1The cloning of Dmc1 cDNAs and a comparative study of its expression in different ploidy cyprinid fishes显示文摘Dmc1 (disrupted meiotic cDNA) is a functionally specific gene, which was firstly discovered in yeast and then found to encode a protein required for homologous chromosome synapsis during the process of meiosis. In this investigation, we cloned the partial cDNAs of Dmc1 of diploid red crucian carp, Japanese crucian carp, common carp, triploid crucian carp and allotetraploid hybrids by using a pair of degenerate primers based on the conservative sequence of amino acids of the DMC1 protein in yeast, mouse and human. The full length cDNAs were then obtained by rapid amplification of cDNA ends (RACE). Our data showed that the full length cDNAs of Dmc1 in the three diploid fishes are all 1375 bp long, while it is 1383 bp long in triploids and 1379 bp long in allotetraploids. And despite of the variation in length, all the cDNAs encode a protein of 342 amino acids. A high homology of 97.3% of the DMC1 protein can be drawn by comparing the amino acid sequences in the three diploids, which is also of 86%, 86% and 95% similarity to human, mouse and zebrafish, respectively. A comparative study of the expression pattern of Dmc1 was carried out by RT-PCR using specific primers against the same se-quences of coding regions in different ploidy cyprinid fishes, from which it was showed that Dmc1 was expressed only in gonads of these five kinds of fishes. The expression pattern of Dmc1 in both ovaries and testes from different ploidy fishes within breeding season was also studied by Real-time PCR, and the results showed that the expression of this gene was greatly different among the three different ploidy fishes, which was the highest of triploid and lowest of allotetraploids. The histological sections data showed matured gonads of both diploid red crucian carp and allotetraploids in breeding season, although the latter demonstrated a higher maturation, and no gonadal maturation could be observed in triploids. In conclusion, we suggest that Dmc1 is specifically expressed in the period of meiosis in all the ploidy cyprinid fishes and directly related with the development of gonad in a manner of ploidy-independent way. 2Evolutionary analysis of allotetraploid hybrids of red crucian carp×common carp,based on ISSR,AFLP molecular markers and cloning of cyclins genes显示文摘The allotetraploid hybrids of red crucian carp × common carp are the first reported artificially cultured polyploid fish with bisexual fertility and stable inheritance in vertebrate. Using ISSR and AFLP markers and the cyclins genes, the genomes and cyclin gene sequence changes were analyzed between the allotetraploid hybrids and their parents. The results indicated that the allotetraploids inherited many genetic characteristics from their parents and the genetic characteristics were stable after 15 generations. However, the allotetraploids had a closer genetic relationship with their original female parents and represented a bias toward the maternal progenitor. DNA fingerprinting analysis showed that the allotetraploids had undergone sequences deletion from their original parents and that the deleted sequences were mostly from the male parent's genome. Some non-parental bands were found in the allotetraploid hybrids. Sequences analysis of the cyclin A1 and B1 genes showed nonsynonymous substitutions of single nucleotides in codons that were different from their original parents, leading to non-parental amino acid loci. 3Influence of wave front healing on seismic tomography显示文摘Wave front healing is a common natural phenomenon.To further investigate wave front healing,we simulated wave propagation in a spherical anomaly surrounded by homogeneous media using a high-order finite difference solution of the acoustic equation.Furthermore,we analyzed the characteristics of the wave propagation in the anomaly,and found that they are related to the dominant frequency of the seismic wave and the dimensions of the anomaly.Through quantitative comparison of the wave front energy of the diffracted wave and transmitted wave,we summarized the influences of the wave front healing on seismic tomography.We conclude that,under the strong scattering condition,only positive anomalies can be inverted by ray-based tomography,only large anomalies can be inverted by finite-frequency tomography,and small negative anomalies cannot be inverted by any first-arrival traveltime tomographic methods.These conclusions are verified by tomographic experiments based on different theoretical models.Finally,we propose that more information besides the first-arrival traveltime should be used to invert the high wave number components of the media.Besides the above acquisitions of wave front healing on seismic tomography,we explain the banana-doughnut phenomena,and offer a new insight into the wave scattering,which should be important for better understanding the wave propagation and seismic inversion.Liu YuZhu Dong LiangGuo 2012Science China Earth Sciences2012,55,11:3
4Cloning of a novel gene associated with human nasopharyngeal carcinoma显示文摘One EST N27741 with high expression in normal adult nasopharynx tissues but low expression in adult poorly differentiated squamous nasopharyngeal carcinoma has been selected out by the high-density cDNA array expression profiling technique. The differential expression has been confirmed by RT-PCR. One novel gene of 1096 bp has been cloned based on this EST. Bioinformatics analysis found that the new gene sequence contains a whole reading frame encoding 256 amino acids. There is a stop codon TAA in front of the 5’ end start codon, and a tailing signal AATAAA and poly A tail at the 3’ end. There is no homologous known gene found after searching by blasting this sequence to non-redundancy nucleotide database. 5Recombinant soluble TRAIL induces apoptosis of cancer cells显示文摘TRAIL is a tumor necrosis factor family member that selectively induces apoptosis of cancer cells but not of normal cells. To develop TRAIL into a potential cancer drug, three different sizes of soluble TRAIL fragments, including sTRAIL(74 - 281), sTRAIL(95 - 281) and sTRAIL(101-281), were expressed in E. coli and purified to homogeneity. Apoptosis assays indicated that sTRAIL(95-281) and sTRAIL(101-281), but not sTRAIL(74-281), can potently induce apoptosis of various cancer cell lines in 6 h, suggesting that the N-terminal fragment of aa101 has inhibitory effect on TRAIL-induced apoptosis. 6abrication of CDs hybrid MIL-68(In) derived In2O_(3)eIn_(2)S_(3) hollow tubular heterojunction and their exceptional self-powered PEC aptasensing properties for ampicillin detecting显示文摘A visible-light-driven self-powered photoelectrochemical(PEC)aptasensor was developed for ampicillin(AMP)detecting based on carbon dots(CDs)hybrid MIL-68(In)-derived In2O_(3)eIn_(2)S_(3) hollow tubular heterojunction(CDs/In2O_(3)eIn_(2)S_(3)).In_(2)S_(3) nanosheets uniformly grew in-situ on the surface of In2O_(3) hollow tubes,forming a close contact heterogeneous interface,which significantly promoted the transfer and separation of photo-generated carriers,and provided a large specific surface area and rich active sites for the PEC aptasensing platform.Furthermore,the CDs/In2O_(3)eIn_(2)S_(3)/ITO electrode,which was obtained by dipping assembly,showed remarkable and stable photoelectric signals at zero-bias under visible light irradiation.The amino-functionalized AMP aptamer was fixed on the working electrode as a biological recognition element,and the concentration of AMP was determined by observing the change in photocurrent intensity caused by the specific capture of AMP molecules in solution.Under optimized conditions,the developed PEC aptasensor displayed a relatively wide linear range from 0.001 ng mL^(-1) to 300 ng mL^(-1),as well as a low detection limit(LOD)of 0.06 pg mL^(-1).Besides,the novel self-powered PEC AMP-aptasensor exhibited excellent reproducibility,good stability and selectivity,which open a potential avenue for antibiotic residues detection in environmental media.Tao Yan Yixuan Feng Xiang Ren Jinkai Li Yizhong Lu Meng Sun Liangguo Yan Qin Wei Huangxian Ju 2021Journal of Materiomics2021,7,4:2
7Profiling gene expression patterns of nasopharyngeal carcinoma and normal nasopharynx tissues with cDNA microarray显示文摘5 μg of total RNAs from normal nasopharynx and nasopharyngeal carcinoma tissue have been labeled with α-32P-dCTP during reverse transcription. The synthesized cDNA probes have been hybridized to high-density cDNA microarray containing 5184 genes or expression sequence tags (ESTs). Then image analysis software has been applied to comparing their expression profiles. Results show that 187 ESTs were of density value above 200 in nasopharyngeal carcinoma tissue while there were 307 such ESTs in normal nasopharynx tissue; 38 ESTs were strongly expressed in nasopharynx, but weakly expressed in nasopharyngeal carcinoma; 48 ESTs were strongly expressed in nasopharyngeal carcinoma, but weakly expressed in normal nasopharynx. These results suggest that there may exist some new differentially expressed genes involved in nasopharyngeal carcinoma development. 20Highly effective visible-photocatalytic hydrogen evolution and simultaneous organic pollutant degradation over an urchin-like oxygen-doped MoS_(2)/ZnIn_(2)S_(4)composite显示文摘Achieving hydrogen production and simultaneous decomposition of organic pollutants through dual-functional photocatalytic reactions has received increasing attention due to the environmentally friendly and cost-effective characteristics of this approach.In this work,an urchin-like oxygen-doped MoS_(2)/ZnIn_(2)S_(4)(OMS/ZIS)composite was fabricated for the first time using a simple solvothermal method.The unique microstructure with abundant active sites and fast charge transfer channels further shortened the charge migration distance and compressed carrier recombination.The obtained composite exhibited an efficient H2 evolution reaction rate of 12.8 mmol/g/h under visible light,which was nearly times higher than pristine ZnIn_(2)S_(4),and the apparent quantum efficiency was 14.9%(420 nm).The results of the simultaneous photocatalytic H2 evolution and organic pollutant decomposition test were satisfactory,resulting in decomposition efficiencies of resorcinol,tetracycline,and bisphenol A that reached 41.5%,63.5%,and 53.0%after 4 h,respectively,and the highest H2 evolution rate was 672.7 umol/g/h for bisphenol A.Furthermore,natural organic matter(NOM)abundantly found in actual water was adopted as an electron donor for H production under simulated sunlight irradiation,indicating the promising practicability of simultaneous hydrogen evolution and NOM decomposition.Moreover,the mechanisms of the dual-purpose photocatalytic reactions,as well as the synergistic effect between the molecular structures of the organic pollutants and the corresponding adsorption behavior on the photocatalyst surface were illustrated in detail.These obtained results may serve as an inspiration for the rational design of highly efficient,dual-functional photocatalysts in the future.Tao Yan Qianqian Yang Rui Feng Xiang Ren Yanxia Zhao Meng Sun Liangguo Yan Qin Wei 2022Frontiers of Environmental Science & Engineering2022,16,10:1
