1永兴岛白穗软珊瑚共附生放线菌筛选及部分活性次级代谢产物的鉴定显示文摘【目的】从白穗软珊瑚中分离和鉴定共附生放线菌,运用PCR技术对所分离的放线菌进行I型聚酮合酶(PKS)筛选,研究其次级代谢产物。【方法】使用11种培养基对白穗软珊瑚共附生放线菌进行分离、鉴定,构建16S rRNA基因系统发育进化树,以基于I型PKS的KS基因设计的简并引物对所分离放线菌进行基因筛选,对阳性菌株用3种培养基发酵检测,对目标菌株进行放大规模发酵分离鉴定次级代谢产物。【结果】从白穗软珊瑚中分离到20株共附生放线菌,包括链霉菌属10株、迪茨氏菌属2株和盐水孢菌属8株,筛选获得18株I型PKS阳性菌株,并从菌株Salinospora arenicola SH04中分离到化合物rifamycin S和rifamycin W。【结论】首次从珊瑚共附生环境中分离得到海洋专属性稀有放线菌盐水孢菌属,并以I型PKS基因筛选为指导,分离鉴定了聚酮类化合物rifamycins,为研究软珊瑚共附生可培养放线菌的多样性和基于基因筛选指导分离次级代谢产物提供了可靠依据。马亮 张文军 朱义广 吴正超 Kumar Saurav 黄晖 张长生 2013微生物学报2013,53,10:10
2Biosorption of Cd (Ⅱ) and Pb (Ⅱ) Ions by Aqueous Solutions of Novel Alkalophillic Streptomyces VITSVK5 spp. Biomass显示文摘Discharge of heavy metals from metal processing industries is known to have adverse effects on the environment.Bio-sorption of heavy metals by metabolically inactive biomass of microbial organisms is an innovative and alternative technology for removal of these pollutants from aqueous solution.The search of marine actinobacteria with potential heavy metal biosorption ability resulted in the identification of a novel alkalophilic Streptomyces VITSVK5 species.The biosorption property of Streptomyces VITSVK5 spp.was investigated by absorbing heavy metals Cadmium (Cd) and Lead (Pb).Physiochemical characteristics and trace metal concentration analysis of the backwater showed the concentrations of different metals were lead 13±2.1 μg L-1,cadmium 3.1±0.3μg L-1,zinc 8.4±2.6μg L-1 and copper 0.3±0.1μg L-1,whereas mercury was well below the detection limit.The effect of pH and biomass dosage on removal efficiency of heavy metal ions was also investigated.The optimum pH for maximal biosorption was 4.0 for Cd (II) and 5.0 for Pb (II) with 41% and 84% biosorption respectively.The biosorbent dosage was optimized as 3 g L-1 for both the trace metals.Fourier transform infrared absorption spectrum results indicated the chemical interactions of hydrogen atoms in carboxyl (–COOH),hydroxyl (–CHOH) and amine (–NH2) groups of biomass with the metal ions.This could be mainly involved in the biosorption of Cd (II) and Pb (II) onto Streptomyces VITSVK5 spp.The results of our study revealed Streptomyces metabolites could be used to develop a biosorbent for adsorbing metal ions from aqueous environments.Kumar Saurav Krishnan Kannabiran 2011Journal of Ocean University of China2011,10,1:6
3Biosorption of Cr(Ⅳ),Cr(Ⅲ),Pb(Ⅱ) and Cd(Ⅱ) from Aqueous Solutions by Sargassum wightii and Caulerpa racemosa Algal Biomass显示文摘Heavy metal pollution is one of the most important environmental problems today.Biosorption is an innovative tech-nology that employs biological materials to accumulate heavy metals from waste water through metabolic process or physicochemi-cal pathways of uptake.Even though several physical and chemical methods are available for removal of heavy metals,currently many biological materials such as bacteria,algae,yeasts and fungi have been widely used due to their good performance,low cost and large quantity of availability.The aim of the present study is to explore the biosorption of toxic heavy metals,Cr(VI),Cr(III),Pb(II) and Cd(II) by algal biomass obtained from algae Sargassum wightii(brown) and Caulerpa racemosa(green).Biosorption of algal biomass was found to be biomass concentration-and pH-dependent,while the maximal biosorption was found at pH 5.0 and with the metal concentration of 100 mg L-1.S.wightii showed the maximal metal biosorption at the biomass concentration of 25 g L-1,followed by C.racemosa with the maximal biosorption at 30 g L-1.S.wightii showed 78% biosorption of Cr(VI),Cr(III),Pb(II) and Cd(II) ions.C.racemosa exhibited 85% biosorption of Cd(II) and Cr(VI),and 50% biosorption of Cr(III) and Pb(II).The results of our study suggest that seaweed biomass can be used efficientlyNarayanaswamyb Tamilselvan Kumar Saurav Krishnan Kannabiran 2012Journal of Ocean University of China2012,11,1:6
4Strength behaviour of sandstone subjected to polyaxial state of stress显示文摘The paper presents an experimental investigation on the strength behaviour of natural rock subjected to polyaxial state of stress. The polyaxial tests were conducted on cubical specimens of sandstone obtained from the Shivpuri district in Madhya Pradesh state of India, The specimens having nominal dimensions of100 mm x 100 mm x 100 mm were tested using a polyaxial testing machine. Twenty-five combinations of intermediate and minor principal stresses were applied and the specimens were loaded till failure occurs. It was observed that the intermediate principal stress has a substantial effect on the strength of the Shivpuri sandstone. A database of rock strength under various combinations of σ_2 and σ_3 was obtained for the Shivpuri sandstone. The database was used to study the predictability of five most commonly used strength criterion. Root mean square error(RMSE), average absolute relative error percentage(AAREP) and coefficient of accordance(COA) were used as indices for the measure of goodness of fit. It was observed that the least error in the prediction was shown by modified Mohr Coulomb criterion followed by modified Weibols and Cook criterion. 5Triaxial strength behaviour of rockmass satisfying Modified Mohr-Coulomb and Generalized Hoek-Brown criteria显示文摘The triaxial strength of twenty rockmass types was predicted using two non-linear triaxial strength criteria for rockmass i.e. Modified Mohr-Coulomb(MMC) criterion and Generalized Hoek-Brown(GHB)criterion. Four different rockmass classification systems were used for the calculation of MMC criterion parameters while only GSI classification system has been used for calculation of GHB parameters. The representative value of the uniaxial compressive strength and elastic modulus of rockmass have been estimated using probabilistic approach. A hypothetical case of an unsupported tunnel has been analyzed considering both MMC and GHB criteria. The analysis was done using the convergence-confinement method with two different approaches. The first approach predicts the tunnel response using GHB criterion directly. The second approach predicts the tunnel response using equivalent Mohr-Coulomb parameters obtained by linearization of triaxial data points obtained from MMC and GHB criteria. The tunnel response has been estimated in terms of radius of plastic zone, tunnel convergence and tunnel convergence strain. For very poor rockmasses the tunnel response predicted by MMC criterion is less than that predicted by GHB criterion. For poor and fair rockmass, the tunnel response estimated considering both the criteria are comparable except for few cases. 6Association of vitamin D and knee osteoarthritis in younger individuals显示文摘BACKGROUND The incidence of primary osteoarthritis knee is gradually increasing among young individuals.The increasing prevalence of obesity,sedentary lifestyle,sporting activity,and vitamin D deficiency(VDD)has been hypothesized for this shifting disease trend.This study was designed to look for the association of serum vitamin D among these young arthritic patients.AIM To look for the association of serum vitamin D in younger knee osteoarthritis(KOA)patients.METHODS In a 2-year observational study,146 non-obese KOA patients of 35-60 years were evaluated clinically(Knee injury and Osteoarthritis Outcome Score,KOOS)and radiologically(Kellegren-Lawrence stage,KL).The serum 25(OH)D level of these patients and 146 normal healthy individuals of same age group were estimated.RESULTS Both the groups were comparable in terms of age and sex.The average serum 25(OH)D level in healthy individuals and KOA patients was 45.83 ng/mL and 34.58 ng/mL,respectively(P<0.001).Inadequate serum 25(OH)D level(<30 ng/mL)was found in 46.57%of KOA patients and 24%of normal healthy participants indicating a significant positive association(odds ratio 2.77,95%CI:1.67-4.54,P<0.001).The 25(OH)D level in KL grade I,II,III and IV was 43.40,30.59,31.56 and 33.93 ng/mL respectively(no difference,P=0.47).Similarly,the KOOS score in sufficient,insufficient and deficient groups were 65.31,60.36 and 65.31,respectively(no difference,P=0.051).CONCLUSION The serum 25(OH)D level is significantly low in younger KOA patients.However,the clinical and radiological severities have no association with serum vitamin D level.Sujit Kumar Tripathy Amrit Gantaguru Saurav Narayan Nanda Sandeep Velagada Anand Srinivasan Manaswini Mangaraj 2020World Journal of Orthopedics2020,11,10:2

10Universal converse flexoelectricity in dielectric materials via varying electric field direction显示文摘Flexoelectricity is a symmetry independent electromechanical cou-pling phenomenon that outperforms piezoelectricity at micro and nanoscales due to its size-dependent behavior arising from gradi-ent terms in its constitutive relations.However,due to this gradient term flexoelectricity,to exhibit itself,requires specially designed geometry or material composition of the dielectric material.First of its kind,the present study put forward a novel strategy of achieving electric field gradient and thereby converse flexoelectri-city,independent of geometry and material composition of the material.The spatial variation of the electric field is established inside the dielectric material,Ba_(0.67)Sr_(0.33)TiO_(3)(BST),by manipulating electrical boundary conditions.Three unique patterns of electrode placement are suggested to achieve this spatial variation.This varying direction of electric field gives rise to electric field gradient,the prerequisite of converse flexoelectricity.A multi-physics cou-pling based theoretical framework is established to solve the flexo-electric actuation by employing isogeometric analysis(IGA).Electromechanically coupled equations of flexoelectricity are solved to obtain the electric field distribution and the resulting displace-ments thereby.The maximum displacements of 0.2 nm and 2.36 nm are obtained with patterns I and II,respectively,while pattern III can yield up to 85 nm of maximum displacement.Saurav Sharma Rajeev Kumar Rahul Vaish 2021International Journal of Smart and Nano Materials2021,12,1:1

14Comparing accuracy of cochlear measurements on magnetic resonance imaging and computed tomography:A step towards radiation-free cochlear implantation显示文摘Objective:Computed Tomography(CT)and Magnetic Resonance Imaging(MRI)are commonly employed in pre-operative evaluation for cochlear implant surgery.However,with a decrease in the age of implantation,even minor radiation exposure can cause detrimental effects on children over their lifetime.The current study compares different cochlear measurements from CT and MRI scans and evaluates the feasibility of using only an MRI scan for radiological evaluation before cochlear implantation.Methods:A longitudinal observational study was conducted on 94 ears/47 children,employing CT and MRI scans.The CT and MRI scan measurements include,A value,B value,Cochlear duct length(CDL),twoturn cochlear length,alpha and beta angles to look for cochlear orientation.Cochlear nerve diameter was measured using MRI.The values were compared.Results:The mean difference between measurements from CT and MRI scans for A value,B value,CDL,and two-turn cochlear length values was 0.567±0.413 mm,0.406±0.368 mm,2.365±1.675 mm,and 2.063±1.477 mm respectively without any significant difference.The alpha and beta angle measures were comparable,with no statistically significant difference.Conclusion:The study suggests that MRI scans can be the only radiological investigation needed with no radiation risk and reduces the cost of cochlear implant program in the paediatric population.There is no significant difference between the measurements obtained from CT and MRI scans.However,observed discrepancies in cochlear measurements across different populations require regionally or race-specific standardized values to ensure accurate diagnosis and precision in cochlear implant surgery.This aspect must be addressed to ensure positive outcomes for patients.Anurita Swarup Vinusree Karakkandy Preetam Chappity Suprava Naik Sanjay Kumar Behera Pradipta Kumar Parida Mohnish Grover Gaurav Gupta Prajna Paramita Giri Saurav Sarkar Pradeep Pradhan Dillip Kumar Samal Aswathi Kallyadan Veetil Asutosh Adhikari Saurav Nayak 2023Journal of Otology2023,18,4:0

17uCARE Chem Suite and uCAREChemSuiteCLI: Tools for bacterial resistome prediction显示文摘In the era of antibiotic resistance,in silico prediction of bacterial resistome pro-files,likely to be associated with inactivation of new potential antibiotics is of utmost impor-tance.Despite this,to the best of our knowledge,no tool exists for such prediction.Therefore,under the rationale that drugs with similar structures have similar resistome profiles,we devel-oped two models,a deterministic model and a stochastic model,to predict the bacterial re-sistome likely to neutralize uncharacterized but potential chemical structures.The current version of the tool involves the prediction of a resistome for Escherichia coli and Pseudomonas aeruginosa.The deterministic model on omitting two diverse but relatively less characterized drug classes,polyketides and polypeptides showed an accuracy of 87%,a sensitivity of 85%,and a precision of 89%,whereas the stochastic model predicted antibiotic classes of the test set compounds with an accuracy of 72%,a sensitivity of 75%,and a precision of 83%.The models have been implemented in both a standalone package and an online server,uCAREChemSuite-CLI and uCARE Chem Suite,respectively.In addition to resistome prediction,the online version of the suite enables the user to visualize the chemical structure,classify compounds in 19 pre-defined drug classes,perform pairwise alignment,and cluster with database compounds using a graphical user interface.Saurav Bhaskar Saha Vijai Kumar Gupta Pramod Wasudeo Ramteke 2021Genes & Diseases2021,8,5:0

20Protein Precipitation to Remove Carbohydrates that Interfere in Protein-Bound Tryptophan Quantification in Soybean Seeds显示文摘Tryptophan is one of the nine essential amino acids in humans that can only be obtained through diets and supplements.It is a precursor to many biological processes,such as serotonin,melatonin,kynurenin,and niacin(nicotinamide)vitamin synthesis.The content of tryptophan in foods,such as soybean is an important indicator of nutritional value.Therefore,accurate quantification of tryptophan in soybean is crucial to soybean nutritional improvement.Quantification of soybean protein-bound amino acids first involves acid hydrolysis of total protein to liberate amino acids.However,tryptophan quantification following acid hydrolysis is difficult or impossible due to its reactions with soybean carbohydrates.Therefore,removal of carbohydrates from soy proteins prior to acid hydrolysis is necessary.In this study,we compared four common protein precipitation methods(i.e.,methanol,acetonitrile,acetone,and trichloroacetic acid(TCA)protein precipitation methods)to determine the best method to separate soy proteins from carbohydrates,and concluded that acetone provided the highest recovery of soy proteins.Tryptophan content in the precipitated proteins was determined after acid hydrolysis of the proteins using liquid chromatography-tandem mass spectrometry multiple reaction monitoring(LC-MS/MS-MRM).No significant difference in the tryptophan content was found among proteins precipitated with methanol,acetonitrile,and TCA,suggesting that these precipitated proteins have similar compositions.A slightly lower,but statistically significant tryptophan content was found in the acetonitrile-precipitated proteins,suggesting that these proteins contain slightly higher glycosylated proteins.Zhentian Lei Santosh Kumar Saurav J.Sarma Ritesh Kumar Barbara W.Sumner Minviluz G.Stacey Lloyd W.Sumner 2020Journal of Analysis and Testing2020,4,3:0
