| 1 | Non-uniform distribution of gas-solid flow through six parallel cyclones in a CFB system: An experimental study显示文摘In large-scale circulating fluidized bed (CFB) boilers, it is common to use multiple cyclones in parallel for the capture of solids, assuming that gas-solid flow to be the same in the cyclones. This article presents a study investigating gas-solid flow through six parallel cyclones in a CFB cold test rig. The six cyclones were located asymmetrically on the left and right walls of the riser. Solid volume fraction and particle velocity profiles at the riser outlets and in the horizontal ducts were measured using a fiber optical probe. Cyclone pressure drop and solid circulating rate were measured for each individual cyclone. Measurements showed good agreement as to the non-uniform distribution of the gas-solid flow, which occurred mainly across the three cyclones on one side: the middle cyclones on both sides had higher particle velocities. Conversely, the solid volume fractions, solid fluxes and solid circulating rates of the middle cyclones were lower than those of the other four cyclones. The apparent reason for the flow non-uniformity among the cyclones is the significant flow non-uniformity at the riser outlets. Under typical operating conditions, the solid volume fractions at the riser outlets had a deviation of up to 26% whereas the solid circulating rates at the stand pipes, 7%. | 2 | Characterizing and annotating the genome using RNA-seq data显示文摘Bioinformatics methods for various RNA-seq data analyses are in fast evolution with the improvement of sequencing technologies. However, many challenges still exist in how to efficiently process the RNA-seq data to obtain accurate and comprehensive results. Here we reviewed the strategies for improving diverse transcriptomic studies and the annotation of genetic variants based on RNA-seq data. Mapping RNA-seq reads to the genome and transcriptome represent two distinct methods for quantifying the expression of genes/transcripts. Besides the known genes annotated in current databases, many novel genes/transcripts(especially those long noncoding RNAs) still can be identified on the reference genome using RNA-seq. Moreover, owing to the incompleteness of current reference genomes, some novel genes are missing from them. Genome-guided and de novo transcriptome reconstruction are two effective and complementary strategies for identifying those novel genes/transcripts on or beyond the reference genome. | 4 | A comprehensive CFD combustion model for supercritical CFB boilers显示文摘A combustion model of a large-scale supercritical circulati ng fluidized bed (CFB) boiler was developed for comprehensive computational-fluid-dynamics analysis. The model incorporates gas-solid hydrodynamics, coal combustion, heat transfer on heat exchange surfaces in the furnace, and heat transfer between fumace and working medium in the heat transfer tubes. In simulating the dense and dilute phases in the fumace, the gas-solid hydrodynamics is based on the Euler-Euler model and energy-minimization multiscale drag model. Coal combustion entails evaporation, devolatilization, char combustion, gas homoge neous reaction, and pollutant emission. The coefficient ofheat transfer between gas-solid and the waterwall is estimated using the cluster renewal model, and for radiation, the discrete ordinate model is used. Moreover, thermohydraulic processes in the membrane wall are also in eluded in the heat transfer process. The model was successfully applied in simulations of a 350-MW supercritical CFB boiler. | 5 | mRNA enrichment protocols determine the quantification characteristics of external RNA spike-in controls in RNA-Seq studies显示文摘RNA-Seq promises to be used in clinical settings as a gene-expression profiling tool;however,questions about its variability and biases remain and need to be addressed.Thus,RNA controls with known concentrations and sequence identities originally developed by the External RNA Control Consortium(ERCC) for microarray and qPCR platforms have recently been proposed for RNA-Seq platforms,but only with a limited number of samples.In this study,we report our analysis of RNA-Seq data from 92 ERCC controls spiked in a diverse collection of 447 RNA samples from eight ongoing studies involving five species(human,rat,mouse,chicken,and Schistosoma japonicum) and two mRNA enrichment protocols,i.e.,poly(A) and RiboZero.The entire collection of datasets consisted of 15650143175 short sequence reads,131603796(i.e.,0.84%) of which were mapped to the 92 ERCC references.The overall ERCC mapping ratio of 0.84% is close to the expected value of 1.0% when assuming a 2.0% mRNA fraction in total RNA,but showed a difference of 2.8-fold across studies and 4.3-fold among samples from the same study with one tissue type.This level of fluctuation may prevent the ERCC controls from being used for cross-sample normalization in RNA-Seq.Furthermore,we observed striking biases of quantification between poly(A) and RiboZero which are transcript-specific.For example,ERCC-00116 showed a 7.3-fold under-enrichment in poly(A) compared to RiboZero.Extra care is needed in integrative analysis of multiple datasets and technical artifacts of protocol differences should not be taken as true biological findings. | QING Tao YU Ying DU TingTing SHI LeMing | 2013 | Science China(Life Sciences)2013,56,2: | 4 |
| 6 | Predictions of NOx/N2O emissions from an ultra-supercritical CFB boiler using a 2-D comprehensive CFD combustion model显示文摘NOx and N2O emissions from an ultra-supercritical circulating fluidized bed(CFB)boiler were predicted using a two dimensional(2-D)comprehensive computational fluid dynamics(CFD)combustion model.This model was developed from a three dimensional model for a supercritical CFB boiler previously constructed by our group.Based on an analysis of the NOx and N2O conversion processes in a CFB boiler,the primary formation and destruction reactions were introduced into the 2-D model and coupled.The resulting model was validated using data from the Baima 600 MW supercritical CFB boiler,and then applied to a 660 MW ultra-supercritical CFB boiler.The effects of excess air,the secondary air(SA)to(primary air(PA)plus SA)ratio and the SA injection height on NOx and N2O emissions were investigated.The results show that a higher excess air volume increases both NOx and N2O emissions,while increasing the SA/(PA+SA)ratio somewhat reduces both the NOx and N2O concentrations.On the basis of the results of this work,optimal locations for SA injection ports so as to lower NOx and N2O emissions are recommended. | Jieqiang Ji Leming Cheng Yangjun Wei Junfeng Wang Xinyu Gao Mengxiang Fang Qinhui Wang | 2020 | Particuology2020,18,2: | 4 |
| 7 | Studies on abacavir-induced hypersensitivity reaction:a successful example of translation of pharmacogenetics to personalized medicine显示文摘Abacavir is an effective nucleoside analog reverse transcriptase inhibitor used to treat human immunodeficiency virus(HIV) infected patients.Its main side effect is hypersensitivity reaction(HSR).The incidence of the HSR is associated with ethnicity among patients exposed to abacavir,and retrospective and prospective studies show a significantly increased risk of abacavir-induced HSR in human leukocyte antigen(HLA)-B*57:01-carrying patients.Immunological studies indicated that abacavir interacts specifically with HLA-B*57:01 and changed the binding specificity between the HLA molecule and the HLA-presented endogenous peptide repertoire,leading to a systemic autoimmune reaction.HLA-B*57:01 screening,combined with patch testing,had clinically predictive value and cost-effective impact in reducing the incidence of abacavir-induced HSR regardless of the HLA-B*57:01 prevalence in the population.Therefore,the US Food and Drug Administration(FDA) and international HIV treatment guidelines recommend a routine HLA-B*57:01 screening prior to abacavir treatment to decrease false positive diagnosis and prevent abacavir-induced HSR.The studies of abacavir-induced HSR and the implementation of the HLA-B*57:01 screening in the clinic represent a successful example of the use of pharmacogenetics for personalized diagnosis and therapy. | GUO YongLi SHI LeMing HONG HuiXiao SU ZhenQiang FUSCOE James NING BaiTang | 2013 | Science China(Life Sciences)2013,56,2: | 3 |

| 9 | Circulating leptin and its muscle gene expression in Nellore cattle with divergent feed efficiency显示文摘Background: Leptin has a strong relation to important traits in animal production, such as carcass composition,feed intake, and reproduction. It is mainly produced by adipose cells and acts predominantly in the hypothalamus.In this study, circulating leptin and its gene expression in muscle were evaluated in two groups of young Nellore bulls with divergent feed efficiency. Individual dry matter intake(DMI) and average daily gain(ADG) of 98 Nellore bulls were evaluated in feedlot for 70 d to determinate the residual feed intake(RFI) and select 20 animals for the high feed efficient(LRFI) and 20 for the low feed efficient(HRFI) groups. Blood samples were collected on d 56 and at slaughter(80 d) to determine circulating plasma leptin. Samples of Longissimus dorsi were taken at slaughter for leptin gene expression levels.Results: DMI and RFI were different between groups and LRFI animals showed less back fat and rump fat thickness,as well as less pelvic and kidney fat weight. Circulating leptin increased over time in all animals. Plasma leptin was greater in LRFI on 56 d and at slaughter(P = 0.0049). Gene expression of leptin were greater in LRFI animals(P = 0.0022) in accordance with the plasma levels. | 12 | Papillary fistulotomy vs conventional cannulation for endoscopic biliary access:A prospective randomized trial显示文摘AIM To compare the cannulation success, biochemical profile, and complications of the papillary fistulotomy technique vs catheter and guidewire standard access.METHODS From July 2010 to May 2017, patients were prospectively randomized into two groups: Cannulation with a catheter and guidewire(Group Ⅰ) and papillary fistulotomy(Group Ⅱ). Amylase, lipase and C-reactive protein at T0, as well as 12 h and 24 h after endoscopic retrograde cholangiopancreatography, and complications(pancreatitis, bleeding, perforation) were recorded.RESULTS We included 102 patients(66 females and 36 males, mean age 59.11 ± 18.7 years). Group Ⅰ and Group Ⅱ had 51 patients each. The successful cannulation rates were 76.5% and 100%, respectively(P = 0.0002). Twelve patients(23.5%) in Group Ⅰ had a difficult cannulation and underwent fistulotomy, which led to successful secondary biliary access(Failure Group). The complication rate was 13.7%(2 perforations and 5 mild pancreatitis) vs 2.0%(1 patient with perforation and pancreatitis) in Groups Ⅰ and Ⅱ, respectively(P = 0.0597). CONCLUSION Papillary fistulotomy was more effective than guidewire cannulation, and it was associated with a lower profile of amylase and lipase.
