1Hepatic differentiation capability of rat bone marrow-derived mesenchymal stem cells and hematopoietic stem cells显示文摘AIM: To investigate the different effects of mesenchymal stem cells (MSCs) and hematopoietic stem cells (HSCs) on hepatic differentiation.METHODS: MSCs from rat bone marrow were isolated and cultured by standard methods. HSCs from rat bone marrow were isolated and purified by magnetic activated cell sorting. Both cell subsets were induced. Morphology, RT-PCR and immunocytochemistry were used to identify the hepatic differentiation grade.RESULTS: MSCs exhibited round in shape after differentiation, instead of fibroblast-like morphology before differentiation. Albumin mRNA and protein were expressed positively in MSCs, without detection of alpha-fetoprotein (AFP). HSCs were polygonal in shape after differentiation. The expression of albumin signal decreased and AFP signal increased. The expression of CK18 was continuous in MSCs and HSCs both before and after induction.CONCLUSION: Both MSCs and HSCs have hepatic differentiation capabilities. However, their capabilities are not the same. MSCs can differentiate into mature hepatocyte-like cells, never expressing early hepatic specific genes, while Thy-1.1^+cells are incllned to differentiate into hepatic stem cell-like cells, with an increasing AFP expression and a decreasing albumin signal. CK18 mRNA is positive in Thy-1.1^+ cells and MSCs, negative in Thy-1.1^+ cells. 4Fluid-melt Inclusions in Fluorite of the Huanggangliang Skarn Iron-Tin Deposit and Their Significance to Mineralization显示文摘For the first time, fluid-melt inclusions are found in fluorite of the Huanggangliang skarn iron-tin deposit (HSID). The fluorite was formed in the main stage of mineralization, named the hydro-skarnization stage. The inclusions contain various components such as Fe, Mg and Cr from deep sources. The melts of primary inclusions are mainly Ca- and F-rich and those of secondary inclusions tend to become Si-rich. During this evolution process, the melts and iron daughter minerals decreased and even vanished. These facts reveal that the evolution of the primary mineralizing fluids and the differentiation of the fluids and melts are the main factors leading to the deposition of Fe, Sn and other elements. This discovery confirms the magmatic genesis of the HSID and has filled in the gaps in the research of magmatogenic skarn deposits and furnished new methods for such research. 5Spaceborne GPS receiver antenna phase center offset and variation estimation for the Shiyan 3 satellite显示文摘In determining the orbits of low Earth orbit(LEO) satellites using spaceborne GPS, the errors caused by receiver antenna phase center offset(PCO) and phase center variations(PCVs) are gradually becoming a major limiting factor for continued improvements to accuracy. Shiyan 3, a small satellite mission for space technology experimentation and climate exploration, was developed by China and launched on November 5, 2008. The dual-frequency GPS receiver payload delivers1 Hz data and provides the basis for precise orbit determination within the range of a few centimeters. The antenna PCO and PCV error characteristics and the principles influencing orbit determination are analyzed. The feasibility of PCO and PCV estimation and compensation in different directions is demonstrated through simulation and in-flight tests. The values of receiver antenna PCO and PCVs for Gravity Recovery and Climate Experiment(GRACE) and Shiyan 3 satellites are estimated from one month of data. A large and stable antenna PCO error, reaching up to10.34 cm in the z-direction, is found with the Shiyan 3 satellite. The PCVs on the Shiyan 3 satellite are estimated and reach up to 3.0 cm, which is slightly larger than that of GRACE satellites. Orbit validation clearly improved with independent k-band ranging(KBR) and satellite laser ranging(SLR) measurements. For GRACE satellites, the average root mean square(RMS) of KBR residuals improved from 1.01 cm to 0.88 cm. 6In vitro resistance to interferon of hepatitis B virus with precore mutation显示文摘AIM: Chronic hepatitis B virus (HBV) infection is predominantly treated with interferon alpha (IFN-α), which results in an efficient reduction of the viral load only in 20-40% of treated patients. Mutations at HBV precore prevail in different clinical status of HBV infection. The roles of precore mutation in the progression of chronic hepatitis and interferon sensitivity are still unknown. The aim of this study was to explore if there was any relationship between HBV precore mutation and sensitivity to interferon in vitro.METHODS: HBV replication-competent recombinant constructs with different patterns of precore mutations were developed. Then the recombinants were transiently transfected into hepatoma cell line (Huh7) by calcium phosphate transfection method. With or without IFN, viral products in culture medium were collected and quantified 3d after transfection.RESULTS: We obtained 4 recombinant constructs by orientation-cloning 1.2-fold-overlength HBV genome into pUC18 vector via the EcoRI and Hind Ⅲ and PCR mediated site-directed mutagenesis method. All the recombinants contained mutations within precore region. Huh7 cells transfected with recombinants secreted HBsAg and HBV particles into the cell culture medium, indicating that all the recombinants were replication-competent. By comparing the amount of HBV DNA in the medium, we found that HBV DNA in medium reflecting HBV replication efficiency was different in different recombinants. Recombinants containing precore mutation had fewer HBV DNAs in culture medium than wild type. This result showed that recombinants containing precore mutation had lower replication efficiency than wild type. HBV DNA was decreased in pUC18-HBV1.2-WT recombinants after IFN was added while others with precore mutations were not, indicating that HBV harboring precore mutation was less sensitive to IFN in cell culture system.CONCLUSION: These data indicate that HBV harboring precore mutation may be resistant to IFN in vitro.YanWang LaiWei DongJiang XuCong RanFei JiangXiao YuWang 2005World Journal of Gastroenterology2005,11,5:10
7The Tuerkubantao ophiolite melange in Xinjiang,NW China:New evidence for the Erqis suture zone显示文摘The extension of the suture zone between the Siberian and Kazakhstan continents in China has been a matter of debate because few outcrops of ophiolitic melange have been found so far.The recently found Tuerkubantao ophiolitic melange,which is located east of the Kekesentao Mt.in the Buerjin County of the Chinese Altay,provides an important clue for this problem.This paper presents the results of field investigation,petrology,U-Pb isotope dating of zircons and bulk-rock geochemistry of the Tuerkubantao melange rocks.The melange consists of fault-contacted ultramafic rocks,gabbro, diabase,basalt,flysch and granitoids.The ultramafic-mafic rocks are Mg-rich(Mg~#=4.25—6.35) and w(SiO_2) spans 38.8%—46.8%.Basalt and gabbro are geochemically similar and are characterized by low w(FeO^t)(10.9%),total alkali w(Na_2O+K_2O)=2.58%) and w(TiO_2)(1.17%) and affinity to the Mg-rich tholeiite series.The flat REE and trace-element patterns of the ultramafic-mafic rocks are indicative of their ophiolite origin,i.e.,formation in a mid-oceanic ridge setting.The fragments of low-K gneissic granite formed in suprasubduction or syn-collisional setting.Zircons from gabbro and gneissic granite yielded U-Pb ages of 363 and 355 Ma.respectively,suggesting Late Devonian mid-oceanic spreading and oceanic subduction accompanied by suprasubduction magmatism.The Tiierkubantao ophiolite together with the Qiaoxiahala and Buergen ophiolites of the Kekesentao belt define an ophiolitic melange belt extending along the Erqis fault.This belt probably belongs to the Ural-Zaisan -South Mongolian suture-shear zone formed during the subduction of the Paleo-Asian Ocean and subsequent collision of the Siberian and Kazakhstan continents.Yuwang Wang Jingbin Wang Lijuan Wang Lingli Long Pingzhi Tang Zhen Liao Huiqiong Zhang Yu Shi 2012Geoscience Frontiers2012,3,5:9
8Dendritic cells from chronic hepatitis B patients can induce HBV antigen-specific T cell responses显示文摘AIM: To determine whether dendritic cells (DCs) from chronic hepatitis B patients could induce HBV antigenspecific T cell responses or not. METHODS: DCs were generated from peripheral blood mononuclear cells of patients with chronic hepatitis B (CHB) infection and healthy donors. We compared the phenotypes of these DCs and their ability to secrete cytokines and to participate in mixed lymphocyte reactions. In addition, autologous lymphocytes were cultured with DCs loaded with HBV core region peptide HBcAg8-27, an epitope recognized by cytotoxic T lymphocytes(CTL), and bearing human leucocyte antigen (HLA)-A2 for 10 d. Cytokine secretion and lytic activity against peptide-pulsed target cells were assessed. RESULTS: DCs with typical morphology were generated successfully by culturing peripheral blood mononuclear cells (PBMCs) from CriB patients with AIM-V containing GM-CSF and IL-4. Compared with DCs from normal donors, the level of CD80 expressed in DCs from CHB patients was lower, and DCs from patients had lower capacity of stimulate T cell proliferation. When PBMCs isolated from patients with chronic or acute hepatitis B infection and from normal donors were cocultured with HBcAg18-27 peptide, the antigen-specific memory response of PBMCs from acute hepatitis B patients was stronger than that of PBMCs from chronic hepatitis B patients or normal donors. PBMCs cocultured with DCs treated with HBcAg18-27 CTL epitope peptide induced an antigen-specific T cell reaction, in which the level of secreted cytokines and lyric activity were higher than those produced by memory T cells. 9Chromic-P32 phosphate treatment of implanted pancreatic carcinoma: Mechanism involved显示文摘AIM: To study the effects of chromic-P32 phosphate (32p colloids) interstitial administration in Pc-3 implanted pancreatic carcinoma, and investigate its anticancer mechanism.METHODS: Ninety-eight tumor bearing nude mice werekilled at different time points after the injection of 32Pcolloids to the tumor core with observed radioactivity. The light microscopy, transmission electron microscopy (TEM) and immuno-histochemistry and flow cytometry were used to study the rates of tumor cell necrosis, proliferating cell nuclear antigen index, the micro vessel density (MVD). The changes of the biological response to the lymphatic transported 32p colloids in the inguinal lymph node (ILN) were dynamically observed, and the percentage of tumor cell apoptosis, and Apo2.7, caspase-3, Bcl-2, Baxrelated gene expression were observed too.RESULTS: The half-life of effective medication is 13 dafter injection of 32P colloids to the tumor stroma, in 1-6groups, the tumor cell necrosis rates were 20%, 45%,65%, 70%, 95% and 4%, respectively (F= 4.14-105.36, P<0.01). MVD were 38.5±4.0, 28.0±2.9, 17.0±2.9, 8.8±1.5,5.7±2.3 and 65.0±5.2 (t= 11.9-26.1, P<0.01), respectively.Under TEM fairly differentiated Pc-3 cells were found. Thirty days after medication, tumors were shrunk and dried with scabs detached, and those in control group increased in size prominently with plenty of hypodermic blood vessels. In all animals the ILN were enlarged but in medicated animals they appeared later and smaller than those in control group. The extent of irradiative injury in ILN was positively correlated to the dosage of medication. Typical tumor cell apoptosis could be found under TEM inanimals with intra-tumoral injection of low dosed 32P colloids. The peak of apoptosis occurred in 2.96 MBq group and 24 h after irradiation. In the course of irradiationinduced apoptosis, the value of Bcl-2/Bax was down regulated; Apo2.7 and caspase-3 protein expression were prominently increased dose dependently. CONCLUSION: 32p colloids intra-tumor injection having prominent anticancer effectiveness may reveal the ability of promoting cell differentiation. 10Ore-forming fluid and mineralization of Caijiaying and Dajing polymetallic ore deposits显示文摘Fluid inclusions from the Dajing and Caijiaying deposits have nearly the same homogenization temperature. Correlation between temperature and salinity shows that both Sn- and Cu-bearing fluids Sn and Cu were present in the Dajing deposit but only one kind of fluids continuously evolved in the Caijiaying deposit. Study on rare earth elements (REE) in ancient fluid from the inclusions indicates that the fluid of Sn mineralizing stage in Dajing was derived from remelting magma of the continental crust, and the fluid of Cu-Pb-Zn mineralizing stage in the Dajing deposit and the fluid of Cu-Pb-Zn mineralization in the Caijiaying deposit were derived from the mantle. It is concluded that the Cu-Pb-Zn mineralizations in the Dajing and Caijiaying deposits resulted from the identical tectono-thermal event of magma-fluids induced by Mesozoic tectonic transition and extension in the eastern part of North China Craton.WANG Lijuan,WANG Jingbin, WANG Yuwang & ZHU Heping1. 11Effect of operation-synchronizing transfusion of apoptotic spleen cells from donor rats on acute rejection of recipient rats after liver transplantation显示文摘AIM: To study effect of operation-synchronizing transfusion of apoptotic spleen cells from donor rats on acute rejection of recipient rats after liver transplantation.METHODS: Two of Wistar rats were chosen randomly for normal liver pathology control and ten of SD rats chosen randomly for liver function control as blank group (no operation). The rest of Wistar and SD rats were divided into four groups: control group (only liver transplantation),Dex group (donors receiving intraperitoneal injection of dexamethasone), SpC group (recipients receiving infusion of spleen cells of donors), Dex-SpC group (recipients receiving infusion of apoptotic spleen cells of donors),with each group except blank group, containing 10 SD rats and 10 Wistar rats, respectively. Wistar rats received liver transplantation from SD rats, in the meantime they received infusion of spleen cells of donors, which were induced by an intraperitoneal injection of dexamethasone The serum alanine transaminase (ALT), total bilirubin (T bili), liver pathological changes and survival time were analysed. Statistical analysis was carried out using SPSS 10.0 for Windows. Differences of the parametric data of ALT in means were examined by one-way ANOVA.Differences of ALT between two groups were examined by LSD. Differences of the nonparametric data of T bili in means and scores of pathology classification for acute rejection were examined by Kruskal-Willis H test. The correlations between ALT and T bili were analysed by Bivariate. Kaplan-Meier curves were used to demonstrate survival distribution. The log-rank test was used to compare the survival data.RESULTS: There were significant differences in ALT of the five groups (F= 23.164 P= 0.000), and ALT in DexSpC group was significantly higher than that in blank control, control, Dex, and SpC groups (P = 0.000), and ALT in SpC group was significantly higher than that in blank control (P = 0.000), control (P = 0.004), and Dex groups (P = 0.02). Results of nonparametric analysis of T bili showed that there were differences in T bili of the five groups (χ2 = 33.265 P= 0.000). T bili in Dex-SpC group was significantly higher than that in blank control, control,Dex, and SpC groups. T bili in SpC group was higher than that in blank control, control, and Dex groups. There were significant differences in scores of pathology classification for acute rejection in each of the groups (χ2 = 25.933,P = 0.000). The pathologically more serious acute rejection was found in Dex-SPC group than in other groups. No sign of acute rejection was observed in the blank control group.Slight acute rejection was observed in the control group.Slight-moderate acute rejection was observed in the Dex group. Moderate-acute rejection was observed in the SpC group. Severe-acute rejection was observed in the DexSpC group. The survival time in Dex-SpC group was shorter than in other groups (statistic = 11.13, P = 0.011). ALT and T bili were positively correlated (r= 0.747, P= 0.000,two-tailed).CONCLUSION: In order to reduce quantity of blood loss from rats after liver transplantation, only one of ALT or T bili is needed for liver function measurement of rats.Simultaneous injection of apoptotic spleen cells from donors induced by dexamethasone to liver transplantation rats aggravates acute rejection. 12Precession–nutation correction for star tracker attitude measurement of STECE satellite显示文摘Space Technology Experiment and Climate Exploration(STECE) is a small satellite mission of China for space technology experiment and climate exploration. A new test star tracker and one ASTRO 10 star tracker have been loaded on the STECE satellite to test the new star tracker's measurement performance. However,there is no autonomous precession–nutation correction function for the test star tracker,which causes an apparent periodic deflection in the inter-boresight angle between the two star trackers with respect to each other of up to ±500 arcsec,so the precession and nutation effect needs to be considered while assessing the test star tracker. This paper researches on the precession–nutation correction for the test star tracker's attitude measurement and presents a precession–nutation correction method based on attitude quaternion data. The periodic deflection of the inter-boresight angle between the two star trackers has been greatly eliminated after the precession and nutation of the test star tracker's attitude data have been corrected by the proposed method and the validity of the proposed algorithm has been demonstrated. 13Physicochemical Control of the Early Permian Xiangshan Fe-Ti Oxide Deposit in Eastern Tianshan(Xinjiang),NW China显示文摘The Xiangshan mafic-ultramafic complex is one of the major Early Permian mafic-ultramafic intrusions in eastern Tianshan(Xinjiang, NW China), and consists of two major intrusive phases. The first intrusive phase is mainly gabbroic rocks hosting ilmenite mineralization, while the second intrusive phase is mainly lherzoilite associated with Ni-Cu sulfide mineralization. The Xiangshan ilmenite orebodies hosted in the Fe-Ti oxide-bearing gabbro occur along the contact between hornblende gabbros and leucogabbros. The hornblende gabbros and Fe-Ti oxide rich gabbros at Xiangshan are newly dated to be Early Permian(280.1 and 279.2 Ma, respectively). Major and trace element compositions of zircons and whole rocks from Xiangshan hornblende gabbro and Fe-Ti oxide gabbro have been measured by in situ excimer laser ablation ICP-MS. Zircon Ce^(4+)/Ce^(3+) ratios based on lattice-strain model and Ti-in-zircon temperatures of hornblende gabbro and Fe-Ti oxide gabbro of the Xiangshan complex are calculated to evaluate the physicochemical variations during the ilmenite mineralization. Whole-rock geochemistry and zircon trace element geochemistry suggest that Fe-Ti oxide gabbros were formed from a basaltic parent magma which had undergone a transfromation from being H_2O-rich to H_2O-poor. During the magmatic evolution, primitive, H_2O-poor basaltic melts may have been replenished into the system, increasing its solidus temperature and decreasing its oxygen fugacity and H_2O contents. 17Protein kinase C-dependent activation of P44/42 mitogen-activated protein kinase and heat shock protein 70 in signal transduction during hepatocyte ischemic preconditioning显示文摘AIM: To investigate the significance of protein kinase C (PKC), P44/42 mitogen-activated protein kinase (MAPKs) and heat shock protein (HSP)70 signal transduction during hepatocyte ischemic preconditioning. METHODS: In this study we used an in vitro ischemic preconditioning (IP) model for hepatocytes and an in vivo model for rat liver to investigate the significance of protein kinase C (PKC), P44/42 mitogen-activated protein kinase (P44/42 MAPKs) and heat shock protein 70 (HSP70) signal transduction in IP. Through a normal liver cell hypoxic preconditioning (HP) model in which cultured normal liver cells were subjected to 3 cycles of 5 rain of incubation under hypoxic conditions followed by 5 rain of reoxygenation and subsequently exposed to hypoxia and reoxygenation for 6 h and 9 h respectively. PKC inhibitor, activator and MEK inhibitor were utilized to analyze the phosphorylation of PKC, the expression of P44/42 MAPKs and HSP70. Viability and cellular ultrastructure were also observed. By using rat liver as an in vivo model of liver preconditioning (3 cycles of 10-min occlusion and 10-min reperfusion), in vivo phosphorylation of PKC and P44/42MAPKs, HSP70 expression were further analyzed. AST/ALT concentration, cellular structure and ultrastruture were also observed. All the data were statistically analyzed. RESULTS: Similar results were obtained in both in vivo and in vitro IP models. Compared with the control withouts IP (or HP), the phosphorylation of PKC and P44/42 MAPKs and the expression of HSP70 were obviously increased in IP (or HP) treated model in which cytoprotection could be found. The effects of preconditioning were mimicked by stimulating PKC with 4β phorobol-12-myristate 13-acetate (PMA). Conversely, inhibiting PKC with chelerythrine abolished the protection given by preconditioning. PD98059, inhibitor of MEK (the upstream kinase of P44/42MAPKs), also reverted the cytoprotection exerted by preconditioning. CONCLUSION: The results demonstrate that preconditioning induces a rapid activation of P44/421VlAPKs and PKC activation plays a pivotal role in the activation of P44/42 MAPKs pathway that participates in the preservation of liver cells. 19Petrogenesis and Metallogenesis of the Niumaoquan Gabbroic Intrusion Associated with Fe-Ti Oxide Ores in the Eastern Tianshan, NW China显示文摘The Niumaoquan layered gabbroic intrusion is in the southern margin of the Central Asian Orogenic Belt in North Xinjiang, China, and hosts a Fe-Ti oxide deposit in its evolved gabbroic phases. In this paper, we report zircon U-Pb age, Sr-Nd-Hf isotopes, plagioclase chemistry, and whole-rock geochemistry of the Niumaoquan layered gabbroic intrusion. Zircon grains separated from an anorthosite sample analyzed by laser ablation inductively coupled plasma mass spectrometry yielded a concordia age of 314.7±0.74 Ma, indicating that the Niumaoquan ore-bearing gabbroic intrusion was emplaced during the Late Carboniferous. The olivine gabbro texture and plagioclase chemistry suggest that plagioclase was an early crystallized silicate phase that crystallized prior to olivine. Fractional crystallization and accumulation of plagioclase significantly controlled the evolution of the Niumaoquan gabbroic intrusion and contributed to the formation of anorthosite layers, causing metallogenic elements to become enriched in the residual melt. The Niumaoquan gabbroic intrusion is characterized by the enrichment of large ion lithophile elements and depletion of high field strength elements, positive zircon εHf(t) values(+2.1 to +12.2), positive εNd(t) values(+3.3 to +5.2), and low initial ^(87)Sr/^(86)Sr ratios(0.7039 to 0.7047), suggesting that the parental magma was produced by interactions between metasomatized lithospheric mantle and depleted asthenospheric melts at an early post-collision stage. 20Sodium-rich volcanic rocks and their relationships with iron deposits in the Aqishan-Yamansu belt of Eastern Tianshan,NW China显示文摘The volcanic rocks hosting the iron deposits in the Aqishan–Yamansu metallogenic belt are sodium-rich.The geochronology,petrography,and geochemistry of minerals and sodium-rich rocks as well as the relationship between these rocks and the iron deposits are studied.Geochemically,the ore-hosting volcanic rocks are sodiumrich(the averages of Na2O and Na2O/K2O are 4.31 wt.%and 8.56,respectively)and belong to the calc-alkaline series.They are enriched in LREEs and LILEs(Ba,U,K,and Sr),but depleted in HFSEs(Nb,Ta,and Ti).SHRIMP zircon U–Pb dating of the crystal tuff in the Aqishan Formation and the dacite in the Tugutu Bulak Formation yields ages of 337.52.3 Ma(n?15,MSWD?0.85)and 313.03.3 Ma(n?13,MSWD?0.74),respectively,indicating that the sodium-rich volcanic rocks formed from the early–late Carboniferous.Electron microprobe data from plagioclases demonstrate that albites and/or oligoclases were formed in the basic–intermediate–acid volcanic rocks.Two stages of albitization are identified,and the latter is likely attributed to the dissolution of iron in the Aqishan–Yamansu belt.The sodium-rich volcanic rocks probably formed by the interaction between volcanic lava and seawater after volcanoes erupted on the seafloor;meanwhile,the albites formed by element substitution in a low-metamorphic environment.The spatiotemporal coupling relationship between sodium-rich volcanic rocks and iron deposits in the Aqishan–Yamansu belt is favorable.Iron dissolved from the dark minerals of basic–intermediate volcanic rocks through sodium metasomatism is one of the material sources for the iron deposits.Zhiyuan Sun WangJingbin Yuwang Wang Lingli Long Zhaohua Luo Xiaohua Deng Qitao Hu Menglong Wang 2020Geoscience Frontiers2020,11,2:2
