1Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 3Superior CO2 uptake on nitrogen doped carbonaceous adsorbents from commercial phenolic resin显示文摘In this study,N-doped porous carbons were produced with commercial phenolic resin as the raw material,urea as the nitrogen source and KOH as the activation agent.Different from conventional carbonization-nitriding-activation three-step method,a facile two-step process was explored to produce N-incorporated porous carbons.The as-obtained adsorbents hold superior CO2 uptake,i.e.5.01 and 7.47 mmol/g at 25℃and 0℃under 1 bar,respectively.The synergistic effects of N species on the surface and narrow micropores of the adsorbents decide their CO2 uptake under 25℃and atmospheric pressure.These phenolic resin-derived adsorbents also possess many extremely promising CO2 adsorption features like good recyclability,quick adsorption kinetics,modest heat of adsorption,great selectivity of CO2 over N2 and outstanding dynamic adsorption capacity.Cheap precursor,easy preparation strategy and excellent CO2 adsorption properties make these phenolic resin-derived N-doped carbonaceous adsorbents highly promising in CO2 capture.Shenfang Liu Linli Rao Pupu Yang Xinyi Wang Linlin Wang Rui Ma Limin Yue Xin Hu 2020Journal of Environmental Sciences2020,32,7:6
4Non-genetic photoacoustic stimulation of single neurons by a tapered fiber optoacoustic emitter显示文摘Neuromodulation at high spatial resolution poses great significance in advancing fundamental knowledge in the field of neuroscience and offering novel clinical treatments.Here,we developed a tapered fiber optoacoustic emitter(TFOE)generating an ultrasound field with a high spatial precision of 39.6 pm,enabling optoacoustic activation of single neurons or subcellular structures,such as axons and dendrites.Temporally,a single acoustic pulse of sub-microsecond converted by the TFOE from a single laser pulse of 3 ns is shown as the shortest acoustic stimuli so far for successful neuron activation.The precise ultrasound generated by the TFOE enabled the integration of the optoacoustic stimulation with highly stable patch-clamp recording on single neurons.Direct measurements of the electrical response of single neurons to acoustic stimulation,which is difficult for conventional ultrasound stimulation,have been demonstrated.By coupling TFOE with ex vivo brain slice electrophysiology,we unveil cell-type-specific responses of excitatory and inhibitory neurons to acoustic stimulation.These results demonstrate that TFOE is a non-genetic single-cell and sub-cellular modulation technology,which could shed new insights into the mechanism of ultrasound neurostimulation.Linli Shi Ying Jiang Fernando R.Fernandez Guo Chen Lu Lan Heng-Ye Man John A.White Ji-Xin Cheng Chen Yang 2021Light(Science & Applications)2021,10,8:4
5An association between the location of white matter changes and the behavioral and psychological symptoms of dementia in Alzheimer's disease patients显示文摘Objective:The frontal lobe may be involved in circuits associated with depression,apathy,aggression,and other psychiatric symptoms.Although white matter changes(WMC)are related to the severity of behavioral and psychological symptoms of dementia(BPSD)in patients with Alzheimer’s disease(AD),it is unclear which part of the WMC may play the most important role in BPSD.This study was designed to investigate the relationship between the location of WMC and the severity of BPSD in AD patients.Methods:Among patients diagnosed with Alzheimer’s disease between 2009 and2014,387 patients were retrospectively reviewed after those with pre‐existing organic brain syndrome,psychiatric diseases,or toxic‐metabolic encephalopathy were excluded.Patients’demographic and laboratory data,WMC measured with brain computed tomography and scored using the age‐related white matter changes(ARWMC)scale,and neuropsychological tests,including the cognitive abilities screening instrument(CASI),the Mini‐Mental State Examination(MMSE),the clinical dementia rating scale with sum‐box(CDR‐SB),and the neuropsychiatric inventory(NPI)were analyzed.Results:There was no significant difference in the NPI between patients with and without a history of stroke,hypertension,and diabetes.No significant difference in the NPI was identified between different sexes or different Apolipoprotein E(APOE)alleles.The NPI score was significantly correlated with the duration of education(r=–0.4515,P=0.0172),CASI(r=–0.2915,P<0.0001),MMSE(r=–0.8476,P<0.0001),and CDR‐SB(r=2.2839,P<0.0001).WMC in the right frontal lobe showed a significant difference in NPI in comparison to those without WMC(P=0.0255).After adjusting for age,duration of education,and CASI,WMC in the right frontal lobe remained significantly associated with the NPI score(β=3.8934,P=0.042).Conclusions:WMC involving the right frontal lobe may play an important role in the BPSD in AD patients during their dementia diagnosis.Further studies are necessary to confirm whether controlling the risk factors of WMC can slow the progression of BPSD.Tzuchou Lin Yihui Lin Linli Kao Yihui Kao Yuanhan Yang Pingsong Chou Mengni Wu 2016Translational Neuroscience and Clinics2016,2,1:3
6Overview to the Hard X-ray Modulation Telescope (Insight-HXMT) Satellite显示文摘As China’s first X-ray astronomical satellite, the Hard X-ray Modulation Telescope (HXMT), which was dubbed as Insight-HXMT after the launch on June 15, 2017, is a wide-band(1-250 ke V) slat-collimator-based X-ray astronomy satellite with the capability of all-sky monitoring in 0.2-3 Me V. 7The β-catenin/YAP signaling axis is a key regulator of melanoma-associated fibroblasts显示文摘β-catenin is a multifunctional protein that plays crucial roles in embryonic development,physiological homeostasis,and a wide variety of human cancers.Previously,we showed that in vivo targeted ablation ofβ-catenin in melanoma-associated fibroblasts after melanoma formation significantly suppressed tumor growth.However,when the expression ofβ-catenin was ablated in melanoma-associated fibroblasts before tumor initiation,melanoma development was surprisingly accelerated.How stromalβ-catenin deficiency leads to opposite biological effects in melanoma progression is not completely understood.Here,we report thatβ-catenin is indispensable for the activation of primary human stromal fibroblasts and the mediation of fibroblast-melanoma cell interactions.Using coimmunoprecipitation and proximity ligation assays,we identified Yes-associated protein(YAP)as an importantβ-catenin-interacting partner in stromal fibroblasts.YAP is highly expressed in the nuclei of cancer-associated fibroblasts(CAFs)in both human and murine melanomas.Mechanistic investigation revealed that YAP nuclear translocation is significantly modulated by Wnt/β-catenin activity in fibroblasts.Blocking Wnt/β-catenin signaling in stromal fibroblasts inhibited YAP nuclear translocation.In the absence of YAP,the ability of stromal fibroblasts to remodel the extracellular matrix(ECM)was inhibited,which is consistent with the phenotype observed in cells withβ-catenin deficiency.Further studies showed that the expression of ECM proteins and enzymes required for remodeling the ECM was suppressed in stromal fibroblasts after YAP ablation.Collectively,our data provide a new paradigm in which theβ-catenin-YAP signaling axis regulates the activation and tumor-promoting function of stromal fibroblasts.Tianyi Liu Linli Zhou Kun Yang Kentaro Iwasawa Ana Luisa Kadekaro Takanori Takebe Thomas Andl Yuhang Zhang 2019Signal Transduction and Targeted Therapy2019,4,1:1

9Optically-generated focused ultrasound for noninvasive brain stimulation with ultrahigh precision显示文摘High precision neuromodulation is a powerful tool to decipher neurocircuits and treat neurological diseases.Current non-invasive neuromodulation methods offer limited precision at the milimeter level.Here,we report opticallygenerated focused ultrasound(OFUS)for non-invasive brain stimulation with ultrahigh precision.OFUS is generated by a soft optoacoustic pad(SOAP)fabricated through embedding candle soot nanoparticles in a curved polydimethylsiloxane film.SOAP generates a transcranial ultrasound focus at 15 MHz with an ultrahigh lateral resolution of 83μm,which is two orders of magnitude smaller than that of conventional transcranial-focused ultrasound(tFUS).Here,we show effective OFUS neurostimulation in vitro with a single ultrasound cycle.We demonstrate submillimeter transcranial stimulation of the mouse motor cortex in vivo.An acoustic energy of 0.6 mJ/cm?,four orders of magnitude less than that of tFUS,is suffcient for successful OFUS neurostimulation.OFUS offers new capabilities for neuroscience studies and disease treatments by delivering a focus with ultrahigh precision noninvasively.Yueming Li Ying Jiang Lu Lan Xiaowei Ge Ran Cheng Yuewei Zhan Guo Chen Linli Shi Runyu Wang Nan Zheng Chen Yang Ji-Xin Cheng 2022Light(Science & Applications)2022,11,11:1

11Enhancing thermostability and activity of sucrose phosphorylase for high-level production of 2-O-α-d-glucosylglycerol显示文摘Sucrose phosphorylase(SPase)can transfer the glucosyl group of sucrose to different compounds and has been widely used in industry.To overcome the low thermostability of the sucrose phosphorylase from Leuconostoc mesenteroides ATCC 12291(LmSP),a method named PROSS was used to construct mutants with increased thermostability.All variants were screened by measuring their residual activities after heating at 50℃.Then,a single point mutant and a combined mutant with improved thermostability and activity were obtained.The half-lives of mutants at 50℃ were approximately twice as high as those of the wild type.In addition,2-O-α-d-glucosylglycerol(αGG)was synthesized by the wild type and the two improved variants,and the reaction conditions were optimized.Under the conditions of glycerol concentration of 3.2 mol/L,sucrose concentration of 1.2 mol/L,and enzyme concentration of 40 U/mL at 37℃ for 60 h,the yield ofαGG reached the maximum,and the sucrose conversion rate of the wild type,the mutant V23L and the combined mutant V23L/S424R were 62.3%,70.7%and 76.3%,respectively.In this study,SPase mutants with higher activity and stability were obtained,and achieved high-level production ofαGG.Linli Yang Lukasz Peplowski Yujuan Shen Haiquan Yang Xianzhong Chen Wei Shen Yuanyuan Xia 2022Systems Microbiology and Biomanufacturing2022,2,4:1

14Influence of ground effect on flow field structure and aerodynamic noise of high-speed trains显示文摘The simulation of the ground effect has always been a technical difficulty in wind tunnel tests of high-speed trains.In this paper,large eddy simulation and the curl acoustic integral equation were used to simulate the flow-acoustic field results of high-speed trains under four ground simulation systems(GSSs):“moving ground+rotating wheel”,“stationary ground+rotating wheel”,“moving ground+stationary wheel”,and“stationary ground+stationary wheel”.By comparing the fluid-acoustic field results of the four GSSs,the influence laws of different GSSs on the flow field structure,aero-acoustic source,and far-field radiation noise characteristics were investigated,providing guidance for the acoustic wind tunnel testing of high-speed trains.The calculation results of the aerodynamic noise of a 350 km/h high-speed train show that the moving ground and rotating wheel affect mainly the aero-acoustic performance under the train bottom.The influence of the rotating wheel on the equivalent sound source power of the whole vehicle was not more than 5%,but that of the moving ground slip was more than 15%.The average influence of the rotating wheel on the sound pressure level radiated by the whole vehicle was 0.3 dBA,while that of the moving ground was 1.8 dBA.Xiaoming TAN Linli GONG Xiaohong ZHANG Zhigang YANG 2024Journal of Zhejiang University-Science A(Applied Physics & Engineering)2024,25,2:0
15Generating homozygous mutant populations of barley microspores by ethyl methanesulfonate treatment显示文摘Induced mutations are important for genetic research and breeding.Mutations induced by physical or chemical mutagenesis are usually heterozygous during the early generations.However,mutations must be fixed prior to phenotyping or field trials,which requires additional rounds of self-pollination.Microspore culture is an effective method to produce double-haploid(DH)plants that are fixed homozygotes.In this study,we conducted ethyl methanesulfonate(EMS)-induced mutagenesis of microspore cultures of barley(Hordeum vulgare)cultivar‘Hua30’and landrace‘HTX’.The EMS concentrations were negatively correlated with the efficiency of callus induction and the frequency of mutant plant regeneration.The two genotypes showed different regeneration efficiencies.The phenotypic variation of the regenerated M1 plants and the presence of genome-wide nucleotide mutations,revealed by whole-genome sequencing,highlight the utility of EMS-induced mutagenesis of isolated microspore cultures for developing DH mutants.Genome-wide analysis of the mutation frequency in the regenerated plants revealed that a considerable proportion of mutations resulted from microspore culture(somaclonal variation)rather than EMS-induced mutagenesis.In addition to producing a population of 1972 homozygous mutant lines that are available for future field trials,this study lays the foundation for optimizing the regeneration efficiency of DH plants and the richness of mutations(mainly by fine-tuning the mutagen dosage).Linli Huang Guangqi Gao Congcong Jiang Guimei Guo Qiang He Yingjie Zong Chenghong Liu Ping Yang 2023aBIOTECH2023,4,3:0

17Discovery and structure-activity relationship studies of novel tetrahydro-β-carboline derivatives as apoptosis initiators for treating bacterial infections显示文摘Developing and excavating new agrochemicals with highly active and safe is an important tactic for protecting crop health and food safety.In this paper,to discover the new bactericide candidates,we designed,prepared a new type of1,2,3,4-tetrahydro-β-carboline(THC)derivatives and evaluated the in vitro and in vivo bioactivities against the Xanthomonas oryzae pv.oryzae(Xoo),Xanthomonas axonopodis pv.citri(Xac),and Pseudomonas syringae pv.actinidiae(Psa).The in vitro bioassay results exhibited that most title molecules possessed good activity toward the three plant pathogenic bacteria,the compound A17 showed the most active against Xoo and Xac with EC50 values of 7.27 and 4.89 mg mL^(-1)respectively,and compound A8 exhibited the best inhibitory activity against Psa with EC50value of 4.87 mg mL^(-1).Pot experiments showed that compound A17 exhibited excellent in vivo antibacterial activities to manage rice bacterial leaf blight and citrus bacterial canker,with protective efficiencies of 52.67 and 79.79%at 200 mgmL^(-1),respectively.Meanwhile,compound A8 showed good control efficiency(84.31%)against kiwifruit bacterial canker at 200 mg mL^(-1).Antibacterial mechanism suggested that these compounds could interfere with the balance of the redox system,damage the cell membrane,and induce the apoptosis of Xoo cells.Taken together,our study revealed that tetrahydro-β-carboline derivatives could be a promising candidate model for novel broadspectrum bactericides.Shanshan Su Hongwu Liu Junrong Zhang Puying Qi Yue Ding Ling Zhang Linli Yang Liwei Liu Xiang Zhou Song Yang 2024Journal of Integrative Agriculture2024,23,4:0

20SBC(Sanhuang Xiexin Tang combined with Baihu Tang plus Cangzhu)alleviates NAFLD by enhancing mitochondrial biogenesis and ameliorating inflammation in obese patients and mice显示文摘In the context of non-alcoholic fatty liver disease (NAFLD), characterized by dysregulated lipid metabolism in hepatocytes, the quest for safe and effective therapeutics targeting lipid metabolism has gained paramount importance. Sanhuang Xiexin Tang (SXT) and Baihu Tang (BHT) have emerged as prominent candidates for treating metabolic disorders. SXT combined with BHT plus Cangzhu (SBC) has been used clinically for Weihuochisheng obese patients. This retrospective analysis focused on assessing the anti-obesity effects of SBC in Weihuochisheng obese patients. We observed significant reductions in body weight and hepatic lipid content among obese patients following SBC treatment. To gain further insights, we investigated the effects and underlying mechanisms of SBC in HFD-fed mice. The results demonstrated that SBC treatment mitigated body weight gain and hepatic lipid accumulation in HFD-fed mice. Pharmacological network analysis suggested that SBC may affect lipid metabolism, mitochondria, inflammation, and apoptosis—a hypothesis supported by the hepatic transcriptomic analysis in HFD-fed mice treated with SBC. Notably, SBC treatment was associated with enhanced hepatic mitochondrial biogenesis and the inhibition of the c-Jun N-terminal kinase (JNK)/nuclear factor-kappa B (NF-κB) and extracellular signal-regulated kinase (ERK)/NF-κB pathways. In conclusion, SBC treatment alleviates NAFLD in both obese patients and mouse models by improving lipid metabolism, potentially through enhancing mitochondrial biogenesis.
