7Prevalence and evolution of drug resistance HIV-1 variants in Henan, China显示文摘To understand the prevalence and evolution of drug resistant HIV strains in Henan China after the implementation of free antiretroviral therapy for AIDS patients. 45 drug nave AIDS patients, 118 AIDS patients who received three months antiretroviral therapy and 124 AIDS patients who received six months antiretroviral treatment were recruited in the southern part of Henan province. Information on general condition, antiretroviral medicines, adherence and clinical syndromes were collected by face to face interview. Meanwhile, 14ml EDTA anticoagulant blood was drawn. CD4/CD8 T cell count, viral load and genotypic drug resistance were tested. The rates of clinical improvement were 55.1% and 50.8% respectively three months and six months after antiretroviral therapy. The mean CD4 cell count after antiretroviral therapy was significantly higher than in drug nave patients. The prevalence rate of drug resistant HIV strains were 13. 9%, 45.4% and 62.7% in drug nave patients, three month treatment patients and six month treatment patients, respectively. The number of resistance mutation codons and the frequency of mutations increased significantly with continued antiretroviral therapy. The mutation sites were primarily at the 103, 106 and 215 codons in the three-month treatment group and they increased to 15 codon mutations in the six-month treatment group. From this result, the evolution of drug resistant strains was inferred to begin with the high level NNRTI resistant strain, and then develop low level resistant strains to NRTIs. The HIV strains with high level resistance to NVP and low level resistance to AZT and DDI were highly prevalent because of the AZT+DDI+NVP combination therapy. These HIV strains were also cross resistant to DLV, EFV, DDC and D4T. Poor adherence to therapy was believed to be the main reason for the emergence and prevalence of drug resistant HIV strains. The prevalence of drug resistant HIV strains was increased with the continu- ation of antiretroviral therapy in the southern part of Henan province. 8Lime and Phosphate Could Reduce Cadmium Uptake by Five Vegetables Commonly Grown in South China显示文摘A pot experiment was conducted in artificially Cd-contaminated (5 mg Cd kg-1) soils to investigate the feasibility of using lime (3 g kg-1) or phosphate (80 mg P kg-1) to mitigate uptake of Cd by vegetables.Five common vegetables in South China,including lettuce (Lactuca sativa L.var.ramosa Hort.),Chinese cabbage [Brassica rapa L.subsp.Chinensis (L.) var.parachinensis (L.H.Bailey) Hanect],Chinese broccoli (Brassica oleracea L.var.albiflora Kuntze),white amaranth (Amaranthus tricolor L.) and purslane (Amaranthus viridis L.),were grown in the soils and harvested after 60 d.The results showed that liming significantly reduced Cd uptake by most vegetables by 40%-50% (or a maximum of 70%),mainly due to immobilization of soil Cd.Increased availability of Ca in the soil might also contribute to the Cd uptake reduction as a result of absorption competition between Ca and Cd.Liming caused biomass reduction in white amaranth and purslane,but did not influence growth of the other vegetables.Phosphate decreased Cd uptake by vegetables by 12%-23%.Compared with lime,phosphate decreased,to a smaller extent,the bioavailability of Cd in most cases.Phosphate markedly promoted growth of vegetables.Changes in soil chemistry by adding lime or phosphate did not markedly influence nutrient uptake of vegetables except that lime increased Ca content and phosphate increased P content in shoots of the vegetables.The results suggested that a proper application of lime could be effective in reducing Cd uptake of vegetables,and phosphate could promote growth of the vegetables as well as alleviate the toxicity of Cd.TAN Wan-Neng LI Zhi-An QIU Jing ZOU Bi LI Ning-Yu ZHUANG Ping WANG Gang 2011Pedosphere2011,21,2:34
9The Association of Maternal Body Composition and Dietary Intake with the Risk of Gestational Diabetes Mellitus during the Second Trimester in a Cohort of Chinese Pregnant Women显示文摘Objective To investigate the association of maternal body composition and dietary intake with the risk of gestational diabetes mellitus(GDM). Methods A total 154 GDM subjects and 981 controls were enrolled in a prospective cohort study in 11 hospitals from May 20, 2012 to December 31, 2013. Bioelectrical impedance analysis and dietary surveys were used to determine body composition and to evaluate the intake of nutrients in subjects at 21-24 weeks' gestation(WG). Logistic regression analysis was applied to explore the relationships of maternal body composition and dietary intake with the risk of GDM morbidity. Results Age, pre-pregnant body weight(BW), and body mass index(BMI) were associated with increased risk of GDM. Fat mass(FM), fat mass percentage(FMP), extracellular water(ECW), BMI, BW, energy, protein, fat, and carbohydrates at 21-24 WG were associated with an increased risk of GDM. In contrast, fat free mass(FFM), muscular mass(MM), and intracellular water(ICW) were associated with a decreased risk of GDM. 12Serum leptin and soluble leptin receptor in non-alcoholic fatty liver disease显示文摘AIM: To determine the role of leptin system in non-al- coholic fatty liver disease (NAFLD) development by deli- neating the changes in serum levels of leptin and soluble leptin receptor (sOB-R). METHODS: Blood samples were collected from 30 consecutive patients with liver-biopsy-proven NAFLD and 30 patients with cholecystolithiasis (stationary phase) as controls. Serum leptin levels were determined by radio- immunoassay and concentration of sOB-R was measured by ELISA. Body mass index (BMI) was calculated for all subjects, and serum insulin, C-peptide, and lipoprotein levels were also detected. RESULTS: Mean serum leptin level and BMI in the NAFLD group were significantly higher than in the con- trols (both P < 0.001), but mean sOB-R level was lower in the NAFLD group when compared to the controls. Both men and women in the NAFLD group had higher mean serum leptin levels and lower sOB-R levels than did the men and women in the control group (all P < 0.001). The- re was a significant negative correlation between serum leptin and sOB-R levels (r = -0.725, P < 0.001). Multiva- riate analysis showed that the percentage of hepatocyte steatosis, sex, BMI, and homeostasis model assessment of insulin resistance (HOMA IR) were independently rela- ted to serum leptin levels. CONCLUSION: Elevated serum leptin seems to be afeature of steatosis, and serum leptin seems to increase as hepatocyte steatosis develops. 13Construction, expression and characterization of the engineered antibody against tumor surface antigen, p185^(c-erbB-2)显示文摘The c-erbB-2 proto-oncogene encodes a 185kDa protein p185, which belongs to epidermal growth factorreceptor family. Amplification of this gene has been shown to correlate with poor clinical prognosis forcertain cancer patients. The monoclonal antibody A21 which directed against p185 specifically inhibitsproliferation of tumor cells overexpressing p185, hence allows it to be a candidate for targeted therapy. Inorder to overcome several drawbacks of murine MAb, we cloned its VH and VL genes and constructed thesingle-chain Fv (scFv) through a peptide linker. The recombinant scFvA21 was expressed in Escherichiacoli and purified by the affinity column. Subsequently it was characterized by ELISA, Western blot, cellimmunohistochemistry and FACS. All these assays showed the binding activity to extracellular domain(ECD) of p185. Based on those properties of scFvA21, we further constructed the scFv-Fc fusion moleculewith a homodimer form and the recombinant product was expressed in mammalian cells. In a series ofsubsequent analysis this fusion protein showed identical antigen binding site and activity with the parentantibody. 14Personal Review:Sources of sulfide in waste streams and current biotechnologies for its removal显示文摘Sulfide-containing waste streams are generated by a number of industries. It is emitted into the environment as dis- solved sulfide (S2- and HS-) in wastewaters and as H2S in waste gases. Due to its corrosive nature, biological hydrogen sulfide removal processes are being investigated to overcome the chemical and disposal costs associated with existing chemically based removal processes. The nitrogen and sulfur metabolism interacts at various levels of the wastewater treatment process. Hence, the sulfur cycle offers possibilities to integrate nitrogen removal in the treatment process, which needs to be further optimized by appropriate design of the reactor configuration, optimization of performance parameters, retention of biomass and optimization of biomass growth. 19Exploring an Integrative Therapy for Treating COVID-19:A Randomized Controlled Trial显示文摘Objectives:To develop a new Chinese medicine(CM)-based drug and to evaluate its safety and effect for suppressing acute respiratory distress syndrome(ARDS)in COVID-19 patients.Methods:A putative ARDS-suppressing drug Keguan-1 was first developed and then evaluated by a randomized,controlled two-arm trial.The two arms of the trial consist of a control therapy(alpha interferon inhalation,50(x g twice daily;and lopinavir/ritonavir,400 and 100 mg twice daily,respectively)and a testing therapy(control therapy plus Keguan-119.4 g twice daily)by random number table at 1:1 ratio with 24 cases each group.After 2-week treatment,adverse events,time to fever resolution,ARDS development,and lung injury on newly diagnosed COVID-19 patients were assessed.Results:An analysis of the data from the first 30 participants showed that the control arm and the testing arm did not exhibit any significant differences in terms of adverse events.Based on this result,the study was expanded to include a total of 48 participants(24 cases each arm).The results show that compared with the control arm,the testing arm exhibited a significant improvement in time to fever resolution(P=0.035),and a significant reduction in the development of ARDS(P=0.048).Conclusions:Keguan-1-based integrative therapy was safe and superior to the standard therapy in suppressing the development of ARDS in COVID-19 patients.(Trial registration No.NCT 04251871 at www.clinicaltrials.gov).WANG Jia-bo WANG Zhong-xia JING Jing ZHAO Peng DONG Jing-hui ZHOU Yong-feng YANG Guang NIU Ming ZHAO Xu JIANG Tian-jun Bl Jing-feng XU Zhe ZHANG Ping WU Dan BAI Zhao-fang GUO Yu-ming YU Si-miao SUN Yong-qiang ZHANG Zi-teng ZHAN Xiao-yan LI Peng-yan DING Jin-biao ZHAO Peng-fei SONG Xue-ai TANG Jian-yuan HE Dong-chu CHEN Zhu QIN En-qiang WANG Rui-lin XIAO Xiao-he 2020Chinese Journal of Integrative Medicine2020,26,9:18
