Chemical screen identifies a geroprotective role of quercetin in premature aging显示文摘Aging increases the risk of various diseases. The main goal of aging research is to find therapies that attenuate aging and alleviate aging-related diseases. In this study, we screened a natural product library for geroprotective compounds using Werner syndrome (WS) human mesenchymal stem cells (hMSCs), a premature aging model that we recently established. Ten candidate compounds were identified and quercetin was investigated in detail due to its leading effects. Mechanistic studies revealed that quercetin alleviated senescence via the enhancement of cell proliferation and restoration of heterochromatin architecture in WS hMSCs. RNA-sequencing analysis revealed the transcriptional commonalities and differences in the geroprotective effects by quercetin and Vitamin C. Besides WS hMSCs, quercetin also attenuated cellular senescence in Hutchinson-Gilford progeria syndrome (HGPS) and physiological-aging hMSCs. Taken together, our study identifies quercetin as a geroprotective agent against accelerated and natural aging in hMSCs, providing a potential therapeutic intervention for treating age-associated disorders. | Lingling Geng Zunpeng Liu Weiqi Zhang Wei Li Zeming Wu Wei Wang Ruotong Ren Yao Su Peichang Wang Liang Sun Zhenyu Ju Piu Chan Moshi Song Jing Qu Guang-Hui Liu | 2019 | Protein & Cell2019,10,6: | 13
Single-cell transcriptomic atlas of primate cardiopulmonary aging显示文摘Aging is a major risk factor for many diseases, especially in highly prevalent cardiopulmonary comorbidities and infectious diseases including Coronavirus Disease 2019(COVID-19).Resolving cellular and molecular mechanisms associated with aging in higher mammals is therefore urgently needed. Here,we created young and old non-human primate single-nucleus/cell transcriptomic atlases of lung, heart and artery, the top tissues targeted by SARS-CoV-2.Analysis of cell type-specific aging-associated transcriptional changes revealed increased systemic inflammation and compromised virus defense as a hallmark of cardiopulmonary aging. With age, expression of the SARS-CoV-2 receptor angiotensin-converting enzyme 2(ACE2)was increased in the pulmonary alveolar epithelial barrier, cardiomyocytes, and vascular endothelial cells. We found that interleukin 7(IL7)accumulated in aged cardiopulmonary tissues and induced ACE2 expression in human vascular endothelial cells in an NF-κB-dependent manner.Furthermore, treatment with vitamin C blocked IL7-induced ACE2 expression.Altogether, our findings depict the first transcriptomic atlas of the aged primate cardiopulmonary system and provide vital insights into age-linked susceptibility to SARS-CoV-2,suggesting that geroprotective strategies may reduce COVID-19 severity in the elderly. | Shuai Ma Shuhui Sun Jiaming Li Yanling Fan Jing Qu Liang Sun Si Wang Yiyuan Zhang Shanshan Yang Zunpeng Liu Zeming Wu Sheng Zhang Qiaoran Wang Aihua Zheng Shuguang Duo Yang Yu Juan Carlos Izpisua Belmonte Piu Chan Qi Zhou Moshi Song Weiqi Zhang Guang-Hui Liu | 2021 | Cell Research2021,31,4: | 10
Stabilization of heterochromatin by CLOCK promotes stem cell rejuvenation and cartilage regeneration显示文摘Accumulating evidence indicates an association between the circadian clock and the aging process. However,it remains elusive whether the deregulation of circadian clock proteins underlies stem cell aging and whether they are targetable for the alleviation of aging-associated syndromes. Here,we identified a transcription factor-independent role of CLOCK,a core component of the molecular circadian clock machinery,in counteracting human mesenchymal stem cell(hMSC)decay.CLOCK expression was decreased during hMSC aging. In addition,CLOCK deficiency accelerated hMSC senescence, whereas the overexpression of CLOCK,even as a transcriptionally inactive form, rejuvenated physiologically and pathologically aged hMSCs.Mechanistic studies revealed that CLOCK formed complexes with nuclear lamina proteins and KAP1,thus maintaining heterochromatin architecture and stabilizing repetitive genomic sequences. Finally, gene therapy with lentiviral vectors encoding CLOCK promoted cartilage regeneration and attenuated age-related articular degeneration in mice. These findings demonstrate a noncanonical role of CLOCK in stabilizing heterochromatin, promoting tissue regeneration, and mitigating aging-associated chronic diseases. | Chuqian Liang Zunpeng Liu Moshi Song Wei Li Zeming Wu Zehua Wang Qiaoran Wang Si Wang Kaowen Yan Liang Sun Tomoaki Hishida Yanning Cai Juan Carlos lzpisua Belmonte Pedro Guillen Piu Chan Qi Zhou Weiqi Zhang Jing Qu Guang-Hui Liu | 2021 | Cell Research2021,31,2: | 8
Super-resolution fluorescence-assisted diffraction computational tomography reveals the threedimensional landscape of the cellular organelle interactome显示文摘The emergence of super-resolution(SR)fluorescence microscopy has rejuvenated the search for new cellular substructures. However,SR fluorescence microscopy achieves high contrast at the expense of a holistic view of the interacting partners and surrounding environment. Thus,we developed SR fluorescence-assisted diffraction computational tomography(SR-FACT),which combines label-free three-dimensional optical diffraction tomography(ODT)with two-dimensional fluorescence Hessian structured illumination microscopy. The ODT module is capable of resolving the mitochondria, lipid droplets, the nuclear membrane, chromosomes, the tubular endoplasmic reticulum, and lysosomes.Using dual-mode correlated live-cell imaging for a prolonged period of time,we observed novel subcellular structures named dark-vacuole bodies, the majority of which originate from densely populated perinuclear regions, and intensively interact with organelles such as the mitochondria and the nuclear membrane before ultimately collapsing into the plasma membrane. This work demonstrates the unique capabilities of SR-FACT,which suggests its wide applicability in cell biology in general. | Dashan Dong Xiaoshuai Huang Liuju Li Heng Mao Yanquan Mo Guangyi Zhang Zhe Zhang Jiayu Shen Wei Liu Zeming Wu Guanghui Liu Yanmei Liu Hong Yang Qihuang Gong Kebin Shi Liangyi Chen | 2020 | Light(Science & Applications)2020,9,1: | 7
High-Temperature Metamorphism, Anataxis and Tectonic Evolution of a Mafic Granulite from the Eastern Himalayan Orogen显示文摘The Himalayan Orogen, resulting from the Tertiary collision of Indian and Asian continents, is a natural laboratory for studying metamorphism, partial melting and granite formation of collisional orogens. However, metamorphic and anatectic conditions and timescales of meta-mafic rocks in the Greater Himalayan Sequences(GHS) in the east-central Himalaya remain controversial. In this paper, we conduct a study of petrology and geochronology of mafic granulite from the Eastern Himalayan Syntaxis(EHS). The mafic granulite with abundant leucosome bands occurs as layers within felsic granulites and is well deformed. The granulite consists of garnet, plagioclase, amphibole and quartz with minor clinopyroxene, orthopyroxene, biotite, rutile, titanite and ilmenite. The garnet has growth compositional zoning and contains abundant mineral inclusions in its core. Peak metamorphic mineral assemblage of the granulite is garnet, amphibole, plagioclase, quartz, clinopyroxene and rutile, recording a high-pressure(HP) and high-temperature(HT) peak-metamorphism under conditions of 14–15.5 kbar and 780–790℃ in the presence of melt. The reconstructed clockwise P-T path is characterized by an early heating and burial prograde metamorphism, and late isothermal and cooling decompression retrogression. The granulite witnessed a long lasting HT metamorphism, partial melting and melt crystallization process which began at ca. 39 Ma and lasted to ca. 11 Ma. The present study shows that various high-grade rocks of the GHS in the EHS core experienced similar metamorphic conditions and P-T-t paths, indicating that they occurred as a coherent slab during the subduction and exhumation of Indian lithosphere. The significant melts generated during the prograde metamorphism of the GHS rocks not only contributed to the formation of the Himalayan leucogranite, but also resulted in the rheological weakening and ductile flow of the thickened lower crust of the Himalayan Orogen. | Zeming Zhang Huixia Ding Xin Dong Zuolin Tian Dongyan Kang Hongchen Mu Shengkai Qin Yuanyuan Jiang Mengmei Li | 2018 | Journal of Earth Science2018,29,5: | 7

Structures and Electrochemical Performances of As-spun RE-Mg-Ni-Mn-based Alloys Applied to Ni-MH Battery显示文摘The La-Mg-Ni-Mn-based AB_2-type La_(1-x)Ce_xMgNi_(3.5)Mn_(0.5)(x = 0, 0.1, 0.2, 0.3, and 0.4) alloys were fabricated by melt spinning technology. The effects of Ce content on the structures and electrochemical hydrogen storage performances of the alloys were studied systematically. The XRD and SEM analyses proved that the experimental alloys consist of a major phase LaMgNi_4 and a secondary phase LaNi_5. The variation of Ce content causes an obvious change in the phase abundance of the alloys without changing the phase composition. Namely, with the increase of Ce content, the LaMgNi_4 phase augments and the LaNi_5 phase declines. The lattice constants and cell volumes of the alloys clearly shrink with increasing Ce content. Moreover, the Ce substitution for La results in the grains of the alloys clearly refined. The electrochemical tests showed that the substitution of Ce for La obviously improves the cycle stability of the as-spun alloys. The analyses on the capacity degradation mechanism demonstrate that the improvement can be attributed to the ameliorated anti-corrosion and antioxidation ability originating from substituting partial La with Ce. The as-spun alloys exhibit excellent activation capability, reaching the maximum discharge capacities just at the first cycling without any activation treatment. The substitution of Ce for La evidently improves the discharge potential characteristics of the as-spun alloys. The discharge capacity of the alloys first increases and then decreases with growing Ce content. Furthermore, a similar trend also exists in the electrochemical kinetics of the alloys, including the high rate discharge ability(HRD), hydrogen diffusion coefficient(D), limiting current density(IL) and charge transfer rate. | 张羊换 SHANG Hongwei LI Yaqin YUAN Zeming HU Feng QI Yan ZHAO Dongliang | 2018 | Journal of Wuhan University of Technology(Materials Science)2018,33,4: | 2
Working-in-tandem mechanism of multi-dopants in enhancing electrocatalytic nitrogen reduction reaction performance of carbon- based materials显示文摘Developing carbon-based electrocatalysts with excellent N2 adsorption and activation capability holds the key to achieve highly efficient nitrogen reduction reaction(NRR)for reaching its practical application. Here,we report a highly active electrocatalyst--metal-free pyrrolic-N dominated N,S co-doped carbon(pyrr-NSC)for NRR.Based on theoretical and experimental results,it is confirmed that the N and S-dopants practice a working-in-tandem mechanism on pyrr-NSC,where the N-dopants are utilized to create electropositive C sites for enhancing N2 adsorption and the S-dopants are employed to induce electron backdonation for facilitating N2 activation. The synergistic effect of the pyrrolic-N and S-dopants can also suppress the irritating hydrogen evolution reaction, further boosting the NRR performance. This work gives an indication that the combination of two different dopants on electrocatalyst can enhance NRR performance by working in the two tandem steps-the adsorption and activation of N2 molecules, providing a new strategy for NRR electrocatalyst design. | Wenqing Zhang Keke Mao Jingxiang Low Hengjie Liu Yanan Bo Jun Ma Qiaoxi Liu Yawen Jiang Jiuzhong Yang Yang Pan Zeming Qi Ran Long Li Song Yujie Xiong | 2021 | Nano Research2021,14,9: | 2
Pan-sharpening:a fast variational fusion approach显示文摘A fast variational fusion model based on partial differential equations (PDEs) is presented for pansharpening. The functional framework consists of several energy terms. The gradient energy term is created by calculating the gradient vector field of the panchromatic image and the geometry of the pan is injected into the multi-spectral bands. The radiometric reduction energy term and the channel correlation energy term are defined to decrease the radiometric distortion and preserve the correlation of multi-spectral channels while enforcing the inter-bands coherence. Inspired by the shock-filtering model, an inverse diffusion term for image enhancement is put to PDEs which are deduced by minimizing the energy functional. In comparison with the state-of-the-art fusion approaches based on `a trous wavelet and non-sampled contourlet, our model can obtain the fused image with a high spatial or spectral quality by adjusting the weight coefficients of the energy terms. It can also achieve a rather good trade-off between the spatial resolution improvement and the spectral quality preserving. Our model's computational complexity for one time step is only O(N). | ZHOU ZeMing LI YuanXiang SHI HanQing MA Ning SHEN Ji | 2012 | Science China(Information Sciences)2012,55,3: | 2
Gaseous hydrogen storage properties of Mg-Y-Ni-Cu alloys prepared by melt spinning显示文摘For purpose of promoting the hydrogen absorption and desorption thermodynamics and kinetics properties of Mg-Ni-based alloys, partially substituting Y and Cu for Mg and Ni respectively and melt spinning technique were applied for getting Mg25-xYxNi9 Cu(χ = 0-7) alloys. Their microstructures and phases were characterized with the help of X-ray diffraction and transmission electron microscopy. Their hydrogen absorbing and desorbing properties were tested by a Sievert apparatus, DSC, and TGA, which were connected with a H2 detector. In order to estimate the dehydrogenation activation energy of alloy hydride, both Arrhenius and Kissinger methods were applied for calculation. It is found that their hydriding kinetics notably declines, however, their hydrogen desorption kinetics conspicuously improves, with spinning rate and Y content increasing. Their hydrogen desorption activation energy markedly decreases under the same constraint, and it is found that melt spinning and Y substituting Mg improve the real driving force for dehydrogenation. As for the tendency of hydrogen absorption capacity,it presents an elevation firstly and soon after a decline with the rising of spinning rate, however, it always lowers with Y content growing. With Y content and spinning rate increasing, their thermodynamic parameters(△H and △S absolute values) visibly decrease, and the starting hydrogen desorption temperatures of alloy hydrides obviously lower. | Yanghuan Zhang Yaqin Li Wei Zhang Zeming Yuan Zhonghui Hou Yan Qi Shihai Guo | 2019 | Journal of Rare Earths2019,37,7: | 2
Microplastic pollution and enrichment of distinct microbiota in sediment of mangrove in Zhujiang River estuary,China显示文摘The microbial communities colonized on microplastics(MPs)have attracted widespread attention. However, few studies focused on the MPs impacts on mangrove ecosystems, particularly on bacterial communities. We investigated the MPs pollution in mangrove of Zhujiang(Pearl)River estuary(ZRE).To study the potential risk posed by MPs to the mangrove ecosystems, the differences in bacterial communities, functions, and complexity between MPs and sediment samples were reported for the first time.Microplastics(2991±1586 items/kg dry weight(dw))in sediment were mainly fibers and polyethylene, mostly transparent, and in size less than 0.5 mm.Bacterial communities and functions significantly differed from MPs in mangrove sediment.Compared with sediment,MPs significantly enriched members of Proteobacteria,Bacteroidetes, and Actinobacteria,as well as the bacteria associated with plastic-degrading and human diseases on their surface, suggesting that microbial communities on MPs may promote MPs degradation and the spread of diseases, posing potential risk to mangrove ecosystems and human health.Although bacteria on MPs exhibited a lower diversity, the co-occurrence network analysis indicated that network of bacteria colonized on MPs was bigger and more complex than those of mangrove sediment, illustrating that MPs can act as a distinct habitat in this special ecosystem. This study provides a new perspective for increasing our understanding of microplastic pollution in mangrove ecosystems. | Muting YAN Xiaofeng CHEN Wei CHU Weixin LI Minqian LI Zeming CAI Han GONG | 2023 | Journal of Oceanology and Limnology2023,41,1: | 1

Enhanced hydrogen storage performance of Mg-Cu-Ni system catalyzed by CeO2 additive显示文摘In this paper, the as-cast Mg85Cu5Ni10 alloy and Mg85Cu5Ni10-x wt%CeO2(x=0,4,8)alloys were prepared successfully by vacuum induction smelting and ball milling. The microstructure, hydrogen absorption/desorption kinetics and thermodynamics performances of the alloys were studied in detail. The results show that the Mg85Cu5Ni10 alloys with CeO2 additive have faster hydrogenation/dehydrogenation kinetics and better thermodynamic properties. The dehydrogenation activation energy is reduced to 81.211 kJ/mol from 119.142 by adding 8 wt%CeO2.CeO2 contributes to producing structural defects, nanocrystallines, grain boundaries, partial amorphous, lattice dislocations and cracks which are favorable to provide more hydrogen diffusion channels during hydriding/dehydriding process.Meanwhile,CeO2 additive weakens the bond energy of Mg-H.These micro structural changes caused by CeO2 additive improve the hydrogen storage performance of Mg85Cu5Ni10 markedly. | Yi Yin Bo Li Zeming Yuan Yan Qi Yanghuan Zhang | 2020 | Journal of Rare Earths2020,38,9: | 1

A Unique Ru-N_(4)-P Coordinated Structure Synergistically Waking Up the Nonmetal P Active Site for Hydrogen Production显示文摘Numerous experiments have demonstrated that the metal atom is the active center of monoatomic catalysts for hydrogen evolution reaction(HER),while the active sites of nonmetal doped atoms are often neglected.By combining theoretical prediction and experimental verification,we designed a unique ternary Ru-N_(4)-P coordination structure constructed by monodispersed Ru atoms supported on N,P dual-doped graphene for highly efficient hydrogen evolution in acid solution. The density functional theory calculations indicate that the charge polarization will lead to the most charge accumulation at P atoms, which results in a distinct nonmetallic P active sites with the moderate H∗adsorption energy.Notably, these P atoms mainly supply highly efficient catalytic sites with ultrasmall absorption energy of 0.007 eV.Correspondingly, the Ru-N_(4)-P demonstrated outstanding HER performance not only in an acidic condition but also in alkaline environment.Notably, the performance of Ru-NPC catalyst at high current is even superior to the commercial Pt/C catalysts, whether in acidic or alkaline medium. Our in situ synchrotron radiation infrared spectra demonstrate that a P-H_(ads) intermediate is continually emerging on the Ru-NPC catalyst, actively proving the nonmetallic P catalytically active site in HER that is very different with previously reported metallic sites. | Chuanqiang Wu Shiqing Ding Daobin Liu Dongdong Li Shuangming Chen Huijuan Wang Zeming Qi Binghui Ge Li Song | 2020 | Research2020,,1: | 1

Exosomes from antler stem cells alleviate mesenchymal stem cell senescence and osteoarthritis显示文摘Dear Editor,Stem cell therapy holds enormous and revolutionary promise to treat various age-related diseases, such as diabetes, heart failure, and Parkinson’s disease. However, low retention and survival rate of delivered stem cells, partially due to immunological rejection, constitute major hurdles for the clinical implementation of stem cell therapy(Lei et al.,2021a).Since mounting evidence showed that several types of stem cells mainly exert their therapeutic effects through the secretion of paracrine effects, exosomes, which are released by stem cells and execute most paracrine functions, have begun to draw attention in the field(Tran and Damaser,2015).Exosomes are membrane-enclosed vesicles with an average diameter of∼100 nanometers secreted by the cells, containing cytokines. | Jinghui Lei Xiaoyu Jiang Wei Li Jie Ren Datao Wang Zhejun Ji Zeming Wu Fang Cheng Yusheng Cai Zheng-Rong Yu Juan Carlos Izpisua Belmonte Chunyi Li Guang-Hui Liu Weiqi Zhang Jing Qu Si Wang | 2022 | Protein & Cell2022,13,3: | 1
