| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 2 | Mathematic principles of interrupted-sampling repeater jamming (ISRJ)显示文摘Coherent jamming is one of the important trends in modern radar electronic war- fare. High-speed sampling of wideband radio frequency (RF) signals and high iso- lation of two receive-transmit antennas are key technologies for the realization of coherent jamming. However, these technologies present significant challenges to engineering application. In this paper, a novel repeater jamming based on inter- rupted sampling technique is presented. For a jammer with a receive-transmit time-sharing antenna, a radar signal is sampled with a low rate by the jammer. Then, a train of false targets will be achieved after the jamming signal feed the matched filter of a pulse compression radar. | 5 | Single-cell RNA-seq highlights intra-tumoral heterogeneity and malignant progression in pancreatic ductal adenocarcinoma显示文摘Pancreatic ductal adenocarcinoma (PDAC) is the most common type of pancreatic cancer featured with high intra-tumoral heterogeneity and poor prognosis. To comprehensively delineate the PDAC intra-tumoral heterogeneity and the underlying mechanism for PDAC progression, we employed single-cell RNA-seq (scRNA-seq) to acquire the transcriptomic atlas of 57,530 individual pancreatic cells from primary PDAC tumors and control pancreases, and identified diverse malignant and stromal cell types, including two ductal subtypes with abnormal and malignant gene expression profiles respectively, in PDAC. We found that the heterogenous malignant subtype was composed of several subpopulations with differential proliferative and migratory potentials. Cell trajectory analysis revealed that components of multiple tumor-related pathways and transcription factors (TFs) were differentially expressed along PDAC progression. Furthermore, we found a subset of ductal cells with unique proliferative features were associated with an inactivation state in tumor-infiltrating T cells, providing novel markers for the prediction of antitumor immune response. | 6 | Formation and destruction processes of upper Sinian oil-gas pools in the Dingshan-Lintanchang structural belt, southeast Sichuan Basin, China显示文摘The lower Cambrian Niutitang Formation hydrocarbon source rocks at the Dingshan-Lintanchang structure in the southeast Sichuan Basin were of medium-good quality with two excellent hydrocarbon-generating centers developed in the periphery areas, with a possibility of forming a medium to large-sized oil-gas field. Good reservoir rocks were the upper Sinian (Dengying Formation) dolomites. The mudstone in the lower Cambrian Niutitang Formation with a good sealing capacity was the cap rock. The widely occurring bitumen in the Dengying Formation indicates that a paleo oil pool was once formed in the study area. The first stage of paleo oil pool formation was maturation of the lower Cambrian source rocks during the late Ordovician. Hydrocarbon generation from the lower Cambrian source rocks stopped due to the Devonian-Carboniferous uplifting. The lower Cambrian source rocks then restarted generation of large quantities of hydrocarbons after deposition of the middle Permian sediments. This was the second stage of the paleo oil pool formation. The oil in the paleo oil pool began to crack during the late Triassic and a paleo gas pool was formed. This paleo gas pool was destroyed during the Yanshan-Himalayan folding, uplifting and denudation. Bitumen can be widely seen in the Dengying Formation in wells and outcrops in the Sichuan Basin and its periphery areas. | 8 | Epidemiological and risk analysis of the H7N9 subtype influenza outbreak in China at its early stage显示文摘Dozens of human cases infected with H7N9 subtype avian influenza virus (AIV) have been confirmed in China since March, 2013. Distribution data of sexes, ages, professions and regions of the cases were analyzed in this report. The results showed that the elderly cases, especially the male elderly, were significantly more than expected, which is different from human cases of H5N1 avian influenza and human cases of the pandemic H1N1 influenza. The outbreak was rated as a Grade Ⅲ (severe) outbreak, and it would evolve into a Grade IV (very severe) outbreak soon, using a method reported previously. The H7N9 AIV will probably circulate in humans, birds and pigs for years. Moreover, with the driving force of natural selection, the virus will probably evolve into highly pathogenic AIV in birds, and into a deadly pandemic influenza virus in humans. Therefore, the H7N9 outbreak has been assumed severe, and it is likely to become very or extremely severe in the future, highlighting the emergent need of forceful scientific measures to eliminate any infected animal flocks. | 11 | Efficient generation of transgenic chickens using the spermatogonial stem cells in vivo and ex vivo transfection显示文摘The highly efficient novel methods to produce transgenic chickens were established by directly in-jecting the recombinant plasmid containing green fluorescent protein (GFP) gene into the cock's testis termed as testis-medianted gene transfer (TMGT), and transplanting transfected spermatogonial stem cells (TTSSCs). For the TMGT approach,four dosages of pEGFP-N1 DNA/cationic polymer complex were injected intratesticularly. The results showed: (1) 48 h after the injection,the percentages of testis cells expressing GFP were 4.0%, 8.7%, 10.2% and 13.6% in the 50, 100, 150 and 200 μg/mL group, re-spectively. The difference from the four dosage groups was significant (P<0.05). On day 25 after the injection, a dosage-dependent and time-dependent increase in the number of transgenic sperm was observed. The percentages of gene expression reached the summit and became stable from day 70 to 160, being 12.7%, 12.8%, 15.9% and 19.1%, respectively. The difference from the four dosage groups was also significant (P<0.05). (2) 70 d after the injection, strong green fluorescent could be observed in the seminiferous tubules by whole-mount in-situ hybridization. (3) 70 d after the injection, the semen was collected and used to artificially inseminate wild-type females. The blastoderms of F1 and F2 transgenic chicken expressed GFP were 56.2% (254/452) and 53.2% (275/517), respectively. The detec-tion of polymerase chain reaction (PCR) of F1 and F2 transgenic chicken blood genomic DNA showed that 56.5% (3/23) of F1 and 52.9% (9/17) of F2 were positive. Southern blot showed GFP DNA was in-serted in their genomic DNAs. (4) Frozen whole mount tissue sections of F1 and F2 transgenic chicken liver, heart, kidney and muscle showed that the rates of green fluorescent positive were between 50.0% and 66.7%. (5) With the TTSSCs method, SSCs ex vivo transfected with GFP were transplanted into recipient roosters whose endogenic SSCs had been resoluted. The donor SSCs settled and GFP ex-pression became readily detectable in the frozen whole mount tissue sections of recepient testes. Moreover, sperms carrying GFP could be produced normally. The results of artificially inseminating wild-type females with these sperms showed 12.5% (8/64) of offspring embryo expressed GFP and 11.1% (2/18) hatched chicks were tested transgenic. In compliance with data standards and structures of the International Nucleotide Sequence Database Collaboration(INSDC), GSA adopts four data objects(Bio Project, Bio Sample,Experiment, and Run) for data organization, accepts raw sequence reads produced by a variety of sequencing platforms, stores both sequence reads and metadata submitted from all over the world,and makes all these data publicly available to worldwide scientific communities. | 13 | Hydrothermal Dolomite in the Upper Sinian (Upper Proterozoic) Dengying Formation, East Sichuan Basin, China显示文摘Hydrothermal Dolomite(HTD) is present in the Upper Sinian(Upper Proterozoic) Dengying Formation, east Sichuan Basin, China. The strata are comprised by primary dolomite. The HTD has various textures, including zebra dolomite, subhorizontal sheet-like cavities filled by saddle dolomite and breccias cemented by saddle dolomites as well occur as a fill of veins and fractures. Also co-occur MVT type lead-zinc ores in the study area. The δ13C and δ18O isotopes of HTD in the Upper Sinian Dengying Formation are lighter than those of the host rocks, while 87Sr/86 Sr is higher. The apparent difference in carbon, oxygen and strontium isotopes, especially the large difference in 87Sr/86 Sr isotopes ratio indicate crystallization from hot basinal and/or hydrothermal fluids. Saddle dolomite was precipitated at temperatures of 270-320°C. The diagenetic parasequences of mineral assemblage deposited in the Dengying Formation are:(1) dolomite host rock →sphalerite-galena-barite-fluorite;(2) dolomite host rock →saddle dolomite →quartz;(3) dolomite host rock → saddle dolomite→bitumen;(4) dolomite host rock → saddle dolomite → barite. The mean chemical composition of the host dolomite matrix and HTD didn't change much during hydrothermal process. The fluids forming the HTDs in the Dengying Formation were mixtures of freshwater from the unconformity at the top of Sinian, fluids from diagenetic compaction and hydrocarbon generation & expulsion from the Lower Cambrian Niutitang Formation mudstones or the Doushantuo Formation silty mudstones, and hydrothermal fluids from the basement. The hydrocarbon reservoirs associated with the HTD were mostly controlled by the basement faults and fractures and karsting processes at the unconformity separating Sinian and Cambrian strata. The hydrocarbon storage spaces of HTD included dissolved cavities and intercrystalline pores. Dissolution cavities are extensive at the top of Dengying Formation, up to about 46 m below the unconformity between Sinian and Cambrian and were generated mainly during karstification. Hydrothermal alteration enhanced the reservoir property of the Dengying Formation dolomites with 3%-5% increase in porosity. No agreement has been reached why zebra dolomite occurs only in the Upper Sinian strata, which would indicate that HTD mineralization occurred during two different periods, each of them related to major extensional tectonic event. The early one related to the Xingkai taphrogenesis(Z2-?1)and the later one to the Emei taphrogenesis(D2-T2). | 14 | Transcriptional Regulation of miR528 by OsSPL9 Orchestrates Antiviral Response in Rice显示文摘Many microRNAs (miRNAs) are critical regulators of plant antiviral defense.However,little is known about how these miRNAs respond to virus invasion at the transcriptional level.We previously show that defense against Rice stripe virus (RSV) invasion entailed a reduction of miR528 accumulation in rice,alleviating miR528-mediated degradation of L-Ascorbate Oxidase (AO) mRNA and bolstering the antiviral activity of AO.Here we show that the miR528-A0 defense module is regulated by the transcription factor SPL9.SPL9 displayed high-affinity binding to specific motifs within the promoter region of miR528 and activated the expression of miR528 gene in vivo.Loss-of-function mutations in SPL9 caused a significant reduction in miR528 accumulation but a substantial increase of AO mRNA,resulting in enhanced plant resistance to RSV.Conversely,transgenic overexpression of SPL9 stimulated the expression of miR528 gene,hence lowering the level of AO mRNA and compromising rice defense against RSV.Importantly,gain in RSV susceptibility did not occur when SPL9 was overexpressed in mir528 loss-of-function mutants,or in transgenic rice expressing a miR528-resistant AO.Taken together,the finding of SPL9-mediated transcriptional activation of miR528 expression adds a new regulatory layer to the miR528-A0 antiviral defense pathway. | Shengze Yao Zhirui Yang Rongxin Yang Yu Huang Ge Guo Xiangyue Kong Ying Lan Tong Zhou He Wang Wenming Wang Xiaofeng Cao Jianguo Wu Yi Li | 2019 | Molecular Plant2019,12,8: | 10 |

The genomic DNA of stool samples were extracted for 16 S r DNA sequencing, and the amplified V4-region was sequenced on the Illumina Miseq platform. Metastats analysis was performed to identify the differential taxa between the groups. Redundancy analysis was used to evaluate the association between gut microbial structure and clinical variables. Thirty NAFLD patients and37 healthy controls were involved. The 16 S r DNA sequencing showed that there was a dramatic variability of the fecal microbiota among all the individuals. Metastats analysis identified eight families and 12 genera with significant differences between the two groups. When some clinical parameters, such as waist-to-hip ratio(WHR) and homeostasis model assessment of insulin resistance(HOMA-IR), were enrolled in Redundancy analysis, the distribution of the two group of samples was obviously changed. The compositional shifts in fecal bacterial communities of NAFLD patients from the healthy controls were mainly at family or genus levels. | 16 | Carbon dioxide capture using polyethylenimine-loaded mesoporous carbons显示文摘A high efficiency sorbent for CO2 capture was developed by loading polyethylenimine (PEI) on mesoporous carbons which possessed well-developed mesoporous structures and large pore volume. The physicochemical properties of the sorbent were characterized by N2 adsorption/desorption, scanning electron microscopy (SEM), thermal gravimetric analysis (TG) and Fourier transform infrared spectroscopy (FT-IR) techniques followed by testing for CO2 capture. Factors that affected the sorption capacity of the sorbent were studied. The sorbent exhibited extraordinary capture capacity with CO2 concentration ranging from 5% to 80%. The optimal PEI loading was determined to be 65 wt.% with a CO2 sorption capacity of 4.82 mmol-CO2 /g-sorbent in 15% CO2 /N2 at 75°C, owing to low mass-transfer resistance and a high utilization ratio of the amine compound (63%). Moisture had a promoting effect on the sorption separation of CO2 . In addition, the developed sorbent could be regenerated easily at 100°C, and it exhibited excellent regenerability and stability. | 20 | Bio-inspired polygonal skeleton structure for vibration isolation:Design,modelling,and experiment显示文摘Inspired by the safe landing of a cat falling from a high altitude,a bio-inspired polygonal skeleton(BIPS) structure is proposed,and its nonlinear characteristics are systematically studied to explore its potential application in the suppression of vibration. The polygon is formed by the skeleton structure of the cat’s entire body and the ground. The BIPS system consists of two symmetrical bionic legs with three robs(as skeleton) and four horizontal springs(as muscle). Two bionic legs are connected through the bearing platform(as spine),which could adjust the distance between the two bionic legs. A theoretical model is developed to characterize its stiffness nonlinearity through geometrical and mechanical analysis. Parameter analysis reveals that the BIPS structure has diverse stiffness,including nonlinear positive stiffness and negative stiffness. By imitating adjustment of leg posture and telescopic function of the spine(control the distance between legs),these flexible stiffness properties can be adjusted by structure parameters. In addition,the load capacity and working range can also be designed by the length of the bars,the initial angle,the mounting position,and the spring stiffness. The experimental setup is established,and the vibration isolation performance under various excitation is tested. The experimental results verify the accuracy of the dynamic model and also show that the proposed BIPS structure can suppress the vibration effectively under a variety of excitations.
