| 5 | Genome sequencing and phylogenetic analysis of Banna virus(genus Seadornavirus,family Reoviridae)isolated from Culicoides显示文摘In an investigation of blood-sucking insects and arboviruses, a virus(YN12243) was isolated from Culicoides samples collected in the Sino-Burmese border region of Yunnan Province, China. The virus caused cytopathic effect(CPE) in C6/36 cells and passaged stably. Polyacrylamide gel analysis showed that the genome of YN12243 was composed of 12 segments of double-stranded RNA(dsRNA), with a distribution pattern of 6-6. The nucleotide and amino acid sequences of the coding region(1.12 segments) were17,803 bp and 5,925 amino acids in length, respectively. The phylogenetic analysis of VP1 protein(RdRp) revealed that YN12243 belonged to genus Seadornavirus of family Reoviridae, and further analysis indicated that YN12243 belongs to the Banna virus(BAV) genotype A2. Additionally, YN12243 was located in the same evolutionary cluster as BAV strains isolated from different mosquito species, suggesting that the BAV isolated from Culicoides does not have species barriers. These results indicate that Culicoides can also be a vector for BAV. | 6 | Effect of Liuweidihuang pill and Jinkuishenqi pill on inhibition of spontaneous breast carcinoma growth in mice显示文摘OBJECTIVE: To investigate the preventing and treating action of Liuweidihuang pill(LP) and Jinkuishenqi pill(JP) on spontaneous breast carcinoma in mice.METHODS: A model of spontaneous breast carcinoma was derived from 11.5-month-old female Kunming breeding mice following the delivery of several litters. The mice were randomly divided into five groups: model control group(C),Liuweidihuang pill high-dose group(LH; 4.6 g·kg1·d1),Liuweidihuang pill low-dose group(LL;2.3 g·kg1·d﹣1),Jinkuishenqi pill high-dose group(JH; 4.6 g·kg﹣1·d1) and Jinkuishenqi pill low-dose group(JL;2.3 g·kg﹣1·d﹣1). Cancer tissue volume was measured by water immersion. Histopathology was analyzed by hematoxylin and eosin staining. Vascular endothelial growth factor(VEGF),extracellular signal-regulated kinase(ERK) and cyclin D1 protein expression in cancer tissue was assayed by western blotting.RESULTS: Compared with the control group,cancer tissue volume and weight were lower in the LP and JP groups,and survival time was longer. The expression of VEGF,ERK and Cyclin D1 were inhibited in the LP and JP groups(P < 0.05),and cell differentiation was increased. | 11 | A cryptic inhibitor of cytokinin phosphorelay controls rice grain size显示文摘Plant hormone cytokinin signals through histidine-aspartic acid(H-D)phosphorelay to regulate plant growth and development.While it is well known that the phosphorelay involves histidine kinases,histidine phosphotransfer proteins(HPs),and response regulators(RRs),how this process is regulated by external components remains unknown.Here we demonstrate that protein phosphatase with kelch-like domains(PPKL1),known as a signaling component of steroid hormone brassinosteroid,is actually a cryptic inhibitor of cytokinin phosphorelay in rice(Oryza sativa).Mutation at a specific amino acid D364 of PPKL1 activates cytokinin response and thus enlarges grain size in a semi-dominant mutant named s48.Overexpression of PPKL1 containing D364,either with the deletion of the phosphatase domain or not,rescues the s48 mutant phenotype.PPKL1 interacts with OsAHP2,one of authentic HPs,and D364 resides in a region resembling the receiver domain of RRs.Accordingly,PPKL1 can utilize D364 to suppress OsAHP2-to-RR phosphorelay,whereas mutation of D364 abolishes the effect.This function of PPKL1 is independent of the phosphatase domain that is required for brassinosteroid signaling.Importantly,editing of the D364-residential region produces a diversity of semi-dominant mutations associated with variously increased grain sizes.Further screening of the edited plants enables the identification of two genotypes that confer significantly improved grain yield.Collectively,our study uncovers a noncanonical cytokinin signaling suppressor and provides a robust tool for seed rational design. | Dapu Liu He Zhao Yunhua Xiao Guoxia Zhang Shouyun Cao Wenchao Yin Yangwen Qian Yanhai Yin Jinsong Zhang Shouyi Chen Chengcai Chu Hongning Tong | 2022 | Molecular Plant2022,15,2: | 3 |
| 12 | Observations and analysis of environment and acoustic field changed by the passage of typhoon Damrey in the Yellow Sea in 2012显示文摘In order to investigate the environment and acoustic filed change induced by typhoon in shallow sea, we conducted two experiments just before and after the passage of typhoon Damrey, which is the strongest to affect the area north of the Yangtze River since 1949, in the Yellow sea in 2012. The data show that the temperature of the whole water column increases dramatically except the sea surface layer after the passage of Damrey while the salinity decreases obviously. The thermocline deepens and weakens, which leads to a change of internal wave activity. The transmission losses(TL) of the two experiments show that the environment change induced by typhoon can increase the TL as large as 8 d B at a distance of 9.2 km and depth of 15 m. The scintillation index(SI) of the sound intensity is simulated to estimate the change of the effect of internal wave activity on acoustic field showing that the SI decreases to a half after the typhoon's passage. The normal mode structures of the two experiments are also significantly different due to the thermocline changes. Redox-active organic molecules have shown promising prospect in the application of RFBs,benefited from their low cost, vast abundance, and high tunability of both potential and solubility. In this review, we discuss the advantages of redox active organic materials over their inorganic compart and the recent progress of organic based aqueous and non-aqueous RFBs. | 14 | Pretreatment with low-frequency repetitive transcranial magnetic stimulation may influence neuronal Bcl-2 and Fas protein expression in the CA1 region of the hippocampus: A possible anti-epilepsy mechanism in a lithium-pilocarpine-induced epileptic rat mod显示文摘BACKGROUND: Bcl-2 and Fas proteins are well known as anti-apoptotic and pro-apoptotic factors, respectively. However, whether the anti-epileptic mechanism of low-frequency repetitive transcranial magnetic stimulation (rTMS) involves an anti-apoptotic effect via regulating Bcl-2 and Fas protein expression remains to be determined. OBJECTIVE: To verify the correlation between the anti-epileptic mechanism following pretreatment of low-frequency rTMS and anti-hippocampal apoptosis. DESIGN, TIME AND SETTING: A randomized controlled animal experiment was performed at Institute of Neurological Disorders, Affiliated Hospital of North Sichuan Medical College between September 2007 and March 2008. MATERIALS: Pilocarpine (053K13011) was provided by Sigma, USA; lithium was provided by Shanghai Biotechnology Co., Ltd., China; Dantec Maglite-r25 rTMS instrument was provided by Dundee, Denmark. METHODS: A total of 21 adult male Wistar rats were randomly divided into control (n = 6), rTMS pretreatment (n = 9), and sham-stimulation (n = 6) groups. The rTMS pretreatment group was pretreated with low-frequency rTMS (0.5 Hz, 75% threshold intensity, 20 times/bundle, and 5 bundles/day), while the sham-stimulation group was sham-stimulated with a similar sound for 7 successive days to establish lithium-pilocarpine-induced epileptic state models. MAIN OUTCOME MEASURES: Epileptic stroke latency; neuronal morphology was observed using hematoxylin and eosin staining; mean positive-reactive cell number and mean absorbance of Bcl-2 and Fas protein in the hippocampal CA1 region was observed using immunohistochemistry. RESULTS: Epileptic latency in the rTMS pretreatment group was significantly enhanced (P < 0.01), and a number of degenerated neurons were observed to be apoptotic. Bcl-2 protein expression increased at each time point, but Fas protein expression decreased (P < 0.01). | 16 | Decreases in Both the Seroprevalence of Serum Antibodies and Seroprotection against Japanese Encephalitis Virus among Vaccinated Children显示文摘The incidence of Japanese encephalitis(JE)has significantly decreased in China due to JE vaccines.In this study,we investigated the post-JE vaccination seroprevalence and protection provided by vaccinated sera against Japanese encephalitis virus(JEV)to elucidate the persistence and waning of antibodies to JEV among JE-SA14-14-2-vaccinated children.A total of 300 serum samples were collected from vaccinated children aged 3-10 years in Zhaotong,Yunnan,China.The seroprevalence of anti-JEV antibodies was determined by enzyme-linked immune sorbent assay and plaque reduction neutralization test.The highest seropositivity of 82%was observed in vaccinated children during the first 0.5-1.5 years after booster vaccination.Then,the seropositivity began to decline and remained lower than the original level observed in the 0.5-1.5-year group.An association was found between the waning of seroprevalence and elapsed time of the post-booster vaccination.Similarly,the neutralizing antibody(nAb)titres gradually decreased over time,and the levels showed a positive correlation with the protective efficacy in mice.This finding suggests that nAbs play an important role in the antiviral process and that the nAb titre is an adequately credible parameter for evaluating the protective efficacy induced by the JE vaccine.Our results provide data that clarify the persistence and waning of antibodies to JEV,which may help elucidate the pathogenesis of JE. | Ran Wang Lyu Xie Na Gao Dongying Fan Hui Chen Peigang Wang Hongning Zhou Jing An | 2019 | Virologica Sinica2019,34,3: | 2 |
| 17 | Dynamic Cluster Key Management Scheme on Wireless Sensor Networks显示文摘Wireless sensor networks are open architectures, so any potential threat can easily intercept, wiretap and counterfeit the information. Therefore, the safety of WSN is very important. Since any single key system cannot guarantee the security of the wireless sensor network for communications, this paper introduces a hierarchical key management scheme based on the different abilities of different sensor nodes in the clustered wireless sensor network. In this scheme, the nodes are distributed into several clusters, and a cluster head must be elected for each cluster. Private communication between cluster heads is realized through the encryption system based on the identity of each head while private communication between cluster nodes in a same cluster head is achieved through the random key preliminary distribution system. Considering the characteristics of WSN, we adopt dynamic means called dynamic cluster key management scheme to deal with master key, so master key will be updated according to the changed dynamic network topology. The precondition of spectrum sharing is to obtain the Spectrum Availability Information(SAI). Energy detection is a typical technology to get SAI. With the mobility of primary users, the energy received by secondary users varies greatly with the distance from the target primary users. Most of the existing energy detection algorithms that use fixed thresholds are not suitable. | 20 | Brassinosteroid-regulated plant growth and development and gene expression in soybean显示文摘Brassinosteroids(BRs) are endogenous phytohormones that play important roles in regulating plant growth and development.In this study, we evaluated the effects of brassinolide(BL, one of the active BRs) on soybean and identified roles of the hormone in regulating multiple aspects of plant growth and development.BL application promoted hypocotyl and epicotyl elongation in the light but blocked epicotyl elongation in the dark.High levels of castasterone and BL accumulated in light-grown plants.BL disrupted shoot negative gravitropism, whereas gibberellin did not.BL delayed leaf senescence.Transcriptome analysis showed that BL induced cell wall-modifying genes and auxin-associated genes but suppressed a class of WRKY genes involved in senescence and stress responses,showing the complex roles of BRs in multiple biological processes. | Wenchao Yin Nana Dong Mei Niu Xiaoxing Zhang Lulu Li Jun Liu Bin Liu Hongning Tong | 2019 | The Crop Journal2019,7,3: | 2 |
