1单味中药提取物对体外培养的黑素细胞酪氨酸酶基因和c-kit基因表达的激活作用显示文摘目的研究补骨脂、白芷、夏枯草、川芎和墨旱莲单味中药提取物对黑素细胞的酪氨酸酶基因和原癌基因c-kit蛋白表达的影响,以及对总蛋白合成的影响。方法采用蛋白印迹法检测了酪氨酸酶蛋白和干细胞因子受体kit蛋白含量的变化;595nm比色标准曲线测定黑素细胞可溶性总蛋白含量。结果白芷、补骨脂、夏枯草、川芎和墨旱莲对黑素细胞可溶性总蛋白合成和c-kit基因蛋白表达有促进作用,其中白芷和补骨脂对c-kit基因蛋白表达的促进作用为69.44%和58.43%;夏枯草、墨旱莲和川芎对酪氨酸酶基因的蛋白表达有一定的促进作用。结论在白癜风治疗中传统中药具有较好的疗效,可能与酪氨酸激酶受体蛋白含量的提高相关,即通过SCF/kit蛋白途径起作用,因此可以通过该途径进一步筛选更多治疗白癜风的单味中药。李永伟 许爱娥 Ruth Halaban 2004中华皮肤科杂志2004,37,10:46
2加味桃红四物汤对黑素细胞酪氨酸酶和酪氨酸激酶受体基因蛋白表达的影响显示文摘目的研究加味桃红四物汤对体外培养的人表皮黑素细胞(MC)可溶性总蛋白以及酪氨酸酶(Tyr)基因和酪氨酸激酶受体基因(c-kit)表达的作用。方法采用免疫印迹法检测了加味桃红四物汤对酪氨酸酶和酪氨酸激酶受体蛋白含量的影响;595nm比色标准曲线测定黑素细胞总蛋白含量;采用细胞数统计和显微镜观察的方法测定加味桃红四物汤对细胞毒性和对细胞增殖的影响。结果加味桃红四物汤对黑素细胞可溶性总蛋白和酪氨酸酶的合成有促进作用,分别为4.76%和1.79%;对酪氨酸激酶受体蛋白的合成具有显著促进作用,高达79.41%。结论加味桃红四物汤治疗白癜风的作用机制之一是与酪氨酸激酶受体蛋白含量的提高相关,而与黑素细胞增殖和酪氨酸酶相关蛋白的含量无明显关系。李永伟 许爱娥 Ruth Halaban 2004中华皮肤科杂志2004,37,12:8
