2Reproductive management through integration of PGD and MPS-based noninvasive prenatal screening/diagnosis for a family with GJB2-associated hearing impairment显示文摘A couple with a proband child of GJB2(encoding the gap junction protein connexin 26)-associated hearing impairment and a previous pregnancy miscarriage sought for a reproductive solution to bear a healthy child. Our study aimed to develop a customized preconception-to-neonate care trajectory to fulfill this clinical demand by integrating preimplantation genetic diagnosis(PGD), noninvasive prenatal testing(NIPT), and noninvasive prenatal diagnosis(NIPD) into the strategy. Auditory and genetic diagnosis of the proband child was carried out to identify the disease causative mutations. The couple then received in-vitro-fertilization treatment, and eight embryos were obtained for day 5 biopsy. PGD was performed by short-tandem-repeat linkage analysis and Sanger sequencing of GJB2 gene. Transfer of a GJB2 c.235del C heterozygous embryo resulted in a singleton pregnancy. At the 13 th week of gestation, genomic DNA(g DNA) from the trio family and cell-free DNA(cf DNA) from maternal plasma were obtained for assessment of fetal chromosomal aneuploidy and GJB2 mutations. NIPT and NIPD showed the absence of chromosomal aneuploidy and GJB2-associated disease in the fetus, which was later confirmed by invasive procedures and postnatal genetic/auditory diagnosis. 4Effect of nitrogen fertilizer on distribution of starch granules in different regions of wheat endosperm显示文摘This study provided visual evidence of a nitrogen effect on starch granules(SGs) in wheat endosperm. Winter wheat(Titicum aestivum L.) cultivar Xumai 30 was cultured under no nitrogen(control) and 240 kg ha-1of nitrogen applied at the booting stage. The number, morphology, and size of Aand B-type SGs in subaleurone of dorsal endosperm(SDE), center of dorsal endosperm(CDE), modified aleurone(MA), subaleurone of ventral endosperm(SVE), and center of ventral endosperm(CVE) were observed under light and electron microscopes.(1) The distribution of SGs in SDE was similar to that in SVE, the distributions of SGs in CDE and CVE were similar, but the distribution of SGs in MA was different from those in the other four endosperm regions. The number of SGs in the five endosperm regions was in the order SDE > CDE > SVE > CVE > MA.(2) Nitrogen increased the number of Aand B-type SGs in SDE and SVE. Nitrogen also increased the number of B-type SGs but decreased the number of A-type SGs in CDE and CVE. Nitrogen decreased the numbers of A-type and B-type SGs in MA. 5Effect of Banxia Xiexin decoction on Helicobacter pylori-related peptic ulcers and its possible mechanism via the TGF-β/Smad signaling pathway显示文摘OBJECTIVE: To investigate the effect of Banxia Xiexin decoction(BXD) on Helicobacter pylori(Hp)-related peptic ulcers(PUs) and the possible mecha-nism underlying BXD actions via the transforming growth factor-β/small mothers against decapentaplegic(TGF-β/Smad) signaling pathway.METHODS: PU patients with cold-heat complex syndrome were randomly assigned to groups that received Chinese or Western medicines with 20 patients in each group. Serum was collected after 7 d of treatment. The healthy group included 20 individuals. Gastric mucosal epithelial cell line GES-1 was cultured in vitro and randomly divided into the following seven groups: control, model, healthy,Western Medicine, prior treatment, low dosage,and high dosage. After 72 h of treatment with the corresponding serum, the m RNA and protein expression levels of TGF-β1, Smad3, and Smad7 were measured by reverse transcription quantitative polymerase chain reaction and western blotting, respectively.RESULTS: The m RNA expression levels of TGF-β1 and Smad3 in GES-1 cells were increased after Hp introduction, and these increased levels were reduced by the BXD-containing serum. The protein levels of p-Smad3, but not TGF-β1 or Smad3, were significantly increased in Hp-treated GES-1 cells,and treatment with the BXD-containing serum markedly decreased the protein levels. 6Dissipative particle dynamics thermostat: a novel thermostat for molecular dynamics simulation of liquid crystals with Gay–Berne potential显示文摘The Gay–Berne(GB) model has been proved to be highly successful in the simulation of liquid crystal phases via both molecular dynamics(MD) and nonequilibrium molecular dynamics(NEMD). However, the conventional thermostats used in the simulations of GB systems, such as Nosé–Hoover and Langevin thermostats, have serious shortcomings especially in NEMD simulations. Recently, dissipative particle dynamics(DPD) has established itself as a useful thermostat for soft matter simulations, whereas the application of DPD thermostat in(NE)MD simulations is limited to the spherically isotropic potential models, such as the Lennard–Jones model. Considering the virtues of the DPD thermostat, that is, local, momentum conserved, and Galilean invariant, we extend the DPD thermostat to the non-spherical GB model. It is interesting to find that the translational DPD and rotational DPD thermostats can be used in the GB system independently and both can achieve the thermostatting effects. Also, we compared the performance of the DPD thermostat with other commonly used thermostats in NEMD simulations by investigating the streaming velocity profiles and the dynamics of phase separation in a typical but simple binary GB mixture under shear field. It is revealed that the known virtues of DPD thermostats, such as Galilean invariant, shear velocity profile-unbiased, and unscreened hydrodynamic interactions, are still intact when applying to GB systems. 7The disbalance of LRP1 and SIRPα by psychological stress dampens the clearance of tumor cells by macrophages显示文摘The relationship between chronic psychological stress and tumorigenesis has been well defined in epidemiological studies;however, the underlying mechanism remains underexplored. In this study, we discovered that impaired macrophage phagocytosis contributed to the psychological stressevoked tumor susceptibility, and the stress hormone glucocorticoid(GC) was identified as a principal detrimental factor. Mechanistically, GC disturbed the balance of the 'eat me' signal receptor(low-density lipoprotein receptor-related protein-1, LRP1) and the 'don’t eat me' signal receptor(signal regulatory protein alpha, SIRPa). Further analysis revealed that GC led to a direct, glucocorticoid receptor(GR)-dependent trans-repression of LRP1 expression, and the repressed LRP1, in turn, resulted in the elevatedgene level of SIRPa by down-regulating mi RNA-4695-3 p. These data collectively demonstrate that stress induces the imbalance of the LRP1/SIRPa axis and entails the disturbance of tumor cell clearance by macrophages. 8Network pharmacology analysis to explore mechanism of Three Flower Tea against nonalcoholic fatty liver disease with experimental support using high-fat diet-induced rats显示文摘Objective:Nonalcoholic fatty liver disease(NAFLD)has become a common chronic liver disease that is harmful to human health.Moreover,there is currently no FDA-approved first-line drug for the treatment of nonalcoholic steatohepatitis(NASH)or NAFLD.Traditional Chinese medicine(TCM)is widely used to ameliorate liver diseases,such as the traditional ancient recipe called Three Flower Tea(TFT),which consists of double rose(Rosa rugosa),white chrysanthemum(Chrysanthemum morifolium),and Daidaihua(Citrus aurantium).However,the mechanisms of the action of TFT are not clear.Therefore,this study aimed to elucidate the mechanisms of TFT against NAFLD in high-fat diet(HFD)-induced rats.Methods:This study utilized bioinformatics and network pharmacology to establish the active and potential ingredient-target networks of TFT.Furthermore,a protein–protein interaction(PPI)network was constructed,and enrichment analysis was performed to determine the key targets of TFT against NAFLD.Furthermore,an animal experiment was conducted to evaluate the therapeutic effect and confirm the key targets of TFT against NAFLD.Results:A total of 576 NAFLD-related genes were searched in Gene Cards,and under the screening criteria of oral bioavailability(OB)≥30%and drug-likeness(DL)≥0.18,a total of 19 active ingredients and 210 targets were identified in TFT.Network pharmacology analysis suggested that 55 matching targets in PPIs were closely associated with roles for NAFLD treatment.Through the evaluation of network topology parameters,four key central genes,PPARγ,SREBP,AKT,and RELA,were identified.Furthermore,animal experiments indicated that TFT could reduce plasma lipid profiles,hepatic lipid profiles and hepatic fat accumulation,improve liver function,suppress inflammatory factors,and reduce oxidative stress.Through immunoblotting and immunofluorescence analysis,PPARγ,SREBP,AKT,and RELA were confirmed as targets of TFT in HFD-induced rats.Conclusion:In summary,our results indicate that TFT can prevent and treat NAFLD via multiple targets,including lipid accumulation,antioxidation,insulin sensitivity,and inflammation.Peixuan Wu Shufei Liang Yanping He Rui Lv Bendong Yang Meng Wang Chao Wang Yong Li Xinhua Song Wenlong Sun 2022Chinese Herbal Medicines2022,14,2:2
9Microglia activation mediated by toll-like receptor-4 impairs brain white matter tracts in rats显示文摘Microglia activation and white matter injury coexist after repeated episodes of mild brain trauma and ischemic stroke. Axon degeneration and demyelination can activate microglia; however, it is unclear whether early microglia activation can impair the function of white matter tracts and lead to injury. Rat corpus callosum(CC) slices were treated with lipopolysaccharide(LPS) or LPS + Rhodobacter sphaeroides(RS)-LPS that is a toll-like receptor 4(TLR-4) antagonist. Functional changes reflected by the change of axon compound action potentials(CAPs) and the accumulation of β-amyloid precursor protein(β-APP) in CC nerve fibers. Microglia activation was monitored by ionized calcium binding adaptor-1 immunofluorescent stain, based on well-established morphological criteria and paralleled proportional area measurement. Input-output(I/O) curves of CAPs in response to increased stimuli were significantly downshifted in a dose-dependent manner in LPS(0.2, 0.5 and 1.0μg/mL)-treated slices, implying that axons neurophysiological function was undermined. LPS caused significant β-APP accumulation in CC tissues,reflecting the deterioration of fast axon transport. LPS-induced I/O curve downshift and P-APP accumulation were significantly reversed by the pre-treatment or co-incubation with RS-LPS. RS-LPS alone did not change the I/O curve.The degree of malfunction was correlated with microglia activation, as was shown by the measurements of proportional areas. 10Hyperactive PI3Kδ predisposes naive T cells to activation via aerobic glycolysis programs显示文摘Activated phosphoinositide 3-kinaseδsyndrome(APDS)is an autosomal-dominant combined immunodeficiency disorder resulting from pathogenic gain-of-function(GOF)mutations in the PIK3CD gene.Patients with APDS display abnormal T cell homeostasis.However,the mechanisms by which PIK3CD GOF contributes to this feature remain unknown.Here,with a cohort of children with PIK3CD GOF mutations from multiple regions of China and a corresponding CRISPR/Cas9 gene-edited mouse model,we reported that hyperactive PI3Kδdisrupted TNaive cell homeostasis in the periphery by intrinsically promoting the growth,proliferation,and activation of TNaive cells.Our results showed that PIK3CD GOF resulted in loss of the quiescence-associated gene expression profile in naive T cells and promoted naive T cells to overgrow,hyperproliferate and acquire an activated functional status.Naive PIK3CD GOF T cells exhibited an enhanced glycolytic capacity and reduced mitochondrial respiration in the resting or activated state.Blocking glycolysis abrogated the abnormal splenic T cell pool and reversed the overactivated phenotype induced by PIK3CD GOF in vivo and in vitro.These results suggest that enhanced aerobic glycolysis is required for PIK3CD GOF-induced overactivation of naive T cells and provide a potential therapeutic approach for targeting glycolysis to treat patients with APDS as well as other immune disorders.Yanjun Jia Qiuyun Yang Yanping Wang Wenyan Li Xuemei Chen Tao Xu Zhirui Tian Minxuan Feng Liang Zhang Wenjing Tang Na Tian Lina Zhou Wenxia Song Xiaodong Zhao 2021Cellular & Molecular Immunology2021,18,7:2

The reproductive performance of sows is different due to different techniques adopted in the production of pig farms, which determines the utilization rate of sows and the number of sows in the pig farms. The article analyzed the reproductive performance of sows from the technical point of view, so that the utilization rate of sows can be paid enough attention in the production of pig farms.It is suggested that the technical training should be strengthened in pig farms to improve the corresponding technical level. 14Fusing Carbocycles of Inequivalent Ring Size to a Bis(imino)pyridine-Iron Ethylene Polymerization Catalyst: Distinctive Effects on Activity, PE Molecular Weight, and Dispersity显示文摘The 4,6-bis(arylimino)-1,2,3,7,8,9,10-heptahydrocyclohepta[b]quinoline-iron(II)chlorides(aryl=2,6-Me_(2)C_(6)H_(3) Fe1;2,6-Et_(2)C_(6)H_(3) Fe_(2);2,6-i-Pr_(2)C_(6)H_(3) Fe_(3);2,4,6-Me3C_(6)H_(2) Fe4;and 2,6-Et_(2)-4-Me_(2)C_(6)H_(2) Fe_(5))have been prepared in good yield by a straightforward one-pot reaction of 2,3,7,8,9,10-hexahydro-1H-cyclohepta[b]quinoline-4,6-dione,FeCl_(2)·4H_(2)O,and the appropriate aniline in acetic acid.All ferrous complexes have been characterized by elemental analysis and FT-IR spectroscopy.In addition,the structure of Fe_(3) has been determined by single crystal X-ray diffraction,which showed the iron center to adopt a distorted square pyramidal geometry with the saturated sections of the fused six-and sevenmembered carbocycles to be cis-configured.In combination with either MAO or MMAO,Fe1–Fe5 exhibited exceptionally high activities for ethylene polymerization(up to 15:86×10^(6) g(PE)mol^(−1)(Fe)h^(−1) at 40℃(MMAO)and 9:60×10^(6) g(PE)mol^(−1)(Fe)h^(−1) at 60℃(MAO))and produced highly linear polyethylene(HLPE,Tm≥128℃)with a wide range in molecular weights;in general,the MMAO-promoted polymerizations were more active.Irrespective of the cocatalyst employed,the 2,6-Me_(2)-substituted Fe1 and Fe4 proved the most active while the more sterically hindered 2,6-i-Pr_(2) Fe_(3) the least but afforded the highest molecular weight polyethylene(Mw:65.6–72.6 kg mol^(-1)).Multinuclear NMR spectroscopic analysis of the polymer formed using Fe4/MMAO at 40℃ showed a preference for fully saturated chain ends with a broad bimodal distribution a feature of the GPC trace(Mw/Mn=13:4).By contrast,using Fe4/MAO at 60℃ a vinyl-terminated polymer of lower molecular weight(Mw=14:2 kg mol^(−1))was identified that exhibited a unimodal distribution(Mw/Mn=3:8).Moreover,the amount of aluminoxane cocatalyst employed,temperature,and run time were also found to be influential on the modality of the polymer.Zheng Wang Gregory A.Solan Yanping Ma Qingbin Liu Tongling Liang Wen-Hua Sun 2019Research2019,,1:1

20Trends of SARS-CoV-2 Infection in Rural Area in Sentinel Community-Based Surveillance——China,December 2022 to January 2023显示文摘Introduction:In late 2022,a rapid transmission of Omicron variants of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)occurred throughout China.The purpose of this study was to provide the latest data and evaluate trends of SARS-CoV-2 infection in rural China among the community population.Methods:Data on SARS-CoV-2 infection among approximately 90,000 participants in rural China were collected by the National Sentinel Community-Based Surveillance(NSCS)system.Participants were tested for SARS-CoV-2 infection(defined as positive for SARS-CoV-2 nucleic acid or antigen)twice weekly from December 16,2022 to January 12,2023.The daily average of newly positive rate and its estimated daily percentage change were calculated to describe the national and regional trends of SARS-CoV-2 infection in rural China.Results:In rural China,the daily average new positive rate of SARS-CoV-2 infection peaked at 4.79%between December 20–22,2022 and then decreased to 0.57%between January 10–12,2023,with an average decrease of 29.95%per round.The peak of new SARS-CoV-2 infection was slightly earlier and lower in North China(5.28%between December 20–22,2022)than in South China(5.63%between December 23–26,2022),and then converged from December 30,2022 to January 2,2023.The peak of 6.09%occurred between December 20–22,2022 in eastern China,while the peak of 5.99%occurred later,between December 27–29,2022,in central China.Conclusions:Overall,the epidemic wave in rural China peaked between December 20–22,2022,and passed quickly following the optimization of prevention and control measures.Currently,SARSCoV-2 infection in community populations in rural China is sporadic.Wenzhan Jing Fan Ding Yanping Zhang Guoqing Shi Jue Liu Yu Wu Fangyuan Li Wenxin Yan Chenyuan Qin Min Liu Qun Li Wannian Liang 2023China CDC weekly2023,5,11:1
