1The effects of probiotics administration on the milk production,milk components and fecal bacteria microbiota of dairy cows显示文摘Probiotics administration can improve host health.This study aims to determine the effects of probiotics(Lactobacillus casei Zhang and Lactobacillus plantarum P-8) administration on milk production,milk functional components,milk composition,and fecal microbiota of dairy cows.Variations in the fecal bacteria microbiota between treatments were assessed based on 16 S rRNA profiles determined by Pac Bio single molecule real-time sequencing technology.The probiotics supplementation significantly increased the milk production and the contents of milk immunoglobulin G(IgG),lactoferrin(LTF),lysozyme(LYS)and lactoperoxidase(LP),while the somatic cell counts(SCC) significantly decreased(P < 0.01).However,no significant difference was found in the milk fat,protein and lactose contents(P > 0.05).Although the probiotics supplementation did not change the fecal bacteria richness and diversity,significantly more rumen fermentative bacteria(Bacteroides,Roseburia,Ruminococcus,Clostridium,Coprococcus and Dorea) and beneficial bacteria(Faecalibacterium prausnitzii) were found in the probiotics treatment group.Meanwhile,some opportunistic pathogens e.g.Bacillus cereus,Cronobacter sakazakii and Alkaliphilus oremlandii,were suppressed.Additionally,we found some correlations between the milk production,milk components and fecal bacteria.To sum up,our study demonstrated the beneficial effects of probiotics application in improving the quality and quantity of cow milk production.Haiyan Xu Weiqiang Huang Qiangchuan Hou Lai-yu Kwok Zhihong Sun Huimin Ma Feiyan Zhao Yuan-Kun Lee Heping Zhang 2017Science Bulletin2017,62,11:21
2Numerical simulation for the coupling effect of local atmospheric circulations over the area of Beijing,Tianjin and Hebei Province显示文摘Using the model system MM5.V3 and multi-layer grid nesting technique, we have done a multi-scale numerical simulation over the area of Beijing, Tianjin and Hebei Province to analyze the temperature and wind field there and study its local circulations. The results show a coupling effect of Urban Heat Island Circulation (UHIC), Mountain Valley Breeze (MVB) and Sea Land Breeze (SLB) occurs in this area when the synoptic system is weak. The SLB can penetrate deep into the mainland for about 200 km when it is blooming. MVB can extend to south and cover almost the whole plain area in Beijing. Both MVB and SLB are diurnal periodical; meanwhile the phase of MVB drops behind that of SLB for about six hours. As a local circulation, the UHIC weakens the two circulations above, and it also has a diurnal period. As a result, the coupling effect of circulations reveals not only different features in spring-summer period and autumn-winter period in a year but also the difference between early morn- ing to noonday and afternoon to night in a day. We noted the diffusion of contamination over the area around Beijing, and found the steady presence of a transport routine of contamination over North-China throughout the year caused by the Coupling Effect mentioned above. 3Respiratory Virus Multiplex RT-PCR Assay Sensitivities and Influence Factors in Hospitalized Children with Lower Respiratory Tract Infections显示文摘Multiplex RT-PCR assays have been widely used tools for detection and differentiation of a panel of respiratory viral pathogens. In this study, we evaluated the Qiagen ResPlex II V2.0 kit and explored factors influencing its sensitivity. Nasopharyngeal swab (NPS) specimens were prospectively collected from pediatric inpatients with lower respiratory tract infections at the time of admission in the Shenzhen Children's Hospital from May 2009 to April 2010. Total nucleic acids were extracted using the EZ1 system (Qiagen, Germany) and 17 respiratory viruses and genotypes including influenza A virus (FluA), FluB, parainfluenza virus 1 (PIV1), PIV2, PIV3, PIV4, respiratory syncytial virus (RSV), human metapneumovirus (hMPV), rhinoviruses (RhV), enteroviruses (EnV), human bocaviruses (hBoV), adenoviruses (AdV), four coronaviruses (229E, OC43, NL63 and HKU1), and FluA 2009 pandemic H1N1(H1N1-p) were detected and identified by the ResPlex II kit. In parallel, 16 real-time TaqMan quantitative RT-PCR assays were used to quantitatively detect each virus except for RhV. Influenza and parainfluenza viral cultures were also performed. Among the total 438 NPS specimens collected during the study period, one or more viral pathogens were detected in 274 (62.6%) and 201(45.9%) specimens by monoplex TaqMan RT-PCR and multiplex ResPlex, respectively. When results from monoplex PCR or cell culture were used as the reference standard, the multiplex PCR possessed specificities of 92.9-100.0%. The sensitivity of multiplex PCR for PIV3, hMPV, PIV1 and BoV were 73.1%, 70%, 66.7% and 55.6%, respectively, while low sensitivities (11.1%-40.0%) were observed for FluA, EnV, OC43, RSV and H1N1. Among the seven viruses/genotypes detected with higher frequencies, multiplex PCR sensitivities were correlated significantly with viral loads determined by the TaqMan RT-PCR in FluA, H1N1-p and RSV (p=0.011-0.000). The Qiagen ResPlex II multiplex RT-PCR kit possesses excellent specificity for simultaneous detection of 17 viral pathogens in NPS specimens in pediatric inpatients at the time of admission. 4Effect of dietary interventions on the intestinal microbiota of Mongolian hosts显示文摘The gut microbiota of Mongolian hosts has distinctive characteristics due to their meat- and dairyoriented daily diets and unique genotype.The aim of the present study was to investigate the effect of switching from the typical high protein and fat Mongolian diets to carbohydrate-rich meals composed principally of wheat,rice and naked oats on the host gut microbiota within 3 weeks.Our study took the advantage of the long sequence reads produced by the Pac Bio single molecule real-time sequencing technology to enable the profiling of subjects' gut microbiota communities along the diet intervention to the species precision.We found that the bacterial richness and diversity decreased apparently along the diet intervention.During the diet intervention,the gut microbiota composition displayed no significant difference at phylum level(with major phyla of Firmicutes,Bacteroidetes,Tenericutes and Proteobacteria).The relative abundances of some genera such as Bacteroidetes,Faecalibacterium,Roseburia,Alistipes,Streptococcus,and Oscillospira were significantly altered after the diet switching started.Notably,significant changes were also observed in the proportions of the species Bacteroides dorei,Bacteroides fragilis,Bacteroides thetaiotaomicron,Ruminococcus albus,Ruminococcus faecis,Roseburia faecis and Eubacterium ventriosum.These results have demonstrated that diet and host gut microbiota is closely linked.Jing Li Haiyan Xu Zhihong Sun Qiangchuan Hou Lai-Yu Kwok Wuri Laga Yanjie Wang Huimin Ma Zhongjie Yu Bilige Menghe Heping Zhang 2016Science Bulletin2016,61,20:14

6Channel noise-induced phase transition of spiral wave in networks of Hodgkin-Huxley neurons显示文摘The phase transition of spiral waves in networks of Hodgkin-Huxley neurons induced by channel noise is investigated in detail.All neurons in the networks are coupled with small-world connections,and the results are compared with the case for regular networks,in which all neurons are completely coupled with nearest-neighbor connections.A statistical variable is defined to study the collective behavior and phase transition of the spiral wave due to the channel noise and topology of the network.The effect of small-world connection networks is described by local regular networks and long-range connection with certain probability p.The numerical results confirm that (1) a stable rotating spiral wave can be developed and maintain robust with low p,where the breakup of the spiral wave and turbulence result from increasing the probability p to a certain threshold;(2) appropriate intensity of the optimized channel noise can develop a spiral wave among turbulent states in small-world connection networks of H-H neurons;and (3) regular connection networks are more robust to channel noise than small-world connection networks.A spiral wave in a small-world network encounters instability more easily as the membrane temperature is increased to a certain high threshold.MA Jun WU Ying YING HePing JIA Ya 2011Chinese Science Bulletin2011,56,2:11
7Fracture Characteristics and Heat Accumulation of Jixianian Carbonate Reservoirs in the Rongcheng Geothermal Field, Xiong'an New Area显示文摘Geothermal energy plays an important role in urban construction of the Xiong’an New Area. Geothermal reservoir fracture distribution of the Mesoproterozoic Jixianian Wumishan Formation(Fm.) carbonate reservoir in the Rongcheng geothermal field are evaluated based on FMI log from Wells D19 and D21. The results show carbonate reservoir fracture density of Well D19 is 15.2/100 m, greater than that of Well D21 with a value of 9.2/100 m. Reservoir porosity and permeability of Well D19 are better than that of Well D21, and the water saturation is bimodally distributed. The movable fluid volume ratio(BVM) of Well D19 is 2% to 8% with some zones exceeding 20%, while the value of Well D21 is less than 4%. Therefore, reservoir fractures in Well D19 are more conducive to fluid flow. Reservoir fractures have a similar occurrence to normal faults, indicating that the tensile stress field controlled the formation of such fractures. Developed reservoir fractures provide a good channel for groundwater convection. 8DJ-1 is involved in the multidrug resistance of SGC7901 gastric cancer cells through PTEN/PI3K/Akt/Nrf2 pathway显示文摘Gastric cancer is a common malignancy worldwide.The occurrence of multidrug resistance(MDR)is the major obstacle for effective gastric cancer chemotherapy.In this study,the in-depth molec-ular mechanism of the DJ-1-induced MDR in SGC7901 gastric cancer cells was investigated.The results showed that DJ-1 expression level was higher in MDR variant SGC7901NCR cells than that in its parental SGC7901 cells.Moreover,DJ-1 overexpression conferred the MDR phenotype to SGC7901 cells,while DJ-1 knockdown in SGC7901NCR cells induced re-sensitization to adriamycin,vincristine,cisplatin,and 5-fluorouracil.These results suggested that DJ-1 mediated the develop-ment of MDR in SGC7901 gastric cancer cells.Importantly,further data revealed that the activation of PI3k/Akt and Nrf2 signaling pathway were required for the DJ-1-induced MDR phenotype in SGC7901 gastric cancer cells.Meanwhile,we found that Pl3k/Akt pathway was activated probably through DJ-1 directly binding to and negatively regulating PTEN,consequently resulting in Nrf2 phosphorylation and activation,and thereby inducing Nrf2-dependent P-glycoprotein(P-gp)and Bcl-2 expressions in the DJ-1-mediated MDR of SGC7901 gastric cancer cells.Overall,these results revealed that activating PTEN/PI3K/Akt/Nrf2 pathway and subsequently upregulating P-gp and Bcl-2 expression could be a critical mechanism by which DJ-1 mediates the development of MDR in SGC7901 gastric cancer cells.The new findings may be helpful for understanding the mechanisms of MDR in gastric cancer cells,prompting its further investigation as a molecular target to overcome MDR.Lejia Qiu Zhaoxia Ma Xiaoran Li Yizhang Deng Guangling Duan Le Zhao Xingwang Xu Lin Xiao Haoyue Liu Zhengming Zhu Heping Chen 2020Acta Biochimica et Biophysica Sinica2020,52,11:5
9Detrital Zircon U-Pb Geochronologyof Sinian–Cambrian Strata in the Eastern Guangxi Area, China显示文摘The Eastern Guangxi area locates in the southwestern part of the transition zone between Yangtze and Cathaysia blocks, which is an important region because the boundary between two blocks probablycrosses there. We determined LA-ICPMS U-Pb ages for detrital zircons extracted from three sandstone samples in the Sinian-Cambrian strata in this region. The resulting ages are in the range of the Archeozoic and Neoproterozoic, with three notable concentrates at 991 Ma, 974 Ma, and 964 Ma, all of which are coeval to the Grenvillian magmatic activity. The new age distribution is similar to the data reported in the Precambrian strata of the adjacent southwestern Cathaysia Block, suggesting that most of our detrital zircons are likely derived from the Cathaysia Block. Combined with others' research, we are more inclined to accept the opinion that there was not an ocean basin between the two blocks during the Sinian-Cambrian period in Eastern Guangxi area if the timing of collision is the Early Neoproterzoic. But if the timing of collision is the Early Paleozoic, we conclude that Luzhai uplift(i.e., the uplift between Guilin-Yongfu faultand Lipu fault) beyond the west of Dayaoshan regoin might be one part of southwestern sedimentation boundary of Cathaysia Block and Yangtze Block. 10Cow, yak, and camel milk diets differentially modulated the systemic immunity and fecal microbiota of rats显示文摘Cow milk is most widely consumed; however,non-cattle milk has gained increasing interest because of added nutritive values. We compared the health effects of yak,cow,and camel milk in rats. By measuring several plasma immune factors,significantly more interferon-c was detected in the camel than the yak(P = 0.0020) or cow(P = 0.0062) milk group. Significantly more Ig M was detected in the yak milk than the control group(P = 0.0071). The control group had significantly less interleukin 6 than the yak(P = 0.0499)and cow(P = 0.0248) milk groups. The fecal microbiota of the 144 samples comprised mainly of the Firmicutes(76.70 ± 11.03%),Bacteroidetes(15.27 ± 7.79%),Proteobacteria(3.61 ± 4.34%),and Tenericutes(2.61 ± 2.53%) phyla. Multivariate analyses revealed a mild shift in the fecal microbiota along the milk treatment. We further identified the differential microbes across the four groups. At day 14,22 and 28 differential genera and species were identified(P = 0.0000–0.0462),while 8 and 11 differential genera and species(P = 0.0000–0.0013) were found at day 28. Some short-chain fatty acid and succinate producers increased,while certain health-concerned bacteria(Prevotella copri,Phascolarctobacterium faecium,and Bacteroides uniformis) decreased after 14 days of yak or camel milk treatment. 11NDUFAB1 confers cardio-protection by enhancing mitochondrial bioenergetics through coordination of respiratory complex and supercomplex assembly显示文摘The impairment of mitochondrial bioenergetics,often coupled with exaggerated reactive oxygen species (ROS) production, is a fundamental disease mechanism in organs with a high demand for energy, including the heart. Building a more robust and safer cellular powerhouse holds the promise for protecting these organs in stressful conditions. Here, we demonstrate that NADH:ubiquinone oxidoreductase subunit AB1 (NDUFAB1), also known as mitochondrial acyl carrier protein, acts as a powerful cardio-protector by conferring greater capacity and efficiency of mitochondrial energy metabolism. In particular, NDUFAB1 not only serves as a complex I subunit, but also coordinates the assembly of respiratory complexes I, II, and III, and supercomplexes, through regulating iron-sulfur biosynthesis and complex I subunit stability. Cardiac-specific deletion of Ndufab1 in mice caused defective bioenergetics and elevated ROS levels, leading to progressive dilated cardiomyopathy and eventual heart failure and sudden death. Overexpression of Ndufab1 effectively enhanced mitochondrial bioenergetics while limiting ROS production and protected the heart against ischemia-reperfusion injury. 12Detecting the breakup of spiral waves in small-world networks of neurons due to channel block显示文摘The breakup of a spiral wave by blockade of sodium and potassium channels in a small-world network of Hodgkin-Huxley neurons is investigated in detail.The influence of ion channel block in poisoned excitable membrane patches of a certain size is measured,by varying channel noise and channel densities resulting from the change in conductance,For example,tetraethylammonium is known to cause a block(poisoning) of potassium channels,while tetrodotoxin blocks sodium channels.We observed the occurrence of spiral waves,which are ordered waves believed to play an important role in facilitating the propagation of electric signals across quiescent regions of the brain.In this paper,the effect of channel block was measured by the factors xK and xNa,which represent the ratios of unblocked,or active,ion channels,to the overall number of potassium or sodium ion channels,respectively.To quantify these observations,we use a simple but robust synchronization measure,which succinctly captures the transition from spiral waves to other collective states,such as broken segments resulting from the breakup of the spiral wave.The critical thresholds of channel block can be inferred from the abrupt changes occurring in plots of the synchronization measure against different values of xK and xNa.Notably,small synchronization factors can be tightly associated with states where the formation of spiral waves is robust to mild channel block.MA Jun HUANG Long YING HePing PU ZhongSheng 2012Chinese Science Bulletin2012,57,17:4
13Reconstruction of the shape of object with near field measurements in a half-plane显示文摘We consider a mathematical problem modelling some characteristics of near field optical microscope.We take a monofrequency line source to illuminate a sample with constant index of refraction and use the scattered field data measured near the sample to reconstruct the shape of it. 14A comparison study of the simulation accuracy between WRF and MM5 in simulating local atmospheric circulations over Greater Beijing显示文摘Several multi-scale numerical simulation experiments were carried out using the mesoscale modeling systems MM5V3.7 and WRFV2.2 for Greater Beijing to estimate the accuracy of WRF and MM5 in simulating the characteristics and variations of mesoscale local circulation in the atmospheric boundary layer of this area. We simulated the horizontal distribution and diurnal variations of temperature and wind fields near the ground and compared them with Automatic Weather System (AWS) data collected from 19 AWS stations in Beijing. Correlation and error analyses were also made. The modeling and statistical results showed that both WRF and MM5 model the temperature field near the ground significantly better than they model the wind field. The temperature field simulated by MM5 is more coincident than that of WRF with the AWS observation records, while WRF does better than MM5 in simulating the wind field, especially under the condition of gusty wind. Neither WRF nor MM5 can capture the fine structure of urban architectural complexity, which is the main error in the wind field simulation. Both models underestimate the land surface temperature at night and overestimate the temperature during the day. 15Emergence of target waves in neuronal networks due to diverse forcing currents显示文摘The electric activities of neurons could be changed when ion channel block occurs in the neurons.External forcing currents with diversity are imposed on the regular network of Hodgkin-Huxley(HH) neuron,and target waves are induced to occupy the network.The forcing current I1 is imposed on neurons in a local region with m 0 ×m 0 nodes in the network,neurons in other nodes are imposed with another forcing current I2.Target wave could be developed to occupy the network when the gradient forcing current(I1-I2) exceeds certain threshold,and the formation of target wave is independent of the selection of boundary condition.It is also found that the developed target wave can decrease the negative effect of ion channel block and suppress the spiral wave,and thus channel noise is also considered.The potential mechanism of formation of target wave could be that the gradient forcing current(I1-I2) generates quasi-periodical signal in local area,and the propagation of quasi-periodical signal induces target-like wave due to mutual coupling among neurons in the network.MA Jun WANG ChunNi YING HePing WU Ying CHU RunTong 2013Science China(Physics,Mechanics & Astronomy)2013,56,6:4
16Desmoglein 2(DSG2) Is A Receptor of Human Adenovirus Type 55 Causing Adult Severe Community-Acquired Pneumonia显示文摘Human adenovirus type 55(HAdV-B55) is a re-emergent acute respiratory disease pathogen that causes adult communityacquired pneumonia(CAP). Previous studies have shown that the receptor of HAdV-B14, which genome is highly similar with HAdV-B55, is human Desmoglein 2(DSG2). However, whether the receptor of HAdV-B55 is DSG2 is undetermined because there are three amino acid mutations in the fiber gene between HAdV-B14 and HAdV-B55. Here, firstly we found the 3T3 cells, a mouse embryo fibroblast rodent cell line which does not express human DSG2, were able to be infected by HAdV-B55 after transfected with pcDNA3.1-DSG2, while normal 3T3 cells were still unsusceptible to HAdV-B55 infection. Next, A549 cells with h DSG2 knock-down by siRNA were hard to be infected by HAdV-B3/-B14/-B55, while the control siRNA group was still able to be infected by all these types of HAdVs. Finally, immunofluorescence confocal microscopy indicated visually that Cy3-conjugated HAdV-B55 viruses entered A549 cells by binding to DSG2 protein.Therefore, DSG2 is a major receptor of HAdV-B55 causing adult CAP. 18Identification of a new isoform of the murine Sh2d1a gene and its functional implications显示文摘Signaling lymphocytic activation molecule(SLAM)-associated protein(SAP)is a Src homology(SH)domain 2-containing intracellular adaptor protein that is predominantly expressed in the hematopoietic system by T lymphocytes and NK cells.SAP protein is encoded by the SH2D1A gene located on the X chromosome.Loss-of-function mutations in SAP cause the X-linked lymphoproliferative disease(XLP),a severe immunodeficiency characterized by heightened susceptibility to Epstein-Barr virus and impaired humoral immunity.Normal individuals express several functional and non-functional isoforms of SAP as a result of alternative splicing.In this study,we identify a cryptic exon in the murine Sh2d1a gene.At the mRNA level,the new isoform of SAP(SAP-2)that includes this new exon is widely expressed in lymphoid tissues by C57BL/6 and 129 strains of inbred mice.SAP-2 accounts for approximately 1%3%of total SAP transcripts,and it is dynamically regulated during lymphocyte activation.At the protein level,the SAP-2 isoform is a 144 amino-acid protein.Compared to the dominant 126 amino-acid SAP-1 isoform,the additional 18 amino acids are inserted into a structural region that is critical for phosphotyrosine binding.Our functional analysis in vitro indicates that SAP-2 is a non-functional isoform due to decreased protein stability.Thus,both human and mouse have multiple SAP splice isoforms that may or may not function.Modulation of relative proportions of these isoforms is potentially a mechanism whereby cells can regulate SAP-mediated biological activities.WU LongYan LU PeiWen MA WeiWei CHU CoCo XU HePing QI Hai 2014Science China(Life Sciences)2014,57,1:2
