| 1 | Inhibition of epidermal growth factor receptor expression by RNA interference in A549 cells显示文摘AIM: To investigate the biological features of A549 cells in which epidermal growth factor (EGF) receptors expression were suppressed by RNA interference (RNAi). METHODS: A549 cells were transfected using short small interfering RNAs (siRNAs) formulated with Lipofectamine 2000. The EGF receptor numbers were determined by Western blotting and flowcytometry. The antiproliferative effects of sequence specific double stranded RNA (dsRNA) were assessed using cell count, colony assay and scratch assay. The chemosensitivity of transfected cells to cisplatin was measured by MTT. RESULTS: Sequence specific dsRNA-EGFR down-regulated EGF receptor expression dramatically. Compared with the control group, dsRNA-EGFR reduced the cell number by 85.0 %, decreased the colonies by 63.3 %, inhibited the migration by 87.2 %, and increased the sensitivity of A549 to cisplatin by four-fold. CONCLUSION: Sequence specific dsRNA-EGFR were capable of suppressing EGF receptor expression, hence significantly inhibiting cellular proliferation and motility, and enhancing chemosensitivity of A549 cells to cisplatin. | 2 | Predicting the spread of nuclear radiation from the damaged Fukushima Nuclear Power Plant显示文摘Japan suffered a M9.0 earthquake and massive tsunami on March 11, 2011, which seriously damaged the Fukushima Nuclear Power Plant and caused a nuclear crisis. The spread of nuclear radiation from the power plant through the atmosphere and ocean was predicted with a short-term climate forecasting model and an ocean circulation model under some idealized assumptions. If nuclear matter were leaked in the near-ground layer of 992 hPa, the climate model results show that the nuclear radiation would cover North America 10 days after the initial leakage, with the concentration at the forefront dramatically reduced to 10 millionths of the initial model concentration at the source. The radiation would span Europe in 15 days and cover much of the Northern Hemisphere in 30 days. If the initial leakage was assumed to occur in the layer 5000-m above the ground, the radiation would cover Europe in 10 days and cover much of the Northern Hemisphere in 15 days. Moreover, under the assumption that the nuclear matter leaked in the 10000-m layer, the radiation would affect much of China after 10 days. The ocean circulation model indicates that the nuclear material would be slowly transported northeast of Fukushima and reach 150°E in 50 days, and the nuclear debris in the ocean would be confined to a narrow band. Compared with the spread in the ocean, the area affected by leaked nuclear radiation in the atmosphere would be very large. | 3 | LjCYC Genes Constitute Floral Dorsoventral Asymmetry in Lotus japonicus显示文摘Previous study shows that LjCYC2,a CYC-like TCP (TB1,CYC and PCFs) gene in the model legume,Lotus japonicus,is involved in dorsal petal development,which together with the other two homologous genes,LjCYC1 and LjCYC3,belongs to an LjCYC gene cluster.In this report,we modified the transformation system in L.japonicus,and constructed different RNAi transgenes to target different LjCYC genes.The expression of three endogenous LjCYC genes was specifically suppressed by different specific RNAi transgenes,and a chimerical RNAi transgene that contains the specific sequences from LjCYC1 and LjCYC2 was found to downregulate the expression of both endogenous genes simultaneously.Effects of silencing three LjCYC genes were mainly restricted on either dorsal or lateral petals,demonstrating their dorsalizing and lateralizing activities during the development of zygomorphic flower.Furthermore,abolishing the expression of three LjCYC genes could give rise to complete loss of dorsoventral (DV) differentiation in the flower whose petals all resembled the ventral one in the wild type and displayed intact organ internal (IN) asymmetry.Our data demonstrate that during zygomorphic flower development,the DV asymmetry is constituted by the LjCYC genes,while the floral organ IN asymmetry is independently determined by other genetic factors. | Jiechen Wang Yumei Wang Da Luo | 2010 | Journal of Integrative Plant Biology2010,52,11: | 8 |
| 4 | Albedo of Coastal Landfast Sea Ice in Prydz Bay,Antarctica:Observations and Parameterization显示文摘The snow/sea-ice albedo was measured over coastal landfast sea ice in Prydz Bay, East Antarctica(off Zhongshan Station)during the austral spring and summer of 2010 and 2011. The variation of the observed albedo was a combination of a gradual seasonal transition from spring to summer and abrupt changes resulting from synoptic events, including snowfall, blowing snow, and overcast skies. The measured albedo ranged from 0.94 over thick fresh snow to 0.36 over melting sea ice. It was found that snow thickness was the most important factor influencing the albedo variation, while synoptic events and overcast skies could increase the albedo by about 0.18 and 0.06, respectively. The in-situ measured albedo and related physical parameters(e.g., snow thickness, ice thickness, surface temperature, and air temperature) were then used to evaluate four different snow/ice albedo parameterizations used in a variety of climate models. The parameterized albedos showed substantial discrepancies compared to the observed albedo, particularly during the summer melt period, even though more complex parameterizations yielded more realistic variations than simple ones. A modified parameterization was developed,which further considered synoptic events, cloud cover, and the local landfast sea-ice surface characteristics. | 5 | Relationship Between Urban Road Traffic Characteristics and Road Grade Based on a Time Series Clustering Model: A Case Study in Nanjing, China显示文摘With the increasing number of vehicles in large-and medium-sized cities challenges in urban traffic management, control, and road planning are being faced. Taxi GPS trajectory data is a novel data source that can be used to study the potential dynamic traffic characteristics of urban roads, and thus identify locations that show a notable lack of road planning. Considering that road traffic characteristics on their own are insufficient for a comprehensive understanding of urban traffic, we develop a road traffic characteristic time series clustering model to analyze the relationship between urban road traffic characteristics and road grade based on existing taxi trajectory data. We select the main urban area of Nanjing as our study area and use the taxi trajectory data of a single month for evaluating our method. The experiments show that the clustering model exhibit good performance and can be successfully used for road traffic characteristic classification. | 6 | Sensitivity of the Arctic sea ice concentration forecasts to different atmospheric forcing: a case study显示文摘A regional Arctic configuration of the Massachusetts Institute of Technology general circulation model(MITgcm) is used as the coupled ice-ocean model for forecasting sea ice conditions in the Arctic Ocean at the National Marine Environmental Forecasting Center of China(NMEFC), and the numerical weather prediction from the National Center for Environmental Prediction Global Forecast System(NCEP GFS) is used as the atmospheric forcing. To improve the sea ice forecasting, a recently developed Polar Weather Research and Forecasting model(Polar WRF) model prediction is also tested as the atmospheric forcing. Their forecasting performances are evaluated with two different satellite-derived sea ice concentration products as initializations:(1) the Special Sensor Microwave Imager/Sounder(SSMIS) and(2) the Advanced Microwave Scanning Radiometer for EOS(AMSR-E). Three synoptic cases, which represent the typical atmospheric circulations over the Arctic Ocean in summer 2010, are selected to carry out the Arctic sea ice numerical forecasting experiments. | 7 | The spatiotemporal patterns of sea ice in the Bohai Sea during the winter seasons of 2000–2016显示文摘In this study,sea ice thickness(SIT)and sea ice extent(SIE)in the Bohai Sea from 2000 to 2016 were investigated.A surface heat balance equation was applied to calculate SIT using ice surface temperatures estimated from the Moderate Resolution Imaging Spectroradiometer(MODIS)data with input from air temperature and wind speed from reanalyzing weather data.No trend was found in SIT during 2000–2016.The mean SIT and SIE during this period were 5.58±0.86 cm and 23×10^(3)±8×10^(3)km^(2),respectively.The largest SIT and SIE periods were observed during the second half of January and the first half of February,respectively.The Spearman correlation coefficient between mean ice thickness and average air temperature from 21 automatic weather stations around the Bohai Sea was–0.94(P<.005),and the coefficient between median ice extent and negative accumulated temperature was–0.503(P<.001).The rate of increase in air temperature around the Bohai Sea is 0.271℃per decade in winter for 1979–2016(P<.05),which is much lower than that in northern polar area(0.648℃per decade).This rate has not resulted in a decreasing trend in SIT and SIE for the past 16 years in the Bohai Sea. | Lunxi Ouyang Fengming Hui Lixian Zhu Xiao Cheng Bin Cheng Mohammed Shokr Jiechen Zhao Minghu Ding Tao Zeng | 2019 | International Journal of Digital Earth2019,12,8: | 3 |

| 9 | Modulation of human enteric epithelial barrier and ion transport function by Peyer's patch lymphocytes显示文摘AIM: To investigate the role of Peyer's patch lymphocytes in the regulation of enteric epithelial barrier and ion transport function in homeostasis and host defense. METHODS: Mouse Peyer's patch lymphocytes were co-cultured with human intestinal epithelial cell line Caco-2 either in the mixed or separated (isolated but permeable compartments) culture configuration. Barrier and transport functions of the Caco-2 epithelial monolayers were measured with short-circuit current (Isc) technique. Release of cytokines was measured by enzyme-linked immunosorbent assay (ELISA) and cytokine mRNA expression was analyzed by semi-quantitative RT-PCR. Barrier and iontransport functions of both culture conditions following exposure to Shigella lipopolysaccharide (LPS) were also examined. RESULTS: The transepithelial resistance (TER) of the epithelial monolayers co-cultured with Peyer's patch lymphocytes was maintained whereas that of the Caco-2 monolayers alone significantly decreased after eight days in culture. The forskolin-induced anion secretion, in either absence or presence of LPS, was significantly suppressed in the both co-cultures as compared with the Caco-2 cells alone. Furthermore, only the mixed co-culture condition induced the expression and release of mIL-6 from Peyer's patch lymphocytes, which could be further enhanced by LPS. However, both co-culture conditions suppressed expression and release of epithelial hIL-8 under the unstimulated conditions, while the treatment with LPS stimulated their hIL-8 expression and release. | 10 | Spatial and temporal evolution of landfast ice near Zhongshan Station, East Antarctica, over an annual cycle in 2011/2012显示文摘Annual observations of first-year ice(FYI) and second-year ice(SYI) near Zhongshan Station, East Antarctica,were conducted for the first time from December 2011 to December 2012. Melt ponds appeared from early December 2011. Landfast ice partly broke in late January, 2012 after a strong cyclone. Open water was refrozen to form new ice cover in mid-February, and then FYI and SYI co-existed in March with a growth rate of 0.8 cm/d for FYI and a melting rate of 2.7 cm/d for SYI. This difference was due to the oceanic heat flux and the thickness of ice,with weaker heat flux through thicker ice. From May onward, FYI and SYI showed a similar growth by 0.5 cm/d.Their maximum thickness reached 160.5 cm and 167.0 cm, respectively, in late October. Drillings showed variations of FYI thickness to be generally less than 1.0 cm, but variations were up to 33.0 cm for SYI in March,suggesting that the SYI bottom was particularly uneven. Snow distribution was strongly affected by wind and surface roughness, leading to large thickness differences in the different sites. Snow and ice thickness in Nella Fjord had a similar 'east thicker, west thinner' spatial distribution. Easterly prevailing wind and local topography led to this snow pattern. Superimposed ice induced by snow cover melting in summer thickened multi-year ice,causing it to be thicker than the snow-free SYI. The estimated monthly oceanic heat flux was ~30.0 W/m2 in March–May, reducing to ~10.0 W/m2 during July–October, and increasing to ~15.0 W/m2 in November. The seasonal change and mean value of 15.6 W/m2 was similar to the findings of previous research. | 11 | Multi-step ahead short-term predictions of storm surge level using CNN and LSTM network显示文摘Storm surges pose significant danger and havoc to the coastal residents’safety,property,and lives,particularly at offshore locations with shallow water levels.Predictions of storm surges with hours of warning time are important for evacuation measures in low-lying regions and coastal management plans.In addition to experienced predictions and numerical models,artificial intelligence(AI)techniques are also being used widely for short-term storm surge prediction owing to their merits in good level of prediction accuracy and rapid computations.Convolutional neural network(CNN)and long short-term memory(LSTM)are two of the most important models among AI techniques.However,they have been scarcely utilised for surge level(SL)forecasting,and combinations of the two models are even rarer.This study applied CNN and LSTM both individually and in combination towards multi-step ahead short-term storm surge level prediction using observed SL and wind information.The architectures of the CNN,LSTM,and two sequential techniques of combining the models(LSTM–CNN and CNN–LSTM)were constructed via a trial-and-error approach and knowledge obtained from previous studies.As a case study,11 a of hourly observed SL and wind data of the Xiuying Station,Hainan Province,China,were organised as inputs for training to verify the feasibility and superiority of the proposed models.The results show that CNN and LSTM had evident advantages over support vector regression(SVR)and multilayer perceptron(MLP),and the combined models outperformed the individual models(CNN and LSTM),mostly by 4%–6%.However,on comparing the model computed predictions during two severe typhoons that resulted in extreme storm surges,the accuracy was found to improve by over 10%at all forecasting steps. | Bao Wang Shichao Liu Bin Wang Wenzhou Wu Jiechen Wang Dingtao Shen | 2021 | Acta Oceanologica Sinica2021,40,11: | 2 |
| 12 | Snow and ice thicknesses derived from Fast Ice Prediction System Version 2.0(FIPS V2.0)in Prydz Bay,East Antarctica:comparison with in-situ observations显示文摘In this paper,snow and ice thickness products derived from an updated Fast Ice Prediction System Version 2.0(FIPS V2.0)in Prydz Bay,East Antarctica,are introduced and compared with in-situ obser-vations.FIPS V2.0 is comprised of a newly-developed snowdrift parameterization compared to the original FIPS V1.0.The simulation domain covers the entire fast ice region in Prydz Bay and is config-ured to 720 grid cells,with a spatial resolution of 0.125°.The ERAInterim reanalysis from the European Centre for Medium-Range Weather Forecasting(ECMWF)were used as the atmospheric forcing.The in-situ observations were obtained near Zhongshan Station by the wintering team,and the measurement frequency of the snow and ice thicknesses was around one week.Both the FIPS V2.0 pro-ducts and in-situ observations introduced in this paper cover the time periods from 2012 to 2016.The primary assessments based on the in-situ observations show that FIPS V2.0 has mean biases of 0.01±0.07 m and 0.23±0.09 m for snow and ice thickness simula-tions,respectively.The results indicate that the updated FIPS V2.0 produces a reasonable snow thickness due to the newly-developed snowdrift parameterization,but it overestimates the ice thickness due to the cold bias in the air temperature forcing.These 2-D snow and ice thickness distributions provide important references for sea ice thermodynamic studies,remote sensing validations,and ice-breaker navigation assessments in this region.The dataset is avail-able at http://gffzza4c772bdab62464fhfffwo95q66uw6cnc.ffgz.tsg.suse.edu.cn/10.11922/sciencedb.j00076.00066. | Jiechen Zhao Jingjing Cheng Zhongxiang Tian Xiaopeng Han Hui Shen Guanghua Hao Honglin Guo Qi Shu | 2022 | Big Earth Data2022,6,4: | 2 |

| 14 | Classification of facial wrinkles among Chinese women显示文摘It is generally recognized that Caucasians and Asians have different skin aging features.The aim of this study was to develop a facial wrinkle grading scale for Chinese women.Standard photographs were taken of 242 Chinese women.Six sets of 0 to 9 wrinkle scales with reference photographs and descriptions were selected,including grading scales for resting and hyperkinetic crow's feet,frontalis lines,glabellar frown lines,and nasolabial folds.To identify the scale by objective quantitative measurement,skin surface measurements from the Visioscan~ VC98 were used.To test the reliability and validity of our wrinkle scale,a multi-rater consensus method was used.A double-blind,randomized,vehicle-controlled 12-week study was conducted to use this clinical photo-score to evaluate the efficacy and safety of Centella triterpenes cream~ in treating crow's feet.A newly developed 10-point photographic and descriptive scale emerged from this study.The final atlas of these photographs contained a total of 6 sets with 10 pictures each.From 0 to 9,surface evaluation of smoothness(SEsm) parametric measurements decreased progressively,indicating that the scale increased inversely.Weighted kappa coefficients for intra-assessor were between 0.75-0.87.The overall Kendall's coefficient is 0.86 on the first rating and 0.87 on the second rating.Thirtysix volunteers were recruited and 35 subjects completed a 12-week trial.Clinical photo-score by investigator showed a significant difference(P < 0.05) between the treatment side and control side after 4 weeks.Use of these scales in clinical settings to evaluate facial wrinkles in Asians individuals is recommended. | Jiechen Zhang Wei Hou Suying Feng Xiangsheng Chen Hongwei Wang | 2017 | The Journal of Biomedical Research2017,31,2: | 2 |
