1In situ boron isotope measurements of natural geological materials by LA-MC-ICP-MS显示文摘There are two main methods to determine boron isotopic composition.One is the solution method,in which boron is purified after the samples are dissolved in solution and the boron isotope ratios are determined by thermal ionization mass spectrometry(P-TIMS and N-TIMS) or multicollector inductively coupled plasma mass spectrometry(MC-ICP-MS).The other is an in-situ analysis method,in which the in-situ boron isotopic ratios in minerals are analyzed directly using secondary ion mass spectrometry(SIMS) or laser ablation multicollector inductively coupled plasma mass spectrometry(LA-MC-ICP-MS).In the in-situ analysis method for boron isotopes,the multifarious chemical purification and separation processes of the solution method are avoided,with increased work efficiency.In addition,the microzones and microbeddings of minerals can be analyzed in-situ to reveal the fine processes and conditions of mineral formation.In this study,using the standard-sample-bracketing(SSB) method,mass bias of the instrument and the fractionation of isotopes were calibrated,and the in-situ determination method of LA-MC-ICP-MS for boron isotopes was established.Through detailed analyses on a series of boron isotope standards and samples,a matrix effect was assessed but not detected,and the analysis results were in accordance with the formerly reported values or P-TIMS determined values,within the error range.The analytical results for IAEA B4 and IMR RB1 with relatively high boron contents were δ 11B =-(8.36±0.58)‰(2σ,n=50) and δ 11B =-(12.96±0.97)‰(2σ,n=57),respectively;the analytical result for IAEA B6 with rela-tively low boron content was δ 11B =-(3.29±1.12)‰(2σ,n=35).In-situ measurements for B isotopes were performed on geo-logical samples such as tourmaline,ulexite,ludwigite,inyoite and ascharite,with the results consistent with those determined by P-TIMS,within the error range.HOU KeJun LI YanHe XIAO YingKai LIU Feng TIAN YouRong 2010Chinese Science Bulletin2010,55,29:13
2Helium Isotope Geochemistry of Ultrahigh-Pressure Metamorphic Eclogites From the Dabie-Sulu Terrane in East China显示文摘The 3He/4He ratios of most eclogites from the Dabie-Sulu terrane range from 0.056 to 0.67 Ra; the data points fall into the mixing part of the crust and the mantle in the 3He-4He diagram. The 3He/4He ratios of eclogites are obviously correlated with the types of their surrounding rocks. The helium isotope composition of the eclogites from the Bixiling complex possesses characters of mantle-derived rocks with the 3He/4He ratio being 5.6 Ra. The 4He concentration of the eclogites exhibits visible inverse correlation with the δ18O value of the quartz in the eclogites from the Sulu area. The δ18O values of the eclogites change synchronously with those of the country rocks. 3Late Mesozoic Ore-forming Events in the Ningwu Ore District, Middle-Lower Yangtze River Polymetallic Ore Belt, East China: Evidence from Zircon U-Pb Geochronology and Hf Isotopic Compositions of the Granodioritic Stocks显示文摘Late Mesozoic volcanic-subvolcanic rocks and related iron deposits, known as porphyry iron deposits in China, are widespread in the Ningwu ore district (Cretaceous basin) of the middle-lower Yangtze River polymetallic ore belt, East China. Two types of Late Mesozoic magmatic rocks are exposed: one is dioritic rocks closely related to iron mineralization as the hosted rock, and the other one is granodioritic (-granitic) rocks that cut the ore bodies. To understand the age of the iron mineralization and the ore-forming event, detailed zircon U-Pb dating and Hf isotope measurement were performed on granodioritic stocks in the Washan, Gaocun-Nanshan, Dongshan and Heshangqiao iron deposits in the basin. Four emplacement and crystallization (typically for zircons) ages of granodioritic rocks were measured as 126.1±0.5 Ma, 126.8±0.5 Ma, 127.3±0.5 Ma and 126.3±0.4 Ma, respectively in these four deposits, with the LA-MC-ICP-MS zircon U-Pb method. Based on the above results combined with previous dating, it is inferred that the iron deposits in the Ningwu Cretaceous basin occurred in a very short period of 131-127 Ma. In situ zircon Hf compositions of εHf(t) of the granodiorite are mainly from -3 to -8 and their corresponding 176Hf/177Hf ratio are from 0.28245 to 0.28265, indicating similar characteristics of dioritic rocks in the basin. We infer that granodioritic rocks occurring in the Ningwu ore district have an original relationship with dioritic rocks. 5Separation and Precise Measurement of Lithium Isotopes in Three Reference Materials Using Multi Collector-Inductively Coupled Plasma Mass Spectrometry显示文摘Lithium separation technique for three reference materials has been established together with precise determination of lithium isotope using a Neptune multi collector-inductively coupled plasma mass spectrometry (MC-ICP-MS). The solutions of lithium element standard reference materials, potassium, calcium, sodium, magnesium and iron single element, were used to evaluate analytical methods applied. Three separate stages of ion-exchange chromatography were carried out using organic cation-exchange resin (AG 50W-X8). Lithium was enriched for the three stages using different eluants, which are 2.8 M HCl, 0.15 M HCl and 0.5 M HCl in 30% ethanol, respectively. The columns for the first and second stages are made of polypropylene, and those for the third stage are made of quartz. Total reagent volume for the entire chemical process was 35 mL for three reference materials. The recovery yielded for the three stages is 98.9-101.2% with an average of 100.0%, 97.6- 101.9% with an average of 99.9%, and 99.8-103.3% with an average of 100.6%, respectively. The precision of this technique is conservatively estimated to be ±0.72-1.04‰ (2σ population), which is similar to the precision obtained by different authors in different laboratories with MC-ICP-MS. The δ 7 Li values ( 7 Li/ 6 Li relative to the IRMM-016 standard) determined for andesite (AGV-2) and basalt (BHVO-2) are 5.68‰ (n=18), 4.33‰ (n=18), respectively. The δ 7 Li value ( 7 Li/ 6 Li relative to the L-SVEC standard) determined for IRMM-016 is -0.01‰ (n=15). All these analytical results are in good agreement with those previously reported. In addition, the results for the same kinds of samples analyzed at the MLR Key Laboratory of Metallogeny and Mineral Assessment, Institute of Mineral Resources, Chinese Academy of Geological Sciences, are consistent with those obtained at the Plasma Laboratory, University of Maryland, within analytical uncertainty. 8Occurrence of the Iron–rich Melt in the Heijianshan Iron Deposit, Eastern Tianshan, NW China: Insights into the Origin of Volcanic Rock–hosted Iron Deposits显示文摘Long-standing controversy persists over the presence and role of iron–rich melts in the formation of volcanic rock-hosted iron deposits. Conjugate iron–rich and silica–rich melt inclusions observed in thin-sections are considered as direct evidence for the presence of iron-rich melt, yet unequivocal outcrop-scale evidence of iron-rich melts are still lacking in volcanic rock-hosted iron deposits. Submarine volcanic rock-hosted iron deposits, which are mainly distributed in the western and eastern Tianshan Mountains in Xinjiang, are important resources of iron ores in China, but it remains unclear whether iron-rich melts have played a role in the mineralization of such iron ores. In this study, we observed abundant iron-rich agglomerates in the brecciated andesite lava of the Heijianshan submarine volcanic rock–hosted iron deposit, Eastern Tianshan, China. The iron-rich agglomerates occur as irregular and angular masses filling fractures of the host brecciated andesite lava. They show concentric potassic alteration with silicification or epidotization rims, indicative of their formation after the wall rocks. The iron-rich agglomerates have porphyritic and hyalopilitic textures, and locally display chilled margins in the contact zone with the host rocks. These features cannot be explained by hydrothermal replacement of wall rocks(brecciated andesite lava) which is free of vesicle and amygdale, rather they indicate direct crystallization of the iron-rich agglomerates from iron-rich melts. We propose that the iron-rich agglomerates were formed by open-space filling of volatile-rich iron-rich melt in fractures of the brecciated andesite lava. The iron-rich agglomerates are compositionally similar to the wall-rock brecciated andesite lava, but have much larger variation. Based on mineral assemblages, the iron-rich agglomerates are subdivided into five types, i.e., albite-magnetite type, albite-K-feldsparmagnetite type, K-feldspar–magnetite type, epidote-magnetite type and quartz-magnetite type, representing that products formed at different stages during the evolution of a magmatic-hydrothermal system. The albite-magnetite type represents the earliest crystallization product from a residual ironrich melt; the albite-K-feldspar-magnetite and K-feldspar-magnetite types show features of magmatichydrothermal transition, whereas the epidote-magnetite and quartz-magnetite types represent products of hydrothermal alteration. The occurrence of iron-rich agglomerates provides macroscopic evidence for the presence of iron-rich melts in the mineralization of the Heijianshan iron deposit. 10N and O isotopes and the ore-forming mechanism of nitrate deposits in the Turpan-Hami Basin,Xinjiang,China显示文摘The recently discovered nitrate ore field in the Turpan-Hami Basin of western China represents an estimated resource of 2.5 billion tons, and is comparable in scale to the Atacama Desert super-scale nitrate deposit in Chile. The research on this area is rarely carried out, and the origin of the deposits remains uncertain. In this study, new methods were used to systematically analyze N and O isotopes in nitrate minerals collected from the Kumutage, Xiaocaohu, Wuzongbulak, Dawadi, Tuyugou, and Shaer ore deposits in the Turpan-Hami Basin. The data showed that the δ15NAir value ranges from 0.7‰ to 27.6‰, but mostly between 2‰ and 6‰, which was similar to atmospheric NO3-. The 18O was highly enriched with δ18OV-SMOW varying from 30.2‰ to 46.7‰. This differs from levels in deposits derived from microbial nitrogen fixation, but is similar to those in atmospheric nitrates. N and O isotopes data indicated that nitrate deposits in Turpan-Hami Basin must be the result of deposition of atmospheric nitrate particles. Although atmospheric nitrate particles are common, the nitrate deposits could form only under the condition of long-term extreme drought climate and very limited biological activity. 12Extreme Enrichment of Tellurium in Deep-Sea Sediments显示文摘Tellurium is a sort of scattered rare element on the earth. Its concentration is very low in earth’s crust, only 1.0 ng/g. However, it has extremely high abundance in Co-rich crusts, marine polymetallic nodules, deep-sea sediments and aerolites. To find out the origin of tellurium enrichment in deep-sea sediments, we analyzed and compared tellurium concentrations and helium isotope compositions in the magnetic parts and those in the bulk parts of deep-sea sediments. The result indicates that the helium content, 3He/4He ratio and tellurium concentration are obviously higher in the magnetic parts than those in the bulk parts. The 3He abundance varies synchronously with the tellurium concentration. 3He and Te have a distinct positive correlation with each other. It is the first time that the paper brings forward that the extreme enrichment of tellurium in deep-sea sediments, like helium isotope anomalies, probably results from the input of interplanetary dust particles (IDPs). 13The origin and evolution of the diosgenin biosynthetic pathway in yam显示文摘Diosgenin,mainly produced by Dioscorea species,is a traditional precursor of most hormonal drugs in the pharmaceutical industry.The mechanisms that underlie the origin and evolution of diosgenin biosynthesis in plants remain unclear.After sequencing the whole genome of Dioscorea zingiberensis,we revealed the evolutionary trajectory of the diosgenin biosynthetic pathway in Dioscorea and demonstrated the de novo biosynthesis of diosgenin in a yeast cell factory.First,we found that P450 gene duplication and neofunctionalization,driven by positive selection,played important roles in the origin of the diosgenin biosynthetic pathway.Subsequently,we found that the enrichment of diosgenin in the yam lineage was regulated by CpG islands,which evolved to regulate gene expression in the diosgenin pathway and balance the carbon flux between the biosynthesis of diosgenin and starch.Finally,by integrating genes fromplants,animals,and yeast,weheterologously synthesized diosgenin to 10mg/l in genetically-engineered yeast.Our study not only reveals the origin and evolutionary mechanisms of the diosgenin biosynthetic pathway in Dioscorea,but also introduces an alternative approach for the production of diosgenin through synthetic biology.Jian Cheng Jing Chen Xiaonan Liu Xiangchen Li Weixiong Zhang Zhubo Dai Lina Lu Xiang Zhou Jing Cai Xueli Zhang Huifeng Jiang Yanhe Ma 2021Plant Communications2021,2,1:2

The proteomes were compared to determine differentially expressed proteins. A novel alkaliphilic bacterium (alkaliphilic Bacillus sp. N16-5 isolated from Wudunur Soda Lake, China) was isolated in media with five different carbon sources (glucose, mannose, galactose, arabinose, and xylose). Comparative proteome analysis identified 61 differentially expressed proteins, which were mainly involved in carbohydrate metabolism, amino acid transport, and metabolism, as well as energy production and conversion. The comparison was based on the draft genome sequence of strain N16-5. The abundance of enzymes involved in central metabolism was significantly changed when exposed to various carbohydrates. Notably, catabolite control protein A (CcpA) was up-regulated under all carbon sources compared with glucose. In addition, pentose exhibited a stronger effect than hexose in CcpA-mediated carbon catabolite repression.
