2Metabarcoding of fecal DNA reveals the broad and flexible diet of a globally endangered bird显示文摘Knowing the diet of endangered wild animals is a prerequisite for species-specific conservation and habitat management.The Sichuan partridge Arborophila rufipectus is a globally endangered Galliformes species endemic to the mountains of southwest China.Existing information on the diet of this species is biased and fragmented owing to traditional observation methods.Little is known about their dietary composition or how they respond to temporal variations in food resources throughout the year.In this study,a dietary analysis was performed on 6o fecal samples using DNA Metabarcoding of invertebrates and plants to determine the primary animal and plant components of the diet across 3 critical periods of adult life history(breeding,postbreeding wandering,and overwintering).Preys from the dipteran order,followed by the lepidopteran and ara-neaen spp.,were the predominant,animal-derived foods.Symplocos,Rubus,Celastrus,Holboellia,and Actinidia spp.supply a large abundance of fruits and seeds for this omnivorous bird.Substantial temporal dietary changes among the 3 periods and a general shift toward lower dietary diversity during the breeding season were observed,suggesting that the Sichuan partridge can adjust their diet according to the availability of food resources and their own needs.Characterizing the composition and seasonal changes in Sichuan partridge diets informs the habitat management of native flora(the plant taxa that can generate berries and seeds,such as Symplocos,Rubus,Celastrus,and Holboellia,which are likely of conservation interest)to achieve full life-cycle conservation.Keyi Tang Yufeng Wang Mengling Wu Shufang Wang Changkun Fu Zhengwang Zhang Yiqiang Fu 2023Current Zoology2023,69,5:0

4CsXDH1 gene promotes caffeine catabolism induced by continuous strong light in tea plant显示文摘Tea plant(Camellia sinensis)is an important cash crop with extensive adaptability in the world.However,complex environmental factors force a large variation of tea quality-related components.Caffeine is essential for the formation of bitter and fresh flavors in tea,and is the main compound of tea that improves human alertness.Continuous strong light stimulation was observed to cause caffeine reduction in tea leaves,but the mechanism is not clear.In this study,the response of tea plant to light intensity was analysed mainly by multi-omics association,antisense oligodeoxynucleotide(asODN)silencing technique,and in vitro enzyme activity assay.The results revealed multiple strategies for light intensity adaptation in tea plant,among which the regulation of chloroplasts,photosynthesis,porphyrin metabolism,and resistance to oxidative stress were prominent.Caffeine catabolism was enhanced in continuous strong light,which may be a light-adapted strategy due to strict regulation by xanthine dehydrogenase(XDH).asODN silencing and enzymatic activity assays confirmed that CsXDH1 is a protein induced by light intensity to catalyze the substrate xanthine.CsXDH1 asODN silencing resulted in significant up-regulation of both caffeine and theobromine in in vitro enzyme activity assay,but not in vivo.CsXDH1 may act as a coordinator in light intensity adaptation,thus disrupting this balance of caffeine catabolism.Qianhui Tang Keyi Liu Chuan Yue Liyong Luo Liang Zeng Zhijun Wu 2023Horticulture Research2023,10,6:0
5Lactobacillus isolates from healthy volunteers exert immunomodulatory effects on activated peripheral blood mononuclear cells显示文摘As probiotics in the gut, Lactobacilli are believed to play important roles in the development and maintenance of both the mucosal and systemic immune system of the host. This study was aimed to investigate the immunomodulatory function of candiate lactobacilli on T cells. Lactobacilli were isolated from healthy human feces and the microbiological characteristics were identified by API 50 CHL and randomly amplified polymorphic DNA (RAPD) assays. Anti-CD3 antibody activated peripheral blood mononuclear cells (PBMCs) were treated by viable, heat-killed lactobacilli and genomic DNA of lactobacilli, and cytokine profiles were tested by ELISA. Isolated lactobacilli C44 and C48 were identified as L. acidophilus and L. paracacei, which have properties of acid and bile tolerance and inhibitor effects on pathogens. Viable and heat-killed C44 and C48 induced low levels of proinflammatory cytokines (TNF-α, IL-6 and IL-8) and high levels of IFN-γ and IL-12p70 in PBMCs. In antiCD3 antibody activated PBMCs, viable and heat-killed C44 increased Th2 cytokine levels (IL-5, IL-6 and IL-10), and simultaneously enhanced Th1 responses by inducing IFN-γ and IL-12p70 production. Different from that of lactabacillus strains, their genomic DNA induced low levels of IL-12p70, IFN-γ and proinflammatory cytokines in PBMCs with or without anti-CD3 antibody activation. 6An AIE probe for long-term plasma membrane imaging and membrane-targeted photodynamic therapy显示文摘Two red-emissive luminogens(TPTH and TPTB) with typical aggregation-induced emission characteristics were developed. By introducing the heavy atom of Br at the end of alkyl chain, TPTB exhibited higher reactive oxygen species generation efficiency through both types I and II pathways. Due to its excellent biocompatibility and proper lipophilicity, TPTB could be used for long-term cell membrane staining and this staining ability was independent of the change of plasma membrane potential.
