1The biological stress of early weaned piglets显示文摘Pigs experience biological stress such as physiological, environmental, and social challenges when weaned from the sow. The process of weaning is one of the most stressful events in the pig's life that can contribute to intestinal and immune system dysfunctions that result in reduced pig health, growth, and feed intake, particularly during the first week after weaning. Technological improvements in housing, nutrition, health, and management have been used to minimize some of the adverse effects of weaning stress, but a greater understanding of the biological impact of stress is needed to improve strategies to overcome weaning stress. 4Sitagliptin in patients with non-alcoholic steatohepatitis: A randomized, placebo-controlled trial显示文摘AIM To evaluate the effect of sitagliptin vs placebo on histologic and non-histologic parameters of nonalcoholic steatohepatitis(NASH).METHODS Twelve patients with biopsy-proven NASH were randomized to sitagliptin(100 mg daily)(n=6)or placebo(n=6)for 24 wk.The primary outcome was improvement in liver fibrosis after 24 wk.Secondary outcomes included evaluation of changes in NAFLD activity score(NAS),individual components of NAS(hepatocyte ballooning,lobular inflammation,and steatosis),glycemic control and insulin resistance[including measurements of glycated hemoglobin(Hb A1C)and adipocytokines],lipid profile including free fatty acids,adipose distribution measured using magnetic resonance imaging(MRI),and thrombosis markers(platelet aggregation and plasminogen activator inhibitor 1 levels).We also sought to determine the correlation between changes in hepatic fat fraction(%)[as measured using the Iterative Decomposition of water and fat with Echo Asymmetry and Least-squares estimation(IDEAL)MRI technique]and changes in hepatic steatosis on liver biopsy.RESULTS Sitagliptin was not significantly better than placebo at reducing liver fibrosis score as measured on liver biopsy(mean difference between sitagliptin and placebo arms,0.40,P=0.82).There were no significant improvements evident with the use of sitagliptin vs placebo for the secondary histologic outcomes of NAS total score as well as for the individual components of NAS.Compared to baseline,those patients who received sitagliptin demonstrated improved Hb A1C(6.7%±0.4%vs 7.9%±1.0%,P=0.02),and trended towards improved adiponectin levels(4.7±3.5μg/m L vs 3.9±2.7μg/m L,P=0.06)and triglyceride levels(1.26±0.43 mmol/L vs 2.80±1.64 mmol/L,P=0.08).However,when compared with placebo,sitagliptin did not cause a statistically significant improvement in Hb A1C(mean difference,-0.7%,P=0.19)nor triglyceride levels(mean difference-1.10mmol/L,P=0.19)but did trend towards improved adiponectin levels only(mean difference,0.60μg/m L,P=0.095).No significant changes in anthropometrics,liver enzymes,other adipocytokines,lipid profile,thrombosis parameters,or adipose distribution were demonstrated.The MRI IDEAL procedure correlated well with steatosis scores obtained on liver biopsy in both groups at baseline and post-treatment,and the Spearman correlation coefficients ranged from r=0.819(baseline)to r=0.878(post-treatment),P=0.002.CONCLUSION Sitagliptin does not improve fibrosis score or NAS after 24 wk of therapy.The MRI IDEAL technique may be useful for non-invasive measurement of hepatic steatosis.Tisha R Joy Charles A McKenzie Rommel G Tirona Kelly Summers Shannon Seney Subrata Chakrabarti Neel Malhotra Melanie D Beaton 2017World Journal of Gastroenterology2017,23,1:18
5Increased hepcidin expression in colorectal carcinogenesis显示文摘AIM:To investigate whether the iron stores regulator hepcidin is implicated in colon cancer-associated anae- mia and whether it might have a role in colorectal car- cinogenesis. METHODS: Mass spectrometry (MALDI-TOF MS and SELDI-TOF MS) was employed to measure hepcidin in urine collected from 56 patients with colorectal cancer. Quantitative Real Time RT-PCR was utilised to determine hepcidin mRNA expression in colorectal cancer tissue. Hepcidin cellular localisation was determined using im- munohistochemistry. RESULTS: We demonstrate that whilst urinary hepcidin expression was not correlated with anaemia it was posi- tively associated with increasing T-stage of colorectal cancer (P < 0.05). Furthermore, we report that hepcidin mRNA is expressed in 34% of colorectal cancer tissue specimens and was correlated with ferroportin repres- sion. This was supported by hepcidin immunoreactivity in colorectal cancer tissue. CONCLUSION: We demonstrate that systemic hepcidin expression is unlikely to be the cause of the systemic anaemia associated with colorectal cancer. 6Heterogeneous molecular catalysts for electrocatalytic CO2 reduction显示文摘This review provides an overview of the literature regarding heterogeneous molecular catalysts for electrochemical CO2 reduction (ECR).Fundamental aspects of the science,including aggregation,electrochemical rate laws,and electrode-catalyst electronic coupling,are discussed to provide a solid foundation on which to design experiments and interpret results.Mechanistic aspects of ECR are presented based on electrokinetic and spectroscopic measurements as well as density functional theory (DFT) calculations.Consensus is improving for electrokinetic measurements,but the redox state of the metal center under reaction conditions and DFT reaction pathways lack agreement in the literature.Concerning the tunable aspects of the molecular catalyst,the impacts of the metal center,ligand substituents,and electrode support on the activity and selectivity toward ECR are presented with an emphasis on those studies that controlled for aggregation and minimized mass-transport limitations.Extended three-dimensional (3D) structures such as polymers,metal-organic frameworks (MOFs),and covalent-organic frameworks (COFs) are discussed as highly tunable architectures that begin to mimic the catalytic pockets of enzyme active sites.To achieve the full potential of these catalysts,design principles must emerge based on a combination of deconvoluting measurements to extract intrinsic catalyst properties and more reliable theoretical calculations to predict reaction pathways.Nathan Corbin Joy Zeng Kindle Williams Karthish Manthiram 2019Nano Research2019,12,9:10

8Advances in Vehicular Ad-hoc Networks(VANETs):Challenges and Road-map for Future Development显示文摘Recent advances in wireless communication technologies and auto-mobile industry have triggered a significant research interest in the field of vehicular ad-hoc networks(VANETs) over the past few years. A vehicular network consists of vehicle-to-vehicle(V2V) and vehicle-to-infrastructure(V2I) communications supported by wireless access technologies such as IEEE 802.11 p. This innovation in wireless communication has been envisaged to improve road safety and motor traffic efficiency in near future through the development of intelligent transportation system(ITS). Hence, governments, auto-mobile industries and academia are heavily partnering through several ongoing research projects to establish standards for VANETs. The typical set of VANET application areas, such as vehicle collision warning and traffic information dissemination have made VANET an interesting field of mobile wireless communication. 10Salivary mycobiome dysbiosis and its potential impact on bacteriome shifts and host immunity in oral lichen planus显示文摘The biodiversity of the mycobiome,an important component of the oral microbial community,and the roles of fungal–bacterial and fungal–immune system interactions in the pathogenesis of oral lichen planus (OLP) remain largely uncharacterized.In this study,we sequenced the salivary mycobiome and bacteriome associated with OLP.First,we described the dysbiosis of the microbiome in OLP patients,which exhibits lower levels of fungi and higher levels of bacteria.Significantly higher abundances of the fungi Candida and Aspergillus in patients with reticular OLP and of Alternaria and Sclerotiniaceae_unidentified in patients with erosive OLP were observed compared to the healthy controls.Aspergillus was identified as an “OLP-associated” fungus because of its detection at a higher frequency than in the healthy controls.Second,the co-occurrence patterns of the salivary mycobiome–bacteriome demonstrated negative associations between specific fungal and bacterial taxa identified in the healthy controls,which diminished in the reticular OLP group and even became positive in the erosive OLP group.Moreover,the oral cavities of OLP patients were colonized by dysbiotic oral flora with lower ecological network complexity and decreased fungal–Firmicutes and increased fungal–Bacteroidetes sub-networks.Third,several keystone fungal genera (Bovista,Erysiphe,Psathyrella,etc.) demonstrated significant correlations with clinical scores and IL-17 levels.Thus,we established that fungal dysbiosis is associated with the aggravation of OLP.Fungal dysbiosis could alter the salivary bacteriome or may reflect a direct effect of host immunity,which participates in OLP pathogenesis.Yan Li Kun Wang Bo Zhang Qichao Tu Yufei Yao Bomiao Cui Biao Ren Jinzhi He Xin Shen Joy D.Van Nostrand Jizhong Zhou Wenyuan Shi Liying Xiao Changqing Lu Xuedong Zhou 2019International Journal of Oral Science2019,11,3:8

12Lubiprostone vs Senna in postoperative orthopedic surgery patients with opioid-induced constipation:A double-blind,active-comparator trial显示文摘AIM:To investigate the efficacy of lubiprostone compared to Senna on bowel symptoms and constipation in post-operative orthopedic patients treated with opioids.METHODS:In this double blind,randomized,active comparator trial,adults who required opioids for analgesia following orthopedic procedures and who were admitted in inpatient rehabilitation were randomized following baseline assessments to lubiprostone(Amitza),orally twice aday or Senna(generic)two capsules administered daily for six days.Subjects were assessed using the patient assessment of constipation(PAC)-symptoms(PAC-SYM)and the PAC-quality of life(PAC-QOL)scales measured at baseline and Day 7;Subjects were assessed daily for secondary measures included the Bristol stool scale bowel consistency,specific bowel symptom score(Nausea,cramping,straining,completeness,abdominal pain,time per lavatory attempt,assistance needed),adverse events and rescue medications required.Function was measured using the functional independence measure(FIM)at admission and discharge;length of stay(LOS)and missed treatments due to gastrointestinal symptoms were also assessed.RESULTS:64 adults were enrolled;56 participants(28 in each group)had baseline and follow up measures and were included in the intention to treat(ITT)analyses.43 participants completed the study,21 in the active lubiprostone and 22 in the active Senna group.The mean age of the participants was 71.5years(SD=11.4 years,range:28-96 years).In the ITT analyses,participants showed significant improvement in bowel symptoms as measured by the PACSYM(mean±SD,-0.28±0.60,range:-1-2.33)and PAC-QOL(mean±SD,0.33±0.81,range:-1.5-2.0)over time,but there were no significant differences between the lubiprostone and Senna groups in mean change in the PAC-SYM(-0.20±0.60 vs-0.36±0.61,P=0.61 respectively)or the PAC-QOL(0.29±0.76 vs0.37±0.87,P=0.61 respectively).The mean change in each bowel symptom also did not significantly differ between treatment groups on ITT analyses,except for completeness of bowel movement,with the Senna group showing greater negative mean change in bowel movement completeness(-0.56±1.01 vs-2.00±1.41,P=0.03)and for reduction of abdominal pain,favoring Senna(-0.14±0.73 vs-0.73±1.08,P=0.04).Fifteen(75%)participants in the lubiprostone and in the Senna group requested rescue treatments.Participants made significant functional improvement from admission to discharge over a median LOS of 12 d,with a mean FIM change of 29.13±13.58 and no significant between group differences(27.0±9.2 vs 31.5±16.6,P=0.27).CONCLUSION:Both lubiprostone and Senna improved constipation-related symptoms and QOL in opioid-induced constipation,with no significant between-group differences.Christina M Marciniak Santiago Toledo Jungwhalia Lee Michael Jesselson Jillian Bateman Benjamin Grover Joy Tierny 2014World Journal of Gastroenterology2014,20,43:7

15胚乳特异sbeⅡa启动子在小麦中的评价及特性分析显示文摘淀粉的合成是一个复杂的生化过程。sbeⅡa基因是淀粉合成过程中的关键酶基因之一,在小麦籽粒成熟过程中对淀粉,尤其对支链淀粉的生物合成调控起重要作用。为了解sbeⅡa基因的调控机理及其胚乳特异表达特性,利用APCR方法克隆了sbeⅡa启动子3094 bp片段,并对该片段进行测序,同时利用GUS瞬间表达体系对sbeⅡa启动子序列进行不同片段的功能缺失研究。研究结果表明,3094 bp序列(存在于sbe.g融合体中)具有稳定的启动子活性,而5'或3'缺失体的启动子活性明显降低。在sbeⅡa启动子中间缺失体中,一些缺失体仅有微弱活性,但sbe.e在缺失-1579—1210 bp片段情况下,却具有比sbe.g(含启动子全长序列)还要高的启动子活性,研究初步证明,一些固定序列(motifs),如-300 bp因子、G盒以及醇溶蛋白盒(Prolamin box)等作为正调控因子在决定启动子胚乳特异表达模式中是必需的,而且在-1579—1210 bp片段中也存在一些负调控因子或固定序列。在瞬间表达检测体系中,小麦胚乳组织年龄对检测结果有着重要影响。苗红梅 Fleming Joy E 吕德彬 韩锦峰 2004科学通报2004,49,5:7
16Mitochondrial dysfunction activates the AMPK signaling and autophagy to promote cell survival显示文摘Autophagy is a cellular self-eating process essential for stress response and maintainingtissue homeostasis by lysosomal degradation of unwanted or damaged proteins and organelles.Here, we show that cells with defective mitochondria induce autophagy to promotecell survival through activating the AMPK pathway. Loss of mitochondrial complex III proteincytochrome b activates the AMPK signaling and induced autophagy. Inhibiting mitochondria energeticsby mitochondria-targeted agents activates the AMPK signaling and induced autophagy.Genetic inhibition of AMPK inhibits autophagy induction in cells with defective mitochondria,while genetic inhibition of autophagy has no effect on AMPK activation. Mitochondria dysfunctionhas no effect of DNA repair of UV-induced DNA damage. However, mitochondria dysfunctionsensitizes cells to apoptosis induced by UV radiation. Genetic inhibition of autophagy orAMPK sensitized cells to apoptosis in cells with defective mitochondria. Our results demonstratethat AMPK and autophagy senses mitochondria dysfunction and serves as a mechanismfor survival. 18The crystal structure of the non-liganded 14-3-3σ protein: insights into determinants of isoform specific ligand binding and dimerization显示文摘Seven different, but highly conserved 14-3-3 proteins are involved in diverse signaling pathways in human cells. It isunclear how the 14-3-3σ isoform, a transcriptional target of p53, exerts its inhibitory effect on the cell cycle in thepresence of other 14-3-3 isoforms, which are constitutively expressed at high levels. In order to identify structuraldifferences between the 14-3-3 isoforms, we solved the crystal structure of the human 14-3-3σ protein at a resolutionof 2.8 ? and compared it to the known structures of 14-3-3ζ and 14-3-3τ. The global architecture of the 14-3-3σ foldis similar to the previously determined structures of 14-3-3ζ and 14-3-3τ: two 14-3-3σ molecules form a cup-shapeddimer. Significant differences between these 14-3-3 isoforms were detected adjacent to the amphipathic groove, whichmediates the binding to phosphorylated consensus motifs in 14-3-3-ligands. Another specificity determining region islocalized between amino-acids 203 to 215. These differences presumably select for the interaction with specific ligands,which may explain the different biological functions of the respective 14-3-3 isoforms. Furthermore, the two 14-3-3σmolecules forming a dimer differ by the spatial position of the ninth helix, which is shifted to the inside of the ligandinteraction surface, thus indicating adaptability of this part of the molecule. In addition, 5 non-conserved residues arelocated at the interface between two 14-3-3σ proteins forming a dimer and represent candidate determinants of homo-and hetero-dimerization specificity.
