| 1 | Relationship between expression and distribution of cyclooxygenase-2 and bcl-2 in human gastric adenocarcinoma显示文摘AIM: To explore expression and distribution features of COX-2 and bcl-2 in human gastric adenocarcinoma tissues and to study its biological significance.METHODS: Totally 36 human gastric carcinoma samples were enrolled in this study (cardiac adenocarcinoma 16 cases, distal gastric adenocarcinoma 20 cases). The expressions of COX-2 and bcl-2 in cancerous tissues and corresponding para-cancerous tissues were investigated by immunohistochemistry using COX-2 polyclonal antibody and bcl-2 monoclonal antibody. The normal gastric mucosa tissues were used as control.RESULTS: The expressions of COX-2 and bcl-2 in gastric carcinoma were significantly higher than that in the paracancerous tissues (77.8% vs 47.2%, P<0.01, 80.56% vs 58.33%, P<0.05). The expression of COX-2 in cardiac adenocarcinoma was remarkably higher than that in the distal gastric carcinoma (93.8% vs 65.0%, P<0.01). The expression of COX-2 was mainly localized in the cytoplasm of tumor cells and partly in the nucleus. There is a transition of the COX-2 cytoplasmic positivity to nucleic in tumor cells with the increase of gastric carcinoma pathological grade. Interstitial macrophages, fibroblasts and vascular endothelial cells also expressed COX-2. The tissues with higher expression of COX-2 also expressed high level of bcl-2 protein.CONCLUSION: Abnormal expression pattern of COX-2within the tissues of human gastric cancer is correlated with tumor location and lymph node metastasis. | 2 | Expression of ORF2 partial gene of hepatitis E virus in tomatoes and immunoactivity of expression products显示文摘AIM: To transfer hepatitis E virus (HEV) ORF2 partial gene to tomato plants, to investigate its expression in transformants and the immunoactivity of expression products, and to explore the feasibility of developing a new type of plantderived HEV oral vaccine.METHODS: Plant binary expression vector p1301E2,carrying a fragment of HEV open reading frame-2 (named HEV-E2), was constructed by linking the fragment to a constitutive CaMV35s promoter and nos terminator, then directly introduced into Agrobacterium tumefaciens EHA105. With leaf-disc method, tomato plants medicated by EHA105 were transformed and hygromycin-resistant plantlets were obtained in selective medium containing hygromycin. The presence and integration of foreign DNA in transgenic tomato genome were confirmed by Gus gene expression, PCR amplification and Southern dot blotting.The immunoactivity of recombinant protein extracted from transformed plants was examined by enzyme-linked immunosorbant assay (ELISA) using a monoclonal antibody specifically against HEV. ELISA was also used to estimate the recombinant protein content in leaves and fruits of the transformants.RESULTS: Seven positive lines of HEV-E2-transgenic tomato plants confirmed by PCR and Southern blotting were obtained and the immunoactivity of recombinant protein could be detected in extracts of transformants. The expression levels of recombinant protein were 61.22 ng/g fresh weight in fruits and 6.37-47.9 ng/g fresh weight in leaves of the transformants.CONCLUSION: HEV-E2 gene was correctly expressed in transgenic tomatoes and the recombinant antigen derived from them has normal immunoactivity. | 4 | Relationship of gastric Helicobacter pylori infection to Barrett's esophagus and gastro-esophageal reflux disease in Chinese显示文摘AIM:To evaluate the relationship of Helicobacter pylori infection to reflux esophagitis (RE), Barrett's esophagus (BE)and gastric intestinal metaplasia (IM).METHODS:RE,BE and gastric IM were determined by upper endoscopy. Patients were divided into 2 groups; those with squamocolumnar junction (SCJ) beyond gastroesophageal junction (GEJ)≥3cm (group A), and those with SCJ beyond GE.1 <3cm (group B). Biopsy specimens were obtainedend escopically from just below the SCJ, gastric antrum along the greater and lesser curvature. Pathological changes and Hpylorr infection were determined by HE staining, Alcian blue staining and Giemsa staining.RESULTS:The prevalence of Hpyloriinfection was 46.93%.There was no difference in the prevalence between males and females.The prevalence of Hpyloriinfection decreased stepwise significantly from RE grade I to Ⅲ.There was no difference in the prevalence between the two groups, and between long-segment and short-segment BE. In distal stomach, prevalence of Hpyloriinfection was significantly higher in patients with IM than those without IM.CONCLUSION: There is a protective role of Hpyloriinfectuion to GERD. There may be no relationship between Hpylori infection of stomach and BE. | 5 | Characteristics of strike-slip inversion structures of the Karatau fault and their petroleum geological significances in the South Turgay Basin,Kazakhstan显示文摘The Karatau fault is one of the important strike-slip faults in central Asia,and the South Turgay Basin is located towards its northern end.Detailed seismic interpretation indicated that the strikeslip tectonism of the Karatau fault weakened gradually from west to east in the South Turgay Basin.Typical flower structures developed on the section,and strike-slip faults showed an echelon pattern on planar view.The Karatau strike-slip fault affected the South Turgay Basin in two periods:(1) The South Turgay strike-slip pull-apart rift basin formed as a result of regional extensive stress in the Early-Middle Jurassic,characterized by the juxtaposition of horsts and grabens.The formation of horsts provided favorable reservoir spaces for later hydrocarbon accumulation,and different filling stages of grabens controlled different reservoir-forming factors in grabens.(2) Two stages of tectonic inversion occurred in the Late Jurassic and Late Cretaceous and played a crucial role in the final shape of the structure in the South Turgay Basin.The oil and gas migrated to form reservoirs and mainly concentrated in the horsts,graben slopes and in both sides of the strike-slip fault zone.In the case of the degree of accumulation of petroleum,the factor explaining why horsts are better than grabens is the strike-slip pull-apart of the South Turgay Basin,and the structure inversion of the South Turgay Basin explains why the west graben is better than the east one.Overall,the Karatau strike-slip fault played a very important role in the formation of the South Turgay Basin and its hydrocarbon accumulations. | Yin Wei Fan Zifei Zheng Junzhang Yin Jiquan Zhang Mingjun Sheng Xiaofeng Guo Jianjun Li Qiyan Lin yaping | 2012 | Petroleum Science2012,9,4: | 11 |

| 7 | Detrital K-feldspar^(40)Ar/^(39)Ar Ages:Source Constraints of the Lower Miocene Sandstones in the Pearl River Mouth Basin,South China Sea显示文摘The South China Sea began to outspread in the Oligocene.A great quantity of terraneous detritus was deposited in the northern continental shelf of the sea,mostly in Pearl River Mouth Basin,which constituted the main paleo-Pearl River Delta.The delta developed for a long geological time and formed a superimposed area.Almost all the oil and gas fields of detrital rock reservoir distribute in this delta.Thirty-three oil sandstone core samples in the Zhujiang Formation,lower Miocene(23-16 Ma),were collected from nine wells.The illite samples with detrital K feldspar(Kfs) separated from these sandstone cores in four sub-structural belts were analysed by the high-precision 40Ar/39Ar laser stepwise heating technique.All 33 illite 40Ar/39Ar data consistently yielded gradually rising age spectra at the low-temperature steps until reaching age plateaus at mid-high temperature steps.The youngest ages corresponding to the beginning steps were interpreted as the hydrocarbon accumulation ages and the plateau ages in mid-high temperature steps as the contributions of the detrital feldspar representing the ages of the granitic parent rocks in the provenances.The ages of the detrital feldspar from the Zhujiang Formation in the four sub-structural belts were different:(1) the late Cretaceous ages in the Lufeng 13 fault structural belt;(2) the late Cretaceous and early Cretaceous-Jurassic ages in the Huizhou 21 buried hill-fault belt;(3) the Jurassic and Triassic ages in the Xijiang 24 buried hill-fault belt;and(4) the early Cretaceous-late Jurassic ages in the Panyu 4 oil area.These detrital feldspar 40Ar/39Ar ages become younger and younger from west to east,corresponding to the age distribution of the granites in the adjacent Guangdong Province,Southern China. | LIU Zaisheng SHI Hesheng ZHU Junzhang QIU Huaning ZHANG Zhilin YUN Jianbing | 2012 | Acta Geologica Sinica(English Edition)2012,86,2: | 9 |
| 8 | Effect of Zhuyeshigao granule on tumor necrosis factor-α and inter leukins serum protein levels in rats with radiation esophagitis显示文摘OBJECTIVE: To investigate the effects of Zhuyeshigao granule(ZSG) on tumor necrosis factor-α(TNF-α), interleukin-1(IL-1), IL-2, IL-6, and IL-8 in rats with radiation esophagitis.METHODS: Fifty Wistar rats were randomly divided into five groups(10 rats in each group): control(without radiation), saline-treated, and low,medium, and high-dose ISG-treated groups. Rats were given normal saline(10 mL/kg) or 1.15, 2.3,or 4.6 g/kg ZSG by intragastric administration once a day for 7 days. A rat model of radiation esophagitis was established by local irradiation of Co60(490.25 cGy/min, totaling 30 Gy). The administration of ZSG was continued for another 7 days and on the 7th day post-irradiation, inferior vena cava blood was collected.The serum was separated, and TNF-α, IL-1, IL-2, IL-6, and IL-8 protein levels were determined.RESULTS: Inflammatory response factors were found in the serum of each group. | 9 | Decreased nitrous oxide emissions associated with functional microbial genes under bio-organic fertilizer application in vegetable fields显示文摘Bio-organic fertilizers enriched with plant growth-promoting microbes(PGPMs)have been widely used in crop fields to promote plant growth and maintain soil microbiome functions.However,their potential effects on N_(2)O emissions are of increasing concern.In this study,an in situ measurement experiment was conducted to investigate the effect of organic fertilizer containing Trichoderma guizhouense(a plant growth-promoting fungus)on soil N_(2)O emissions from a greenhouse vegetable field.The following four treatments were used:no fertilizer(control),chemical fertilizer(NPK),organic fertilizer derived from cattle manure(O),and organic fertilizer containing T.guizhouense(O+T,referring to bio-organic fertilizer).The abundances of soil N cycling-related functional genes(amoA)from ammonium-oxidizing bacteria(AOB)and archaea(AOA),as well as nirS,nirK,and nosZ,were simultaneously determined using quantitative PCR(qPCR).Compared to the NPK plot,seasonal total N_(2)O emissions decreased by 11.7%and 18.7%in the O and O+T plots,respectively,which was attributed to lower NH_(4)^(+)-N content and AOB amoA abundance in the O and O+T plots.The nosZ abundance was significantly greater in the O+T plot,whilst the AOB amoA abundance was significantly lower in the O+T plot than in the O plot.Relative to the organic fertilizer,bio-organic fertilizer application tended to decrease N_(2)O emissions by 7.9%and enhanced vegetable yield,resulting in a significant decrease in yield-scaled N_(2)O emissions.Overall,the results of this study suggested that,compared to organic and chemical fertilizers,bio-organic fertilizers containing PGPMs could benefit crop yield and mitigate N_(2)O emissions in vegetable fields. | Yajun GENG Yiming YUAN Yingcheng MIAO Junzhang ZHI Mengyuan HUANG Yihe ZHANG Hong WANG Qirong SHEN Jianwen ZOU Shuqing LI | 2021 | Pedosphere2021,31,2: | 7 |
| 10 | Barrett's esophagus and its correlation with gastroesophageal reflux in Chinese显示文摘AIM- To study the prevalence of Barrett's esophagus in Chinese and its correlation with gastroesophageal reflux. METHODS: This study was carded out in a large prospective series of 391 patients who had undergone upper endoscopy. The patients were divided into 3 groups according to the position of squamocolumnar junction (SC3). Reflux esophagitis (RE) and its degree were recorded. Intestinal metaplasia (IM) in biopsy specimen was typed according to histochemistry and HE and alcian blue (pH2.5) staining separately. Results correlating with clinical, endoscopic, and pathological data were analysed. RESULTS: The prevalence of IM endoscopically appearing Long-segment Barrett's Esophagus (LSBE) was 26.53%, Short-segment Barrett's Esophagus (SSBE) was 33.85% and gastroesophageal junction (GEJ) was 34.00%. IM increased with age of above 40 years old and no difference was found between male and female. Twelve were diagnosed as dysplasia (7 low -grade, 5 high-grade), 16 were diagnosed as cardiac adenocarcinoma and 1 as esophageal adenocarcinoma. The more far away the SCJ moved upward above GEJ, the higher the prevalence and the more severe the RE were. CONCLUSION: There was no difference of the prevalence of IM in different places of SCJ, and IM increased with age of above 40 years old. It is important to pay attention to dysplasia in the distal esophagus and gastro-esophageal junction, and adenocarcinoma is more common in cardia than in esophagus. | 11 | A rapid and efficient method to express target genes in mammalian cells by baculovirus显示文摘AIM: To investigate the modification of baculovirus vector and the feasibility of delivering exogenous genes into mammalian cells with the culture supernatant of Spodoptera frugiperta (Sf9) cells infected by recombinant baculoviruses. METHODS: Two recombinant baculoviruses (BacV-CMV-EGFPA, BacV-CMV-EGFPB) containing CMV-EGFP expression cassette were constructed. HepG2 cells were directly incubated with the culture supernatant of Sf9 cells infected by recombinant baculoviruses, and reporter gene transfer and expression efficiencies were analyzed by flow cytometry (FCM). The optimal transduction conditions were investigated by FCM assay in HepG2 cells. Gene-transfer and expression efficiencies in HepG2 or CV1 cells by baculovirus vectors were compared with lipofectAMINE, recombinant retrovirus and vaccinia virus expression systems. Twenty different mammalian cell lines were used to investigate the feasibility of delivering exogenous genes into different mammalian cells with the culture supernatant of infected Sf9 cells. RESULTS: CMV promoter could directly express reporter genes in Sf9 cells with a relatively low efficiency. Target cells incubated with the 1:1 diluted culture supernatant (moi=50) for 12 h at 37 ℃ could achieve the highest transduction and expression efficiencies with least impairment to cell viability. Under similar conditions the baculovirus vector could achieve the highest gene-transfer and expression efficiency than lipofectAMINE, recombinant retrovirus and vaccinia virus expression systems. Most mammalian cell lines could be transduced with recombinant baculovirus. In primate adherent culture cells the recombinant baculovirus could arrive the highest infection and expression efficiencies, but it was not very satisfactory in the cell lines from mice and suspended culture cells. | 12 | Long-term outcome of esophageal myotomy for achalasia显示文摘AIM:Modified Heller's myotomy is still the first choice for achalasia and the assessment of surgical outcomes is usually made based on the subjective sensation of patients.This study was to objectively assess the long-term outcomes of esophageal myotomy for achalasia using esophageal manometry, 24-hour pH monitoring,esophageal scintigraphy and fiberoptic esophagoscopy.METHODS:From February 1979 to October 2000, 176 patients with achalasia underwent modified Heller's myotomy, including esophageal myotomy alone in 146 patients, myotomy in combination with Gallone or Dor antirefiux procedure in 22 and 8 patients, respectively. Clinical score,pressure of the lower esophageal sphincter (LES),esophageal clearance rate and gastroesophageal reflux were determined before and i to 22 years after surgery.RESULTS: After a median follow-up of 14 years, 84.5% of patients had a good or excellent relief of symptoms,and clinical scores as well as resting pressures of the esophageal body and LES were reduced compared with preoperative values (P<0.001).However,there was no significant difference in DeMeester score between pre-and postoperative patients(P=0.51).Esophageal transit was improved in postoperative patients, but still slower than that in normal controls. The incidence of gastroesophageal reflux in patients who underwent esophageal myotomy alone was 63.6% compared to 27.3% in those who underwent myotomy and antirefiux procedure (P=-0.087). Three (1.7%) patients were complicated with esophageal cancer after surgery.CONCLUSION: Esophageal myotomy for achalasia can reduce the resting pressures of the esophageal body and LES and improve esophageal transit and dysphagia. | 13 | Transcription factor Sp1 expression in gastric cancer and its relationship to long-term prognosis显示文摘AIM: To explore the expression of Sp1 in gastric carcinoma as well as its association with other clinicopathologic features, and to evaluate the role of Sp1 as a prognostic Indicator of gastric carcinoma.METHODS: By using immunohistochemistry, we examined the Sp1 expression patterns in 65 cases of human gastric cancer, and 40 normal gastric mucosa specimens.Simultaneously, the correlation between Sp1 expression and clinical outcome or clinicopathologic features was investigated.RESULTS: The percentage of Sp1 expression was 12.5%(5/40) in normal gastric mucosa, and the Sp1 protein was mainly expressed in the nuclei of cells located in the mucous neck region. In sharp contrast, strong Sp1 expression was detected in tumor cells, whereas no or faint Sp1 staining was detected in stromal cells and normal glandular cells surrounding the tumors. The expression rate of Sp1 in gastric cancer lesions was 53.85% (35/65).The medium survival duration in patients who had a tumor with negative, weak and strong Sp1 expressions was 1 700, 1 560 and 1 026 d, respectively (P<0.05). Sp1 protein expression was closely related to the depth of tumor infiltration (χ2 = 13.223, P<0.01) and TNM stage (χ2= 11.009, P<0.05), but had no relationship with the number of lymph nodes and Lauren's classification (P>0.05). Cox regression model for multivariate analysis revealed that high Sp1 expression (P<0.05) and advanced stage (P<0.01) were independent predictors of poor survival.CONCUSION: Normal and malignant gastric tissues have unique Sp1 expression patterns. | 15 | Hydrophobicity of reactive site loop of SCCA1 affects its binding to hepatitis B virus显示文摘AIM: To investigate the role of SCCA2 and other SCCA1 molecules in the process of hepatitis B virus (HBV) binding to mammalian cells.METHODS: SCCA1 and SCCA2 were isolated from HepG2. Binding protein (BP) genes were obtained through PCR. Recombinant baculoviruses expressing SCCA1, SCCA2, BP, and different mutants were constructed and utilized to infect mammalian cells to investigate the binding ability of infected cells to HBV.RESULTS: A SCCA1 gene (A1) was isolated from HepG2, but it appeared to lack the binding ability of infected cells to HBV. Two mutants, A1-BP and BP-A1, were constructed by interchanging the carboxyl terminal of A1 and BP. Cells expressing A1-BP showed an increased virus bindingcapacity, but not BP-A1. Comparison of A1 sequence with the sequence of BP indicated the presence of only three amino acid changes in the carboxyl terminal, two of them were found in the reactive site loop (RSL) of SCCA1. Primary structure assay revealed that the hydrophobicity of BP and AJ515706 in this domain was strong, but A1 was relatively weak. Changing the aa349 of A1 from low hydrophobic glutamic acid to high hydrophobic valine enhanced HBV binding. In contrast, HBV binding was reduced by changing the aa349 of BP from valine to glutamic acid. | 16 | Gene expression profiling in Barrett's esophagus and cardia intestinal metaplasia: A comparative analysis using cDNA microarray显示文摘AIM: To study the difference of gene expression profile changes in Barrett's esophagus (BE) and cardia intestinal metaplasia (CIM) and to screen the novel genes in the early stage by cDNA microarray.METHODS: cDNA retrotranscribed from an equal amount of mRNA from BE and CIM epithelial tissues was labeled with Cy3 and Cy5 fluorescence as probes. The mixed probe was hybridized with three pieces of BiostarH-40 s double dot human whole gene chip. The chips were scanned with a ScanArray 4000. The acquired images were analyzed using GenePix Pro 3.0 software.RESULTS: A total of 141 genes were screened out that exhibited different expression in all three chips. There were 74 upregulated and 67 downregulated genes in gene expression profiles of BE which were two times of that in CIM.CONCLUSION: There is a difference in gene expression level between BE and CIM epithelia. | 17 | Expression and immunoactivity of chimeric particulate antigens of receptor binding site-core antigen of hepatitis B virus显示文摘AIM: To improve the immunogenicity of receptor binding site of hepatitis B virus (HBV) on preS1 antigen using HBV core antigen as an immuno-carrier.METHODS: One to 6 tandem copies of HBV preS1 (21-47)fragment were inserted into HBcAg at the sites of aa 78 and 82, and expressed in E. coli. ELISA, Western blot and animal immunization were used to analyze the antigenicity and immmunogenicity of purified particulate antigens. The ability to capture HBV by antibodies elicited by chimeric partides was detected with immuno-capture PCR.RESULTS: Recombinant antigens CⅠ, CⅡ, CⅢ carrying 1-3 copies of HBV preS1 (21-47) individually could form viruslike particles (VLPs), similar to HBcAg in morphology. But recombinant antigens carrying 4-6 copies of HBV preS1 (21-47) were poorly expressed in E.coli. Chimeric antigens were lacking of immunoreactivity with anti-HBc monoclonal antibodies (McAbs), but still reserved good immunoreactivity with anti-HBe McAbs. CⅠ, CⅡ, CⅢ could strongly react with anti-preS1 McAb, suggesting that preS1 (21-47) fragment was well exposed on the surface of chimeric VLPs. Three chimeric VLP antigens (CⅠ, CⅡ and CⅢ) could stimulate mice to produce high-level antibody responses, and their immunogenicity was stronger than non-particulate antigen 21-47*6, containing 6 copies of preS1 (21-47). Mouse antibodies to CⅠ, CⅡ and CⅢ were able to capture HBV virions in immuno-capture PCR assay in vitro.CONCLUSION: Chimeric particulate antigens of receptor binding site-core antigen of HBV can elicit strong antibody responses to preS1. | 18 | Selection of a peptide mimicking neutralization epitope of hepatitis E virus with phage peptide display technology显示文摘AIM: To select the peptide mimicking the neutralization epitope of hepatitis E virus which bound to non-type-specific and conformational monoclonal antibodies (mAbs) 8Cll and 8H3 fromed 7-peptide phage display library, and expressed the peptide recombinant with HBcAg in E.coli and to observe whether the recombinant HBcAg could still form virus like particle (VLP) and to test the activation of the recombinant polyprotein and chemo-synthesized peptide that was selected by mAb 8H3. METHODS: 8Cll and 8H3 were used to screen for binding peptides through a 7-peptide phage display library. After 4 rounds of panning, monoclonal phages were selected and sequenced. The obtained dominant peptide coding sequences was then synthesized and inserted into amino acid 78 to 83 of hepatitis B core antigen (HBcAg), and then expressed in E.coli Activity of the recombinant proteins was detected by Western blotting, VLPs of the recombinant polyproteins were tested by transmission electron microscopy and binding activity of the chemo-synthesized peptide was confirmed by BIAcore biosensor. RESULTS: Twenty-one positive monoclonal phages (10 for 8C11, and 11 for 8H3) were selected and the inserted fragments were sequenced. The DNA sequence coding for the obtained dominant peptides 8Cll (N'-His-Pro-Thr-Leu-Leu-Arg-Ile-C, named 8C11A) and 8H3 (N'-Ser-Ile-Leu-Pro- Tyr-Pro-Tyr-C, named 8H3A) were then synthesized and cloned to the HBcAg vector, then expressed in E.coli The recombinant proteins aggregated into homodimer or polymer on SDS-PAGE, and could bind to mAb 8Cll and 8H3 in Western blotting. At the same time, the recombinant polyprotein could form virus like particles (VLPs), which could be visualized on electron micrograph. The dominant peptide 8H3A selected by mAb 8H3 was further chemosynthesized, and its binding to mAb 8H3 could be detected by BIAcore biosensor. | 19 | Correlation between the Sensitivity to TRAIL and the Expression Level of DR5 on the Surface of Tumor Cells显示文摘OBJECTIVE To investigate the correlation between the sensitivity to the tumor necrosis factor-related apoptosis inducing ligand (TRAIL) and the level of expression of the death receptor 5 (DR5) on the surface of tumor cells.METHODS Anti-DR5 mAbs were used to directly detect the level of expression of DR5 on the surface of tumor cells. Using a TRAIL apoptosis kit and flow cytometry, the sensitivity of the tumor cells to TRAIL-induced apoptosis was determined and the correlation between DR5 expression and sensitivity to TRAI L analyzed.RESULTS The expression level of DR5 on the surface of different tumor cells was as follows: 97.9% in U937 cells, 95.1% in Jurkat cells, 93.8% in SW480 cells, 86.2% in HCT116 cells, 64.2% in HL-60 cells, 46.6% in Hela cells and 13.1% in K562 cells. The TRAIL-induced apoptotic rate was 72.6% in U937 cells, 85.2% in Jurkat cells, 78.6% in SW480 cells, 70.2% in HCT116 cells,60.1% in HL-60 cells, 45.4% in Hela cells and 12.3% in K562 cells. Statistical analysis showed there was a significant positive correlation (r=0.997, P<0.001) between DR5 expression and sensitivity to TRAIL.CONCLUSION The sensitivity of tumor cells to TRAIL is related to the level of expression of DR5 on the surface of tumor cells.
