1The nucleocapsid protein of rice stripe virus in cell nuclei of vector insect regulates viral replication显示文摘Rice stripe virus(RSV)transmitted by the small brown planthopper causes severe rice yield losses in Asian countries.Although viral nuclear entry promotes viral replication in host cells,whether this phenomenon occurs in vector cells remains unknown.Therefore,in this study,we systematically evaluated the presence and roles of RSV in the nuclei of vector insect cells.We observed that the nucleocapsid protein(NP)and viral genomic RNAs were partially transported into vector cell nuclei by utilizing the importin a nuclear transport system.When blocking NP nuclear localization,cytoplasmic RSV accumulation significantly increased.In the vector cell nuclei,NP bound the transcription factor YY1 and affected its positive regulation to FAIM.Subsequently,decreased FAIM expression triggered an antiviral caspase-dependent apoptotic reaction.Our results reveal that viral nuclear entry induces completely different immune effects in vector and host cells,providing new insights into the balance between viral load and the immunity pressure in vector insects.Wan Zhao Junjie Zhu Hong Lu Jiaming Zhu Fei Jiang Wei Wang Lan Luo Le Kang Feng Cui 2022Protein & Cell2022,13,5:2
2Optimization of Extraction Process and Determination of Antioxidant Activity of Myrica ruba Leaf Pigment by Orthogonal Design显示文摘[Objectives] To determine the optimum conditions and antioxidant activity of Myrica ruba leaf pigment,and provide reference for further development and use of Myrica ruba leaves. [Methods]Pigment of Myrica ruba leaves was extracted by ethanol extraction method. Absorbance was measured at wavelength of 300 nm. Effects of ethanol volume fraction,solid-to-liquid ratio,temperature,and extraction time were studied. L_9( 3~3) orthogonal experiment was carried out to optimize the extraction process,and DPPH scavenging ability was measured.[Results]The optimum extraction process of Myrica ruba leaf pigment was as follows: the ethanol volume fraction was 50%; the solid-to-liquid ratio was 1: 60( w/v),extraction temperature 70℃; extraction time was 60 min; after the concentration of purified Myrica ruba leaf pigment reached 0. 4455 mg/mL,the DPPH scavenging ability was basically the same as the same concentration Vc.[Conclusions] The experiment indicates that the optimized extraction process has a high yield rate,and Myrica ruba leaf pigment has high anti-oxidant activity.Gongxiao MO Mengyao LIN Jiaming WANG Yuandan LAN Xunjie LIANG Xiang WANG Qiumeng WEI Suoyi HUANG 2017Medicinal Plant2017,8,5:1

5Hepatic steatosis is associated with dysregulated cholesterol metabolism and altered protein acetylation dynamics in chickens显示文摘Background Hepatic steatosis is a prevalent manifestation of fatty liver, that has detrimental effect on the health and productivity of laying hens, resulting in economic losses to the poultry industry. Here, we aimed to systematically investigate the genetic regulatory mechanisms of hepatic steatosis in laying hens.Methods Ninety individuals with the most prominent characteristics were selected from 686 laying hens according to the accumulation of lipid droplets in the liver, and were graded into three groups, including the control, mild hepatic steatosis and severe hepatic steatosis groups. A combination of transcriptome, proteome, acetylome and lipidome analyses, along with bioinformatics analysis were used to screen the key biological processes, modifications and lipids associated with hepatic steatosis.Results The rationality of the hepatic steatosis grouping was verified through liver biochemical assays and RNA-seq. Hepatic steatosis was characterized by increased lipid deposition and multiple metabolic abnormalities. Integration of proteome and acetylome revealed that differentially expressed proteins(DEPs) interacted with differentially acetylated proteins(DAPs) and were involved in maintaining the metabolic balance in the liver. Acetylation alterations mainly occurred in the progression from mild to severe hepatic steatosis, i.e., the enzymes in the fatty acid oxidation and bile acid synthesis pathways were significantly less acetylated in severe hepatic steatosis group than that in mild group(P < 0.05). Lipidomics detected a variety of sphingolipids(SPs) and glycerophospholipids(GPs) were negatively correlated with hepatic steatosis(r ≤-0.5, P < 0.05). Furthermore, the severity of hepatic steatosis was associated with a decrease in cholesterol and bile acid synthesis and an increase in exogenous cholesterol transport.Conclusions In addition to acquiring a global and thorough picture of hepatic steatosis in laying hens, we were able to reveal the role of acetylation in hepatic steatosis and depict the changes in hepatic cholesterol metabolism. 6Multifunctional polyeugenol-based nanoparticles with antioxidant and antibacterial properties显示文摘Over the past few decades,extensive scientific has been dedicated to polymer synthesis employing renewable resources.In this study,we devised and synthesized multifunctional polyeugenol-based nanoparticles,exhibiting remarkable antioxidant and antibacterial properties.The grafting of eugenol onto the nanoparticle surface was achieved via a thiol-ene chemical reaction with a grafting rate of 3.5%.To comprehend the properties of the synthesized nanoparticles,we employed Fourier transform infrared spectroscopy,X-ray photoelectron spectroscopy and pyrolysis-gas chromatography mass spectrometry.Subsequent scanning electron microscopic and transmission electron microscopic analyses revealed the presence of a crosslinked structure within the polyeugenol-based nanoparticles,as well as a heteroge-neous microsphere structure on the surface.Due to the inherent crosslinking structure,the polyeugenol-based nanoparticles demonstrated robust the resistance to solvents,as ascertained through thermog-ravimetric analysis and solvent resistance tests.Notably,nitrogen adsorption/desorption studies confirmed the adsorption capacity of the polyeugenol-based nanoparticles,rendering them potentially suitable for drug transport applications.Moreover,the assays for assessing cytocompatibility and reactive oxygen species scavenging activities demonstrated better performance of the eugenol-based nano-particles compared to eugenol.Furthermore,the polyeugenol-based nanoparticles exhibited certain bactericidal activities against Escherichia coli,Staphylococcus aureus,and Fusobacterium nucleatum.Consequently,these observations indicated the nontoxic nature and expansive application prospects of the polyeugenol-based nanoparticles in the domains of medicine and food preservation.This work presented a pioneering concept for the development of antioxidant and antibacterial multifunctional polymer materials derived from eugenol.Qiaoqiao Lan Xuying Zhang Jiaming Liang Wei Wang Leili Lv Jiamin He Zefeng Wang Chunsheng Qu 2024Particuology2024,87,4:0

10Application of animal models to compare and contrast the virulence of current and future potential SARS-CoV-2 variants显示文摘Since severe acute respiratory syndrome coronavirus 2(SARS‐CoV‐2)was first identified during late 2019,the sustained spread of this pathogen within the human population has caused worldwide disruption with staggering infection rates and death tolls.Due to the accumulation of mutations in SARS‐CoV‐2,the virus has evolved into many variants,five of which have been listed as variants of concern VOCs by the World Health Organization(WHO).Multiple animal models of SARS‐CoV‐2 have been developed to evaluate vaccines and drugs and to assess the pathogenicity,transmissibility and antiviral measures of these VOCs.Here,we review the cutting‐edge research based on mouse,hamster,ferret and non‐human primate models for evaluating SARS‐CoV‐2 with a focus on the Omicron variant,and highlight the importance of updating vaccines in a timely manner in order to mitigate the negative effects of SARS‐CoV‐2 infections in the human population.Zhe Ding Tong Chen Jiaming Lan Gary Wong 2022Biosafety and Health2022,4,3:0
