| 2 | Inhalation treatment of primary lung cancer using liposomal curcumin dry powder inhalers显示文摘Lung cancer is the leading cause of cancer-related deaths. Traditional chemotherapy causes serious toxicity due to the wide bodily distribution of these drugs. Curcumin is a potential anticancer agent but its low water solubility, poor bioavailability and rapid metabolism significantly limits clinical applications. Here we developed a liposomal curcumin dry powder inhaler(LCD) for inhalation treatment of primary lung cancer. LCDs were obtained from curcumin liposomes after freeze-drying. The LCDs had a mass mean aerodynamic diameter of 5.81 μm and a fine particle fraction of 46.71%, suitable for pulmonary delivery. The uptake of curcumin liposomes by human lung cancer A549 cells was markedly greater and faster than that of free curcumin. The high cytotoxicity on A549 cells and the low cytotoxicity of curcumin liposomes on normal human bronchial BEAS-2B epithelial cells yielded a high selection index partly due to increased cell apoptosis. Curcumin powders, LCDs and gemcitabine were directly sprayed into the lungs of rats with lung cancer through the trachea. LCDs showed higher anticancer effects than the other two medications with regard to pathology and the expression of many cancer-related markers including VEGF, malondialdehyde, TNF-α, caspase-3 and BCL-2. | 3 | Influence of magma intrusion on gas outburst in a low rank coal mine显示文摘The effect of magma intrusion on gas outburst is illustrated by a case study of the exposed magma intrusion in the 313 mining area,upper coal seam Number 3,in the Qiwu Mine located in Shandong province.Vitrinite reflectance,mercury injection,and maceral statistical analysis are used to characterize the coal.The aspects of coal metamorphism include changes in micro-components as well as in coal structure,the formation of new substances,and changes in gas absorption and storage.The results show that vitrinite reflectance increases within the region influenced by magma intrusion.The metamorphosed region may be divided into a weakly affected belt,a medium affected belt,a strongly affected belt,and a completely affected belt.Compared to the unaffected coal the total pore volume,as well as the amount of big and middle sized holes,increases while the number of transition holes and micro-pores decreases.This diminishes the absorption capacity of the matrix but enlarges the total gas storage space.Vitrinite content initially decreases slightly but then increases rapidly while the inertinite content increases at first but then decreases.Exinite content decreases,then increases,and finally drops to zero.Higher vitrinite,and a lower inertinite,content increase gas absorption ability.This balances reduced adsorption caused by changes to pore structure.Consequently,gas adsorption capacity is not substantially reduced as the coal rank increases.Thermal metamorphism of the coal produces CH4 and other hydrocarbons that increase the total gas content in the coal seam.Asphaltene migrates into the medium and weakly affected regions filling in the pores and fractures there.This plugs the pathway for gas transport.A barrier is formed that hinders gas flow.CO2,H2S,N2,and other gases carried in by the magma react to produce CO2,which increases in relative concentration and enhances the risk of gas outburst.The two barriers,magma intrusion on one side and the medium and weakly affected belts on the other,as well as the unaffected coal seam itself,trap a large amount of gas during the thermal activity.This is the basic reason for gas outburst.These conclusions can enlighten activities related to gas prevention and control in a low rank coal mine affected by magma intrusion. | Chen Shangbin Zhu Yanming Li Wu Wang Hui | 2012 | International Journal of Mining Science and Technology2012,22,2: | 11 |
| 4 | Effect of initial material bulk density and easily-degraded organic matter content on temperature changes during composting of cucumber stalk显示文摘To inactivate the potentially pathogenic microorganisms and safely utilize vegetable waste compost, ultra-high temperatures(> 70°C) should be maintained during the composting without having an inhibitory effect on maturity. This study investigated the influence of bulk density(part 1) and easily-degraded organic matter content(EDOMC, part 2) on temperature evolution during vegetable waste composting: Part 1: corn straw with different particle sizes was used to achieve different bulk densities in the composting material(BD1–BD3); Part 2: partial or total substitution of the corn straw by corn starch was carried out to obtain different EDOMC(ED1–ED4). The composting experiments were conducted in a labscale reactor(1.75 kg material) and lasted for 30 d. Temperature and CO2 emission were recorded daily, and the organic matter, lignocellulose, microbial activity, germination index(GI) and C/N of the samples were measured at different stages. The highest temperature(65.7°C) in part 1 occurred in the treatment with the bulk density of 0.35 g/cm3, which also had the longest thermophilic phase. Bulk density was found to seriously influence the utilization efficiency of O2 and heat transfer through materials, rather than heat production from organic matter degradation. In experiment part 2, the highest temperature was obtained with EDOMC of 45%(71.4°C). | 6 | Effect of naringenin in Qianggu capsule on population pharmacokinetics in Chinese women with primary osteoporosis显示文摘OBJECTIVE:To characterize naringenin(NAR) population pharmacokinetics(PPK) in Chinese women with primary osteoporosis.METHODS:Ninety-eight female patients with primary osteoporosis from the Jingshan,Beixinqiao,Jiaodaokou,Chaoyangmen,and Donghuamen communities in Beijing,China,aged 40 to 80 years,received oral Qianggu capsules(250 mg).Blood samples were collected before and at 0.5,1,2,3,4,6,8,10,12,and 24 h after administration.The concentration of NAR in the blood samples was measured using high performance liquid chromatography-tandem mass spectrometry.PPK analyses were performed with nonlinear mixed-effect modeling software(version 7.1.2,PsN3.2.12).The clearance(C1),central distribution volume(V),absorption rate constant(Ka1),peripheral distribution volume(VII),and inter-compartmental clearance(CLII) were set as parameters and estimated by the base model,covariate model,and final model.Kidney-Yang deficiency[Shenyangxu(SYAX)]and liver-kidney-Yin deficiency(Ganshenyinxu) are patterns of symptoms in Traditional Chinese Medicine that were set as covariates,along with age,height,blood urea nitrogen,serum creatinine,alanine transaminase,aspartate transaminase,and hyperlipidemia.Both stepwise forward and backward procedures were accomplished to build models.The final model was evaluated by internal and external validation,visual predictive check,bootstrap,and leverage analysis.RESULTS:A one compartment open model with first order degradation was the best fitted to the concentration-time profiles following oral administration of NAR.The mean of population parameters of the final model,C1,SYAX on C1,V,Ka1,CLII,and VII,were measured to be 37.6 L/h,0.427 L,123 L/h,0.12/h,0.3056,and 1.446,respectively.Inter-individual variability was estimated and SYAX was identified as a significant covariate.CONCLUSION:The population pharmacokinetic model described in this study could effectively characterize the pharmacokinetic profile of NAR following administration of a single dose of oral Qianggu capsules in Chinese women with primary osteoporosis.Among the tested covariates,only SYAX was a significant factor. | Wang Jiannong Jiang Junjie Xie Yanming Wei Xu Li Jianpeng Duan Jingli Xiong Xin | 2015 | Journal of Traditional Chinese Medicine2015,35,2: | 9 |
| 7 | Precise base editing of non-allelic acetolactate synthase genes confers sulfonylurea herbicide resistance in maize显示文摘Single-nucleotide polymorphisms contribute to phenotypic diversity in maize. Creation and functional annotation of point mutations has been limited by the low efficiency of conventional methods based on random mutation. An efficient tool for generating targeted single-base mutations is desirable for both functional genomics and precise genetic improvement. The objective of this study was to test the efficiency of targeted C-to-T base editing of two non-allelic acetolactate synthase(ALS) in generating sulfonylurea herbicide-resistant mutants. A CRISPR/Cas9 nickase-cytidine deaminase fused with uracil DNA glycosylase inhibitor(UGI) was employed to achieve targeted conversion of cytosine to thymine in ZmALS1 and ZmALS2. Both protoplasts and recovered mutant plants showed the activity of the cytosine base editor, with an in vivo efficiency of up to 13.8%. Transgene-free edited plants harboring a homozygous ZmALS1 mutation or a ZmALS1 and ZmALS2 double mutation were tested for their resistance at a dose of up to 15-fold the recommended limit of chlorsulfuron, a sulfonylurea herbicide widely used in agriculture. | 9 | Fluxes of methane,carbon dioxide and nitrous oxide in an alpine wetland and an alpine grassland of the Tianshan Mountains,China显示文摘Methane(CH4), carbon dioxide(CO2) and nitrous oxide(N2O) are known to be major greenhouse gases that contribute to global warming. To identify the flux dynamics of these greenhouse gases is, therefore, of great significance. In this paper, we conducted a comparative study on an alpine grassland and alpine wetland at the Bayinbuluk Grassland Eco-system Research Station, Chinese Academy of Sciences. By using opaque, static, manual stainless steel chambers and gas chromatography, we measured the fluxes of CH4, N2O and CO2 from the grassland and wetland through an in situ monitoring study from May 2010 to October 2012. The mean flux rates of CH4, N2O and CO2 for the experimental alpine wetland in the growing season(from May to October) were estimated at 322.4 μg/(m2?h), 16.7 μg/(m2?h) and 76.7 mg/(m2?h), respectively; and the values for the alpine grassland were –88.2 μg/(m2?h), 12.7 μg/(m2?h), 57.3 mg/(m2?h), respectively. The gas fluxes showed large seasonal and annual variations, suggesting weak fluxes in the non-growing season. The relationships between these gas fluxes and environmental factors were analyzed for the two alpine ecosystems. | 12 | Carbon Storage and Vertical Distribution in Three Shrubland Communities in Gurbantünggüt Desert,Uygur AutonomousRegion of Xinjiang,Northwest China显示文摘This study was carried out in the Gurbantünggüt Desert,Uygur Autonomous Region of Xinjiang,Northwest China in August,2009.To quantify the storage,contribution and vertical distribution patterns of plant biomass carbon (PBC)and soil organic carbon(SOC)in the study area,we investigated the carbon concentrations and its vertical distribution in three different desert shrubland communities dominated by Reaumuria soongorica,Haloxylon ammodendron+R.soongorica and Tamarix ramosissima+R.soongorica,respectively.We analyzed vertical distribution of root biomass carbon and soil carbon contents by excavating soil profiles for each dominated community.The results show that SOC is considerably the larger carbon pool in the soil layers of 1.0-3.0 m(the mean value of three shrubland communities is 38.46%)and 3.0-5.0 m(the mean value is 40.24%).In contrast,70.74%of belowground biomass carbon storage in 0-1.0 m layer,and its content decrease with increasing soil depth.The Haloxylon ammodendron+R. soongorica shrubland community has the highest belowground biomass carbon among three selected communities. | 13 | Characterization of miRNomes in Acute and Chronic Myeloid Leukemia Cell Lines显示文摘Myeloid leukemias are highly diverse diseases and have been shown to be associated with microRNA(miRNA) expression aberrations. The present study involved an in-depth miRNome analysis of two human acute myeloid leukemia(AML) cell lines, HL-60 and THP-1, and one human chronic myeloid leukemia(CML) cell line, K562, via massively parallel signature sequencing. mRNA expression profiles of these cell lines that were established previously in our lab facilitated an integrative analysis of miRNA and mRNA expression patterns. miRNA expression profiling followed by differential expression analysis and target prediction suggested numerous miRNA signatures in AML and CML cell lines. Some miRNAs may act as either tumor suppressors or oncomiRs in AML and CML by targeting key genes in AML and CML pathways. Expression patterns of cell type-specific miRNAs could partially reflect the characteristics of K562, HL-60 and THP-1 cell lines, such as actin filament-based processes, responsiveness to stimulus and phagocytic activity. miRNAs may also regulate myeloid differentiation, since they usually suppress differentiation regulators. Our study provides a resource to further investigate the employment of miRNAs in human leukemia subtyping, leukemogenesis and myeloid development. | 14 | Pore Structure Heterogeneity of the Xiamaling Formation Shale Gas Reservoir in the Yanshan Area of China: Evaluation of Geological Controlling Factors显示文摘Micro-heterogeneity is an integral parameter of the pore structure of shale gas reservoir and it forms an essential basis for setting and adjusting development parameters.In this study,scanning electron microscopy,high-pressure mercury intrusion and low-temperature nitrogen adsorption experiments were used to qualitatively and quantitatively characterize the pore structure of black shale from the third member of the Xiamaling Formation in the Yanshan area.The pore heterogeneity was studied using fractal theory,and the controlling factors of pore development and heterogeneity were evaluated in combination with geochemical parameters,mineral composition,and geological evolution history.The results show that the pore structure of the reservoir was intricate and complicated.Moreover,various types of micro-nano scale pores such as dissolution pores,intergranular pores,interlayer pores,and micro-cracks are well developed in member 3 of the Xiamaling Formation.The average porosity was found to be 6.30%,and the mean value of the average pore size was 4.78 nm.Micropores and transition pores provided most of the storage space.Pore development was significantly affected by the region and was mainly related to the total organic carbon content,vitrinite reflectance and mineral composition.The fractal dimension,which characterizes the heterogeneity,is 2.66 on average,indicating that the pore structure is highly heterogeneous.Fractal dimension is positively correlated with maturity and clay mineral content,while it is negatively correlated with brittle mineral content and average pore size.These results indicate that pore heterogeneity is closely related to thermal history and material composition.Combined with the geological background of this area,it was found that the pore heterogeneity was mainly controlled by the Jurassic magmatism.The more intense the magma intrusion,the stronger the pore heterogeneity.The pore structure and its heterogeneity characteristics present today are a general reflection of the superimposed geological processes of sedimentary-diagenetic-late transformation.The influence of magmatic intrusion on the reservoir is the main geological factor that should be considered for detailed evaluation of the Xiamaling Formation shale gas reservoir in the Yanshan area. | LI Xueyuan CHEN Shangbin WANG Xiaoqi ZHU Yanming CHANG Minghua Clementine UWAMAHORO | 2019 | Acta Geologica Sinica(English Edition)2019,93,3: | 5 |

The result showed that the semi - exchange time was about more than 6 d in the bay end, and about 1~2 d in the bay mouth. Besides, based on the calculation of the semi - exchange time before and after the Xuanmen Dam pro- ject, a comparison between them was further carried out. And the same work was also done with the recent reclamation projects in the Yueqing Bay as well. The results showed that the change in semi - exchange time caused by the Xuanmen Dam project was a- bout 6 d increase near the dam and 4.5 d increase at the bay end. | 17 | Lock threshold deterioration induced by antenna vibration and signal coupling effects in hypersonic vehicle carrier tracking system of Ka band显示文摘The envelope of a hypersonic vehicle is affected by severe fluctuating pressure, which causes the airborne antenna to vibrate slightly. This vibration mixes with the transmitted signals and thus introduces additional multiplicative phase noise. Antenna vibration and signal coupling effects as well as their influence on the lock threshold of the hypersonic vehicle carrier tracking system of the Ka band are investigated in this study. A vibration model is initially established to obtain phase noise in consideration of the inherent relationship between vibration displacement and electromagnetic wavelength. An analytical model of the Phase-Locked Loop(PLL), which is widely used in carrier tracking systems, is established. The coupling effects on carrier tracking performance are investigated and quantitatively analyzed by imposing the multiplicative phase noise on the PLL model. Simulation results show that the phase noise presents a Gaussian distribution and is similar to vibration displacement variation. A large standard deviation in vibration displacement exerts a significant effect on the lock threshold. A critical standard deviation is observed in the PLL of Binary Phase Shift Keying(BPSK) and Quadrature Phase Shift Keying(QPSK) signals. The effect on QPSK signals is more severe than that on BPSK signals. The maximum tolerable standard deviations normalized by the wavelength of the carrier are 0.04 and 0.02 for BPSK and QPSK signals,respectively. It makes the protein obtain more complicated structures, perfect functions, more accurate regulations and more spe-cific operations. The most common protein post- translational modifications include ubiquitylation, phosphorylation, glycosylation, lipodation, methyla-tion, and acetylation and so on. Ubiquitylation plays an essential role in cellular functions such as cellular differentiation, apoptosis, DNA repair, antigen proc-essing, and stress response. Phosphorylation is re-lated to physiological and pathological processes including cellular signal conduction, nervous activity, muscle contraction and proliferation, development and differentiation of cells. Protein glycosylation is of great importance for many cell processes like im-munoprotection, virus replication, cell growth, and occurrence of inflammation and so on. Lipodation is vital to signal conduction. Histone methylation and acetylation are responsible for transcription regula-tion. In vivo, different post-translational modifications do not occur isolatedly, but influence each other’s function and cooperate with each other. | 20 | Targeting PFKL with penfluridol inhibits glycolysis and suppresses esophageal cancer tumorigenesis in an AMPK/FOXO3a/BIM-dependent manner显示文摘As one of the hallmarks of cancer,metabolic reprogramming leads to cancer progression,and targeting glycolytic enzymes could be useful strategies for cancer therapy.By screening a small molecule library consisting of 1320 FDA-approved drugs,we found that penfluridol,an antipsychotic drug used to treat schizophrenia,could inhibit glycolysis and induce apoptosis in esophageal squamous cell carcinoma(ESCC).Gene profiling and Ingenuity Pathway Analysis suggested the important role of AMPK in action mechanism of penfluridol.By using drug affinity responsive target stability(DARTS)technology and proteomics,we identified phosphofructokinase,liver type(PFKL),a key enzyme in glycolysis,as a direct target of penfluridol.Penfluridol could not exhibit its anticancer property in PFKL-deficient cancer cells,illustrating that PFKL is essential for the bioactivity of penfluridol.High PFKL expression is correlated with advanced stages and poor survival of ESCC patients,and silencing of PFKL significantly suppressed tumor growth.Mechanistically,direct binding of penfluridol and PFKL inhibits glucose consumption,lactate and ATP production,leads to nuclear translocation of FOXO3a and subsequent transcriptional activation of BIM in an AMPK-dependent manner.Taken together,PFKL is a potential prognostic biomarker and therapeutic target in ESCC,and penfluridol may be a new therapeutic option for management of this lethal disease. | Cancan Zheng Xiaomei Yu Yiyao Liang Yidong Zhu Yan He Long Liao Dingkang Wang Yanming Yang Xingfeng Yin Ang Li Qingyu He Bin Li | 2022 | Acta Pharmaceutica Sinica B2022,12,3: | 4 |
