2Ultra-sensitive Nanoprobe Modified with Tumor Cell Membrane for UCL/MRI/PET Multimodality Precise Imaging of Triple-Negative Breast Cancer显示文摘Triple-negative breast cancer(TNBC)is a subtype of breast cancer in which the estrogen receptor and progesterone receptor are not expressed,and human epidermal growth factor receptor 2 is not amplified or overexpressed either,which make the clinical diagnosis and treatment very challenging.Molecular imaging can provide an effective way to diagnose TNBC.Upconversion nanoparticles(UCNPs),are a promising new generation of molecular imaging probes.However,UCNPs still need to be improved for tumor-targeting ability and biocompatibility.This study describes a novel probe based on cancer cell membrane-coated upconversion nanoparticles(CCm-UCNPs),owing to the low immunogenicity and homologous-targeting ability of cancer cell membranes,and modified multifunctional UCNPs.This probe exhibits excellent performance in breast cancer molecular classification and TNBC diagnosis through UCL/MRI/PET tri-modality imaging in vivo.By using this probe,MDA-MB-231 was successfully differentiated between MCF-7 tumor models in vivo.Based on the tumor imaging and molecular classification results,the probe is also expected to be modified for drug delivery in the future,contributing to the treatment of TNBC.The combination of nanoparticles with biomimetic cell membranes has the potential for multiple clinical applications.Hanyi Fang Mengting Li Qingyao Liu Yongkang Gai Lujie Yuan Sheng Wang Xiao Zhang Min Ye Yongxue Zhang Mingyuan Gao Yi Hou Xiaoli Lan 2020Nano-Micro Letters2020,12,5:5
3Operation of offshore wind farms connected with DRU-HVDC transmission systems with special consideration of faults显示文摘The diode rectifier unit(DRU)-based high-voltage DC(DRU-HVDC) system is a promising solution for offshore wind energy transmission thanks to its compact design, high efficiency, and strong reliability. Herein we investigate the feasibility of the DRU-HVDC system considering onshore and offshore AC grid faults, DC cable faults, and internal DRU faults. To ensure safe operation during the faults, the wind turbine(WT) converters are designed to operate in either current-limiting or voltage-limiting mode to limit potential excessive overcurrent or overvoltage. Strategies for providing fault currents using WT converters during offshore AC faults to enable offshore overcurrent and differential fault protection are investigated. The DRU-HVDC system is robust against various faults, and it can automatically restore power transmission after fault isolation. 4Mass flows and river response in rapid uplifting regions-A case of lower Yarlung Tsangpo basin, southeast Tibet, China显示文摘The fluvial geomorphology in tectonically active(particularly rapid uplift)regions often undergoes continuous change.The rapid uplift is coincident with high erosion rates;consequently,incised valleys are formed.Mass flows(for example,avalanches,landslides,and debris flows)in incised valleys can markedly influence fluvial processes and even reshape valley geomorphology.However,these processes and long-term evolution corresponding to mass flows require further clarification.Field campaigns were carried out in the region near the Yigong Tsangpo and Palong Tsangpo Rivers(hereafter the Yigong and Palong Rivers),the two largest tributaries of the lower Yarlung Tsangpo River Tibet,China,to examine the feedback between fluvial processes and mass flows.Remote sensing images from recent decades were used to compare the channel morphology before and after typical mass flows(particularly catastrophic ones).The morphology of the lower Yigong River has evidently been impacted by landslides,while that of the Palong River has mainly been shaped by glacial processes and debris flows.At present,the morphology of the latter consists of alternating sections of gorges and wide valleys,with a staircase-like longitudinal profile.The gorge sections exhibit single and deeply incised channels with a high-gradient channel bed and terraces.In contrast,the wide valley sections consist of lakes,braided or anabranching channels,gentle bed gradients,and thick alluvial deposits.Debris flows occur more frequently in gullies in the reaches of the gorge sections and rarely in gullies along the wide valley sections.The occurrence of mass flow events has resulted in an imbalance of the previous(quasi-)equilibrium in the river morphology;however,this has triggered negative feedback that is driving the transient river morphology to a new state of(quasi-)equilibrium.Guo-An Yu Jianyin Lu Liqun Lyu Lujie Han Zhaoyin Wang 2020International Journal of Sediment Research2020,35,6:3
5Preparation of pitch precursor with excellent spinnability for general-purpose carbon fibre using coal tar pitch as raw material显示文摘Tetrahydrofuran(THF) extract of coal tar pitch(CTP) was used instead of blending CTP with pretreated pyrolysis fuel oil to prepare an isotropic pitch precursor with excellent spinnability for general-purpose carbon fibre through bromination-dehydrobromination. The feasibility and effectiveness of synthesising an isotropic pitch precursor derived from THF-soluble(CTP-THFs) is demonstrated in this study.The results show that CTP-THFs contains more light components than CTP;CTP-THFs and CTP monomer proportions were 62.52% and 45.32%, respectively. However, based on comparisons of CTP-THFsBr0 and CTPBr0 characterisations, CTP-THFs exhibits better polycondensation than CTP. Bromination-dehydrobro mination promotes polycondensation of pitch precursors, leading to greater carbon aromaticity in CTP-THFsBr5, CTP-THFsBr10, and CTP-THFsBr15 than that in CTP-THFsBr0 and CTPBr0. CTP-THFsBr5 and CTP-THFsBr10 have excellent spinnability even with softening points as high as 230 ℃. The pericondensed carbon and carbon aromaticity of CTP-THFsBr5 and CTP-THFsBr10 are high owing to the higher degree of polycondensation;however, they still possess a more linear molecular structure. 6Persistence of Cry1Ac Protein from Transgenic Bt Cotton Cultivation and Residue Returning in Fields and Its Effect on Functional Diversity of Soil Microbial Communities显示文摘The persistence of Cry1 Ac protein in the soil and its effect on soil microbial communities are a core issue in assessing the ecological risk of transgenic Bacillus thuringiensis(Bt) cotton. In this study a field experiment was conducted on the cultivation of transgenic Bt cotton(Jin 26 and BtJ i 668) with the immediate returning of residues to the fields, in order to quantify the Cry1 Ac protein content in the fields and investigate its effects on the functional diversity of soil microbial communities. Cry1 Ac protein in the residue-soil mixture was gradually degraded in the transgenic Bt cotton fields. After transgenic Bt cotton straw was returned to the fields for 30 d, 63.73%and 58.33% of the initial amounts of Cry1 Ac protein were degraded in the Jin 26 and BtJ i 668 fields, respectively. Before the crops were sown in the following year(180 d after returning the straw), no Cry1 Ac protein was detected in the fields. After returning the cotton straw to the fields for 30 d, the Shannon-Wiener and McIntosh indices of soil microbial communities in the transgenic Bt cotton fields were significantly higher than those in the non-transgenic cotton fields. Meanwhile, the utilization of carbon sources including amino acids, amines, and carbohydrates by the soil microbial communities significantly increased. Both the McIntosh index and the utilization of carbohydrates increased until 180 d. Principal component analysis revealed that amino acids, amides, and carbohydrates were the main carbon sources distinguishing the two principal component factors. 7Effect of ALA-PDT on the expressions of MMP-9, MMP-13 and TIMP-1 of hypertrophic scar model in rabbit ears显示文摘Objective: To investigate the effect of 5-aminolevulinic(ALA)-photodynamic therapy(PDT) on the expressions of MMP-9, MMP-13 and TIMP-1 of hypertrophic scar model in rabbit ears, and analyze the possible therapeutic mechanisms of ALA-PDT treatment to hypertrophic scars of rabbit ears. Methods: The experimental animals were randomly divided into normal control, negative control, high concentration of ALA-PDT, low concentration of ALA-PDT and PDT groups. The latter three groups received ALA-PDT treatment or PDT treatment once a week for 3 weeks. The specimens of the rabbits were collected respectively 1, 2 and 3 months after treatment to be used for RT-PCR and Western-blot test. Results: 1, 2 and 3 months after PDT treatment, the expressions of MMP-9 and MMP-13(including mRNA and protein) in hypertrophic scar tissues of three treatment groups were significantly higher than those of the negative control group(P<0.01), and the expression of TIMP-1 mRNA and protein of three treatment groups were significantly lower than that of the negative control group(P<0.01). There were also significant differences between high-concentration ALA-PDT treatment group and the low one(P<0.05). Conclusion: ALA-PDT is effective in treating hypertrophic scars of rabbit ears, and its possible therapeutic mechanisms are that ALA-PDT treatment generates oxidation activation effect to activate the activity of MMPs and induces the photoaging of fibroblasts of hypertrophic scar tissues of rabbit ears to inhibit the activity of TIMPs, which causes the up-regulation of MMPs and the down-regulation of TIMPs. 11Key problems and solutions in supercritical CO2 fracturing technology显示文摘Supercritical CO2 fracturing is considered to be a new method for efficient exploitation of unconventional reservoirs,such as shale gas,coal bed methane,and tight sand stone gas.Supercritical CO2 has many special properties including low viscosity,high diffusion coefficient,and lack of surface tension,which brings about great advantages for fracturing.However,these properties also cause several problems,such as difficulty in proppant transportation,high friction loss,and high pump displacement.In this paper,the above problems were analyzed by combining field test with laboratory study and specific solutions to these problems are given.The high frictionloss in the pipeline could be reduced by developing a new drag reducing agent and selecting large-size casing.Besides,for the problem of poor capacity in proppant carrying and sand plug,the methods of adding tackifier into supercritical CO2,increasing pump displacement and selecting ultralow density proppants are proposed.Moreover,for the problem of fast leak-off and high requirement for pump displacement,the displacement can be increased or the pad fluid can be injected into the reservoir.After solving the above three problems,the field test of supercritical CO2 fracturing can be conducted.The research results can promote the industrialization process of supercritical CO2 fracturing.Haizhu WANG Gensheng LI Bin ZHU Kamy SEPEHRNOORI Lujie SHI Yong ZHENG Xiaomei SHI 2019Frontiers in Energy2019,13,4:2

20Multiomics approach reveals the ubiquitination-specific processes hijacked by SARS-CoV-2显示文摘The Coronavirus Disease 2019(COVID-19)caused by Severe Acute Respiratory Syndrome Coronavirus 2(SARS-CoV-2)is a global pandemic that seriously threatens health and socioeconomic development,but the existed antiviral drugs and vaccines still cannot yet halt the spread of the epidemic.Therefore,a comprehensive and profound understanding of the pathogenesis of SARS-CoV-2 is urgently needed to explore effective therapeutic targets.Here,we conducted a multiomics study of SARS-CoV-2-infected lung epithelial cells,including transcriptomic,proteomic,and ubiquitinomic.Multiomics analysis showed that SARS-CoV-2-infected lung epithelial cells activated strong innate immune response,including interferon and inflammatory responses.Ubiquitinomic further reveals the underlying mechanism of SARS-CoV-2 disrupting the host innate immune response.In addition,SARS-CoV-2 proteins were found to be ubiquitinated during infection despite the fact that SARS-CoV-2 itself didn’t code any E3 ligase,and that ubiquitination at three sites on the Spike protein could significantly enhance viral infection.Further screening of the E3 ubiquitin ligases and deubiquitinating enzymes(DUBs)library revealed four E3 ligases influencing SARS-CoV-2 infection,thus providing several new antiviral targets.This multiomics combined with high-throughput screening study reveals that SARS-CoV-2 not only modulates innate immunity,but also promotes viral infection,by hijacking ubiquitination-specific processes,highlighting potential antiviral and anti-inflammation targets.Gang Xu Yezi Wu Tongyang Xiao Furong Qi Lujie Fan Shengyuan Zhang Jian Zhou Yanhua He Xiang Gao Hongxiang Zeng Yunfei Li Zheng Zhang 2022Signal Transduction and Targeted Therapy2022,7,10:1
