyu Wang Qiangguo Ao Qingli Cheng 2024Nano Research2024,17,5:0

15CDK11 negatively regulates Wnt/β-catenin signaling in the endosomal compartment by affecting microtubule stability显示文摘Objectives:Improper activation of Wnt/β-catenin signaling has been implicated in human diseases.Beyond the well-studied glycogen synthase kinase 3p(GSK3p)and casein kinase 1(CK1),other kinases affecting Wnt/β-catenin signaling remain to be defined.Methods:To identify the kinases that modulate Wnt/β-catenin signaling,we applied a kinase small interfering RNA(siRNA)library screen approach.Luciferase assays,immunoblotting,and real-time polymerase chain reaction(PCR)were performed to confirm the regulation o f the Wnt/β-catenin signaling pathway by cyclin-dependent kinase 11(CDK11)and to investigate the underlying mechanism.Confocal immunofluorescence,coimmunoprecipitation(co-IP),and scratch wound assays were used to demonstrate colocalization,detect protein interactions,and explore the function of CDK11.Results:CDK11 was found to be a significant candidate kinase participating in the negative control of Wnt/P-catenin signaling.Down-regulation of CDK11 led to the accumulation of Wnt/β-catenin signaling receptor complexes,in a manner dependent on intact adenomatosis polyposis coli(APC)protein.Further analysis showed that CDK11 modulation of Wnt/P-catenin signaling engaged the endolysosomal machinery,and CDK11 knockdown enhanced the colocalization of Wnt/β-catenin signaling receptor complexes with early endosomes and decreased colocalization with lysosomes.Mechanistically,CDK11 was found to function in Wnt/β-catenin signaling by regulating microtubule stability.Depletion of CDK11 down-regulated acetyl-a-tubulin.Moreover,co-IP assays demonstrated that CDK11 interacts with the a-tubulin deacetylase SIRT2,whereas SIRT2 down-regulation in CDK11-depleted cells reversed the accumulation of Wnt/(3-catenin signaling receptor complexes.CDK11 was found to suppress cell migration through altered W nt/β-catenin signaling.Conclusions:CDK11 is a negative modulator of Wnt/β-catenin signaling that stabilizes microtubules,thus resulting in the dysregulation of receptor complex trafficking from early endosomes to lysosomes.Danmin Ou Lin Chen Jiang He Zhuoxian Rong Jie Gao Liyu Liu Feiyu Tang Jiang Li Yuezhen Deng Lunquan Sun 2020Cancer Biology & Medicine2020,17,2:0
16Examined lymph node numbers influence prognosis in rectal cancer treated with neoadjuvant therapy显示文摘Background:The number of lymph nodes examined(LNe)is often insufficient in patients with rectal cancer(RC)treated with neoadjuvant therapy;however,its prognostic value remains controversial.Thus,we retrospectively explored whether LNe had an influence on staging and prognosis and investigated whether there was a cut-off value for better prognosis in patients with RC treated with neoadjuvant therapy.Methods:Data were collected from seven prospective hospital databases in China from July 2002 to May 2018.Binary logistic regression models were used to predict lymph node metastasis.The cut-off value for LNe was determined using X-tile 3.6.1.Survival outcomes and risk factors were analyzed using the log-rank test and Cox regression model.Results:A total of 482 patients were included,of whom 459 had complete overall survival(OS)information.Using the percentile method,the total number of lymph nodes examined(TLNe)was 14-16(40th-60th percentile),and the proportion of patients with lymph node metastasis reached a maximum of 48.1%.Cox multivariate analysis showed that the odds ratio(OR)remained the highest when TLNe was 14-16(OR=3.379,P=0.003).The 3-year and 5-year OS were 85.4% and 77.8%,respectively.Negative lymph nodes examined(NLNe)of≤6 was an independent risk factor for 3-year and 5-year OS(3-year OS 71.1%vs.85.9%,P=0.004;5-year OS 66.3%vs.74.3%,P=0.035).Subgroup analysis for patients with ypN+showed that higher 3-year and 5-year OS were achieved when the TLNe was>10,78.8%vs.54.0%(P=0.005),and 60.8%vs.36.0%(P=0.012),respectively.Patients with ypN0M0 had a higher 5-year OS when the TLNe was>19(P=0.055).Conclusion:The TLNe and NLNe influenced the staging accuracy and demonstrated prognostic value in patients with RC treated with neoadjuvant therapy.Liyu Zhu Lin Wang Zhidong Gao Yujian Zeng Kaixiong Tao Quan Wang Xinming Li Huanhu Zhang Zhanlong Shen Jing Zhou Kai Shen Yingjiang Ye Aiwen Wu 2023Cancer Pathogenesis and Therapy2023,1,3:0
