1Whole genome sequencing of 10K patients with acute ischaemic stroke or transient ischaemic attack: design, methods and baseline patient characteristics显示文摘Background and purpose Stroke is the second leading cause of death worldwide and the leading cause of mortality and long-term disability in China,but its underlying risk genes and pathways are far from being comprehensively understood.We here describe the design and methods of whole genome sequencing(WGS)for 10914 patients with acute ischaemic stroke or transient ischaemic attack from the Third China National Stroke Registry(CNSR-III).Methods Baseline clinical characteristics of the included patients in this study were reported.DNA was extracted from white blood cells of participants.Libraries are constructed using qualified DNA,and WGS is conducted on BGISEQ-500 platform.The average depth is intended to be greater than 30×for each subject.Afterwards,Sentieon software is applied to process the sequencing data under the Genome Analysis Toolkit best practice guidance to call genotypes of single nucleotide variants(SNVs)and insertion-deletions.For each included subject,21 fingerprint SNVs are genotyped by MassARRAY assays to verify that DNA sample and sequencing data originate from the same individual.The copy number variations and structural variations are also called for each patient.All of the genetic variants are annotated and predicted by bioinformatics software or by reviewing public databases.Results The average age of the included 10914 patients was 62.2±11.3 years,and 31.4%patients were women.Most of the baseline clinical characteristics of the 10914 and the excluded patients were balanced.Conclusions The WGS data together with abundant clinical and imaging data of CNSR-III could provide opportunity to elucidate the molecular mechanisms and discover novel therapeutic targets for stroke.Si Cheng Zhe Xu Yang Liu Jinxi Lin Yong Jiang Yilong Wang Xia Meng Anxin Wang Xinying Huang Zhimin Wang Guohua Chen Songdi Wu Zhengchang Jia Yongming Chen Xuerong Qiu Jun Wu Binbin Song Weizhong Ji Zhongping An Wenjun Xue Lili Zhao Yu Geng Hongyan Li Hao Li Yongjun Wang 2021Stroke & Vascular Neurology2021,6,2:8
23-year Treatment of Tenofovir Alafenamide vs.Tenofovir Disoproxil Fumarate for Chronic HBV Infection in China显示文摘Background and Aims:Tenofovir alafenamide(TAF)has similar efficacy to tenofovir disoproxil fumarate(TDF)but with improved renal and bone safety in chronic hepatitis B patients studied outside of China.We report 3-year results from two phase 3 studies with TAF in China(Clinicaltrials.gov:NCT02836249 and NCT02836236).Methods:Chinese hepatitis B e antigen(HBeAg)-positive and-negative chronic hepatitis B patients with viremia and elevated alanine aminotransferase were randomized 2:1 to TAF or TDF treatment groups and treated in a double-blind fashion for 144 weeks(3 years).Efficacy responses were assessed by individual study while safety was assessed by a pooled analysis.Results:Of the 334 patients(180 HBeAg-positive and 154 HBeAg-negative)randomized and treated,baseline characteristics were similar between groups.The overall mean age was 38 years and 73%were male.The mean HBV DNA was 6.4 log10 IU/mL.The median alanine aminotransferase was 88 U/L,and 37%had a history of antiviral use.At week 144,the proportion with HBV DNA<29 IU/mL was similar among the two groups,with TAF at 83%vs.TDF at 79%,and TAF at 93%vs.TDF at 92%for the HBeAg-positive and-negative patients,respectively.In each study,higher proportions of TAF than TDF patients showed normalized alanine aminotransferase(via the American Association for the Study of Liver Diseases and the China criteria)and showed loss of HBsAg;meanwhile,the HBeAg seroconversion rates were similar.Treatment was well-tolerated among the TAF patients,who showed a smaller median decline in creatinine clearance(−0.4 vs.−3.2 mL/min;p=0.014)and less percentage change in bone mineral density vs.TDF at hip(−0.95%vs.−1.93%)and spine(+0.35%vs.−1.40%).Conclusions:In chronic hepatitis B patients from China,TAF treatment provided efficacy similar to TDF but with better renal and bone safety at 3 years.Jinlin Hou Qin Ning Zhongping Duan You Chen Qing Xie Fu-Sheng Wang Lunli Zhang Shanming Wu Hong Tang Jun Li Feng Lin Yongfeng Yang Guozhong Gong John FFlaherty Anuj Gaggar Shuyuan Mo Cong Cheng Gregory Camus Chengwei Chen Yan Huang Jidong Jia Mingxiang Zhang GS-US-320-0110 and GS-US-320-0108 China Investigators 2021Journal of Clinical and Translational Hepatology2021,9,3:7
3Expression of ipt gene driven by tomato fruit specific promoter and its effects on fruit development of tomato显示文摘Fruit specific promoter (2A12) from Lycopersi-com esculentum and cDNA of isopentenyl-transferase (ipt) from Ti plasmid of Agrobacterium tumerfaciens C58 were cloned by PCR procedure respectively. Two plant expression vectors with 2A12/gus or 2A12/ipt were respectively constructed. These two chimeric genes were transferred into tomato by Agrobacterium mediated procedure. The results of Southern hybridization showed that the fusion genes had been integrated into tomatoes. The result of gus histochemi-cal staining showed that 2A12 had high fruit specific expressive capability in transgenic tomato. The ipt expression resulted in accumulation of high level of cytokinins (CTKs) in fruit lead to developmental changes in fruits and seeds. The fruit of ipt transformed tomato had the hyperplastic placenta with very few seeds or even seedless. The shelf life of transgenic fruits elongated for 1-2 weeks. 4Technology for high performance TRIP steel显示文摘Thermodynamics of martensite transformation in low alloy TRIP steel was evaluated.It follows from the calculation that ε phase might be formed at above room temperature in the low Mn steel.Tested steel was therefore prepared according to the estimation result.High volume fraction of retained austenite(above 20%) containing 1.37%C was obtained in the steel after continuous annealing treatment.Tensile test showed that the ultimate strength of the steel was 1248 MPa,elongation rate 25%,the product of strength and elongation rate 31200 MPa%,reaching the grade of the third generation automobile steel.XRD detection indicated that after tensile test there existed ' martensite while TEM revealed that ε martensite also existed in the steel.Strengthening-toughening of the steel was thus raised due to the effect of transformation induced plasticity of two kinds of martensite.In addition,the phase stability of carbonitrates containing V was estimated and the complex micro-alloying effect was applied to the steel to control the size of austenite(about 1 μm) and enhance the stability of austenite.LI Lin GAO Yi ZHU NaQiong HE YanLin LIU RenDong HE ZhongPing SHI Wen ZHANG Mei 2012Science China(Technological Sciences)2012,55,7:5
5Cloning and expression pattern of a dehydrin-like BDNI gene from drought-tolerant Boea crassifolia Hemsl.显示文摘A 500-bp cDNA fragment was amplified via RT-PCR from drought-induced total RNA of the drought-tolerant B. crassifolia Hemsl. using primers based on the sequence of published dehydrin conserved region. By using 5’ RACE, full-length coding region (1 148 bp) of BDN1 gene was produced. It is a new member of the dehydrin gene family. Southern analysis indicated that BDN1 is present in the 6. crassifolia genome as a single-copy gene. 7Enhanced late blight resistance of transgenic potato expressing glucose oxidase under the control of pathogen-inducible promoter显示文摘To engineer crop disease resistance by utilizing natural defense mechanism that was expressed in the incompatible host-pathogen interactions is expected to result in a durable and broad-spectrum resistance. In order to prove this viewpoint, we amplified the coding region of the glucose oxidase (GO) gene from Aspergillus nigervia PCR and fused it to the pathogen-inducible promoter, Prp1-1. The chimeric gene was cloned into a plant expression vector and conjugated into Agrobacterium. Twenty-three transgenic potato plants were obtained by Agrobacterium-mediated transformation. The integration of GO gene was confirmed by Southern hybridization and the GO gene expression was identified with Kl-starch color reaction. Phytophthora infestans inoculation revealed that the expression of the chimeric transgene was induced by pathogen infection. Most of the transgenic plants exhibited various degrees of enhanced disease resistance. 11Computer Simulation of Fe-Al-Si System Diffusion Couples显示文摘Diffusion couples, Fe-6.8 wt% Al-1.0 wt% Si/Fe and Fe-6.3 wt% Al-0.9 wt% Si/Fe were constructed and separately annealed at 1050°C for 3 h and at 1000°C for 64 h. The concentration profiles of Fe, Al and Si atoms in these couples were measured by electron probe micro-analysis (EPMA), while the diffusion behavior was also simulated by coupling thermodynamic and kinetic properties of Fe-Al-Si system. The simulation results were in good agreement with the measured concentration profiles showing the validity of dynamic parameters of Fe-Al-Si system. Calculation was made for Fe-7 wt% Al-1 wt% Si/Fe diffusion couples at 1000?C with different diffusion time. 20Transcriptome and metabolome analysis reveal that oral secretions from Helicoverpa armigera and Spodoptera litura influence wound-induced host response in cotton显示文摘Cotton(Gossypium hirsutum)is an important fiber crop worldwide.Insect attack causes cotton yield and quality losses.However,little is known about the mechanism of cotton response to insect attack.We simulated insect feeding by applying insect oral secretions(OS)to wounds,and combined transcriptome and metabolome analysis to investigate how OS from two major pest species(Helicoverpa armigera and Spodoptera litura)affect cotton defense responses.We found that respectively 12,668 and 13,379 genes were differentially expressed in comparison with wounding alone.On addition of OS,the jasmonic acid signaling pathway was rapidly and strongly induced,whereas genes involved in salicylic acid biosynthesis were downregulated.On constructing a coexpression gene network,we identified a hub gene encoding a leucine-rich repeat receptor kinase that may play an important role in early signal recognition and transduction.OS from the two insect species altered the abundance of flavonoid-related compounds in different patterns.Gossypol remained in lower concentration after OS application than after wounding alone,suggesting a suppressive effect of OS on cotton defense response.This study illustrated transcriptional and metabolic changes of cotton in responding to OS from two chewing insect species,identified potential key response genes,and revealed evidence for OS inhibition of wounding-induced cotton defense response.Huan Si Hongling Liu Yiwen Sun Zhongping Xu Sijia Liang Bo Li Xiao Ding Jianying Li Qiongqiong Wang Lin Sun Xianlong Zhang Shuangxia Jin 2020The Crop Journal2020,8,6:1
