2G-Quadruplex(G4) Motifs in the Maize(Zea mays L.) Genome Are Enriched at Specific Locations in Thousands of Genes Coupled to Energy Status,Hypoxia,Low Sugar,and Nutrient Deprivation显示文摘The G-quadruplex(G4) elements comprise a class of nucleic acid structures formed by stacking of guanine base quartets in a quadruple helix.This G4 DNA can form within or across single-stranded DNA molecules and is mutually exclusive with duplex B-form DNA.The reversibility and structural diversity of G4 s make them highly versatile genetic structures,as demonstrated by their roles in various functions including telomere metabolism,genome maintenance,immunoglobulin gene diversification,transcription,and translation.Sequence motifs capable of forming G4 DNA are typically located in telomere repeat DNA and other non-telomeric genomic loci.To investigate their potential roles in a large-genome model plant species,we computationally identified 149,988 non-telomeric G4 motifs in maize(Zea mays L.,B73 AGPv2),29%of which were in non-repetitive genomic regions.G4 motif hotspots exhibited non-random enrichment in genes at two locations on the antisense strand,one in the 5'UTR and the other at the 5'end of the first intron.Several genic G4 motifs were shown to adopt sequence-specific and potassium-dependent G4 DNA structures in vitro.The G4 motifs were prevalent in key regulatory genes associated with hypoxia(group VII ERFs),oxidative stress(DJ-1/GATase1),and energy status(AMPK/SnRK) pathways.They also showed statistical enrichment for genes in metabolic pathways that function in glycolysis,sugar degradation,inositol metabolism,and base excision repair.Collectively,the maize G4 motifs may represent conditional regulatory elements that can aid in energy status gene responses.Such a network of elements could provide a mechanistic basis for linking energy status signals to gene regulation in maize,a model genetic system and major world crop species for feed,food,and fuel.Carson M.Andorf Mykhailo Kopylov Drena Dobbs Karen E.Koch M.Elizabeth Stroupe Carolyn J.Lawrence Hank W.Bass 2014Journal of Genetics and Genomics2014,41,12:2

12S2B-2 Comorbidity of Anxiety and Depression Induced by Liang’s Contextual-Stress Box in Mice显示文摘Rationale:Comorbidity of depression and anxiety is one of the most common mental disorders in practice.Few experimental procedures,however,have focused on identifying mixed depression-and anxiety-like behaviors in animal models.Objectives:To develop a novel testing paradigm,the so-called Liang’s contextual-stress Box(briefly,Liang’s Box),for evaluating depression-and/or anxiety-like behaviors in mice.Methods:Liang’s Box consisted of a central area(CA)with three peripheral arms(PA).Mice were placed individually in the end of a peripheral arm facing to the central area and were allowed to travel around Liang’s Box.During a test period,behavioral parameters were recorded and collected by a video-tracking system named SMART 3.0(Panlab,Spain),including①Latency to CA,②Time spent in CA(CA-time),③Distance traveled in CA(CA-distance),④Distance traveled in PA(PA-distance),⑤Transition number between CA and PA,⑥Global activity,⑦CA-time/cm2,⑧PA-time/cm2,⑨CAdistance/cm2,and⑩PA-distance/cm2.In order to assess the reliability and validity of Liang’s Box and get baseline information,we undertook a series of experiment,including verifying consistency and stability in the various strains of mice(KM,ICR,and C57 BL/6 J mice),imposing high illumination,restraint and tail suspension stress,pre-treating with an anxiolytic(diazepam,buspirone)or antidepressant(desipramine,mianserin).Results:①There were not remarkable differences in all the parameters among KM,ICR,and C57 BL/6 J mice;②High illumination,restraint and tail suspension significantly increased the latency to CA and decreased the CA-time,CA-distance and transition number.However,these three types of stresses evoked different effect on global activity.High illumination increased activity,but restraint stress had no significant influence.In contrast with high illumination,tail suspension stress markedly reduced activity in mice;③The anxiolytics diazepam(0.25 mg·kg-1 and 0.5 mg·kg-1,i.p.)and buspirone(0.5 mg·kg-1 and 1.0 mg·kg-1,i.p.)significantly decreased latency to CA and transition number,and dosedependently increased the CA-time.However,they did not affect CA-or PA-distance,or global activity;④The antidepressants desipramine(10.0 mg·kg-1 and 30.0 mg·kg-1,i.p.)and mianserin(0.25 mg·kg-1 and 0.5 mg·kg-1,i.p.)significantly decreased latency to CA,and dose-dependently enhanced CA-time,CA-distance and transition number,while neither affected PA-distance or global activity.Conclusion:Our data suggest that Liang’s Box is a useful experimental instrument and method for screening and assessing potential antidepressants and/or anxiolytics.Moreover,depression-and anxiety-like behaviors induced by Liang’s Box in mice may be a new hybrid animal model to investigate the pathogenic mechanisms underlying the comorbidity of anxiety and depression.LIANG Jian-hui LIANG hui CHENG tao LIU Xiao-yan Simon M.Y.Lee TANG Ben-qin WANG Xiufen CHEN Feng Andrew J.Lawrence 2018神经药理学报2018,8,4:0
13Influence of supraphysiological cortisol manipulation on predator avoidance behaviors and physiological responses to a predation threat in a wild marine teleost fish显示文摘The stress axis in teleost fish attempts to maintain internal homeostasis in the face of allostatic loading.However,stress axis induction has been associated with a higher predation rate in fish.To date,the physiological and behavioral factors associated with this outcome are poorly understood.The purpose of the present study was to investigate the impact of experimental cortisol elevation on anti-predator behavior and physiological responses to predator presence.We hypothesized that semi-chronic cortisol elevation would increase susceptibility to predation by increasing stress-induced risk-taking behaviors.To test this hypothesis,schoolmaster snapper were given cocoa butter implants without cortisol(sham)or with cortisol(50 mg/kg body weight)and tethered to cover.Fish were exposed to either a lemon shark or control conditions for 15-min.Space use and activity were recorded throughout and fish were terminally sampled for blood.Cortisol implantation,relative to shams,resulted in higher blood glucose and plasma cortisol concentrations with a lower plasma lactate concentration.Shark exposure,relative to controls,elicited higher blood glucose and lactate concentrations but had no effect on plasma cortisol concentration.No interactions were detected between shark exposure and cortisol treatment for any physiological trait.Behavioral metrics,including shelter use and activity,were unaffected by either cortisol implantation or shark exposure.Physiological responses to cortisol implantation likely resulted from enhanced gluconeogenic activity,whereas alterations under predator exposure may have been the product of catecholamine mobilization.Further work should address context-specific influences of stress in mediating behavioral responses to predation.Michael J.LAWRENCE Erika J.ELIASON Jacob W.BROWNSCOMBE Kathleen M.GILMOUR John W.MANDELMAN Lee F.G.GUTOWSKY Steven J.COOKE 2018Integrative Zoology2018,13,2:0
