1Embryonic stem cells generated by nuclear transfer of human somatic nuclei into rabbit oocytes显示文摘To solve the problem of immune incompatibility, nuclear transplantation has been envisaged as a means to produce cells or tissues for human autologous transplantation. Here we have derived embryonic stem cells by the transfer of human somatic nuclei into rabbit oocytes. The number of blastocysts that developed from the fused nuclear transfer was comparable among nuclear donors at ages of 5, 42, 52 and 60 years, and nuclear transfer (NT) embryonic stem cells (ntES cells) were subsequently derived from each of the four age groups. These results suggest that human somatic nuclei can form ntES cells independent of the age of the donor. The derived ntES cells are human based on karyotype, isogenicity, in situ hybridization, PGR and immunocytochemistry with probes that distinguish between the various species. The ntES cells maintain the capability of sustained growth in an undifferentiated state, and form embryoid bodies, which, on further induction, give rise to cell types such as neuron and muscle, as well as mixed cell populations that express markers representative of all three germ layers. 2Synthesis of Metal Porphyrins Tailed with Salicylic Acid and their Interaction with Bovine Serum Albumin显示文摘A synthetic method of porphyrins tailed with salicylic substituents is described.Reaction of bromoalkoxyphenyl porphyrin 1 with salicylic acid gave porphyrins 2-5. 3A Statistical Approach Designed for Finding Mathematically Defined Repeats in Shotgun Data and Determining the Length Distribution of Clone-Inserts显示文摘The large amount of repeats, especially high copy repeats, in the genomes of higher animals and plants makes whole genome assembly (WGA) quite difficult. In order to solve this problem, we tried to identify repeats and mask them prior to assembly even at the stage of genome survey. It is known that repeats of different copy number have different probabilities of appearance in shotgun data, so based on this principle, we constructed a statistical model and inferred criteria for mathematically defined repeats (MDRs) at different shotgun coverages. According to these criteria, we developed software MDRmasker to identify and mask MDRs in shotgun data. With repeats masked prior to assembly, the speed of assembly was increased with lower error probability. In addition, clone-insert size affects the accuracy of repeat assembly and scaffold construction. We also designed length distribution of clone-inserts using our model. In our simulated genomes of human and rice, the length distribution of repeats is different, so their optimal length distributions of clone-inserts were not the same. 13Chrysanthemum indicum linné extracts attenuate the mitogenic effect of epinephrine on human HCC cells显示文摘Objective: To investigate the stimulatory effect of epinephrine(Epi) and the antagonistic effect of Chrysanthemum indicum Linné extracts (CILE) on Epi-induced growth of human hepatocellular carcinoma(HCC) cells. Methods: The stimulatory effect of Epi and inhibitory effect of CILE on the growth of HepG2 and MHCC97H cells were investigated using a proliferation assay in correlation with βadrenergic receptor(β 2-AR) blockade, a MEK1/MEK2 inhibitor, and assessment of MAPK/ERK1/2 intracellular activity. Results: Epi transiently activated MAPK/ERK1/2 in HepG2 and MHCC97H cells, resulting in a burst of growth. The effect of Epi was significantly attenuated by ICI 118551 and U0126. CILE exhibited a dose-dependent attenuation of the stimulatory effect of Epi on the growth of both cell lines and inhibited the Epi-induced activation of MAPK/ERK1/2.
