Sodium oligomannate therapeutically remodels gut microbiota and suppresses gut bacterial amino acids-shaped neuroinflammation to inhibit Alzheimer's disease progression显示文摘Recently, increasing evidence has suggested the association between gut dysbiosis and Alzheimer's disease(AD)progression, yet the role of gut microbiota in AD pathogenesis remains obscure.Herein,we provide a potential mechanistic link between gut microbiota dysbiosis and neuroinflammation in AD progression.Using AD mouse models,we discovered that, during AD progression, the alteration of gut microbiota composition leads to the peripheral accumulation of phenylalanine and isoleucine, which stimulates the differentiation and proliferation of pro-inflammatory T helper 1(Thl)cells.The brain-infiltrated peripheral Th1 immune cells are associated with the Ml microglia aaivation, contributing to AD-associated neuroinflammation.Importantly, the elevation of phenylalanine and isoleucine concentrations and the increase of Th1 cell frequency in the blood were also observed in two small independent cohorts of patients with mild cognitive impairment(MCI)due to AD.Furthermore,GV-971,a sodium oligomannate that has demonstrated solid and consistent cognition improvement in a phase 3 clinical trial in China, suppresses gut dysbiosis and the associated phenylalanine/isoleucine accumulation, harnesses neuroinflammation and reverses the cognition impairment.Together, our findings highlight the role of gut dysbiosis-promoted neuroinflammation in AD progression and suggest a novel strategy for AD therapy by remodelling the gut microbiota.Xinyi Wang Guangqiang Sun Teng Feng Jing Zhang Xun Huang Tao Wang Zuoquan Xie Xingkun Chu Jun Yang Huan Wang Shuaishuai Chang Yanxue Gong Lingfei Ruan Guanqun Zhang Siyuan Yan Wen Lian Chen Du Dabing Yang Qingli Zhang Feifei Lin Jia Liu Haiyan Zhang Changrong Ge Shifu Xiao Jian Ding Meiyu Geng 2019Cell Research2019,29,10:192
Terrestrial carbon sinks in China and around the world and their contribution to carbon neutrality显示文摘Enhancing the terrestrial ecosystem carbon sink(referred to as terrestrial C sink) is an important way to slow down the continuous increase in atmospheric carbon dioxide(CO_(2)) concentration and to achieve carbon neutrality target. To better understand the characteristics of terrestrial C sinks and their contribution to carbon neutrality, this review summarizes major progress in terrestrial C budget researches during the past decades, clarifies spatial patterns and drivers of terrestrial C sources and sinks in China and around the world, and examines the role of terrestrial C sinks in achieving carbon neutrality target.According to recent studies, the global terrestrial C sink has been increasing from a source of (-0.2±0.9) Pg C yr^(-1)(1 Pg=1015g)in the 1960s to a sink of (1.9±1.1) Pg C yr^(-1) in the 2010s.By synthesizing the published data,we estimate terrestrial C sink of 0.20–0.25 Pg C yr^(-1) in China during the past decades, and predict it to be 0.15–0.52 Pg C yr^(-1) by 2060.The terrestrial C sinks are mainly located in the mid-and high latitudes of the Northern Hemisphere, while tropical regions act as a weak C sink or source. The C balance differs much among ecosystem types:forest is the major C sink;shrubland, wetland and farmland soil act as C sinks;and whether the grassland functions as C sink or source remains unclear.Desert might be a C sink, but the magnitude and the associated mechanisms are still controversial.Elevated atmospheric CO_(2) concentration, nitrogen deposition, climate change, and land cover change are the main drivers of terrestrial C sinks, while other factors such as fires and aerosols would also affect ecosystem C balance. The driving factors of terrestrial C sink differ among regions.Elevated CO_(2) concentration and climate change are major drivers of the C sinks in North America and Europe, while afforestation and ecological restoration are additionally important forcing factors of terrestrial C sinks in China. For future studies,we recommend the necessity for intensive and long-term ecosystem C monitoring over broad geographic scale to improve terrestrial biosphere models for accurately evaluating terrestrial C budget and its dynamics under various climate change and policy scenarios.Yuanhe Yang Yue Shi Wenjuan Sun Jinfeng Chang Jianxiao Zhu Leiyi Chen Xin Wang Yanpei Guo Hongtu Zhang Lingfei Yu Shuqing Zhao Kang Xu Jiangling Zhu Haihua Shen Yuanyuan Wang Yunfeng Peng Xia Zhao Xiangping Wang Huifeng Hu Shiping Chen Mei Huang Xuefa Wen Shaopeng Wang Biao Zhu Shuli Niu Zhiyao Tang Lingli Liu Jingyun Fang 2022Science China(Life Sciences)2022,65,5:32
Toxicity of graphene oxide and multi-walled carbon nanotubes against human cells and zebrafish显示文摘Graphene possesses unique physical and chemical properties, which have inspired a wide range of potential biomedical applications. However, little is known about the adverse effects of graphene on the human body and ecological environment. The purpose of our work is to make assessment on the toxicity of graphene oxide (GO) against human cell line (human bone marrow neuroblastoma cell line and human epithelial carcinoma cell line) and zebrafish (Danio rerio) by comparing the toxic effects of GO with its sister, multi-walled carbon nanotubes (MWNTs). The results show that GO has a moderate toxicity to organisms since it can induce minor (about 20%) cell growth inhibition and slight hatching delay of zebrafish embryos at a dosage of 50 mg/L, but did not result in significant increase of apoptosis in embryo, while MWNTs exhibit acute toxicity leading to a strong inhibition of cell proliferation and serious morphological defects in developing embryos even at relatively low concentration of 25 mg/L. The distinctive toxicity of GO and MWNTs should be ascribed to the different models of interaction between nanomaterials and organisms, which arises from the different geometric structures of nanomaterials. Collectively, our work suggests that GO does actual toxicity to organisms posing potential environmental risks and the result is also shedding light on the geometrical structure-dependent toxicity of graphitic nanomaterials.CHEN LiQiang HU PingPing ZHANG Li HUANG SiZhou LUO LingFei HUANG ChengZhi 2012Science China Chemistry2012,55,10:12
Investigation on microstructure and mechanical properties of Al-Zn-Mg-Cu alloys with various Zn/Mg ratios显示文摘The effects of Zn/Mg ratios on microstructure and mechanical properties of Al-Zn-Mg-Cu alloys aged at 150℃have been investigated by using tensile tests, optical metallography, scanning electron microscopy, transmission electron microscopy and atom probe tomography analyses. With increasing Zn/Mg ratios, the ageing process is significantly accelerated and the time to peak ageing is reduced.T’phase predominates in alloys of lower Zn/Mg ratios whileη’phase predominates in alloys with a Zn/Mg ratio over 2.86.Co-existence of T’phase andη’phase with a large number density is beneficial to the high strength of alloys.Such precipitates together with narrow precipitate free zones cause a brittle intergranular fracture.A strength model has been established to predict the co-strengthening effect of T’phase andη’phase in Al-Zn-Mg-Cu alloys, including the factors of the grain boundary, solid solution and precipitation.Yan Zou Xiaodong Wu Songbai Tang Qianqian Zhu Hui Song Mingxing Guo Lingfei Cao 2021Journal of Materials Science & Technology2021,,26:12

Jasmonic acid carboxyl methyltransferase regulates development and herbivory-induced defense response in rice显示文摘Jasmonic acid(JA) and related metabolites play a key role in plant defense and growth. JA carboxyl methyltransferase(JMT) may be involved in plant defense and development by methylating JA to methyl jasmonate(Me JA) and thus influencing the concentrations of JA and related metabolites. However, no JMT gene has been well characterized in monocotyledon defense and development at the molecular level. After we cloned a rice JMT gene,Os JMT1, whose encoding protein was localized in the cytosol, we found that the recombinant Os JMT1 protein catalyzed JA to Me JA. Os JMT1 is up-regulated in response to infestation with the brown planthopper(BPH; Nilaparvata lugens). Plants in which Os JMT1 had been overexpressed(oeJMT plants) showed reduced height and yield. These oe-JMT plants also exhibited increased Me JA levels but reduced levels of herbivore-induced JA and jasmonoyl-isoleucine(JAIle). The oe-JMT plants were more attractive to BPH female adults but showed increased resistance to BPH nymphs, probably owing to the different responses of BPH female adults and nymphs to the changes in levels of H_2O_2 and Me JA in oe-JMT plants. These results indicate that Os JMT1,by altering levels of JA and related metabolites, plays a role in regulating plant development and herbivore-induced defense responses in rice.Jinfeng Qi Jiancai Li Xiu Han Ran Li Jianqiang Wu Haixin Yu Lingfei Hu Yutao Xiao Jing Lu Yonggen Lou 2016Journal of Integrative Plant Biology2016,58,6:10
Treatment of Phenol Wastewater Using High Gravity Electrochemical Reactor with Multi-concentric Cylindrical Electrodes显示文摘A novel high gravity electrochemical reactor with multi-concentric cylindrical electrodes was used in the electrochemical treatment of 5 000 mg/L phenol-containing wastewater at a petrochemical plant, which can operate continuously and process in a large scale. The results show that the high gravity technology used in electrochemical treatment of phenol-containing wastewater can shorten the electrolysis time, decrease the electrolysis voltage, and reduce the energy consumption. The COD removal efficiency was high in the high-gravity field, and reached up to about 48%, which was about 2 times the value achieved in the normal gravity field at a processing capacity of 6 L, a high gravity factor of 80, a voltage of 12 V, an electrolysis time of 40 min, and a wastewater flowrate of 80 L/h.Gao Jing Liu Youzhi Chang Lingfei 2012China Petroleum Processing & Petrochemical Technology2012,14,2:10
Recent progress in NIR-Ⅱ emitting lanthanide-based nanoparticles and their biological applications显示文摘The second near-infrared(NIR-Ⅱ,1000-1700 nm)window provides a superior optical platform with high resolution, deep penetration and high signal-to-noise ratios(SNRs),which results from the intrinsic low scattering and auto fluorescence in biological tissues. As one of the promising NIR-Ⅱemitting probes, lanthanide based nanoparticles(LnNPs)exhibit high photo stability and chemostability, long photoluminescence lifetimes, low long-term cytotoxicity and narrow emission bandwidths. All these merits have spurred the evolution of related bio-optics and a variety of biomedical applications of LnNPs.This mini-review discusses the most recent advances in both the design-the composition and surface modifications-and the applications of NIR-Ⅱemitting LnNPs in bioimaging, disease diagnosis and therapy. We also summarize the current limits and challenges facing the applications of LnNPs as well as discuss the directions of future development.Suwan Ding Lingfei Lu Yong Fan Fan Zhang 2020Journal of Rare Earths2020,38,5:9

Histological, hormonal and transcriptomic reveal the changes upon gibberellin-induced parthenocarpy in pear fruit显示文摘Phytohormones play crucial roles in fruit set regulation and development.Here,gibberellins(GA4+7),but not GA3,induced pear parthenocarpy. To systematically investigate the changes upon GA4+7 induced pear parthenocarpy, dynamic changes in histology, hormone and transcript levels were observed and identified in unpollinated, pollinated and GA4+7-treated ovaries.Mesocarp cells continued developing in both GA4+7-treated and pollinated ovaries. In unpollinated ovaries, mesocarp cells stopped developing 14 days after anthesis.During fruit set process,GA4+7,but not GA1+3,increased after pollination.Abscisic acid(ABA)accumulation was significantly repressed by GA4+7 or pollination, but under unpollinated conditions,ABA was produced in large quantities.Moreover,indole-3-acetic acid biosynthesis was not induced by GA4+7 or pollination treatments.Details of this GA–auxin–ABA cross-linked gene network were determined by a comparative transcriptome analysis. The indole-3-acetic acid transport-related genes, mainly auxin efflux carrier component genes, were induced in both GA4+7-treated and pollinated ovaries.ABA biosynthetic genes of the 9-cis-epoxycarotenoid dioxygenase family were repressed by GA4+7 and pollination. Moreover, directly related genes in the downstream parthenocarpy network involved in cell division and expansion(upregulated),and MADS-box family genes(downregulated),were also identified. Thus,a model of GA-induced hormonal balance and its effects on parthenocarpy were established.Lulu Liu Zhigang Wang Jianlong Liu Fengxia Liu Rui Zhai Chunqin Zhu Huibin Wang Fengwang Ma Lingfei Xu 2018Horticulture Research2018,5,1:7
The R2R3-MYB transcription factor GhMYB1a regulates flavonol and anthocyanin accumulation in Gerbera hybrida显示文摘Anthocyanins and flavonols have vital roles in flower coloration, plant development, and defense.Because anthocyanins and flavonols share the same subcellular localization and common biosynthetic substrates, these pathways may compete for substrates. However, the mechanism regulating this potential competition remains unclear. Here,we identified GhMYB1a,an R2R3-MYB transcription factor involved in the regulation of anthocyanin and flavonol accumulation in gerbera(Gerbera hybrida).GhMYB1a shares high sequence similarity with that of other characterized regulators of flavonol biosynthesis. In addition,GhMYB1a is also phylogenetically grouped with these proteins. The overexpression of GhMYB1a in gerbera and tobacco(Nicotiana tabacum)resulted in decreased anthocyanin accumulation and increased accumulation of flavonols by upregulating the structural genes involved in flavonol biosynthesis. We further found that GhMYB1a functions as a homodimer instead of interacting with basic helix-loop-helix cofactors. These results suggest that GhMYB1a is involved in regulating the anthocyanin and flavonol metabolic pathways through precise regulation of gene expression. The functional characterization of GhMYB1a provides insight into the biosynthesis and regulation of flavonols and anthocyanins.Chunmei Zhong Yi Tang Bin Pang Xukun Li Yuping Yang Jing Deng Chengyong Feng Lingfei Li Guiping Ren Yaqin Wang Jianzong Peng Shulan Sun Shan Liang Xiaojing Wang 2020Horticulture Research2020,7,1:7
Aircraft Engine Gas Path Fault Diagnosis Based on Hybrid PSO-TWSVM显示文摘Twin support vector machine(TWSVM)is a new development of support vector machine(SVM)algorithm. It has the smaller computation scale and the stronger ability to cope with unbalanced problems. In this paper,TWSVM is introduced into aircraft engine gas path fault diagnosis. The generalization capacity of Gauss kernel function usually used in TWSVM is relatively weak.So a mixed kernel function is used to improve performance to ensure that the TWSVM algorithm can better balance a strong generalization ability and a good learning ability.Experimental results prove that the cross validation training accuracy of TWSVM using the mixed kernel function averagely increases 2%.Grid search is usually applied in parameter optimization of TWSVM,but it heavily depends on experience. Therefore, the hybrid particle swarm algorithm is introduced. It can intelligently and rapidly find the global optimum.Experiments prove that its training accuracy is better than that of the classical particle swarm algorithm by 5%.Du Yanbin Xiao Lingfei Chen Yusheng Ding Runze 2018Transactions of Nanjing University of Aeronautics and Astronautics2018,35,2:6
PbGA2ox8 induces vascular-related anthocyanin accumulation and contributes to red stripe formation on pear fruit显示文摘Fruit with stripes, which are generally longitudinal, can occur naturally, but the bioprocesses underlying this phenomenon are unclear.Previously,we observed an atypical anthocyanin distribution that caused red-striped fruit on the spontaneous pear bud sport“Red Zaosu”(Pyrus bretschneideri Rehd.).In this study, comparative transcriptome analysis of the sport and wild-type“Zaosu”revealed that this atypical anthocyanin accumulation was tightly correlated with abnormal overexpression of the gene-encoding gibberellin(GA)2-beta-dioxygenase 8,PbGA2ox8.Consistently,decreased methylation was also observed in the promoter region of PbGA2ox8 from“Red Zaosu”compared with“Zaosu”.Moreover, the GA levels in“Red Zaosu”seedlings were lower than those in“Zaosu”seedlings, and the application of exogenous GA4 reduced abnormal anthocyanin accumulation in“Red Zaosu”.Transient overexpression of PbGA2ox8 reduced the GA4 level and caused anthocyanin accumulation in pear fruit skin. Moreover, the presence of red stripes indicated anthocyanin accumulation in the hypanthial epidermal layer near vascular branches(VBs)in“Red Zaosu”.Transient overexpression of PbGA2ox8 resulting from vacuum infiltration induced anthocyanin accumulation preferentially in calcium-enriched areas near the vascular bundles in pear leaves. We propose a fruit-striping mechanism,in which the abnormal overexpression of PbGA2ox8 in“Red Zaosu”induces the formation of a longitudinal array of anthocyanin stripes near vascular bundles in fruit.Rui Zhai Zhigang Wang Chengquan Yang Kui Lin-Wang Richard Espley Jianlong Liu Xieyu Li Zhongying Wu Pengmin Li Qingmei Guan Fengwang Ma Lingfei Xu 2019Horticulture Research2019,6,1:5
The interplay of aging, genetics and environmental factors in the pathogenesis of Parkinson’s disease显示文摘Background:Parkinson’s disease(PD)is characterized by dopaminergic neuronal loss in the substantia nigra pars compacta and intracellular inclusions called Lewy bodies(LB).During the course of disease,misfoldedα-synuclein,the major constituent of LB,spreads to different regions of the brain in a prion-like fashion, giving rise to successive non-motor and motor symptoms.Etiology is likely multifactorial, and involves interplay among aging, genetic susceptibility and environmental factors.Main body:The prevalence of PD rises exponentially with age,and aging is associated with impairment of cellular pathways which increases susceptibility of dopaminergic neurons to cell death. However, the majority of those over the age of 80 do not have PD,thus other factors in addition to aging are needed to cause disease.Discovery of neurotoxins which can result in parkinsonism led to efforts in identifying environmental factors which may influence PD risk.Nevertheless, the causality of most environmental factors is not conclusively established, and alternative explanations such as reverse causality and recall bias cannot be excluded. The lack of geographic clusters and conjugal cases also go against environmental toxins as a major cause of PD.Rare mutations as well as common variants in genes such as SNCA,LRRK2 and GBA are associated with risk of PD,but Mendelian causes collectively only account for 5%of PD and common polymorphisms are associated with small increase in PD risk.Heritability of PD has been estimated to be around 30%.Thus,aging,genetics and environmental factors each alone is rarely sufficient to cause PD for most patients. Conclusion:PD is a multifactorial disorder involving interplay of aging, genetics and environmental factors. This has implications on the development of appropriate animal models of PD which take all these factors into account.Common converging pathways likely include mitochondrial dysfunction, impaired autophagy, oxidative stress and neuroinflammation, which are associated with the accumulation and spread of misfoldedα-synuclein and neurodegeneration.Understanding the mechanisms involved in the initiation and progression of PD may lead to potential therapeutic targets to prevent PD or modify its course.Shirley Yin-Yu Pang Philip Wing-Lok Ho Hui-Fang Liu Chi-Ting Leung Lingfei Li Eunice Eun Seo Chang David Boyer Ramsden Shu-Leong Ho 2019Translational Neurodegeneration2019,8,1:5

Using zebrafish as the model organism to understand organ regeneration显示文摘The limited regenerative capacity of several organs, such as central nervous system(CNS), heart and limb in mammals makes related major diseases quite difficult to recover. Therefore, dissection of the cellular and molecular mechanisms underlying organ regeneration is of great scientific and clinical interests. Tremendous progression has already been made after extensive investigations using several model organisms for decades. Unfortunately, distance to the final achievement of the goal still remains. Recently, zebrafish became a popular model organism for the deep understanding of regeneration based on its powerful regenerative capacity, in particular the organs that are limitedly regenerated in mammals. Additionally, zebrafish are endowed with other advantages good for the study of organ regeneration. This review summarizes the recent progress in the study of zebrafish organ regeneration, in particular regeneration of fin, heart, CNS, and liver as the representatives. We also discuss reasons of the reduced regenerative capacity in higher vertebrate, the roles of inflammation during regeneration, and the difference between organogenesis and regeneration.SHI WenChao FANG ZhiBing LI Li LUO LingFei 2015Science China(Life Sciences)2015,58,4:5
Enhancement of femtosecond laser-induced surface ablation via temporal overlapping double-pulse irradiation显示文摘This paper reports the physical phenomenon of the temporal overlapping double femtosecond laser-induced ablation enhancement at different time delays.Detailed thermodynamic modeling demonstrates the ablation enhancement is highly dependent on the first pulse's laser fluence. In the case of the first pulse laser fluence being higher than material's ablation threshold, the ablation enhancement is attributed to optical absorption modification by the first pulse ablation. While the first pulse's laser fluence is lower than the material's ablation threshold, the first pulse-induced melting leads to much higher absorption of the second pulse. However, for the case of the first pulse's laser fluence even lower than melting threshold, the ablation enhancement decreases obviously with time delay. The results of the temporal overlapping double femtosecond laser ablation of poly(ε-caprolactone)are in good agreement with the theoretical predictions.ZHENYUAN LIN LINGFEI JI MINGHUI HONG 2020Photonics Research2020,8,3:5

Apple SERRATE negatively mediates drought resistance by regulating MdMYB88 and MdMYB124 and microRNA biogenesis显示文摘The function of serrate(SE)in miRNA biogenesis in Arabidopsis is well elucidated, whereas its role in plant drought resistance is largely unknown. In this study,we report that MdSE acts as a negative regulator of apple(Malus×domestica)drought resistance by regulating the expression levels of MdMYB88 and MdMYB124 and miRNAs,including mdm-miR156,mdm-miR166,mdm-miR172,mdm-miR319,and mdm-miR399.MdSE interacts with MdMYB88 and MdMYB124,two positive regulators of apple drought resistance.MdSE decreases the transcript and protein levels of MdMYB88 and MdMYB124,which directly regulate the expression of MdNCED3,a key enzyme in abscisic acid(ABA)biosynthesis.Furthermore,MdSE is enriched in the same region of the MdNECD3 promoter where MdMYB88/MdMYB124 binds.Consistently,MdSE RNAi transgenic plants are more sensitive to ABA-induced stomatal closure, whereas MdSE OE plants are less sensitive. In addition, under drought stress,MdSE is responsible for the biogenesis of mdm-miR399,a negative regulator of drought resistance, and negatively regulates miRNAs,including mdm-miR156,mdm-miR166,mdm-miR172,and mdm-miR319,which are positive regulators of drought resistance.Taken together,by revealing the negative role of MdSE,our results broaden our understanding of the apple drought response and provide a candidate gene for apple drought improvement through molecular breeding.Xuewei Li Pengxiang Chen Yinpeng Xie Yan Yan Liping Wang Huan Dang Jing Zhang Lingfei Xu Fengwang Ma Qingmei Guan 2020Horticulture Research2020,7,1:4
