2Genetic diversity of Harpins from Xanthomonas oryzae and their activity to induce hypersensitive response and disease resistance in tobacco显示文摘Three hrfA (hypersensitive response-functioning faction A) homologues (hrf1, hrf2 and hrf3) are cloned from 12 strains of Xanthomonas oryzae using PCR based techniques. Hrf1, hrf2 and hrf3 are derived from strains belonging to X. o. pv. oryzae, X. o. pv. oryzicola and X. o. pv. oryzae respectively. Sequence analysis shows that all three genes encode glycine-rich pro-teins with various numbers of GGG-GG motifs. They all share a conserved cysteine residue at position 45 or 47. Hrf1 and hrf3 encode HarpinXoo while hrf2 encodes Harpinxooc. Hrf1 and hrf3 encodes two different types of HarpinXoo proteins. Hrf1 from X. o. pv. oryzae strains ( JxoIII, JxoIV, Jxov, Pxo61, Pxo76, Pxo79, Pxo99, Pxo99 and Pxo124) encodes a 15.6 kD HarpinXoo with 3 GGG-GG motifs while Hrf3 from strain Pxo86 and Pxo112 encodes a 15.9 kD Harpinxoo with 4 GGG-GG motifs. Harpinxooc encoded by hrf2 from X. o. pv. oryzicola (strain RS105) has the mo-lecular weight of 15.3 kD and contains 2 GGG-GG motifs. Cluster analysis is performed using deduced sequences of hrf1, hrf2 and hrf3 as well as previously reported Hpa1 and XopI protein sequence. The results indicated that Harpinxoo and Harpinxooc belong to two closely related sub-groups. Hrf, hrf2 and hrf3 are expressed in E. coli strain BL21 successfully. Under the same condition, hrf1, hrf2 and hrf3 are expressed at the level of 0.389, 0.530 and 0.083 mg/mL re-spectively. All expressed hrf1, hrf2 and hrf3 proteins (Harpins) are shown to be able to induce hypersensitive reaction and TMV resistance on tobacco. 3Signal-Based Intelligent Hydraulic Fault Diagnosis Methods: Review and Prospects显示文摘Hydraulic systems have the characteristics of strong fault concealment,powerful nonlinear time-varying signals,and a complex vibration transmission mechanism;hence,diagnosis of these systems is a challenge.To provide accurate diagnosis results automatically,numerous studies have been carried out.Among them,signal-based methods are commonly used,which employ signal processing techniques based on the state signal used for extracting features,and further input the features into the classifier for fault recognition.However,their main deficiencies include the following:(1)The features are manually designed and thus may have a lack of objectivity.(2)For signal processing,feature extraction and pattern recognition are conducted using independent models,which cannot be jointly optimized globally.(3)The machine learning algorithms adopted by these methods have a shallow architecture,which limits their capacity to deeply mine the essential features of a fault.As a breakthrough in artificial intelligence,deep learning holds the potential to overcome such deficiencies.Based on deep learning,deep neural networks(DNNs)can automatically learn the complex nonlinear relations implied in a signal,can be globally optimized,and can obtain the high-level features of multi-dimensional data.In this paper,the main technology used in an intelligent fault diagnosis and the current research status of hydraulic system fault diagnosis are summarized and analyzed.The significant prospect of applying deep learning in the field of intelligent fault diagnosis is presented,and the main ideas,methods,and principles of several typical DNNs are described and summarized.The commonality between a fault diagnosis and other issues regarding typical pattern recognition are analyzed,and research ideas for applying DNNs for hydraulic fault diagnosis are proposed.Meanwhile,the research advantages and development trend of DNNs(both domestically and overseas)as applied to an intelligent fault diagnosis are reviewed.Furthermore,the fault characteristics of a complex hydraulic system are summarized and discussed,and the key problems and possible research ideas of applying DNNs to an intelligent hydraulic fault diagnosis are presented and comprehensively analyzed.Juying Dai Jian Tang Shuzhan Huang Yangyang Wang 2019Chinese Journal of Mechanical Engineering2019,32,5:11
4Outcomes in refractory diffuse large B-cell lymphoma:results fromamulticenter real-world study in China显示文摘Background:Diffuse large B-cell lymphoma(DLBCL)patients refractory to rituximab-based immunochemotherapy have a dismal prognosis.However,the definition of refractory DLBCL remains inconsistent and no large cohort study data is available from Asian countries.To validate the definition and outcomes of refractory DLBCL in China,we conducted a multicenter,retrospective cohort study.Methods:The REtrospective AnaLysis of Treatment REspoNse of refractory DLBCL(REAL-TREND)study was performed using real-world data from 8 centers in China.DLBCL patients with curative intent were included in the REAL-TREND dataset.Overall survival(OS)was estimated using the Kaplan-Meier method and compared by the log-rank test.Due to heterogeneity in response rates among different centers,the response rates of refractory patients were pooled using random-effect models.Multivariate survival analysis was performed using the Cox regression model.Results:A total of 2778 DLBCL patients diagnosed between January,2010 and December,2015 were enrolled to this study.After validating previous definitions,the SCHOLAR-1 study was most suitable to define refractory DLBCL.The estimated 5-year cumulative incidence of refractory patients was 20%(95% confidence Interval[CI]=18%-22%).After the determination of refractory disease,overall response rate and complete remission rate were 30%(95%CI=22%-38%)and 9%(95%CI=4%-15%),respectively.Patients with either no response to immunochemotherapy or relapse within 12 months after stem-cell transplantation had inferior survival with a median OS of 5.9 months(95%CI=5.5-7.1 months)and 2-year OS rate of 16%(95%CI=12%-20%).International prognostic index score 4-5(hazard ratio[HR]=2.22;95%CI=1.47-3.35),central nervous systemrelapse(HR=1.43;95%CI=1.04-1.97),and best response status(HR=2.68;95%CI=1.42-5.03 for partial remission.HR=5.97,95%CI=3.21-11.11 for stable disease/progressive disease)were independent unfavorable prognostic factors.Conclusions:This is the first large-scale Asian cohort study focusing on outcomes of refractory DLBCL.The definition of the SCHOLAR-1 study identifies patients with homogenously inferior survival,thus is appropriate to select refractory DLBCL.Due to poor clinical outcomes in the rituximab era,patients with refractory DLBCL may be potential candidates for novel treatment modalities.Shuo Wang Li Wang Jianda Hu Wenbin Qian Xi Zhang Yu Hu Qi Zhu Bobin Chen Depei Wu Chung-Chou H.Chang Pengpeng Xu Xiaoyun Zheng Juying Wei Yao Liu Guohui Cui Yong Tang Yan Ma Haiwen Huang Hongmei Yi Weili Zhao 2021Cancer Communications2021,41,3:10

6Active greeting technique: a mother-and-child catheter based technique to facilitate retrograde wire externalization in recanalization of coronary chronic total occlusion显示文摘Although retrograde approach has greatly improved the success rate of percutaneous coronary intervention(PCI) for coronary chronic total occlusion(CTO), retrograde wire externalization still remains challenging and time-consuming in some cases. Cases utilizing ‘‘Active Greeting Technique(AGT)', a mother-and-child catheter based technique to facilitate retrograde wire externalization, were extracted from Chronic Total Occlusion Club, China(CTOCC) database. AGT was performed by deep intubation a mother-and-child catheter(GuidezillaTMextension, 4 or 5 Fr inner catheter, and etc.) in combination with either reverse controlled antegrade or retrograde subintimal tracking(CART) technique or retrograde wire crossing technique. A total of 111 patients with 112 CTO lesions treated with this technique were retrospectively analyzed. Reverse CART technique and retrograde wire crossing technique were performed in 90.2% and 9.8% of all procedures. The utilization of GuidezillaTMextension, 4 Fr, and 5 Fr inner catheter accounted for 94.6%, 3.6%, and 1.8%, respectively. Externalization of retrograde wire was successful in all cases. No procedural complications were adjudicated to AGT. Complications independent of AGT included two target vessel perforations and two collateral perforations. No in-hospital major adverse cardiac events were found. 7miR-210 over-expression enhances mesenchymal stem cell survival in an oxidative stress environment through antioxidation and c-Met pathway activation显示文摘microRNA-210(miR-210)has generally been reported to be associated with cell survival under hypoxia.However,there are few data regarding the role of miR-210 in the survival of mesenchymal stem cells(MSCs)under oxidative stress conditions.Thus,we sought to investigate whether miR-210 over-expression could protect MSCs against oxidative stress injury and what the primary mechanisms involved are.The results showed that over-expression of miR-210 significantly reduced the apoptosis of MSCs under oxidative stress,accompanied by obvious increases in cell viability and superoxide dismutase activity and remarkable decreases in malonaldehyde content and reactive oxygen species production,resulting in a noticeable reduction of apoptotic indices when compared with the control.Moreover,the above beneficial effects of miR-210 could be significantly reduced by c-Met pathway repression.Collectively,these results showed that miR-210 over-expression improved MSC survival under oxidative stress through antioxidation and c-Met pathway activation,indicating the potential development of a novel approach to enhance the efficacy of MSC-based therapy for injured myocardium.XU JianFeng HUANG ZheYong LIN Li FU MingQiang GAO YanHua SHEN YunLi ZOU YunZeng SUN AiJun QIAN JuYing GE JunBo 2014Science China(Life Sciences)2014,57,10:10
8Changes in sediment discharge in a sediment-rich region of the Yellow River from 1955 to 2010: implications for further soil erosion control显示文摘The well-documented decrease in the discharge of sediment into the Yellow River has attracted considerable attention in recent years. The present study analyzed the spatial and temporal variation of sediment yield based on data from 46 hydrological stations in the sediment-rich region of the Yellow River from 1955 to 2010. The results showed that since 1970 sediment yield in the region has clearly decreased at different rates in the 45 sub-areas controlled by hydrological stations. The decrease in sediment yield was closely related to the intensity and extent of soil erosion control measures and rainstorms that occurred in different periods and sub-areas. The average sediment delivery modulus(SDM) in the study area decreased from 7,767.4 t/(km2?a) in 1951–1969 to 980.5 t/(km2?a) in 2000–2010. 10Soil erosion and sediment interception by check dams in a watershed for an extreme rainstorm on the Loess Plateau,China显示文摘The magnitude of soil erosion and sediment load reduction efficiency of check dams under extreme rainstorms is a long-standing concern.The current paper aims to use check dams to deduce the amount of soil erosion under extreme rainstorms in a watershed and to identify the difference in sediment interception efficiency of different types of check dams.Based on the sediment deposition at 12 check dams with 100% sediment interception efficiency and sub-catchment clustering by taking 12 damcontrolled catchments as clustering criteria,the amount of soil erosion resulting from an extreme rainstorm event on July 26,2017(named '7·26' extreme rainstorm) was estimated in the Chabagou watershed in the hill and gully region of the Loess Plateau.The differences in the sediment interception efficiency among the check dams in the watershed were analyzed according to field observations at 17 check dams.The results show that the average erosion intensity under the '7-26' extreme rainstorm was approximately 2.03×10~4 t/km^2,which was 5 times that in the second largest erosive rainfall in 2017(4.15×10~3 t/km^2) and 11-384 times that for storms in 2018(0.53×10~2 t/km^2-1.81×10~3 t/km^2).Under the '7-26' extreme rainstorm,the amount of soil erosion in the Chabagou watershed above the Caoping hydrological station was 4.20 × 106 t.The sediment interception efficiency of the check dams with drainage canals(including the destroyed check dams) and with drainage culverts was 6.48 and 39.49%,respectively.The total actual sediment amount trapped by the check dams was 1.11 × 106 t,accounting for 26.36% of the total amount of soil erosion.In contrast,3.09×106 t of sediment were input to the downstream channel,and the sediment deposition in the channel was 2.23×10~6 t,accounting for 53.15%of the total amount of soil erosion.The amount of sediment transport at the hydrological station was8.60×10~5 t.The Sediment Delivery Ratio(SDR) under the '7·26' extreme rainstorm was 0.21.The results indicated that the amount of soil erosion was huge,and the sediment interception efficiency of the check dams was greatly reduced under extreme rainstorms.It is necessary to strengthen the management and construction technology standards of check dams to improve the sediment interception efficiency and flood safety in the watershed.Leichao Bai Nan Wang Juying Jiao Yixian Chen Bingzhe Tang Haolin Wang Yulan Chen Xiqin Yan Zhijie Wang 2020International Journal of Sediment Research2020,35,4:7

13Effect of different levels of nitrogen deficiency on switchgrass seedling growth显示文摘Switchgrass(Panicum virgatum L.) is a warm-season rhizomatous perennial grass that can tolerate diverse abiotic stresses while yielding relatively high biomass, and is considered a leading biofuel feedstock for marginal lands. Nitrogen(N) is crucial for the growth and development of switchgrass, and its tolerance to low N supply and high N use efficiency are very important for its production under poor conditions. The large-scale planting of switchgrass on marginal lands could be an effective approach to solving the problem of feedstock supply for biomass energy. This study used a hydroponic experiment to evaluate the effect of N deficiency on switchgrass seedlings. Three N treatments(0, 0.15, and 1.50 mmol L-1Hoagland's solution)and six cultivars were used, three of each ecotype(upland and lowland). The results showed that biomass, leaf area, root surface area, net photosynthesis, and total chlorophyll content significantly decreased under low N treatments compared with those in full strength Hoagland's nutrient solution. However, once established, all plants survived extreme N stress(0 mmol L-1) and, to some extent, were productive. Cultivar Kanlow performed best of the six cultivars under stress. Significant interactions between stress treatment and cultivars showed that breeding for cultivars with high yield and superior performance under N deficiency is warranted. 14Antiviral activity of Jinchai capsule against influenza virus显示文摘OBJECTIVE:To evaluate the effect on influenza virus of Jinchai,a capsule made of Traditional Chinese Medicine.METHODS:Madin-darby canine kidney(MDCK) cells were infected with the FM1 strain of influenza virus A(subtype H1N1) in vitro.They were used to explore how Jinchai affected cell adsorption,cell membrane fusion,transcription and replication of the influenza virus.Hemagglutinin(HA) protein,intracellular pH,and influenza virus protein acid(PA) polymerase subunit were detected with confocal microscopy and real-time fluorescent quantitative polymerase chain reaction.RESULTS:Jinchai significantly reduced the expression of HA and PA polymerase subunit mRNA in infected MDCK cells.Jinchai also significantly decreased intracellular pH in infected cells.CONCLUSIONS:Jinchai had strong anti-influenza activity against the influenza virus.It weakened the ability of the influenza virus to adsorb to cell wall and fuse with cell membranes in the early infection stage,and inhibited the transcription and replication of the virus.Juying Zhong Xiaolan Cui Yujing Shi Yingjie Gao Hongxin Cao 2013Journal of Traditional Chinese Medicine2013,33,2:4

19Propofol blocks apoptosis and Bcl-2 and Bax expression induced by hypoxia-reoxygenation in primary cultures of rat hippocampal astrocytes显示文摘BACKGROUND:Cerebral hippocampal astrocytes are more sensitive to ischemic injury than neurons.Hypoxic-ischemic brain injury induces profound astrocyte apoptosis,and propofol may protect against astrocyte apoptosis. OBJECTIVE:To verify the protective effects of propofol against astrocyte apoptosis and to investigate anti-apoptotic Bcl-2 and pro-apoptotic Bax expression in primary cultures of rat hippocampal astrocytes exposed to hypoxia-reoxygenation for different periods of time following propofol treatment. DESIGN,TIME,AND SETTING:In vitro neural immunocytochemistry was performed at the Central Laboratory of Yunyang Medical College between September 2007 and March 2008. MATERIALS:A total of 30 Wistar rats,aged 1-3 days,with equal numbers of males and females, were included for isolation and culture of hippocampal astrocytes. METHODS:Hippocampal astrocytes were purified and cultured for 3 weeks and treated with four culture conditions:50μL Hank's solution(normal control);0.2 mL/L Intralipid;50μL Hank's solution for 10 minutes followed by hypoxic incubation for 4 hours and normoxic incubation for 12,24,36,48, 60 or 72 hours;propofol(250μmol/L final) for 10 minutes followed by hypoxic incubation for 4 hours and normoxic incubation for 12,24,36,48,60 and 72 hours. MAIN OUTCOME MEASURES:(1) Morphologic changes in hippocampal astrocytes.(2) Levels of astrocyte apoptosis and Bcl-2 and Bax expression. RESULTS:Hypoxia and reoxygenation increased apoptosis over time,with Bcl-2 expression peaking at 24 hours and decreasing gradually(P<0.01);Bax expression peaked at 72 hours(P< 0.01);the ratio of Bcl-2/Bax was 1.4,0.8,and 0.6,respectively,at 24,48 and 72 hours. Non-apoptotic astrocytes showed significant proliferation and swelling.Propofol treatment decreased apoptosis after hypoxia-reoxygenation(P<0.01),as well as Bcl-2 and Bax expression (P<0.05,P<0.01),with Bcl-2/Bax ratios of 1.6-1.8.Propofol treatment also blocked astrocyte proliferation and swelling.No apoptotic cells or Bcl-2/Bax expression was detected in astrocytes cultured in Hank's or Intralipid solution.
