2Manipulating mRNA splicing by base editing in plants显示文摘Precursor-mRNAs(pre-mRNA) can be processed into one or more mature m RNA isoforms through constitutive or alternative splicing pathways. Constitutive splicing of pre-mRNA plays critical roles in gene expressional regulation, such as intronmediated enhancement(IME), whereas alternative splicing(AS) dramatically increases the protein diversity and gene functional regulation. However, the unavailability of mutants for individual spliced isoforms in plants has been a major limitation in studying the function of mRNA splicing. Here, we describe an efficient tool for manipulating the splicing of plant genes. Using a Cas9-directed base editor, we converted the 5′ splice sites in four Arabidopsis genes from the activated GT form to the inactive AT form. Silencing the AS of HAB 1.1(encoding a type 2 C phosphatase) validated its function in abscisic acid signaling, while perturbing the AS of RS31 A revealed its functional involvement in plant response to genotoxic treatment for the first time. Lastly,altering the constitutive splicing of Act2 via base editing facilitated the analysis of IME. 3Application of magnetic particles modified with amino groups to adsorb copper ions in aqueous solution显示文摘A magnetic adsorbent can be easily recovered from treated water by magnetic force,without requiring further downstream treatment.In this research,amine-functionalized silica magnetite has been synthesized using N-[3-(trimethoxysilyl)propyl]-ethylenediamine(TPED) as a surface modification agent.The synthesized magnetic amine adsorbents were used to adsorb copper ions in an aqueous solution in a batch system,and the maximum adsorption was found to occur at pH 5.5 ± 0.1.The adsorption equilibrium data fitted the Langmuir isotherm equation reasonably well,with a maximum adsorption capacity of 10.41 mg/g.A pseudo second-order model could best describe the adsorption kinetics,and the derived activation energy was 26.92 kJ/mol.The optimum condition to desorb Cu2+ from NH2/SiO2/Fe3O4 was provided by a solution with 0.1 mol/L HNO3.Yafen Lin Huawei Chen Kaelong Lin Boryann Chen Chyowsan Chiou 2011Journal of Environmental Sciences2011,23,1:4
4Identification of the varietal origin of processed loose-leaf tea based on analysis of a single leaf by SNP nanofluidic array显示文摘Tea is an important cash crop, representing a $40 billion-a-year global market. Differentiation of the tea market has resulted in increasing demand for tea products that are sustainably and responsibly produced. Tea authentication is important because of growing concerns about fraud involving premium tea products. Analytical technologies are needed for protection and value enhancement of high-quality brands. For loose-leaf teas, the challenge is that the authentication needs to be established on the basis of a single leaf, so that the products can be traced back to the original varieties. A new generation of molecular markers offers an ideal solution for authentication of processed agricultural products. Using a nanofluidic array to identify variant SNP sequences, we tested genetic identities using DNA extracted from single leaves of 14 processed commercial tea products. Based on the profiles of 60 SNP markers, the genetic identity of each tea sample was unambiguously identified by multilocus matching and ordination analysis. Results for repeated samples of multiple tea leaves from the same products(using three independent DNA extractions) showed 100% concordance, showing that the nanofluidic system is a reliable platform for generating tea DNA fingerprints with high accuracy. The method worked well on green, oolong, and black teas, and can handle a large number of samples in a short period of time. 5Protective effects and potential underlying mechanisms of sodium copper chlorophyllin against ethanol-induced gastric ulcer in mice显示文摘In study,we aimed to determine the mechanisms underlying the gastroprotective effects of sodium copper chlorophyllin(SCC)against ethanol-induced gastric ulcer injury in mice.First,the gastroprotective effects of SCC against gastric ulcer induced by ethanol were assessed.Then,biochemical,histopathological,immunohistochemistry assays,and western blot analysis were conducted to determine the possible mechanisms of action underlying the effects of SCC.Compared to the effects of omeprazole(OME)in a confirmed mouse model of ethanol-induced gastric ulcer injury,treatment with various doses of SCC resulted in up-regulation of Bcl-2 and down-regulation of the pro-apoptotic protein Bax.Significant decreases in the levels of the malondialdehyde(MDA),myeloperoxidase(MPO),and NO in the gastric tissues were observed.Furthermore,inflammatory cytokine analysis revealed that SCC treatment inhibited the expressions of TNF-αand IL-6,greatly reduced the phosphorylation level of IκB,and repressed the nuclear translocation of NF-κB p65,which demonstrated that SCC inhibited the activation of the NF-κB pathway.The present findings suggest that the protective effects of SCC may be beneficial as a potential preventive and therapeutic agent for gastric ulcer through the NF-κB pathway.Taken together,SCC administration significantly decreased the levels of MPO,NO,and MDA in gastric tissue and exerted a powerful anti-inflammatory activity as demonstrated by reduction in the secretions of proinflammatory mediators such as IL-6 and TNF-α in the serum of mice exposed to ethanol.Huawei Lv Yan Lin Peigang Liu Weiqing Liang Kemin Wei Jinbao Pu Hongjian Zhang 2019Acta Biochimica et Biophysica Sinica2019,51,9:2
6Changes in Protein Content and Chitinase and β-1,3- glucanase Activities of Rice with Blast Resistance Induced by Ag-antibiotic 702显示文摘[Objective] The paper was to explore the effect of rice blast resistance induced by Ag-antibiotics 702 on protein content and chitinase and β-1,3-glucanase activities in rice. [Method]At the fourth leaf stage of Luliangyou 996,15 μg/m L Ag-antibiotic 702 was sprayed,while Validamycin and distilled water were sprayed as positive control and negative control,respectively. All treatments were inoculated with spore fluid of Magnaporthe grisea at 48 h post spraying,and the rice inoculated with only distilled water was used as blank control. The enzymes activities( endochitinase,exochitinase and β-1,3-glucanase) and total protein content in rice leaves were determined every 24 h within 168 h post spraying. [Result] Compared with the blank control,the rice inoculated with spore fluid of M. grisea could significantly increase the total protein content and the activities of β-1,3-glucanase and chitinase. The induction effect of Ag-antibiotics 702 exceeded that of Validamycin treatment. And the changes in activities of β-1,3-glucanase and chitinase had obvious synchronicity. 7The Impact of Green Finance on the Ecologicalization of Urban Industrial Structure-Based on GMM of Dynamic Panel System显示文摘Although a number of studies have been published in the general area on various factors affecting the ecologicalization of urban industrial structure,little work has been carried out for empirical studies quantitatively analyzing the relevance between green finance development and the ecologicalization of urban industrial structure.Therefore,based on a comprehensive index of green finance development,this research employs panel data of target cities1 for the period 2012–2020 to explore the influence of green finance on the ecologicalization of urban industrial structure.The empirical results show that green finance development significantly improves the ecologicalization level of urban industrial structure.In addition,it is found that green finance plays a stronger role in promoting the ecologicalization of industrial structure in economically developed regions than in economically underdeveloped regions2.The research results can provide a valuable policy reference for urban green financial market planning and green product innovation.Kai Lin Huawei Zhao 2022Journal of Artificial Intelligence and Technology2022,2,3:1
8Neurogranin as an important regulator in swimming training to improve the spatial memory dysfunction of mice with chronic cerebral hypoperfusion显示文摘Background:Vascular cognitive impairment caused by chronic cerebral hypoperfusion(CCH)has become a hot issue worldwide.Aerobic exercise positively contributes to the preservation or restoration of cognitive abilities;however,the specific mechanism has remained inconclusive.And recent studies found that neurogranin(Ng)is a potential biomarker for cognitive impairment.This study aims to investigate the underlying role of Ng in swimming training to improve cognitive impairment.Methods:To test this hypothesis,the clustered regularly interspaced short palindromic repeats(CRISPR)-associated protein 9(Cas9)system was utilized to construct a strain of Ng conditional knockout(Ng cKO)mice,and bilateral common carotid artery stenosis(BCAS)surgery was performed to prepare the model.In Experiment 1,2-month-old male and female transgenic mice were divided into a control group(wild-type littermate,n=9)and a Ng cKO group(n=9).Then,2-month-old male and female C57BL/6 mice were divided into a sham group(C57BL/6,n=12)and a BCAS group(n=12).In Experiment 2,2-month-old male and female mice were divided into a sham group(wild-type littermate,n=12),BCAS group(n=12),swim group(n=12),BCAS+Ng cKO group(n=12),and swim+Ng cKO group(n=12).Then,7 days after BCAS,mice were given swimming training for 5 weeks(1 week for adaptation and 4 weeks for training,5 days a week,60 min a day).After intervention,laser speckle was used to detect cerebral blood perfusion in the mice,and the T maze and Morris water maze were adopted to test their spatial memory.Furthermore,electrophysiology and Western blotting were conducted to record long-term potential and observe the expressions of Ca^(2+)pathway-related proteins,respectively.Immunohistochemistry was applied to analyze the expression of relevant markers in neuronal damage,inflammation,and white matter injury.Results:The figures showed that spatial memory impairment was detected in Ng cKO mice,and a sharp decline of cerebral blood flow and an impairment of progressive spatial memory were observed in BCAS mice.Regular swimming training improved the spatial memory impairment of BCAS mice.This was achieved by preventing long-term potential damage and reversing the decline of Ca^(2+)signal transduction pathway-related proteins.At the same time,the results suggested that swimming also led to improvements in neuronal death,inflammation,and white matter injury induced by CCH.Further study adopted the use of Ng cKO transgenic mice,and the results indicated that the positive effects of swimming training on cognitive impairments,synaptic plasticity,and related pathological changes caused by CCH could be abolished by the knockout of Ng.Conclusion:Swimming training can mediate the expression of Ng to enhance hippocampal synaptic plasticity and improve related pathological changes induced by CCH,thereby ameliorating the spatial memory impairment of vascular cognitive impairment.Huawei Lin Jiayong Zhang Yaling Dai Huanhuan Liu Xiaojun He Lewen Chen Jing Tao Chaohui Li Weilin Liu 2023Journal of Sport and Health Science2023,12,1:1

11Classification of aviation incident causes using LGBM with improved cross-validation显示文摘Aviation accidents are currently one of the leading causes of significant injuries and deaths worldwide. This entices researchers to investigate aircraft safety using data analysis approaches based on an advanced machine learning algorithm.To assess aviation safety and identify the causes of incidents, a classification model with light gradient boosting machine (LGBM)based on the aviation safety reporting system (ASRS) has been developed. It is improved by k-fold cross-validation with hybrid sampling model (HSCV), which may boost classification performance and maintain data balance. The results show that employing the LGBM-HSCV model can significantly improve accuracy while alleviating data imbalance. Vertical comparison with other cross-validation (CV) methods and lateral comparison with different fold times comprise the comparative approach. Aside from the comparison, two further CV approaches based on the improved method in this study are discussed:one with a different sampling and folding order, and the other with more CV. According to the assessment indices with different methods, the LGBMHSCV model proposed here is effective at detecting incident causes. 15Effect of Mn Doping ZnO on Morphology and Magnetism Synthesized by Hydrothermal Method显示文摘The diluted magnetic semiconductor Zn1-xMnxO crystals were synthesized at 430 ℃ for 24 h by hydrothermal method. The mineralizer is 3 mol·L-1 KOH, and fill factor is 35%. The Mn concentration in crystal was determined by electron probe. The atomic percentage of Mn in crystal increases as the concentration of MnCl2·6H2O in precursor increases. The maximum of the Mn concentration in crystal is above 8%. The morphology of the crystal varies with the concentration of MnCl2·6H2O in precursor increases. And they all show the hexagonal feature of ZnO wurtzite. The measurement of magnetism was carried out by using a superconducting quantum interference device (SQUID).
