| 2 | Expression of a New hemA Gene from Agrobacterium radiobacter in Escherichia coli for 5-Aminolevulinate Production显示文摘A new hemA gene encoding 5-aminolevulinate (ALA) synthase was cloned from Agrobacterium ra- diobacter zju-0121. The ALA synthase catalyzes the pyridoxal phosphate-dependent condensation of succinyl coen- zyme A (succinyl-CoA) and glycine to produce ALA. Four plasmids carrying the A, radiobacter hemA gene were transformed into different E. coli strains. The effects of both genetic and physiological factors on the expression of ALA synthase and ALA production were studied. The results indicated that the final intracellular activity of ALA synthase and the production of ALA in different expression systems varied largely. Among them, the recombinant E. coli BL21 (DE3) harboring the expression plasmid pET28-A. R-hemA was the most suitable one. The effects of isopropyl-β-D-thiogalactopyranoside (IPTG) addition time, IPTG concentration, culture temperature and the initial concentration of precursors and glucose on the ALA production were also evaluated. The expressed ALA synthase accounted for about 23.7% of the intracellular soluble protein. The highest specific activity of ALA syn- thase was 13.8nmol·min-1·mg-1 of intracellular soluble protein. | 3 | Dietary energy intake affects fetal survival and development during early and middle pregnancy in Large White and Meishan gilts显示文摘This experiment was designed to determine the effects of variations in dietary energy intake on reproductive performance and gene expression of luteal and endometrium tissues in Large White(LW)and Meishan(MS) gilts during early and middle pregnancy. After insemination, 32 LW gilts were assigned to high and low(HE_Land LE_L, 14.23 and 12.56 MJ DE/kg, respectively) diet treatment groups,while 32 MS gilts were allocated to HE_M and LE_M(12.56 and 10.88 MJ DE/kg) groups. Gilts were slaughtered on days 35, 55 and 90 of gestation. The fetal survival and luteal progesterone(P_4) concentration in the HE_Lgroup were higher on day 35 but lower on day 90 of gestation compared with the LE_L group(P < 0.05) for LW gilts. However, fetal survival and luteal P_4 concentration on day 35 of gestation were greater(P < 0.05) in the LE_M group than in the HE_M group for MS gilts, but no significant difference in mid-gestation was showed. The fetal weights of both breeds were higher for the high energy diets compared with the respective control group on day 90 of gestation(P < 0.05). In addition,the m RNA levels of P_4 synthesis-related proteins had correlated with luteal P_4 concentration in both breeds. Further, endometrial levels of uteroferrin(ACP5), retinol-binding protein 4(RBP_4) and secreted phosphoprotein 1(SPP1) m RNA were upregulated in the HE_Lgroup on day 35 of gestation but ACP5 and SPP1 were downregulated on day 55 of gestation compared with the LE_Lgroup(P < 0.05) for LW gilts. In MS gilts, diet only affected the expression of SPP1(P < 0.05). Our results revealed the differential sensitivity of LW and MS breeds to variations in dietary energy intake. For LW gilts, the HE_Lgroup improved fetal survival on day 35 but a sustained high energy diet decreased fetal survival on day 90 of gestation. The differences in dietary energy intake did not influence fetal survival on day 90 of gestation but the higher energy diet did increase fetal weight in the MS breed compared with the lower energy intake diet. | 11 | The dynamic changes of genes revealed that persistently overexpressed genes drive the evolution of cyflumetofen resistance in Tetranychus cinnabarinus显示文摘Changes in gene expression are associated with the evolution of pesticide resis-tance in arthropods.In this study,transcriptome sequencing was performed in 3 different resistance levels(low,L;medium,M;and high,H)of cyflumetofen-resistant strain(YN-CyR).A total of 1685 genes,including 97 detoxification enzyme genes,were upregulated in all 3 stages,of which 192 genes,including 11 detoxification enzyme genes,showed a continuous increase in expression level with resistance development(L to H).RNA in-terference experiments showed that overexpression of 7 genes(CYP392A1,TcGSTd05,CCE06,CYP389A1,TcGSTz01,CCE59,and CYP389C2)is involved in the development of cyflumetofen resistance in Tetranychus cinnabarinus.The recombinant CYP392A1 can effectively metabolize cyflumetofen,while CCE06 can bind and sequester cyflumetofen in vitro.We compared 2 methods for rapid screening of resistance molecular markers,in-cluding short-term induction and 1-time high-dose selection.Two detoxification enzyme genes were upregulated in the field susceptible strain(YN-S)by induction with 20%lethal concentration(LC2o)of cyflumetofen.However,16 detoxification enzyme genes were up-regulated by 1-time selection with LCso of cyflumetofen.Interestingly,the 16 genes were overexpressed in all 3 resistance stages.These results indicated that 1685 genes that were upregulated at the L stage constituted the basis of cyflumetofen resistance,of which 192 genes in which upregulation continued to increase were the main driving force for the de-velopment of resistance.Moreover,the 1-time high-dose selection is an efficient way to rapidly obtain the resistance-related genes that can aid in the development of resistance markers and resistance management in mites. | Kaiyang Feng Jialu Liu Mingyu Zhao Zhixin Jiang Peilin Liu Peng Wei Wei Dou Lin He | 2023 | Insect Science2023,30,4: | 0 |
