| 1 | Hepatic differentiation capability of rat bone marrow-derived mesenchymal stem cells and hematopoietic stem cells显示文摘AIM: To investigate the different effects of mesenchymal stem cells (MSCs) and hematopoietic stem cells (HSCs) on hepatic differentiation.METHODS: MSCs from rat bone marrow were isolated and cultured by standard methods. HSCs from rat bone marrow were isolated and purified by magnetic activated cell sorting. Both cell subsets were induced. Morphology, RT-PCR and immunocytochemistry were used to identify the hepatic differentiation grade.RESULTS: MSCs exhibited round in shape after differentiation, instead of fibroblast-like morphology before differentiation. Albumin mRNA and protein were expressed positively in MSCs, without detection of alpha-fetoprotein (AFP). HSCs were polygonal in shape after differentiation. The expression of albumin signal decreased and AFP signal increased. The expression of CK18 was continuous in MSCs and HSCs both before and after induction.CONCLUSION: Both MSCs and HSCs have hepatic differentiation capabilities. However, their capabilities are not the same. MSCs can differentiate into mature hepatocyte-like cells, never expressing early hepatic specific genes, while Thy-1.1^+cells are incllned to differentiate into hepatic stem cell-like cells, with an increasing AFP expression and a decreasing albumin signal. CK18 mRNA is positive in Thy-1.1^+ cells and MSCs, negative in Thy-1.1^+ cells. | 2 | In vitro resistance to interferon of hepatitis B virus with precore mutation显示文摘AIM: Chronic hepatitis B virus (HBV) infection is predominantly treated with interferon alpha (IFN-α), which results in an efficient reduction of the viral load only in 20-40% of treated patients. Mutations at HBV precore prevail in different clinical status of HBV infection. The roles of precore mutation in the progression of chronic hepatitis and interferon sensitivity are still unknown. The aim of this study was to explore if there was any relationship between HBV precore mutation and sensitivity to interferon in vitro.METHODS: HBV replication-competent recombinant constructs with different patterns of precore mutations were developed. Then the recombinants were transiently transfected into hepatoma cell line (Huh7) by calcium phosphate transfection method. With or without IFN, viral products in culture medium were collected and quantified 3d after transfection.RESULTS: We obtained 4 recombinant constructs by orientation-cloning 1.2-fold-overlength HBV genome into pUC18 vector via the EcoRI and Hind Ⅲ and PCR mediated site-directed mutagenesis method. All the recombinants contained mutations within precore region. Huh7 cells transfected with recombinants secreted HBsAg and HBV particles into the cell culture medium, indicating that all the recombinants were replication-competent. By comparing the amount of HBV DNA in the medium, we found that HBV DNA in medium reflecting HBV replication efficiency was different in different recombinants. Recombinants containing precore mutation had fewer HBV DNAs in culture medium than wild type. This result showed that recombinants containing precore mutation had lower replication efficiency than wild type. | 3 | Dendritic cells from chronic hepatitis B patients can induce HBV antigen-specific T cell responses显示文摘AIM: To determine whether dendritic cells (DCs) from chronic hepatitis B patients could induce HBV antigenspecific T cell responses or not. METHODS: DCs were generated from peripheral blood mononuclear cells of patients with chronic hepatitis B (CHB) infection and healthy donors. We compared the phenotypes of these DCs and their ability to secrete cytokines and to participate in mixed lymphocyte reactions. In addition, autologous lymphocytes were cultured with DCs loaded with HBV core region peptide HBcAg8-27, an epitope recognized by cytotoxic T lymphocytes(CTL), and bearing human leucocyte antigen (HLA)-A2 for 10 d. Cytokine secretion and lytic activity against peptide-pulsed target cells were assessed. RESULTS: DCs with typical morphology were generated successfully by culturing peripheral blood mononuclear cells (PBMCs) from CriB patients with AIM-V containing GM-CSF and IL-4. Compared with DCs from normal donors, the level of CD80 expressed in DCs from CHB patients was lower, and DCs from patients had lower capacity of stimulate T cell proliferation. When PBMCs isolated from patients with chronic or acute hepatitis B infection and from normal donors were cocultured with HBcAg18-27 peptide, the antigen-specific memory response of PBMCs from acute hepatitis B patients was stronger than that of PBMCs from chronic hepatitis B patients or normal donors. PBMCs cocultured with DCs treated with HBcAg18-27 CTL epitope peptide induced an antigen-specific T cell reaction, in which the level of secreted cytokines and lyric activity were higher than those produced by memory T cells. | 4 | Association of interleukin-12 p40 gene 3'-untranslated region polymorphism and outcome of HCV infection显示文摘AIM: To investigate the effect of interleukin-12 p40 gene (IL12E 3'-untranslated region polymorphism on the outcome of HCV infection.METHODS: A total of 133 patients who had been infected with HCV for 12-25 (18.2±3.8) years, were enrolled in this study. Liver biochemical tests were performed with an automated analyzer and HCV RNA was detected by fluorogenic quantitative polymerase chain reaction. B-mode ultrasound was used for liver examination. Polymerase chain reactionrestriction fragment length polymorphism (PCR-RFLP) was used for the detection of IL12B (1188A/C) polymorphism.RESULTS: Self-limited infection was associated with AC genotype (OR = 3.48; P = 0.001) and persistent infection was associated with AA genotype (OR = 0.34; P = 0.014)at site 1188 of IL12B. In patients with persistent HCV infection, no significant differences were found regarding the age, gender, duration of infection and biochemical characteristics (P>0.05). According to B-mode ultrasound imaging and clinical diagnosis, patients with persistent infection were divided into groups based on the severity of infection. | 5 | A Novel Resource Allocation Algorithm based on Downlink and Uplink Decouple Access Scheme in Heterogeneous Networks显示文摘Traditional cellular network requires that a user equipment(UE) should associate to the same base station(BS) in both the downlink(DL) and the uplink(UL). Based on dual connectivity(DC) introduced in LTE-Advanced R12, DL/UL decouple access scheme has been proposed, which is especially suitable for heterogeneous networks(Het Nets). This paper is the pioneer to take the DL/UL decouple access scheme into consideration and develop a novel resource allocation algorithm in a two-tier Het Net to improve the total system throughput in the UL and ease the load imbalance between macro base stations(MBSs) and pico base stations(PBSs). A model is formulated as a nonlinear integer programming, and the proposed algorithm is a sub-optimal algorithm based on the graph theory. First, an undirected and weighted interference graph is obtained. Next, the users are grouped to let users with large mutual interferences to be assigned to different clusters. Then, the users in different clusters are allocated to different resource blocks(RBs) by using the Hungarian algorithm. | 10 | Transparent characterization and quantitative analysis of broken gangue's 3D fabric under the bearing compression显示文摘Broken gangue has been extensively used in rockfill dams,subgrade,embankment,foundation cushion and other engineering construction.The deformation characteristics of broken gangue under the bearing compression play a decisive role in the firmness,stability and safety of these structures(buildings),and the meso-fabric change of broken gangue under the bearing compression significantly affects its macro deformation.In this study,the transparent characterization and quantitative analysis of 3D fabric of broken gangue under the bearing compression were performed through CT scanning test,image processing and 3D reconstruction technology,and the influence mechanism of internal fabric of broken gangue on its macro deformation was revealed.The results show that:In the loading stage of 0–2 MPa,the sharp corners,thin edges on the blocks and the bar-shaped and blade-shaped blocks with poor regularity are broken first under the bearing compression;in the loading stage of 2–8 MPa,a large number of larger particles in the sample are crushed in the mode of fragmentation;in the loading stage of 8–10 MPa,the breakage degree of samples is relieved.The axial displacement of the block inside the sample occurs,as well as the lateral displacement of the block converging to the central axis of the sample.In the rapid deformation stage,the macro deformation of the broken gangue is mainly caused by the rearrangement and adjustment of the block structure and the breakage of the block;in the slow deformation stage,it is mainly caused by the breakage of the block;in the stable deformation stage,it is mainly caused by the optimization and adjustment of the bearing skeleton in the sample. | Junmeng Li Yanli Huang Shenyang Ouyang Yachao Guo Huadong Gao Laiwei Wu Yibing Shi Lei Zhu | 2022 | International Journal of Mining Science and Technology2022,32,2: | 0 |
