3Hazardous elements and amorphous nanoparticles in historical estuary coal mining area显示文摘In Brazil, intense coal exploitation activities have led to environmental deterioration, including soil mortification, water contamination, loss of ecosystem, and atmospheric contamination. In addition,considerable quantities of sulfur-rich residues are left behind in the mining area; these residues pose grave environmental issues as they undergo sulfide oxidation reactions. When sulfur oxides come in contact with water, extreme acid leachate is produced with great proportions of sulfate, and hazardous elements(HEs), which are identified as coal drainage(CMD). CMD is an environmental pollution challenge, particularly in countries with historic or active coal mines. To prevent CMD formation or its migration, the source must be controlled; however, this may not be feasible at many locations. In such scenarios, the mine water should be collected, treated, and discharged. In this study, data from 2005 to2010 was gathered on the geochemistry of 11 CMD discharges from ten different mines. There are several concerns and questions on the formation of nanominerals in mine acid drainage and on their reactions and interfaces. The detailed mineralogical and geochemical data presented in this paper were derived from previous studies on the coal mine areas in Brazil. Oxyhydroxides, sulfates, and nanoparticles in these areas possibly go through structural transformations depending on their size and formation conditions. The geochemistry of Fe-precipitates(such as jarosite, goethite, and hematite) existent in the CMD-generating coal areas and those that could be considered as a potential source of hazardous elements(HEs)(e.g., Cr) were also studied because these precipitates are relatively stable in extremely low pH conditions. To simplify and improve poorly ordered iron, strontium, and aluminum phase characterization, field emission scanning electron microscopy(FE-SEM), high-resolution transmission electron microscopy(HR-TEM), micro-Raman spectroscopy, and X-ray diffraction(XRD) and sequential extraction(SE) studies were executed on a set CMD samples from the Brazilian mines. 4Role of the HLA-DQ locus in the development of chronic gastritis and gastric carcinoma in Mexican patients显示文摘AIM: To determine the HLA-DQ locus in Mexican patients with Chronic gastritis and gastric adenocarcinoma. METHODS: Oligotyping for HLA-DQ locus was performed in 45 Mexican patients with chronic gastritis and 13 Mexican patients with diffuse-type gastric adenocarcinoma, and was then compared with 99 clinically healthy unrelated individuals. H pylori infection and CagA status were assessed in patients by enzyme-linked immunosor-bent assay (ELISA) method. RESULTS: We found a significant increased frequency of HLA-DQB1*0401 allele in H pylori-positive patients with chronic gastritis when compared with healthy subjects [19 vs 0%, P= 1 × 10-7, odds ratio (OR) = 4.96; 95% confidence interval (95% CI), 3.87-6.35]. We also found a significant increased frequency of HLA-DQB1*0501 in patients with diffuse-type gastric carcinoma in comparison with healthy individuals (P = 1 × 10-6, OR = 13.07; 95% CI, 2.82-85.14). 5A family study of endophenotypes for psychosis within an early intervention programme in Hong Kong: Rationale and preliminary findings显示文摘The study of endophenotypes may be a viable strategy to tackle the genetic complexity and phenotypic heterogeneity of psychosis, but this research direction is relatively under-developed in China as compared to Western countries. We have recently initiated one of the first family studies of endophenotypes for psychosis in China. Patients entering an established early psychosis intervention service are recruited into this research project for phenotyping, endophenotyping and genotyping. At the endophenotypic level, four domains (neurological soft signs, neurocognition of prospective memory, social cognition of facial emotion recognition, and affective cognition of anticipatory and consummatory pleasure) are studied in the sample of patients with psychosis and their unaffected siblings. 7Is artificial intelligence the final answer to missed polyps in colonoscopy?显示文摘Lesions missed by colonoscopy are one of the main reasons for post-colonoscopy colorectal cancer,which is usually associated with a worse prognosis.Because the adenoma miss rate could be as high as 26%,it has been noted that endoscopists with higher adenoma detection rates are usually associated with lower adenoma miss rates.Artificial intelligence(AI),particularly the deep learning model,is a promising innovation in colonoscopy.Recent studies have shown that AI is not only accurate in colorectal polyp detection but can also reduce the miss rate.Nevertheless,the application of AI in real-time detection has been hindered by heterogeneity of the AI models and study design as well as a lack of long-term outcomes.Herein,we discussed the principle of various AI models and systematically reviewed the current data on the use of AI on colorectal polyp detection and miss rates.The limitations and future prospects of AI on colorectal polyp detection are also discussed.Thomas K L Lui Wai K Leung 2020World Journal of Gastroenterology2020,26,35:2
8Hepatocellular carcinoma xenograft supports HCV replication:A mouse model for evaluating antivirals显示文摘AIM: To develop a hepatocellular carcinoma (HCC) xenograft model for studying hepatitis C virus (HCV) replication in a mice, and antiviral treatment.METHODS: We developed a stable S3-green fluorescence protein (GFP) cell line that replicated the GFP-tagged HCV sub-genomic RNA derived from a highly efficient JFH1 virus. S3-GFP replicon cell line was injected subcutaneously into γ-irradiated SCID mice. We showed that the S3-GFP replicon cell line formed human HCC xenografts in SCID mice. Cells were isolated from subcutaneous tumors and then serially passaged multiple times in SCID mice by culturing in growth medium supplemented with G-418. The mouse-adapted S3-GFP replicon cells were implanted subcutaneously and also into the liver of SCID mice via intrasplenic infusion to study the replication of HCV in the HCC xenografts. The tumor model was validated for antiviral testing after intraperitoneal injection of interferon-α (IFN-α). RESULTS: A highly tumorigenic S3-GFP replicon cell line was developed that formed subcutaneous tumors within 2 wk and diffuse liver metastasis within 4 wk in SCID mice. Replication of HCV in the subcutaneous and liver tumors was confirmed by cell colony assay, detection of the viral RNA by ribonuclease protection assay and real-time quantitative reverse transcription polymerase chain reaction. High-level replication of HCV sub-genomic RNA in the tumor could be visualized by GFP expression using fluorescence microscopy. IFN-α cleared HCV RNA replication in the subcutaneous tumors within 2 wk and 4 wk in the liver tumor model. CONCLUSION: A non-infectious mouse model allows us to study replication of HCV in subcutaneous and metastatic liver tumors.
