| 2 | B7-H7 (HHLA2) inhibits T-cell activation and proliferation in the presence of TCR and CD28 signaling显示文摘Modulation of T-cell responses has played a key role in treating cancers and autoimmune diseases.Therefore,understanding how different receptors on T cells impact functional outcomes is crucial.The influence of B7-H7(HHLA2)and CD28H(TMIGD2)on T-cell activation remains controversial.Here we examined global transcriptomic changes in human T cells induced by B7-H7.Stimulation through TCR with OKT3 and B7-H7 resulted in modest fold changes in the expression of select genes;however,these fold changes were significantly lower than those induced by OKT3 and B7-1 stimulation.The transcriptional changes induced by OKT3 and B7-H7 were insufficient to provide functional stimulation as measured by evaluating T-cell proliferation and cytokine production.Interestingly,B7-H7 was coinhibitory when simultaneously combined with TCR and CD28 stimulation.This inhibitory activity was comparable to that observed with PD-L1.Finally,in physiological assays using T cells and APCs,blockade of B7-H7 enhanced T-cell activation and proliferation,demonstrating that this ligand acts as a break signal.Our work defines that the transcriptomic changes induced by B7-H7 are insufficient to support full costimulation with TCR signaling and,instead,B7-H7 inhibits T-cell activation and proliferation in the presence of TCR and CD28 signaling. | Sadiye Amcaoglu Rieder Jingya Wang Natalie White Ariful Qadri Catherine Menard Geoffrey Stephens Jodi LKarnell Christopher ERudd Roland Kolbeck | 2021 | Cellular & Molecular Immunology2021,18,6: | 9 |

This 1 kW average power diode-pumped solid-state laser(DPSSL) is based on a master oscillator power amplifier(MOPA) design, which includes two cryogenic gas cooled amplifier stages based on DiPOLE multi-slab ceramic Yb:YAG amplifier technology developed at the CLF. The laser will produce pulses between 2 and 15 ns in duration with precise, arbitrarily selectable shapes, at pulse repetition rates up to 10 Hz, allowing real-time shape optimization for compression experiments. | 11 | An overview of the latest developments in facial imaging显示文摘Facial imaging is a term used to describe methods that use facial images to assist or facili-tate human identification. This pertains to two craniofacial identification procedures that use skulls and faces—facial approximation and photographic superimposition—as well as face-only methods for age progression/regression, the construction of facial graphics from eye-witness memory (including composites and artistic sketches), facial depiction, face mapping and newly emerging methods of molecular photofitting. Given the breadth of these facial imaging techniques, it is not surprising that a broad array of subject-matter experts partici-pate in and/or contribute to the formulation and implementation of these methods (includ-ing forensic odontologists, forensic artists, police officers, electrical engineers, anatomists, geneticists, medical image specialists, psychologists, computer graphic programmers and software developers). As they are concerned with the physical characteristics of humans, each of these facial imaging areas also falls in the domain of physical anthropology, although not all of them have been traditionally regarded as such. This too offers useful opportunities to adapt established methods in one domain to others more traditionally held to be disciplines within physical anthropology (e.g. facial approximation, craniofacial super-imposition and face photo-comparison). It is important to note that most facial imaging methods are not currently used for identification but serve to assist authorities in narrowing or directing investigations such that other, more potent, methods of identification can be used (e.g. DNA). Few, if any, facial imaging approaches can be considered honed end-stage scientific methods, with major opportunities for physical anthropologists to make meaningful contributions. Some facial imaging methods have considerably stronger scientific underpin-nings than others (e.g. facial approximation versus face mapping), some currently lie entirely within the artistic sphere (facial depiction), and yet others are so aspirational that realistic capacity to obtain their aims has strongly been questioned despite highly advanced tech-nical approaches (molecular photofitting). All this makes for a broad-ranging, dynamic and energetic field that is in a constant state of flux. | 15 | Current management of gastro-oesophageal reflux disease—treatment costs,safety profile,and effectiveness:a narrative review显示文摘Background The purpose of this study was to review the current management of gastro-oesophageal reflux disease(GORD),including treatment costs,safety profile and effectiveness.Methods A literature review was performed of randomized-controlled trials,systematic reviews,Cochrane reports and National/Societal guidelines of themedical,endoscopic and surgicalmanagement of GORD.Proton pump inhibitor(PPI)prescribing patterns and expenditure were reviewed in different countries,including Australia,Canada,New Zealand,UK and USA.Results Proton pump inhibitors(PPIs)are primarily indicated for control of GORD,Helicobacter pylori eradication(combined with antibiotics),preventing NSAID-induced gastrointestinal bleeding and treating peptic ulcer disease.There is widespread overprescribing of PPIs in Western and Eastern nations in terms of indication and duration,with substantial expense for national health providers.Despite a favourable short-term safety profile,there are observational associations of adverse effects with long-term PPIs.These include nutrient malabsorption,enteric infections and cardiovascular events.The prevalence of PPI use makes their long-termsafety profile clinically relevant.Cost-benefit,symptom control and quality-oflife outcomes favour laparoscopic fundoplication rather than chronic PPI treatment.Laparoscopic fundoplication in long-termmanagement of PPI-responsive GORD is supported by SAGES,NICE and ACG,and PPI-refractory GORD by AGA and SAGES guidelines.The importance of establishing a definitive diagnosis prior to invasive management is emphasized,especially in PPI-refractory heartburn.Conclusions We examined evidence-based guidelines for PPI prescribing and deprescribing in primary care and hospital settings and the need for PPI stewardship and education of health professionals.This narrative review presents the advantages and disadvantages of surgical,endoscopic and medical management of GORD,which may assist in shared decision making and treatment choice in individual patients. | Tahmina Lata Jodie Trautman Philip Townend Robert B.Wilson | 2023 | Gastroenterology Report2023,11,1: | 3 |

| 18 | AIF-regulated oxidative phosphorylation supports lung cancer development显示文摘Cancer is a major and still increasing cause of death in humans. Most cancer cells have a fundamentally different metabolic profile from that of normal tissue. This shift away from mitochondrial ATP synthesis via oxidative phosphorylation towards a high rate of glycolysis, termed Warburg effect, has long been recognized as a paradigmatic hallmark of cancer, supporting the increased biosynthetic demands of tumor cells. Here we show that deletion of apoptosis-inducing factor (AIF) in a Kras^G12D-driven mouse lung cancer model resulted in a marked survival advantage, with delayed tumor onset and decreased malignant progression. Mechanistically, Aif deletion leads to oxidative phosphorylation (OXPHOS) deficiency and a switch in cellular metabolism towards glycolysis in non-transformed pneumocytes and at early stages of tumor development. Paradoxically, although Aif-deficient cells exhibited a metabolic Warburg profile, this bioenergetic change resulted in a growth disadvantage of Kras^G12D-driven as well as Kras wild-type lung cancer cells. Cell-autonomous re-expression of both wild-type and mutant AIF (displaying an intact mitochondrial, but abrogated apoptotic function) in Aif-knockout Kras^G12D mice restored OXPHOS and reduced animal survival to the same level as AIF wild-type mice. In patients with non-small cell lung cancer, high AIF expression was associated with poor prognosis. | 20 | Health Disparities among the Western, Central and Eastern Rural Regions of China after a Decade of Health Promotion and Disease Prevention Programming显示文摘Health disparities between the western, central and eastern regions of rural China, and the impact of national health improvement policies and programming were assessed. A total of 400 counties were randomly sampled. ANOVA and Logistic regression modeling were employed to estimate differences in health outcomes and determinants. Significant differences were found between the western, central and eastern rural regions in community infrastructure and health outcomes. From 2000 to 2010, health indicators in rural China were improved significantly, and the infant mortality rate(IMR), maternal mortality rate(MMR) and under 5 mortality rate(U5MR) had fallen by 62.79%, 71.74% and 61.92%, respectively. Central rural China had the greatest decrease in IMR(65.05%); whereas, western rural China had the greatest reduction in MMR(72.99%) but smallest reduction in U5MR(57.36%). Despite these improvements, Logistic regression analysis showed regional differences in key health outcome indicators(odds ratios): IMR(central: 2.13; western: 5.31), U5MR(central: 2.25; western: 5.69), MMR(central: 1.94; western: 3.31), and prevalence of infectious diseases(central: 1.62; western: 3.58). The community infrastructure and health outcomes of the western and central rural regions of China have been improved markedly during the first decade of the 21 st century. However, health disparities still exist across the three regions.
