1Expression,deleton and mnutation of ρ16 gene in human gastric cancer显示文摘AIM To investigate the relationship between the expression of p16 gene and the gastric carcinogenesis,depth of invasion and lymph node metastases, and to evaluate the deletion and mutation of exon 2 in p16 gene in gastric carcinoma.METHODS The expression of P16 protein was examined by streptavidin-peroxidase conjugated method (S-P); the deletion and mutation of p16 gene were respectively examined by polymerase chain reaction (PCR) and polymerase chain reaction single-strand conformation polymorphism analysis (PCR-SSCP) in gastric carcinoma.RESULTS Expression of P16 protein was detected in 96.25% (77/80) of the normal gastric mucosa, in 92.00% (45/50) of the dysplastic gastric mucosa and in 47.54% (58/122) of the gastric carcinoma. The positive rate of P16 protein expression in gastric carcinoma was significantly lower than that in normal gastric mucosa and dysplastic gastric mucosa (P<0.05). The positive rate of P16 protein expression in mucoid carcinoma 10.00% (1/ 10) was significantly lower than that in poorly differentiated carcinoma 51.22% ( 21/ 41 ),undifferentiated carcinoma 57.69% (15/26) and signet ring cell carcinoma 62.50% (10/ 16) (P<0.05). The positive rate of p16 protein in 30 cases paired primary and lymph node metastatic gastric carcinoma: There was 46.67% (14/30) in primary gastric carcinoma, 16.67% (5/30) in lymph node metastatic gastric carcinoma. The positive rate of lymph node metastatic carcinoma was significantly lower than that of primary carcinoma (P<0.05). There was of p16 gene mutation in exon 2, but 5 cases displayed deletion of p16 gene in exon 2 in the 25 primary gastric carcinomas.CONCLUSIONS The expression loss of P16 protein related to the gastric carcinogenesis, gastric carcinoma histopathological subtypes and lymph metastasis. The mutation of p16 gene in exon 2 may not be involved in gastric carcinogenesis. 2Interleukin-33 drives hepatic fibrosis through activation of hepatic stellate cells显示文摘Liver fibrosis is a consequence of chronic liver disease,causing morbidity and mortality.Interleukin-33(IL-33)is a critical mediator of inflammation,which may be involved in the development of liver fibrosis.Here,we investigated the role of IL-33 in human patients and experimental bile-duct ligation(BDL)-induced fibrosis in mice.We report increased hepatic IL-33 expression in the murine BDL model of fibrosis and in surgical samples obtained from patients with liver fibrosis.Liver injury,inflammatory cell infiltration and fibrosis were reduced in the absence of the IL-33/ST2 receptor,and the activation of hepatic stellate cells(HSCs)was decreased in ST2-deficient mice.Recombinant IL-33 activated HSCs isolated from C57BL/6 mice,leading to the expression of IL-6,TGF-β,α-SMA and collagen,which was abrogated in the absence of ST2 or by pharmacological inhibition of MAPK signaling.Finally,administration of recombinant IL-33 significantly increased hepatic inflammation in sham-operated BL6 mice but did not enhance BDL-induced hepatic inflammation and fibrosis.In conclusion,BDL-induced liver inflammation and fibrosis are dependent on ST2 signaling in HSCs,and therefore,the IL-33/ST2 pathway may be a potential therapeutic target in human patients with chronic hepatitis and liver fibrosis.Zhongming Tan Qianghui Liu Runqiu Jiang Long Lv Siamak S Shoto Isabelle Maillet Valerie Quesniaux Junwei Tang Wenjie Zhang Beicheng Sun Bernhard Ryffel 2018Cellular & Molecular Immunology2018,15,4:24
3The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 4Establishment and evaluation of the system of extracorporeal liver perfusion in pigs显示文摘BACKGROUND; In recent years, extracorporeal liver per- fusion (ECLP) has been regarded as a treatment of acute liver failure ( ALF ); but the system of ECLP has many problems. The purpose of this experiment was to detect the factors affecting the system of ECLP and to establish a sta- ble and effective system of ECLP. METHODS; Livers were harvested from health pigs, ac- cording to the different styles of perfusion and oxygena- tion, which were randomly divided into 3 groups. The liv- ers in group A (n =4) were subjected to single portal vein perfusion, oxygenating perfusion blood; the livers in group B (n =4) to dual ( portal vein and hepatic artery) vessel perfusion, oxygenating blood, together; and the livers in group C ( n = 4) to dual (portal vein and hepatic artery) vessel perfusion, but oxygenating blood, separately. The perfusion time, the data of bile production, and hemody- namic parameters of extracorporeal livers in each group were tested. The histological examination of liver tissues from each group was performed at the end of perfusion. RESULTS: The perfusion time of the liver in group A is significantly shorter than in groups B and C (P<0.05). At 1 , 3 , 6 hours after perfusion, the data of bile production and hemodynamic parameters of livers in group A were sta- tistically different from those of livers in groups B and C (P<0.05). At 1, 3, 6 hours after perfusion, the data of group B were not statistically different from those of livers in group C (P>0.05). But at 12 hours after perfusion, the data of group B were statistically different from those of liv- ers in group C (P<0.01). 5Functional distinction of rat liver natural killer cells from spleen natural killer cells under normal and acidic conditions in vitro显示文摘BACKGROUND: The microenvironment within solid tumors has often been shown to exhibit an acidic extracellular pH. Although the morphologic and functional differences in natural killer (NK) cells of the liver and spleen have been reported previously under physiological conditions, the difference under acidic conditions is still unclear. This study was to investigate the differences in the morphological and functional characteristics between rat liver and spleen NK cells under normal and acidic conditions in vitro. METHODS: Liver and spleen NK cells were isolated and purified from Sprague-Dawley rats by density gradient centrifugation and the Dynabeads FlowComp TM Flexi system, and stimulated for 4 days with or without IL-2 or treated with low pH or control for different times. Morphology was examined by scanning electron microscopy (SEM) and transmission electron microscopy (TEM), cell death and proliferation assays were performed by flow cytometry, IFN-γ production was tested by ELISA, and cytotoxic activity was evaluated by lactate dehydrogenase (LDH) release assay. RESULTS: Liver NK cells had significantly higher levels of cytotoxic activity than spleen NK cells under normal and acidic conditions, and the maximum difference was observed at pH 5.6. Further analysis revealed that the cytotoxic activity of NK cells was correlated with morphology, cell death, proliferative activity and IFN-γ production. By TEM, liver NK cells contained a greater number of electron-dense granules per cell at pH 5.6.Moreover, a modest elevation of cell death and reduction of proliferation of liver NK cells occurred within a range of 5.6-7.2. Interestingly, an acidic extracellular pH only marginally, and not significantly, suppressed IFN-γ production by liver NK cells. 6Origin of pit chains in the floor of lunar Copernican craters显示文摘There are small pit chains in the floor of lunar Copernican craters. They are usually so small in scale that there are few lunar spacecrafts to detect their detailed morphology. Combining camera data from Lunar Orbiter, Lunar Reconnaissance Orbiter (LRO), Kaguya and Chang’e-1 missions, 5 representative large Copernican craters on various terrains of the lunar surface are chosen to study the origin of the pit chains in the crater floor. The morphology and distribution characteristic of the pit chains are referred by the high resolution images in this research. It is suggested that it is the magma activities from the subsurface magma layer combining with the existence of fractures and faults under the crater floor that leaded the formation of the pit chains. The model is further verified and discussed using the regolith thickness data in the crater floor. Our model suggests that the pit chains are still developing in the floor of the Copernican craters and the Moon may not be totally cold. 7Th1 cytokines promote T-cell binding to antigen-presenting cells via enhanced hyaluronan production and accumulation at the immune synapse显示文摘Hyaluronan(HA)production by dendritic cells(DCs)is known to promote antigen presentation and to augment T-cell activation and proliferation.We hypothesized that pericellular HA can function as intercellular‘glue’directly mediating T cell–DC binding.Using primary human cells,we observed HA-dependent binding between T cells and DCs,which was abrogated upon pre-treatment of the DCs with 4-methylumbelliferone(4-MU),an agent which blocks HA synthesis.Furthermore,T cells regulate HA production by DCs via T cell-derived cytokines in a T helper(Th)subset-specific manner,as demonstrated by the observation that cell-culture supernatants from Th1 but not Th2 clones promote HA production.Similar effects were seen upon the addition of exogenous Th1 cytokines,IL-2,interferon c(IFN-c)and tumor necrosis factor a(TNF-a).The critical factors which determined the extent of DC–T cell binding in this system were the nature of the pre-treatment the DCs received and their capacity to synthesize HA,as T-cell clones which were pre-treated with monensin,added to block cytokine secretion,bound equivalently irrespective of their Th subset.These data support the existence of a feedforward loop wherein T-cell cytokines influence DC production of HA,which in turn affects the extent of DC–T cell binding.We also document the presence of focal deposits of HA at the immune synapse between T-cells and APC and on dendritic processes thought to be important in antigen presentation.These data point to a pivotal role for HA in DC–T cell interactions at the IS.Paul L Bollyky Stephen P Evanko Rebecca P Wu Susan Potter-Perigo S Alice Long Brian Kinsella Helena Reijonen Kelly Guebtner Brandon Teng Christina K Chan Kathy R Braun John A Gebe Gerald T Nepom Thomas N Wight 2010Cellular & Molecular Immunology2010,7,3:3

9饲料中添加蛋白酶对肉鸡性能、养分利用率、肠道形态和胴体品质的影响显示文摘试验在饲喂玉米、高粱为主的肉鸡日粮中补充大豆,并添加蛋白酶,研究蛋白酶对肉鸡性能、养分消化率、肠道形态和胴体品质的影响。试验选取1日龄AA肉仔鸡(43.8±1.31 g)256只,均为公鸡,随机分配到2×2试验设计方阵中,方阵包括正常组(玉米组和高粱组)和蛋白酶组(空白组和添加蛋白酶150 mg·kg^(-1)的试验组)。试验1~21 d为第一阶段,22~42 d为第二阶段。试验结果表明,第二阶段中,日粮中添加蛋白酶极显著提高肉鸡的平均日增重(ADG,P<0.01),在整个试验过程中显著提高(P<0.05)。在各个时期,正常组对肉鸡生产性能均无显著影响(P>0.05)。19~21 d时,添加蛋白酶可显著提高肉鸡的表观代谢能(AME),干物质(DM)、总能量(GE)和养分消化率(P<0.05)。此外,日粮以高粱为主时,AME、GE消化率有降低趋势(P=0.05),氮沉积率也有降低趋势(P=0.08)。40~42 d时,日粮中添加蛋白酶可显著提高AME,DM、GE和氮沉积率(P<0.05)。21 d时,日粮中添加蛋白酶极显著增加肠绒毛高度、肠绒毛与隐窝深度比值,极显著降低十二指肠、空肠、回肠的隐窝深度(P<0.01)。42 d时,日粮中添加蛋白酶显著增加空肠肠绒毛高度、绒毛高度与隐窝深度比值,显著降低隐窝深度(P<0.05)。日粮中添加蛋白酶显著增加胸肌重、24 h p H,显著降低胸肌的滴水损失(P<0.05)。试验结果表明,饲养阶段日粮中添加复合蛋白酶可提高肉鸡ADG;在第一阶段可改善AME、DM、GE和氮沉积率和肠道形态。补充蛋白酶可提高出栏重、胸肌重和24 h pH,降低胸肌的滴水损失。低单宁高粱日粮与玉米日粮相比,对肉鸡性能的影响相近。Xu X Wang H L Pan L Ma X K Tian Q Y Xu Y T Long S F Zhang Z H Piao X S 2017饲料博览2017,0,1:2

14“Trends” and variations of global oceanic evaporation data sets from remote sensing显示文摘The variability in global oceanic evaporation data sets was examined for the period 1988-2000.These data sets are satellite estimates based on bulk aerodynamic formulations and include the NASA/Goddard Space Flight Center Satellite-based Surface Turbulent Flux version 2(GSSTF2),the Japanese-ocean flux using remote sensing observations(J-OFURO),and the Hamburg Ocean-Atmosphere Parameters and Fluxes from Satellite version 2(HOAPS2).The National Center for Environmental Prediction(NCEP)reanalysis is also included for comparison.An increase in global average surface latent heat flux(SLHF)can be observed in all the data sets.Empirical mode decomposition(EMD)shows long-term increases that started around 1990 for all remote sensing data sets.The effect of Mt.Pinatubo eruption in 1991 is clearly evident in HOAPS2 but is independent of the long-term increase.Linear regression analyses show increases of 9.4%,13.0%,7.3%,and 3.9% for GSSTF2,J-OFURO,HOAPS2 and NCEP,for the periods of the data sets.Empirical orthogonal function(EOF)analyses show that the pattern of the first EOF of all data sets is consistent with a decadal variation associated with the enhancement of the tropical Hadley circulation,which is supported by other satellite observations.The second EOF of all four data sets is an ENSO mode,and the correlations between their time series and an SOI are 0.74,0.71,0.59,and 0.61 for GSSTF2,J-OFURO,HOAPS2,and NCEP in that order.When the Hadley modes are removed from the remote sensing data,the residue global increases are reduced to 2.2%,7.3%,and <1% for GSSTF2,J-OFURO and HOAPS,respectively.If the ENSO mode is used as a calibration standard for the data sets,the Hadley mode is at least comparable to,if not larger than,the ENSO mode during our study period.CHIU Long S CHOKNGAMWONG R XING Yukun YANG Ruixin SHIE Chung-Lin 2008Acta Oceanologica Sinica2008,27,3:2

18Digital spiral object identification using random light显示文摘Photons that are entangled or correlated in orbital angular momentum have been extensively used for remote sensing,object identification and imaging.It has recently been demonstrated that intensity fluctuations give rise to the formation of correlations in the orbital angular momentum components and angular positions of random light.Here we demonstrate that the spatial signatures and phase information of an object with rotational symmetries can be identified using classical orbital angular momentum correlations in random light.The Fourier components imprinted in the digital spiral spectrum of the object,as measured through intensity correlations,unveil its spatial and phase information.Sharing similarities with conventional compressive sensing protocols that exploit sparsity to reduce the number of measurements required to reconstruct a signal,our technique allows sensing of an object with fewer measurements than other schemes that use pixel-by-pixel imaging.One remarkable advantage of our technique is that it does not require the preparation of fragile quantum states of light and operates at both low-and high-light levels.In addition,our technique is robust against environmental noise,a fundamental feature of any realistic scheme for remote sensing.Zhe Yang Omar S Magaña-Loaiza Mohammad Mirhosseini Yiyu Zhou Boshen Gao Lu Gao Seyed Mohammad Hashemi Rafsanjani Gui-Lu Long Robert W Boyd 2017Light(Science & Applications)2017,6,1:2
