| 4 | Isoliquiritigenin inhibits TGF-β1-induced fibrogenesis through activating autophagy via PI3K/AKT/mTOR pathway in MRC-5 cells显示文摘Isoliquiritigenin(ISL),a natural flavonoid derived from the root of liquorice,has been reported to possess anti-inflammatory and antioxidant activities.Previous studies have found that ISL plays a crucial role in anti-fibrosis of adipose tissue and renal tissue;however,its effect on pulmonary fibrogenesis has not been demonstrated.In this study,we aimed to explore the roles and the underlying mechanisms of ISL in TGF-β1-induced fibrogenesis using human lung fibroblast-derived MRC-5 cells.Cell proliferation and migration were determined by MTT and wound healing assay,respectively.The expression levels of alpha-smooth muscle actin(α-SMA),collagen type I alpha 1(COLIA1)and fibronectin(FN),microtubule-associated protein light chain 3(LC3)and related signaling molecules were detected by quantitative real-time PCR,western blot and immunofluorescence assay,correspondingly.EGFP-LC3 transfection was used for autophagy analysis.The results showed that ISL inhibited the TGF-β1-induced proliferation and migration,and down-regulated the expressions ofα-SMA,COLIA1 and FN.ISL treatment led to up-regulation of LC3 in TGF-β1-treated MRC-5 cells,accompanied by significant decrease in the phosphorylation levels of phosphatidylinositol 3-kinase(PI3K),protein kinase B(AKT),and mammalian target of rapamycin(mTOR).In addition,the inhibitory effects of ISL on TGF-β1-induced fibrogenic features in MRC-5 cells were enhanced by pretreatment with autophagy activator Rapmycin and PI3K/AKT inhibitor LY294002 and reversed by autophagy inhibitor 3-methyladenine and PI3K/AKT activator IGF-1.Taken together,our results demonstrated that ISL could attenuate the fibrogenesis of TGF-β1-treated MRC-5 cells by activating autophagy via suppressing the PI3K/AKT/mTOR pathway.Therefore,ISL holds a great potential to be developed as a novel therapeutic agent for the treatment of pulmonary fibrosis. | Jinjuan He Hao Peng Meifang Wang Ying Liu Xingrong Guo Bin Wang Longjun Dai Xueqin Cheng Zhongji Meng Leyong Yuan Fenglin Cai Yijun Tang | 2020 | Acta Biochimica et Biophysica Sinica2020,52,8: | 16 |

| 8 | Enumeration and molecular characterization of circulating tumor cell using an in vivo capture system in squamous cell carcinoma of head and neck显示文摘Objective: Detection rate and isolation yield of circulating tumor cell(CTC) are low in squamous cell carcinoma of head and neck(SCCHN) with in vitro approaches due to limited sample volumes. In this study, we applied the Cell Collector to capture CTC in vivo from peripheral blood.Methods: In total, the study included 22 cases with 37 times of detection. All of the patients were newly diagnosed with locally advanced or metastatic SCCHN, including laryngocarcinoma(40.9%, 9/22) and hypopharyngeal carcinoma(59.1%, 13/22). All patients received CTC analysis before treatment. Three patients received induction chemotherapy. Sixteen patients received surgical therapy, of which 13 patients received postoperative detection. Two patients received both induction chemotherapy and surgery treatment. Patients underwent two successive Cell Collector applications 24 h before and 7 d after surgical therapy. Nine healthy volunteers were enrolled as the control group. Epidermal growth factor receptor variant type Ⅲ(EGFRⅧ)expression was analyzed with fluorescent dye labeled antibody.Results: With Cell Collector isolation, 72.7%(16/22) of the patients were positive for ≥1 CTC(CTC; range,1–17 cells) before treatments and 46.7%(7/15) of patients were CTC positive for ≥1 CTC(CTC; range, 1–29 cells) after surgical therapy. Moreover, the detection rate of Cell Collector(82.4%, 14/17; CTC count range, 0–17)in advanced SCCHN(stage Ⅲ–Ⅳ) was much higher than that in early stages(stage Ⅰ–Ⅱ, 40.0%, 2/5; CTC count range, 0–2)(P<0.05). EGFRⅧ expression of CTC was also analyzed with fluorescence staining. | 12 | Amphibian-derived peptide homodimer OA-GL17d promotes skin wound regeneration through the miR-663a/TGF-β1/Smad axis显示文摘Background:Amphibian-derived peptides exhibit considerable potential in the discovery and development of new therapeutic interventions for clinically challenging chronic skin wounds.MicroRNAs(miRNAs)are also considered promising targets for the development of effective therapies against skin wounds.However,further research in this field is anticipated.This study aims to identify and provide a new peptide drug candidate,as well as to explore the underlying miRNA mechanisms and possible miRNA drug target for skin wound healing.Methods:A combination of Edman degradation,mass spectrometry and cDNA cloning were adopted to determine the amino acid sequence of a peptide thatwas fractionated from the secretion of Odorrana andersonii frog skin using gel-filtration and reversed-phase high-performance liquid chromatography.The toxicity of the peptide was evaluated by Calcein-AM/propidium iodide(PI)double staining against human keratinocytes(HaCaT cells),hemolytic activity against mice blood cells and acute toxicity against mice.The stability of the peptide in plasma was also evaluated.The prohealing potency of the peptide was determined by MTS,scratch healing and a Transwell experiment against HaCaT cells,full-thickness injury wounds and scald wounds in the dorsal skin of mice.miRNA transcriptome sequencing analysis,enzyme-linked immunosorbent assay,real-time polymerase chain reaction and western blotting were performed to explore the molecular mechanisms.Results:A novel peptide homodimer(named OA-GL17d)that contains a disulfide bond between the 16th cysteine residue of the peptide monomer and the sequence‘GLFKWHPRCGEEQSMWT’was identified.Analysis showed that OA-GL17d exhibited no hemolytic activity or acute toxicity,but effectively promoted keratinocyte proliferation and migration and strongly stimulated the repair of full-thickness injury wounds and scald wounds in the dorsal skin of mice.Mechanistically,OA-GL17d decreased the level of miR-663a to increase the level of transforming growth factor-β1(TGF-β1)and activate the subsequent TGF-β1/Smad signaling pathway,thereby resulting in accelerated skin wound re-epithelialization and granular tissue formation.Conclusions:Our results suggest that OA-GL17d is a new peptide drug candidate for skin wound repair.This study emphasizes the importance of exogenous peptides as molecular probes for exploring competing endogenous RNA mechanisms and indicates that miR-663a may be an effective target for promoting skin repair. | Yue Zhang Ying Wang Lin Zeng Yixiang Liu Huiling Sun Shanshan Li Siyu Wang Longjun Shu Naixin Liu Saige Yin Junsong Wang Dan Ni Yutong Wu Ying Yang Li He Buliang Meng Xinwang Yang | 2022 | Burns & Trauma2022,10,1: | 0 |
