| 2 | Vitamin D deficiency in cirrhosis relates to liver dysfunction rather than aetiology显示文摘AIM: To examine the vitamin D status in patients with alcoholic cirrhosis compared to those with primary biliary cirrhosis. METHODS: Our retrospective case series comprised 89 patients with alcoholic cirrhosis and 34 patients with primary biliary cirrhosis who visited our outpatient clinic in 2005 and underwent a serum vitamin D status assessment. RESULTS: Among the patients with alcoholic cirrhosis, 85% had serum vitamin D levels below 50 nmol/L and 55% had levels below 25 nmol/L, as compared to 60% and 16% of the patients with primary biliary cirrhosis, respectively (P < 0.001). In both groups, serum vitamin D levels decreased with increasing liver disease severity, as determined by the Child-Pugh score. | 3 | Extrahepatic complications to cirrhosis and portal hypertension: Haemodynamic and homeostatic aspects显示文摘In addition to complications relating to the liver, patients with cirrhosis and portal hypertension develop extrahepatic functional disturbances of multiple organ systems. This can be considered a multiple organ failure that involves the heart, lungs, kidneys, the immune systems, and other organ systems. Progressive fibrosis of the liver and subsequent metabolic impairment leads to a systemic and splanchnic arteriolar vasodilatation. This affects both the haemodynamic and functional homeostasis of many organs and largely determines the course of the disease. With the progression of the disease, the circulation becomes hyperdynamic with cardiac, pulmonary as well as renal consequences for dysfunction and reduced survival. | 4 | Multi-scale phylogenetic structure in coastal dune plant communities across the globe显示文摘Aims Studies integrating phylogenetic history and large-scale community assembly are few,and many questions remain unanswered.Here,we use a global coastal dune plant data set to uncover the important factors in community assembly across scales from the local filtering processes to the global long-term diversification and dispersal dynamics.Coastal dune plant communities occur worldwide under a wide range of climatic and geologic conditions as well as in all biogeographic regions.However,global patterns in the phylogenetic composition of coastal dune plant communities have not previously been studied.Methods The data set comprised vegetation data from 18463 plots in New Zealand,South Africa,South America,North America and Europe.The phylogenetic tree comprised 2241 plant species from 149 families.We calculated phylogenetic clustering(Net Relatedness Index,NRI,and Nearest Taxon Index,NTI)of regional dune floras to estimate the amount of in situ diversification relative to the global dune species pool and evaluated the relative importance of land and climate barriers for these diversification patterns by geographic analyses of phylogenetic similarity.We then tested whether dune plant communities exhibit similar patterns of phylogenetic structure within regions.Finally,we calculated NRI for local communities relative to the regional species pool and tested for an association with functional traits(plant height and seed mass)thought to vary along sea–inland gradients.Important Findings Regional species pools were phylogenetically clustered relative to the global pool,indicating regional diversification.NTI showed stronger clustering than NRI pointing to the importance of especially recent diversifications within regions.The species pools grouped phylogenetically into two clusters on either side of the tropics suggesting greater dispersal rates within hemispheres than between hemispheres.Local NRI plot values confirmed that most communities were also phylogenetically clustered within regions.NRI values decreased with increasing plant height and seed mass,indicating greater phylogenetic clustering in communities with short maximum height and good dispersers prone to wind and tidal disturbance as well as salt spray,consistent with environmental filtering along sea–inland gradients.Height and seed mass both showed significant phylogenetic signal,and NRI tended to correlate negatively with both at the plot level.Low NRI plots tended to represent coastal scrub and forest,whereas high NRI plots tended to represent herb-dominated vegetation.We conclude that regional diversification processes play a role in dune plant community assembly,with convergence in local phylogenetic community structure and local variation in community structure probably reflecting consistent coastal-inland gradients.Our study contributes to a better understanding of the globally distributed dynamic coastal ecosystems and the structuring factors working on dune plant communities across spatial scales and regions. | Ane Kirstine Brunbjerg Jeannine Cavender-Bares Wolf L.Eiserhardt Rasmus Ejrnæs Lonnie W.Aarssen Hannah L.Buckley Estelle Forey Florian Jansen Jens Kattge Cynthia Lane Roy A.Lubke Angela T.Moles Ana Laura Monserrat Robert K.Peet Julissa Roncal Louise Wootton Jens-Christian Svenning | 2014 | Journal of Plant Ecology2014,7,2: | 9 |
| 5 | Impaired contractility and remodeling of the upper gastrointestinal tract in diabetes mellitus type-1显示文摘AIM: To investigate that both the neuronal function of the contractile system and structural apparatus of the gastrointestinal tract are affected in patients with longstanding diabetes and auto mic neuropathy. METHODS: The evoked esophageal and duodenal contractile activity to standardized bag distension was assessed using a specialized ultrasound-based probe. Twelve type-1 diabetic patients with autonomic neuropathy and severe gastrointestinal symptoms and 12 healthy controls were studied. The geometry and biomechanical parameters (strain, tension/stress, and stiffness) were assessed. RESULTS: The diabetic patients had increased frequency of distension-induced contractions (6.0 ± 0.6 vs 3.3 ± 0.5, P < 0.001). This increased reactivity was correlated with the duration of the disease (P = 0.009). Impaired coordination of the contractile activity in diabetic patients was demonstrated as imbalance between the time required to evoke the first contraction at the distension site and proximal to it (1.5 ± 0.6 vs 0.5 ± 0.1, P = 0.03). The esophageal wall and especially the mucosa-submucosa layer had increased thickness in the patients (P < 0.001), and the longitudinal and radial compressive stretch was less in diabetics (P <0.001). The esophageal and duodenal wall stiffness and circumferential deformation induced by the distensions were not affected in the patients (all P > 0.14). CONCLUSION: The impaired contractile activity with an imbalance in the distension-induced contractions likely reflects neuronal abnormalities due to autonomic neuropathy. However, structural changes and remodeling of the gastrointestinal tract are also evident and may add to the neuronal changes. | 8 | Brain changes in diabetes mellitus patients withgastrointestinal symptoms显示文摘Diabetes mellitus is a common disease and its prevalence is increasing worldwide. In various studies up to 30%-70% of patients present dysfunction and complications related to the gut. To date several clinical studies have demonstrated that autonomic nervous system neuropathy and generalized neuropathy of the central nervous system(CNS) may play a major role. This systematic review provides an overview of the neurodegenerative changes that occur as a consequence of diabetes with a focus on the CNS changes and gastrointestinal(GI) dysfunction. Animal models where diabetes was induced experimentally support that the disease induces changes in CNS. Recent investigations with electroencephalography and functional brain imaging in patients with diabetes confirm these structural and functional brain changes. Encephalographic studies demonstrated that altered insular processing of sensory stimuli seems to be a key player in symptom generation. In fact one study indicated that the more GI symptoms the patients experienced, the deeper the insular electrical source was located. The electroencephalography was often used in combination with quantitative sensory testingmainly showing hyposensitivity to stimulation of GI organs. Imaging studies on patients with diabetes and GI symptoms mainly showed microstructural changes,especially in brain areas involved in visceral sensory processing. | 11 | Trait variation in response to resource availability and plant diversity modulates functional dissimilarity among species in experimental grasslands显示文摘Aims Functional trait differences among species are thought to be a pre-requisite for niche differentiation.Plant traits are known to vary plastically in response to environmental conditions and different plant neighbours.However,it is not clear to which extent the direc-tion and magnitude of trait variation differ among species repre-senting different growth forms or varying dominance in different environments and how this trait variation affects community-level trait dissimilarity.Methods In a field experiment,we studied shoot and leaf traits of eight peren-nial grassland species assigned to two functional groups(grasses vs.forbs)and varying in growth stature(small vs.tall)in monocultures,two-and four-species mixtures that were exposed to two levels of light(shaded vs.full light)and nutrient availability(fertilized vs.non-fertilized)to address this gap of knowledge.Important Findings Fertilization increased leaf greenness and specific leaf area(SLA)as well as tissue nitrogen concentrations.Under shading,changes in morphological and physiological light-acquisition traits as well as increased nitrogen and decreased carbon concentrations in plant tissue indicated larger efforts to acquire light and carbon limita-tion of plant growth.Similar changes in light-acquisition traits as observed under shading and decreasing shoot biomass also revealed an accelerated carbon limitation at higher species rich-ness(SR)irrespective of the external manipulation of light and nutrient supply.Overall,the direction of trait variation in response to resource availability(shade,fertilization)and SR did not differ between functional groups or growth statures.The magnitude of variation in several traits at different resource availability,however,was larger in grasses than in forbs as well as in small-statured than in tall-statured species.In general,dominant species in terms of above-ground biomass production had taller shoots with higher carbon and lower nitrogen concentrations and allocated less biomass into leaves than subordinates.The expression of leaf traits of dominants(lower SLA and higher leaf greenness than subordinates)indicated their competitive advantage in light acquisition.Shading,as well as fertilization,accentuated trait differences between dominants and subordinates.Fertilization increased community-level dissimilarity in tissue nitrogen concentrations due to differential responses of functional groups and growth statures.However,resource supply did not alter community-level dissimilarity in other traits.Our study shows that a varying extent of trait variation of species belonging to different functional groups or growth statures and varying in their ability to achieve dominance may modulate community-level trait dissimilarity at different resource availability.These results empha-size the importance to consider the context-dependency of trait-based approaches. | Alrun Siebenkäs Jens Schumacher Christiane Roscher | 2017 | Journal of Plant Ecology2017,10,6: | 2 |

| 16 | Antibiotic-loaded phosphatidylcholine inhibits staphylococcal bone infection显示文摘AIM: To test antibiotic-loaded coating for efficacy in reducing bacterial biofilm and development of osteomyelitis in an orthopaedic model of implant infection. METHODS: Phosphatidylcholine coatings loaded with 25% vancomycin were applied to washed and sterilized titanium wires 20 mm in length. A 10 mm segment was removed from rabbit radius(total = 9; 5 coated, 4 uncoated), and the segment was injected with 1 × 106 colony forming units(CFUs) of Staphylococcus aureus(UAMS-1 strain). Titanium wires were inserted throughthe intramedullary canal of the removed segment and into the proximal radial segment and the segment was placed back into the defect. After 7 d, limbs were removed, X-rayed, swabbed for tissue contamination. Wires were removed and processed to determine attached CFUs. Tissue was swabbed and streaked on agar plates to determine bacteriological score.RESULTS: Antibiotic-loaded coatings resulted in significantly reduced biofilm formation(4.7 fold reduction in CFUs; P < 0.001) on titanium wires and reduced bacteriological score in surrounding tissue(4.0 ± 0 for uncoated, 1.25 ± 0.5 for coated; P = 0.01). Swelling and pus formation was evident in uncoated controls at the 7 d time point both visually and radiographically, but not in antibiotic-loaded coatings.CONCLUSION: Active antibiotic was released from coated implants and significantly reduced signs of osteomyelitic symptoms. Implant coatings were well tolerated in bone. Further studies with additional control groups and longer time periods are warranted.
