1Restorative effect and mechanism of mecobalamin on sciatic nerve crush injury in mice显示文摘Mecobalamin,a form of vitamin B12 containing a central metal element(cobalt),is one of the most important mediators of nervous system function.In the clinic,it is often used to accelerate recovery of peripheral nerves,but its molecular mechanism remains unclear.In the present study,we performed sciatic nerve crush injury in mice,followed by daily intraperitoneal administration of mecobalamin(65 μg/kg or 130 μg/kg) or saline(negative control).Walking track analysis,histomorphological examination,and quantitative real-time PCR showed that mecobalamin significantly improved functional recovery of the sciatic nerve,thickened the myelin sheath in myelinated nerve fibers,and increased the cross-sectional area of target muscle cells.Furthermore,mecobalamin upregulated m RNA expression of growth associated protein 43 in nerve tissue ipsilateral to the injury,and of neurotrophic factors(nerve growth factor,brain-derived nerve growth factor and ciliary neurotrophic factor) in the L4–6 dorsal root ganglia.Our findings indicate that the molecular mechanism underlying the therapeutic effect of mecobalamin after sciatic nerve injury involves the upregulation of multiple neurotrophic factor genes.Lin Gan Minquan Qian Keqin Shi Gang Chen Yanglin Gu Wei Du Guoxing Zhu 2014Neural Regeneration Research2014,9,22:11
2TMEM43-S358L mutation enhances NF-κBTGFp signal cascade in arrhythmogenic right ventricular dysplasia/cardiomyopathy显示文摘Arrhythmogenic right ventricular dysplasia/cardiomyopathy(ARVD/C)is a genetic cardiac muscle disease that accounts for approximately 30%sudden cardiac death in young adults.The Ser358Leu mutation of transmembrane protein 43(TMEM43)was commonly identified in the patients of highly lethal and fully penetrant ARVD subtype,ARVD5.Here,we generated TMEM43 S358L mouse to explore the underlying mechanism.This mouse strain showed the classic patholo.gies of ARVD patients,including structural abnormalities and cardiac fibrofatty.TMEM43 S358L mutation led to hyper-activated nuclear factor kB(NFkB)activation in heart tissues and primary cardiomy.ocyte cells.Importantly,this hyper activation of NF-κB directly drove the expression of pro-fibrotic gene,transforming growth factor beta(TGFβ),and enhanced downstream signal,indicating that TMEM43 S358L mutation up-regulates NF-κB-TGFβ signal cascade during ARVD cardiac fibrosis.Our study partially reveals the regulatory mechanism of ARVD development.Guoxing Zheng Changying Jiang Yulin Li Dandan Yang Youcai Ma Bing Zhang Xuan Li Pei Zhang Xiaoyu Hu Xueqiang Zhao Jie Du Xin Lin 2019Protein & Cell2019,10,2:5

19Proteomic analysis of a chronic obstructive pulmonary disease mouse model to determine the efficacy of treatment using Guben Zhike decoction显示文摘Background:Guben Zhike decoction(GBZKD)is derived from the experience of Professor Enxiang Chao,an esteemed master of Chinese medicine,while treating chronic obstructive pulmonary disease(COPD).GBZKD reinforces the healthy qi and consolidates defensive qi.This study explored the efficacy and potential mechanism of action of GBZKD in a COPD mouse model using proteomics.Methods:A COPD mouse model was established through cigarette smoke exposure and intranasal lipopolysaccharide administration.The model was verified through lung function test and lung histopathological observation.Label-free quantitative proteomics was used to detect the lung tissue proteins of mice from the GBZKD,COPD,and control groups.Results:GBZKD markedly improved the lung function and associated pathological conditions in the COPD mouse model.Proteomic analysis identified 4316 proteins,of which 3696 were quantitative proteins.We highlighted 287 and 184 proteins with significant regulatory roles in the lung tissues of COPD mice and GBZKD-treated mice,respectively.These proteins participated in multiple functions,including complement/coagulation cascade,immune response,and metabolic pathways.Conclusion:GBZKD exhibits multitarget and multipathway therapeutic effects in a COPD mouse model.Mingzhe Wang Guoxing Liu Yao Xiao Zhe Cai Chang Liu Lin Pan Ying Liu Mengchao Liu Hongchun Zhang 2021Journal of Traditional Chinese Medical Sciences2021,8,1:0
20Influence of cerium and yttrium addition on strength and electrical conductivity of pure aluminum alloys显示文摘To develop pure aluminum alloys with high conductivity and strength, Al-0.2Ce and Al-0.2Ce-0.1Y alloys were prepared by rolling and annealing processes in this study. The effects of trace rare earth elements on the strength and electrical conductivity of the pure aluminum alloys were investigated. It is revealed that the addition of Ce and Y to pure aluminum can effectively enhance the strength and electrical conductivity of the alloys. In Al-0.2Ce, the addition of Ce can effectively refine the grain size of a-Al, with an average grain size of 90.68 μm in the as-cast state. The grain size of the alloy is further reduced to 87.55 μm by the simultaneous addition of Y. The synergistic addition of Ce and Y changes the properties of the alloy. The addition of Ce and Y also produces the Al_(11)Ce_(3) and Al_(3)Y second phases, which have coherent relationship with a-Al. The two-dimensional mismatch degree was calculated to be only 4.43%and 0.85%, respectively, which can provide a certain amount of nucleation substrate for a-Al in the incubation stage. The interfacial match between the L1_(2)structure of Al_(3)Y and a-Al was calculated using first-principles simulations. The results indicate that Al_(3)Y has a strong bonding effect with a-Al.
