1Immobilization of functional nano-objects in living engineered bacterial biofilms for catalytic applications显示文摘Nanoscale objects feature very large surface-area-to-volume ratios and are now understood as powerful tools for catalysis,but their nature as nanomaterials brings challenges including toxicity and nanomaterial pollution.Immobilization is considered a feasible strategy for addressing these limitations.Here,as a proof-of-concept for the immobilization of nanoscale catalysts in the extracellular matrix of bacterial biofilms,we genetically engineered amyloid monomers of the Escherichia coli curli nanofiber system that are secreted and can self-assemble and anchor nano-objects in a spatially precise manner.We demonstrated three scalable,tunable and reusable catalysis systems:biofilm-anchored gold nanoparticles to reduce nitro aromatic compounds such as the pollutant p-nitrophenol,biofilm-anchored hybrid Cd0.9Zn0.1S quantum dots and gold nanoparticles to degrade organic dyes and biofilm-anchored CdSeS@ZnS quantum dots in a semi-artificial photosynthesis system for hydrogen production.Our work demonstrates how the ability of biofilms to grow in scalable and complex spatial arrangements can be exploited for catalytic applications and clearly illustrates the design utility of segregating high-energy nano-objects from injury-prone cellular components by engineering anchoring points in an extracellular matrix.Xinyu Wang Jiahua Pu Yi Liu Fang Ba Mengkui Cui Ke Li Yu Xie Yan Nie Qixi Mi Tao Li Lingli Liu Manzhou Zhu Chao Zhong 2019National Science Review2019,6,5:9
2Wheat powdery mildew resistance gene Pm64 derived from wild emmer (Triticum turgidum var.dicoccoides) is tightly linked in repulsion with stripe rust resistance gene Yr5显示文摘Stripe rust and powdery mildew are both devastating diseases for durum and common wheat.Pyramiding of genes conferring resistance to one or more diseases in a single cultivar is an important breeding approach to provide broader spectra of resistances in wheat improvement. A new powdery mildew resistance gene originating from wild emmer(Triticum turgidum var.dicoccoides) backcrossed into common wheat(T. aestivum) line WE35 was identified. It conferred an intermediate level of resistance to Blumeria graminis f. sp. tritici isolate E09 at the seedling stage and a high level of resistance at the adult plant stage. Genetic analysis showed that the powdery mildew resistance in WE35 was controlled by a dominant gene designated Pm64. Bulked segregant analysis(BSA) and molecular mapping indicated that Pm64 was located in chromosome bin 2 BL4-0.50–0.89. Polymorphic markers were developed from the corresponding genomic regions of Chinese Spring wheat and wild emmer accession Zavitan to delimit Pm64 to a 0.55 cM genetic interval between markers WGGBH1364 and WGGBH612, corresponding to a 15 Mb genomic region on Chinese Spring and Zavitan 2 BL, respectively. The genetic linkage map of Pm64 is critical for fine mapping and cloning. Pm64 was completely linked in repulsion with stripe rust resistance gene Yr5. 3Application of approximate entropy on dynamic characteristics of epileptic absence seizure显示文摘Electroencephalogram signals are time-varying complex electrophysiological signals. Existing studies show that approximate entropy, which is a nonlinear dynamics index, is not an ideal method for electroencephalogram analysis. Clinical electroencephalogram measurements usually contain electrical interference signals, creating additional challenges in terms of maintaining robustness of the analytic methods. There is an urgent need for a novel method of nonlinear dynamical analysis of the electroencephalogram that can characterize seizure-related changes in cerebral dynamics. The aim of this paper was to study the fluctuations of approximate entropy in preictal, ictal, and postictal electroencephalogram signals from a patient with absence seizures, and to improve the algorithm used to calculate the approximate entropy. The approximate entropy algorithm, especially our modified version, could accurately describe the dynamical changes of the brain during absence seizures. We could also demonstrate that the complexity of the brain was greater in the normal state than in the ictal state. 4Physicochemical Control of the Early Permian Xiangshan Fe-Ti Oxide Deposit in Eastern Tianshan(Xinjiang),NW China显示文摘The Xiangshan mafic-ultramafic complex is one of the major Early Permian mafic-ultramafic intrusions in eastern Tianshan(Xinjiang, NW China), and consists of two major intrusive phases. The first intrusive phase is mainly gabbroic rocks hosting ilmenite mineralization, while the second intrusive phase is mainly lherzoilite associated with Ni-Cu sulfide mineralization. The Xiangshan ilmenite orebodies hosted in the Fe-Ti oxide-bearing gabbro occur along the contact between hornblende gabbros and leucogabbros. The hornblende gabbros and Fe-Ti oxide rich gabbros at Xiangshan are newly dated to be Early Permian(280.1 and 279.2 Ma, respectively). Major and trace element compositions of zircons and whole rocks from Xiangshan hornblende gabbro and Fe-Ti oxide gabbro have been measured by in situ excimer laser ablation ICP-MS. Zircon Ce^(4+)/Ce^(3+) ratios based on lattice-strain model and Ti-in-zircon temperatures of hornblende gabbro and Fe-Ti oxide gabbro of the Xiangshan complex are calculated to evaluate the physicochemical variations during the ilmenite mineralization. Whole-rock geochemistry and zircon trace element geochemistry suggest that Fe-Ti oxide gabbros were formed from a basaltic parent magma which had undergone a transfromation from being H_2O-rich to H_2O-poor. During the magmatic evolution, primitive, H_2O-poor basaltic melts may have been replenished into the system, increasing its solidus temperature and decreasing its oxygen fugacity and H_2O contents. 5Petrogenesis and Metallogenesis of the Niumaoquan Gabbroic Intrusion Associated with Fe-Ti Oxide Ores in the Eastern Tianshan, NW China显示文摘The Niumaoquan layered gabbroic intrusion is in the southern margin of the Central Asian Orogenic Belt in North Xinjiang, China, and hosts a Fe-Ti oxide deposit in its evolved gabbroic phases. In this paper, we report zircon U-Pb age, Sr-Nd-Hf isotopes, plagioclase chemistry, and whole-rock geochemistry of the Niumaoquan layered gabbroic intrusion. Zircon grains separated from an anorthosite sample analyzed by laser ablation inductively coupled plasma mass spectrometry yielded a concordia age of 314.7±0.74 Ma, indicating that the Niumaoquan ore-bearing gabbroic intrusion was emplaced during the Late Carboniferous. The olivine gabbro texture and plagioclase chemistry suggest that plagioclase was an early crystallized silicate phase that crystallized prior to olivine. Fractional crystallization and accumulation of plagioclase significantly controlled the evolution of the Niumaoquan gabbroic intrusion and contributed to the formation of anorthosite layers, causing metallogenic elements to become enriched in the residual melt. The Niumaoquan gabbroic intrusion is characterized by the enrichment of large ion lithophile elements and depletion of high field strength elements, positive zircon εHf(t) values(+2.1 to +12.2), positive εNd(t) values(+3.3 to +5.2), and low initial ^(87)Sr/^(86)Sr ratios(0.7039 to 0.7047), suggesting that the parental magma was produced by interactions between metasomatized lithospheric mantle and depleted asthenospheric melts at an early post-collision stage. 7RNA-induced liquid phase separation of SARS-CoV-2 nucleocapsid protein facilitates NF-kB hyper-activation and inflammation显示文摘The ongoing 2019 novel coronavirus disease(COVID-19)caused by SARS-CoV-2 has posed a worldwide pandemic and a major global public health threat.The severity and mortality of COVID-19 are associated with virus-induced dysfunctional inflammatory responses and cytokine storms.However,the interplay between host inflammatory responses and SARS-CoV-2 infection remains largely unknown.Here,we demonstrate that SARS-CoV-2 nucleocapsid(N)protein,the major structural protein of the virion,promotes the virus-triggered activation of NF-kB signaling.After binding to viral RNA,N protein robustly undergoes liquid-liquid phase separation(LLPS),which recruits TAK1 and IKK complex,the key kinases of NF-kB signaling,to enhance NF-kB activation.Moreover,1,6-hexanediol,the inhibitor of LLPS,can attenuate the phase separation of N protein and restrict its regulatory functions in NF-kB activation.These results suggest that LLPS of N protein provides a platform to Induce NF-kB hyper-activation,which could be a potential therapeutic target against COVID-19 severe pneumonia.Yaoxing Wu Ling Ma Sihui Cai Zhen Zhuang Zhiyao Zhaou Shouheng Jin Weihong Xie Lingli Zhou Lei Zhang Jincun Zhao Jun Cui 2021Signal Transduction and Targeted Therapy2021,6,5:2
8New techniques of on-line biological sample processing and their application in the field of biopharmaceutical analysis显示文摘Biological sample pretreatment is an important step in biological sample analysis. Due to the diversity of biological matrices, the analysis of target substances in these samples presents significant challenges to sample processing. To meet these emerging demands on biopharmaceutical analysis, this paper summarizes several new techniques of on-line biological sample processing: solid phase extraction, solid phase micro-extraction, column switching, limited intake filler, molecularly imprinted solid phase extraction,tubular column, and micro-dialysis. 9Intracellular pharmacokinetic study of zidovudine and its phosphorylated metabolites显示文摘Zidovudine(AZT), the first drug approved by the US Food and Drug Administration for the treatment of human immunodeficiency virus(HIV) infection, is metabolized in the host cells to 5′-AZT triphosphate(AZT-TP) which inhibits HIV reverse transcriptase. As the pharmacokinetics of AZT and its phosphorylated metabolites in human peripheral blood mononuclear cells(h PBMCs) is limited, the aim of this study was to determine the pharmacokinetic parameters of AZT and its phosphorylated metabolites in h PBMCs from 12 healthy Chinese male subjects after a single oral dose of 600 mg of AZT. Blood samples were collected prior to drug administration, then at 0.5, 1, 1.5, 2, 2.5, 3, 3.5, 4, 5, 6, 8 and 10 h after drug administration. Mononuclear cells collected by Ficoll-Hypaque density gradient centrifugation were used for determination of AZT and metabolites [AZT monophosphate(AZT-MP), AZT diphosphate(AZT-DP) and AZT-TP] and the plasma was used to evaluate the pharmacokinetics of AZT. Plasma concentration of AZT peaked within 0.583 h and intracellular concentrations of AZT, AZT-MP, AZT-DP and AZT-TP peaked within 1.083, 1.500, 1.417 and 1.583 h, respectively. AZT in plasma was eliminated rapidly with t1/2of 2.022 h, and AZT-MP, AZT-DP and AZT-TP were eliminated with t1/2of 13.428,8.285 and 4.240 h, respectively. 11Identification of gut microbes associated with feed efficiency by daily-phase feeding strategy in growing-finishing pigs显示文摘Feed efficiency is one of the most important issues for sustainable pig production.Daily-phase feeding(DPF)is a form of precision feeding that could improve feed efficiency in pigs.Gut microbiota can regulate host nutrient digestion,absorption,and metabolism.However,which key microbes may play a vital role in improving the feed efficiency during DPF remains unclear.In the present study,we used a DPF program compared to a three-phase feeding(TPF)program in growing-finishing pigs to investigate the effects of gut microbiota on feed efficiency.A total of 204 Landrace×Yorkshire pigs(75 d)were randomly assigned into 2 treatments.Each treatment was replicated 8 times with 13 to 15 pigs per replicate pen.Pigs in the TPF group were fed with a commercial feeding program that supplied fixed feed for phasesⅠ,Ⅱ,andⅢ,starting at 81,101,and 132 d of age,respectively,and pigs in the DPF group were fed a blend of adjacent phase feed from 81 to 155 d at a gradual daily ratio and phaseⅢfeed from 155 to 180 d of age.Daily feed intake and body weight were recorded by a computerized device in the feeders.Feces and blood samples were collected from 1 pig per replicate at 155 and 180 d of age.The results showed that the DPF program remarkably improved the feed efficiency at 155 d(P<0.001)and 180 d of age(P<0.001),with a significant reduction of the intake of crude protein(P<0.01),net energy(P<0.001),crude fiber(P<0.001),ether extract(P<0.01),and ash(P<0.001).The daily-phase feeding program increased the abundance of Prevotella copri(P<0.05)and Paraprevotella clara(P<0.05),while it decreased the abundance of Ocilibacter(P<0.05)at 155 d of age.The results of correlation analysis indicated that the differentially abundant microbiota communities were closely associated with 20 metabolites which enriched amino acid and phenylalanine metabolism.Our results suggest that 2 key microbes may contribute to feed efficiency during daily-phase feeding strategies in pigs.Qin Jiang Chunlin Xie Lingli Chen Hongli Xiao Zhilian Xie Xiaoyan Zhu Libao Ma Xianghua Yan 2023Animal Nutrition2023,,1:0

13A 36 Mb terminal deletion of chromosome 2BL is responsible for a wheat semi-dwarf mutation显示文摘Reduced plant height is one of the most important traits related to lodging resistance and crop yield. The use of reduced height genes has been one of the main features in breeding modern high-yielding wheat varieties with less lodging. A spontaneous dwarf mutant DD399 was identified in a high yielding, gibberellic acid(GA)-insensitive, lodging-resistant variety Nongda 399(ND399). Significant differences in upper internode lengths between mutant DD399 and wild type ND399 were caused by reduced cell elongation. The plant height of ND399 × DD399 F_(1) hybrids was intermediate between the parents, indicating incomplete dominance or a dose–response effect of a reduced height gene. Plant height showed continuous distribution in the F_(2) population, and segregation distortion was observed among the 2292 F_(2:3) progenies. The reduced height mutation was characterized by Illumina 90 K iSelect SNP genotyping and bulked segregant RNA-Seq(BSR-Seq) analysis of the segregating population. A concentrated cluster of polymorphic SNPs associated with the reduced height phenotype was detected in the distal region of chromosome arm 2 BL. 14Penetration of foliar-applied Zn and its impact on apple plant nutrition status:in vivo evaluation by synchrotron-based X-ray fluorescence microscopy显示文摘The absorption of foliar fertilizer is a complex process and is poorly understood.The ability to visualize and quantify the pathway that elements take following their application to leaf surfaces is critical for understanding the science and for practical applications of foliar fertilizers.By the use of synchrotron-based X-ray fluorescence to analyze the in vivo localization of elements,our study aimed to investigate the penetration of foliar-applied Zn absorbed by apple(Malus domestica Borkh.)leaves with different physiological surface properties,as well as the possible interactions between foliar Zn level and the mineral nutrient status of treated leaves.The results indicate that the absorption of foliar-applied Zn was largely dependent on plant leaf surface characteristics.High-resolution elemental maps revealed that the high binding capacity of the cell wall for Zn contributed to the observed limitation of Zn penetration across epidermal cells.Trichome density and stomatal aperture had opposite effects on Zn fertilizer penetration:a relatively high density of trichomes increased the hydrophobicity of leaves,whereas the presence of stomata facilitated foliar Zn penetration.Low levels of Zn promoted the accumulation of other mineral elements in treated leaves,and the complexation of Zn with phytic acid potentially occurred owing to exposure to high-Zn conditions.The present study provides direct visual evidence for the Zn penetration process across the leaf surface,which is important for the development of strategies for Zn biofortification in crop species.Ruohan Xie Jianqi Zhao Lingli Lu Patrick Brown Jiansheng Guo Shengke Tian 2020Horticulture Research2020,7,1:0
